Efficacy and toxic action of the natural product natamycin against Sclerotinia sclerotiorum

Yuxuan Cao,Xu Zhang,Xiaoning Song,Wenkui Li,Zheng Ren,Juntao Feng,Zhiqing Ma,Xili Liu,Yong Wang

BACKGROUND Sclerotinia stem rot caused by Sclerotinia sclerotiorum seriously endangers oilseed rape production worldwide, and the occurrence of fungicide-resistant mutants of S. sclerotiorum leads to control decline. Thus, it is critical to explore new green substitutes with different action mechanisms and high antifungal activity. Herein, the activity and the action mechanism of natamycin against S. sclerotiorum were evaluated. RESULTS Natamycin showed potent inhibition on the mycelial growth of S. sclerotiorum , and half-maximal effective concentration (EC 50 ) values against 103 S. sclerotiorum strains ranged from 0.53 to 4.04 μg/mL (mean 1.44 μg/mL). Natamycin also exhibited high efficacy against both carbendazim- and dimethachlone-resistant strains of S. sclerotiorum on detached oilseed rape leaves. No cross-resistance was detected between natamycin and carbendazim. Natamycin markedly disrupted hyphal form, sclerotia formation, integrity of the cell membrane, and reduced the content of oxalic acid and ergosterol, whereas it increased the reactive oxygen species (ROS) and malondialdehyde content. Interestingly, exogenous addition of ergosterol could reduce the inhibition of natamycin against S. sclerotiorum . Importantly, natamycin significantly inhibited expression of the Cyp51 gene, which is contrary to results for the triazole fungicide flusilazole, indicating a different action mechanism from triazole fungicides. CONCLUSION Natamycin is a promising effective candidate for the resistance management of S. sclerotiorum . © 2023 Society of Chemical Industry.

Bioassay-guided isolation of antifungal amides against Sclerotinia sclerotiorum from the seeds of Clausena lansium

He Yan,Zi Xiong,Na Xie,Shanzhi Liu,Lianlian Zhang,Fang Xu,Wenhui Guo,Juntao Feng

Clausena lansium ( Lour.) Skeels is an attractive shrub or small tree, a member of the Rutaceae family. The seeds, leaves and roots of this plant have been used as herbal medicines for a long time. In this study, seven amides were isolated from the seeds of Clausena lansium . The antifungal assay indicated that lansiumamide B ( 4 ) and lansiumamide C ( 2 ) displayed the highest antifungal activity against Sclerotinia sclerotiorum with EC 50 values of 4.95 and 13.24 μg/mL, which were both lower than that of carbendazim (EC 50  = 0.64 μg/mL). The structure-activity relationships of these amides were investigated using assays to measure their in vitro antifungal activities against Sclerotinia sclerotiorum . The effect of the most potent compound lansiumamide B on morphology and physiology of S. sclerotiorum were also evaluated. Lansiumamide B caused cell rupture and mycelial abnormalities of S. sclerotiorum , and its curative efficacy (75.17%) against S. sclerotiorum infection was better than that of carbendazim (56.57%). This study demonstrated that amides isolated from C. lansium possessed the potential to be exploited as botanical fungicides for commercial application or can be used as templates for designing new fungicides with novel action modes.

Maleic acid and malonic acid reduced the pathogenicity of Sclerotinia sclerotiorum by inhibiting mycelial growth, sclerotia formation and virulence factors

Fei Yu-chen,Cheng Qin,Zhang Huan,Han Chuang,Wang Xu,Li Yan-feng,Li Shi-qian,Zhao Xiao-hu

Sclerotinia sclerotiorum is a necrotrophic plant pathogenic fungus with broad distribution and host range. Bioactive compounds derived from plant extracts have been proven to be effective in controlling S. sclerotiorum . In this study, the mycelial growth of S. sclerotiorum  was effectively inhibited by maleic acid, malonic acid, and their combination at a concentration of 2 mg/mL, with respective inhibition rates of 32.5%, 9.98%, and 67.6%. The treatment of detached leaves with the two acids resulted in a decrease in lesion diameters. Interestingly, maleic acid and malonic acid decreased the number of sclerotia while simultaneously increasing their weight. The two acids also disrupted the cell structure of sclerotia, leading to sheet-like electron-thin regions. On a molecular level, maleic acid reduced oxalic acid secretion, upregulated the expression of Ss-Odc2 and downregulated CWDE10 , Ss-Bi1 and Ss-Ggt1 . Differently, malonic acid downregulated CWDE2 and Ss-Odc1 . These findings verified that maleic acid and malonic acid could effectively inhibit S. sclerotiorum , providing promising evidence for the development of an environmentally friendly biocontrol agent.

