Nicotiana tabacum leaves are mainly used in the production of cigarettes and cigars all over the world, and a large amount of waste materials will be produced in the process. High-value scopoletin can be extracted and separated from waste materials, but there are some problems, such as low purity of products and inability of large-scale production. In this study, we established an efficient and stable method for producing high-purity scopoletin from waste tobacco by containing acetone reflux extracting with dynamic axial compression-high performance liquid chromatography (DAC-HPLC) separating. In particular, the extraction system makes the scopoletin fully extracted through continuous reflux, the DAC-HPLC system makes the high-purity scopoletin repeatedly obtained, and the DAC-HPLC system was first used for scopoletin. Finally, related to the waste tobacco power, the yield of scopoletin is 1.9%, and its HPLC purity is 99.94%. Additionally, a preparation method for the colorless needle crystal of scopoletin was established. Meanwhile, it has a strong antioxidant activity by testing the scavenging activity of the DPPH free radical and the Fe 3+ reducing power. And its spatial structure was identified by an X-ray diffractometer, which laid a good foundation for in-depth study of scopoletin. Graphical Abstract

Background: Scoparone, the principal natural active ingredient of Artemisia capillaries (Yin Chen), can effectively treat cholestatic diseases, but the pharmacokinetic properties of scoparone are rarely studied in intrahepatic cholestatic rats. Objective: A sensitive
and rapid LC-MS/MS method was established to detect scoparone and its metabolite of scopoletin in rat plasma and then compare their plasma pharmacokinetic differences between the normal and ANITinduced cholestasis rats. Methods: Positive ionization was used to separate scoparone and scopoletin
using acetonitrile and 0.1 % formic acid water as the mobile phase on a Hypersil ODS-BP column. Results: The calibration curves presented good linearity (R=0.9983 and 0.9989) in the concentration range of 10- 10000 ng/mL and 0.5-500 ng/mL for scoparone and scopoletin, respectively. The
precision of ≤ 9.4% and the accuracy ranged from -6.4% to 6.8% were recorded over three validation runs, and the recovery was higher than 83.9%. Under different storage conditions, scoparone and scopoletin were stable. Therefore, we studied the pharmacokinetic properties of scoparone and
scopoletin in rats after a single oral administration with the above method. According to the results, the pharmacokinetic parameters of AUC, t1/2, and Cmax values of scoparone in the ANIT group were increased by 106%, 75%, and 44%, respectively,
while these values of scopoletin were increased by 142%, 62%, and 65%. Conclusion: The findings indicated that the pharmacokinetic properties of scoparone and scopoletin were significantly different between the normal and ANIT-induced cholestasis rats, which suggested that the clinical
application dosage of scoparone should be adjusted according to the liver function of patients.

Scopoletin, a plant-derived coumarin, exhibits diverse pharmacological activities. Nowadays, scopoletin is mainly produced by plant extraction. In this study, a Saccharomyces cerevisiae strain capable of de novo scopoletin biosynthesis was constructed by successively integrating ten heterologous genes into a p -coumaric acid producing strain. The following strategies were employed to improve the yield of scopoletin: (1) A high-yield caffeic acid platform strain (301.98 mg/L) was established by increasing the HpaBC copy number, integration of EcaroL and RgTAL , feedback-resistant mutant expression cassette ( ARO3 D154N / ARO4 G141S / ARO7 K229L ), and knockout competing pathway genes ( ARO10 / PDC5 ); (2) Two distinct pathways from caffeic acid to scopoletin were introduced and selected; (3) Integration of heterologous genes AtHCT and AtC3’H , to construct the pathway from p -coumaroyl-CoA to caffeoyl-CoA; (4) Attenuating PYK1 by introducing the mutant gene of PYK1 D146N to increase PEP supply; (5) Overexpression of cofactor regeneration genes ( ADO1 , SAH1 , and ZWF1 ) to strengthen the supply of SAM and NADPH; (6) Adjustment of the fermentation medium. The engineered strain produced scopoletin of 55.32 mg/L in shake flasks. To our knowledge, this is the first report of de novo scopoletin biosynthesis in S. cerevisiae . These results establish new insights for microbial production of scopoletin and coumarin derivatives.