Broad-spectrum antifungal activity of dichloromethane extract of Waltheria indica stems and isolated compounds

Cai Liang,Li Yang,Yu Shao,Xinyue Zhu,Hongliang Zhao,Biwei Chen,Wen Song,Xiangmin Song,Xiaofan Ding,Ranfeng Sun

The secondary metabolites produced by plants are important sources of fungicides with intriguing bioactivities and environmentally benign attributes (eg. low residual toxicity to mammals and readily biodegradable in soil). In this study, the dichloromethane extract of stems of Waltheria indica L. was found to exhibit broad-spectrum antifungal activity toward eight phytopathogenic fungi, particularly against carbendazim-resistant Sclerotinia sclerotiorum with inhibition rate of 100% at 500 μg/mL. Two quinoline alkaloids, antidesmone and waltherione C, were isolated and identified as fungicidal compounds by bioassay-guided isolation and spectroscopic analyses. Antidesmone displayed superior antifungal activities against eight phytopathogenic fungi to commercial botanical fungicides (osthole, carvacrol, and eugenol). Moreover, antidesmone was found to have significant negative cross-resistance with carbendazim to Sclerotinia sclerotiorum for the first time and exhibited much better antifungal activity (EC 50  = 0.60 μg/mL) against carbendazim-resistant strains than carbendazim-sensitive strain (EC 50  = 9.61 μg/mL) of Sclerotinia sclerotiorum , which was further supported by the molecular modelling result. In addition, the dichloromethane extract of the Waltheria indica stems could effectively control the development of eleven species of phytopathogenic fungi in vivo. These results suggest that Waltheria indica dichloromethane extract and antidesmone can be developed as promising broad-spectrum botanical fungicides.

Baseline sensitivity and physiological characteristics of natural product hinokitiol against Sclerotinia sclerotiorum

Mengwei Zhang,Xingyu Ren,Yuying Li,Yaqiang Wang,Yi Li,Zhiqing Ma,Yong Wang,Juntao Feng

BACKGROUND Sclerotinia sclerotiorum , a pathogenic fungus of oilseed rape, poses a severe threat to the oilseed rapeseed industry. In this study, we evaluated the potential of the natural compound hinokitiol against S. sclerotiorum by determining its biological activity and physiological characteristics. RESULTS Our results showed that hinokitiol strongly inhibited the hyphae expansion of S. sclerotiorum , and its effective concentration of hyphae growing inhibition by 50% (EC 50 ) against 103 S. sclerotiorum strains varied from 0.36 to 3.45 μg/mL, with an average of 1.23 μg/mL. Hinokitiol possessed better protective efficacy than therapeutic effects, and it exhibited no cross-resistance between carbendazim. After treatment with hinokitiol, many vesicular protrusions developed on the mycelium with rough surface and thickened cell wall. Moreover, the cell membrane permeability and glycerol content increased, while the oxalic acid declined after hinokitiol treatment. In addition, hinokitiol induced membrane lipid peroxidation and improved the production of reactive oxygen species (ROS) in S. sclerotiorum . Importantly, real-time quantitative polymerase chain reaction showed that cell wall and ROS synthesis-related genes were significantly up-regulated after hinokitiol treatment. CONCLUSION This study revealed that hinokitiol has good biological activity against S. sclerotiorum and could be considered as an alternative bio-fungicide for the resistance management in controlling sclerotinia stem rot infected by S. sclerotiorum . These investigations provided new insights into understanding the toxic action of hinokitiol against pathogenic fungi. © 2024 Society of Chemical Industry.

Synthesis of N-substituted dimethylmaleimides and their antifungal activities against Sclerotinia sclerotiorum

Shen Zhenzhong,Fan Yongxian,Li Fuge,Chen Xiaolong,Shen Yinchu

Compounds with maleimide, both natural and synthesized, have good biological activities, especially the antifungal activity. In order to investigate the antifungal activity of dimethylmaleimides, 17 N -substituted dimethylmaleimides were prepared from the reactions of 2,3-dimethyl maleic anhydride and amines using a facile synthetic method in this paper. These compounds were evaluated for antifungal activities against Sclerotinia sclerotiorum by the mycelium growth rate method. They exhibited minimum inhibitory concentrations (MICs) ranging from 0.01–50.0 μg/mL, with N -(2-benzimidazole)-3,4-dimethylmaleimide being the most active one with an MIC of 0.01 μg/mL. The structure and activity relationship on these compounds indicated that the hydrophobicity of the N -substituents is associated with their antifungal activity. Compared to current antifungals, most of N -substituted dimethylmaleimides have a perfect activity for S. sclerotiorum control and low toxicity.

Antifungal activities against Sclerotinia sclerotiorum by Cinnamomum cassia oil and its main components

Zhenhua Jiang,Hong Jiang,Pengfei Xie

The plant disease produced by Sclerotinia sclerotiorum is a big problem in agriculture. This paper tested the effects of cinnamon oil and its components on the growth of S. sclerotiorum. The cinnamon oil was first separated into carbonyl and noncarbonyl parts by treatment with sodium bisulfite. Then two main cinnamaldehyde derivatives, that is 2′-methoxycinnamaldehyde and coniferyl aldehyde, were further separated by column chromatography from the carbonyl parts. Cinnamon oil demonstrated a significant antifungal effect against S. sclerotiorum with a minimum inhibitory concentration (MIC) of 256 μg/mL in agar and 64 μg/mL air, respectively. trans-Cinnamaldehyde exhibited the highest antifungal activity among all the three cinnamaldehydes tested. In addition, thymol and carvacrol had an additive effect with trans-cinnamaldehyde in preventing the mycelial growth of S. sclerotiorum.

Expression of the Histidine Kinase Gene Sshk Correlates with Dimethachlone Resistance in Sclerotinia sclerotiorum

Jinli Li,Fuxing Zhu,Jianhong Li

Histidine kinases (HK) are implicated in virulence, vegetative mycelial growth, and osmotic and oxidative responses in pathogenic fungi. Our previous work showed that transcriptional levels of the group III HK gene Sshk are higher in field dimethachlone-resistant isolates of Sclerotinia sclerotiorum compared with sensitive isolates. However, it is not clear whether the overexpression of Sshk is the major mechanism for resistance to dimethachlone. In this study, we constructed Sshk silencing and overexpression vectors and assessed dimethachlone resistance levels, virulence, mycelial growth, and sensitivity to osmotic stress for the Sshk -silenced and -overexpression transformants. Overexpression of Sshk resulted in resistance to dimethachlone and increased sensitivity to various stresses and to the cell-wall-perturbing agents sodium dodecyl sulfate (SDS) and Congo red (CR). Compared with the parent isolate, Sshk -silenced transformants had reduced resistance to dimethachlone, significantly higher ( P < 0.05) mycelial growth and virulence, and lower sclerotium production, and were less sensitive to various exogenous stresses such as sodium chloride. Compared with the parent sensitive isolate HLJMG1, dimethachlone resistance ratios of the three overexpression transformants ∆C101, ∆C21, and ∆C10 increased 168.1-, 189.5-, and 221.2-fold, respectively. The three overexpression transformants were more sensitive to CR and SDS than their parent isolate. These findings suggest that overexpression of Sshk is a major mechanism for dimethachlone resistance in some isolates of S. sclerotiorum , and that Sshk plays an important role in maintaining the integrity of the cell wall. Our findings reveal a novel molecular mechanism for dimethachlone resistance in plant-pathogenic fungi.

Antifungal effects of 3-(2-pyridyl)methyl-2-(4-chlorphenyl) iminothiazolidine against Sclerotinia sclerotiorum

Xianfei Zhang,Jiuyong Xu,Solange Muhayimana,Hui Xiong,Xuefeng Liu,Qingchun Huang

BACKGROUND Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum threatens oilseed rape cultivation, and the emergence of fungicide-resistant strains has led to control failures worldwide. Identifying novel chemical alternatives with different modes of action and high antifungal activities is thus crucial. Herein we evaluated the antifungal effects of 3-(2-pyridyl)methyl-2-(4-chlorphenyl)imino- thiazolidine (PMAS) on S. sclerotiorum to determine its efficacy for SSR management. RESULTS PMAS had an inhibitory effect on mycelial growth; the EC 50 values were 17.83 and 21.15 μg mL −1 for the carbendazim-susceptible strain Ss01 and carbendazim-resistant strain Hm25, respectively. PMAS treatment changed the color of inhibited mycelia to green, and the hyphae were sustained in the undifferentiated stage. Cysteine supplementation made this green color disappear, whereas methionine enhanced the color. Moreover, PMAS treatment markedly inhibited oxalic acid biogenesis, increased free thiol content in mycelia, and weakened the activities of oxaloacetase and malate dehydrogenase, but had little effect on the activity of glyoxylate dehydrogenase. Cysteine could reverse the inhibitory effects of PMAS on mycelial morphogenesis and biochemical constituents, except thiol production. In the pot-culture experiment, PMAS showed a good protective effect, with the control efficacy being >91% on SSR. CONCLUSION PMAS appears to be an effective fungicide for SSR management. © 2020 Society of Chemical Industry

pH-responsive and NIR-triggered nanopesticide for precision control of sclerotinia sclerotiorum in oilseed rape