The detection of organophosphorus pesticides (OPs) has received considerable attention for their great harm to human beings. Herein, a novel ratiometric fluorescence biosensor was constructed for the determination of OPs by using Scopoletin (SC) and Amplex Red (AR) as probe pairs that have opposite responses to MnO 2 nanosheets (MnO 2 NS). MnO 2 NS possess peroxidase-like catalytic activity , which could quench the fluorescence of SC as well as enhance the fluorescence of the non-fluorescent substance AR by oxidation . In the absence of OPs, acetylcholinesterase (AChE) hydrolyzed acetylcholine chloride (ATCh) into choline (TCh) and acetate. TCh led the decomposition of MnO 2 NS to manganese ions (Mn 2+ ), increasing signal of SC and decreasing signal of AR. In the presence of OPs, the activity of AChE was inhibited and the decomposition of MnO 2 NS was hindered, therefore the fluorescence intensity of SC was weak and the fluorescence intensity of AR had an obvious increase. Moreover, under the optimal conditions, the ratio of fluorescence intensity response recorded on the AR/SC increases with increasing the concentration of DDVP. The method has wider linear range of 5.0 pg/mL ∼500 ng/mL with a detection limit of 1.6 pg/mL, which is superior to previously reported methods. This strategy has also been applied to a visual observation based on the color change of the solution under UV light.

Scopoletin, a bioactive compound with important physiological and pharmacological effects, lacks a simple and rapid method for real-world detection. In this paper, graphdiyne quantum dots (GDYQDs) were prepared via a one-step ultrasonication method and subsequently combined with N-doped graphene (NGR). The GDYQDs/NGR composite was drop-coated onto a screen-printed electrode (SPE) to fabricate GDYQDs/NGR-modified SPEs, integrated with a portable electrochemical workstation and Bluetooth-enabled smartphone for real-time, wireless detection of scopoletin. Beyond experimental performance, density functional theory (DFT) calculations and partial density of states (PDOS) analyses were conducted for three model substrates (I: GDYQDs, II: NGR, III: GDYQDs/NGR) and three adsorption configurations (1: O–H adsorption, 2: carbonyl O toward substrate, 3: parallel π-adsorption). Adsorption energies indicate the π-adsorption configuration as the most stable across all substrates. PDOS analysis shows a pronounced downshift of the molecular p-state upon adsorption, most notably on the GDYQDs/NGR substrate, indicating stronger electronic coupling and stabilization of adsorption complexes. Collectively, these results demonstrate a cooperative GDYQDs–NGR effect that enhances scopoletin adsorption via π–π interactions and electronic coupling, providing molecular-level guidance for designing high-performance GDYQDs/NGR-based on-site electrochemical sensors.

It is known that sunflower (Helianthus annuusL.) calathide enzymatically hydrolyzed extract (SCHE) contributes to the regulation of serum uric acid (UA); however, evidence regarding its bioactive components and mechanism are lacking. We identified two water-soluble components (scopoletin and chlorogenic acid) that are abundant in sunflower calathide, especially evaluated for the inhibition of xanthine oxidase (XO) and the expression levels of urate transporters with SCHE. Molecular docking of a chlorogenic acid–XO complex was more stable than that of the Scopoletin–XO, and its binding pockets, which closed the Mo = S center, was similar to xanthine pockets. Moreover, chlorogenic acid exhibited stronger inhibition than that of the scopoletin below 260μM, despite the IC50of scopoletin (577.7μM) being lower than that chlorogenic acid (844.7μM) on the UA generation assessed by a spectrophotometer in vitro. It revealed that chlorogenic acid and scopoletin were competitive inhibitors of XO. In addition, the SCHE (300μg/mL) and chlorogenic acid (0.75 mM) obviously inhibited urate transporter 1 (URAT1) and glucose transporter 9 (GLUT9) expression levels, while scopoletin significantly upregulated the expression of GLUT9. To summarize, chlorogenic acid served a crucial role in UA regulation consistent with the SCHE and functioned as an important ingredient of SCHE. The strategic analysis of SCHE combined with scopoletin and chlorogenic acid may contribute to the development of food supplemental alternatives on UA metabolism and the reduction of agricultural byproduct waste. Keywords:sunflower calathide;scopoletin;chlorogenic acid;urate transporters;xanthine oxidase inhibitor;molecular docking

A chemometrics-assisted excitation-emission matrix (EEM) fluorescence method is presented for simultaneous determination of umbelliferone and scopoletin in Tibetan medicine Saussurea laniceps (SL) and traditional Chinese medicine Radix angelicae pubescentis (RAP). Using the strategy of combining EEM fluorescence data with second-order calibration method based on the alternating trilinear decomposition (ATLD) algorithm, the simultaneous quantification of umbelliferone and scopoletin in the two different complex systems was achieved successfully, even in the presence of potential interferents. The pretreatment is simple due to the “second-order advantage” and the use of “mathematical separation” instead of awkward “physical or chemical separation”. Satisfactory results have been achieved with the limits of detection (LODs) of umbelliferone and scopoletin being 0.06 ng mL − 1 and 0.16 ng mL − 1 , respectively. The average spike recoveries of umbelliferone and scopoletin are 98.8 ± 4.3% and 102.5 ± 3.3%, respectively. Besides, HPLC-DAD method was used to further validate the presented strategy, and t- test indicates that prediction results of the two methods have no significant differences. Satisfactory experimental results imply that our method is fast, low-cost and sensitive when compared with HPLC-DAD method.