Rao Qin,Yuhan Jing,Tianqi Wan,Guangliang Lu,Dabiao Zhai,Xingyu Li,Feng Zhang,Jiangang Jiang,Dan Zhang,Ping Zhang,Yiping Wang,Yong Wang,He Wang

A composite consisting of upconversion nanoparticles (UCNPs) and methylene blue (MB)-loaded zeolitic imidazolate framework-8 (ZIF-8), referred to as UCNP@ZIF-8@MB, was synthesized via a simple hydrothermal and one-pot approach for the targeted control of Sclerotinia sclerotiorum ( S. sclerotiorum ) in oilseed rape ( Brassica napus L. ). Taking advantage of the pH-responsive nature of ZIF-8, the composite responds selectively to the acidic microenvironment associated with pathogen infection. Upon 980 nm excitation, UCNPs emit at 650 nm, which activates MB to generate reactive oxygen species (ROS) via photolysis, thereby exerting antifungal effects. The physicochemical properties of UCNP@ZIF-8@MB, including its optical behavior, MB release profile, and adhesion capacity, were systematically characterized. Under mildly acidic conditions (pH 5.5–4.0), MB release exceeded 80 %, indicating efficient responsiveness to the infection milieu. In vitro antifungal assays demonstrated a minimum inhibitory concentration (MIC) of approximately 10 μg/mL under 980 nm laser irradiation. Pot experiments further confirmed significant antifungal activity under light exposure. Additionally, UCNP@ZIF-8@MB exhibited targeted release and favorable biocompatibility with plant tissues and the surrounding environment. These results underscore the potential of UCNP@ZIF-8@MB as an environmentally friendly nanopesticide for the precise management of sclerotinia disease, offering a promising strategy for sustainable agriculture.

A cyclodextrin/ROS-responsive Pydiflumetofen nanoparticle for enhanced fungicidal activity against Sclerotinia sclerotiorum in soybean

Qian Zhao,Liyan Zhang,Ziyu Wang,Weibo Han,Jianzhong Wu

Sclerotinia stem rot (SSR), caused by Sclerotinia sclerotiorum , significantly reduces soybean yields. Conventional SSR-based pesticide application presents environmental and health hazards, whereas ROS-responsive nanocarrier systems demonstrate superior efficacy as a sustainable alternative. In this study, PYD@OxbCD NPs, encapsulating Pydiflumetofen (PYD), were synthesized and showed ROS-triggered release and strong in vitro activity against SSR. This targeted release mechanism reduces ecological risk by minimizing pesticide exposure to non-target organisms. PYD@OxbCD delivers PYD more precisely to infected soybean tissues, outperforming traditional Pydiflumetofen suspension concentrate (SC) in antifungal efficacy. It also upregulates defense-related genes and reduces disease incidence and severity in soybean plants. PYD@OxbCD also mitigates the phytotoxic effects of PYD SC on seedling growth. Overall, PYD@OxbCD shows strong potential as an effective nanoplatform for managing SSR in soybeans, enhancing disease control and pesticide utilization for sustainable agriculture.

Discovery of Novel Stilbene-Containing Nicotinamide Derivatives as Potential SDHIs against Rhizoctonia solani and Sclerotinia sclerotiorum

Luo Bo,Hu Yihan,Zhao Yi,Tao Yuchen,Feng Xiaoyu,Li Dan,Wang Yinge,Hu Xinyue,Wu Yanbing

As part of an ongoing effort to develop novel succinate dehydrogenase inhibitors (SDHIs), a range of innovative nicotinamide derivatives featuring stilbene scaffolds were synthesized and assessed for their antifungal properties. Lead compounds 6g and 6c demonstrated significant antifungal efficacies against Rhizoctonia solani and Sclerotinia sclerotiorum , with EC 50 (half maximal effective concentration) values of 0.008 and 0.034 μg/mL, respectively, surpassing Boscalid (EC 50 = 0.573 and 0.455 μg/mL, respectively) and Fluxapyroxad (EC 50 = 0.026 and 0.037 μg/mL, respectively). Detached leaf assays further confirmed their excellent protective and curative effects, outperforming Boscalid and Fluxapyroxad. Further studies revealed that compound 6g operated through a mechanism akin to Boscalid and Fluxapyroxad. These results indicated that both 6g and 6c showed great potential as novel SDHIs for effectively managing R. solani and S. sclerotiorum , underscoring the necessity for further research to fully explore their capabilities.
