Identification of the Anti-ischemia-reperfusion Injury Effect of Baicalein from Scutellaria baicalensis Georgi in Liver Transplantation

Junyan Liu,Shanshan Guo,Junhua Gong,Lue Cheng,Jiefu Luo,Mingxiang Cheng,Shengwei Li,Jianping Gong,Degong Jia

BackgroundThe complex etiology of Ischemia-Reperfusion Injury (IRI) induced by liver transplantation (LT) and the “one-target-focused” method limit the development of effective therapeutic interventions. We aimed to reveal the specific active ingredients and mechanisms involved in the Chinese herb Scutellaria baicalensis Georgi (SBG) in alleviating IRI in LT.MethodsThe active ingredients and potential macromolecular targets of SBG were screened through related databases. The differentially expressed genes of LT were obtained from GSE151648. The protein-protein interaction network was constructed by the STRING database, and Cytoscape 3.7.1 was used to construct a compound-target-disease network. GO and KEGG enrichment analyses were performed on the DAVID database. Finally, the main active components of SBG and the corresponding mechanisms were verified in a donation after circulatory death (DCD) rat LT model.ResultsThirty-two active ingredients of SBG and their targets were identified, and a total of 38 intersection targets were obtained. GO function and KEGG pathway enrichment analyses demonstrated that the plasma membrane and its components play an important role. Molecular docking showed baicalein, the core component of SBG, had a strong binding ability to all hub targets. Next, in DCD rats, baicalein was proven to improve liver function, alleviate pathological injury and apoptosis, and increase the survival rate. Baicalein also significantly affected the expression of 7 hub genes. Furthermore, baicalein could inhibit ferroptosis by inhibiting phospholipid peroxidation.ConclusionBaicalein, the main component of SBG, could alleviate IRI, affect the expression of hub genes, and inhibit ferroptosis in LT.

Application of enzyme-assisted extraction of baicalin from Scutellaria baicalensis Georgi

Xiao-Di Ma,Xin-Guo Zhang,Si-Jia Guo,Guo-Yan Ma,Wen-Jie Liu,Nan Wang,Ming Feng,Yu Su

Endophytes may depend on degrading the plant cell wall with cellulases for their survival. Therefore, cellulase produced by endophytes may be useful in releasing the active ingredient of medicinal plants. Scutellaria baicalensis Georgi is a traditional Chinese medicinal plant widely used in China and baicalin is one of its main active ingredients. In this study, fresh S. baicalensis Georgi was used to isolate endophytes, Congo red staining was used to screen cellulase-producing strains, and HPLC was used to determine the content of baicalin in S. baicalensis Georgi. As a result, a highly active strain of endophyte capable of the extraction of high levels of baicalin was obtained. The strain was named HG-5 and identified as Bacillus sp. Scanning electron microscopy analysis confirmed that the enzyme better promotes the dissolution of plant active ingredients. After optimizing the enzyme production and extraction processes, we found that when compared with the traditional extraction method, the baicalin yield was increased 79.31% after extraction with the HG-5 enzyme. The current study provides a novel approach and method for the use of endophyte cellulase to improve the extraction of compounds from medicinal plants.

Cold atmospheric plasma pretreatment on the extraction of bioactive compounds from Scutellaria baicalensis Georgi

Zhang Jiaming,Chu Jiangnan,Wu Zhengwei

Scutellaria baicalensis Georgi (SBG) is a traditional medicinal plant for its anti-inflammatory, antibacterial and antiviral properties. Nevertheless, the current conventional means of extracting the bioactive compounds of SBG are less efficient, and this study aimed to improve the extraction efficiency of flavonoids from SBG by pretreatment with cold atmospheric pressure plasma (CAP) technique. The results showed that CAP pretreatment could facilitate an increase in the specific surface area, a reduction in particle size, thus increasing the area in contact to the extraction solvent. The reactive oxygen of CAP can also degrade cell wall components. Ultimately, the extraction efficiency of flavonoids was improved by 8.25%–59.77%. In conclusion, this study demonstrated that CAP is an effective method to enhance the extraction efficiency of SBG bioactive compounds, indicating the potential to improve the nutritional and functional properties of plants in the food industry.

Scutellaria baicalensis Georgi regulates REV-ERBα/BMAL1 to protect against skin aging in mice

Sun Guanghui,Dang Yongkang,Lin Yanke,Zeng Wanying,Wu Zongjian,Zhang Xingwang,Dong Dong,Wu Baojian

Scutellaria baicalensis Georgi (SBG) is a traditional Chinese medicine widely used to treat disorders such as hypertension, dysentery and hemorrhaging. Here, we aimed to assess the pharmacological effects of SBG on skin aging and to investigate the underlying mechanisms. Mice with skin aging were established by treatment with D-galactose and ultraviolet-B. SBG (topical application) showed a protective effect on skin aging in mice, as evidenced by less formation of skin wrinkles, higher levels of SOD (superoxide dismutase) and HYP (hydroxyproline) as well as a lower level of MDA (malondialdehyde). In the meantime, skin MMP-1 and p53 expression were lower, epidermis was thinner and collagen amount was higher in SBG-treated mice. Anti-skin aging effects of SBG were also confirmed in NIH3T3 and HaCaT cells, as well as in mouse primary dermal fibroblasts and human primary epidermal keratinocytes. Furthermore, we found that loss of Rev-erbα (a known repressor of Bmal1) up-regulated skin BMAL1 (a clock component and a known anti-aging factor) and ameliorated skin aging in mice. Moreover, SBG dose-dependently increased the expression of BMAL1 in the skin of aged mice and in senescent NIT3H3 cells. In addition, based on a combination of Gal4 chimeric, luciferase reporter and expression assays, SBG was identified as an antagonist of REV-ERBα and thus an inducer of BMAL1 expression. In conclusion, SBG antagonizes REV-ERBα to up-regulate BMAL1 and to protect against skin aging in mice.

A spatially-resolved approach to visualize the distribution and biosynthesis of flavones in Scutellaria baicalensis Georgi

Chenglong Sun,Minmin Zhang,Hongjing Dong,Wei Liu,Lanping Guo,Xiao Wang

Imaging the spatial distributions and dynamics of flavones in heterogeneous plant tissues is significant for our understanding of plant metabolism. Here, we proposed a spatially-resolved approach to map the locations and biosynthesis of flavones in S. baicalensis . A total of 11 flavones, 5 flavone glycosides, 6 carbohydrates, and a variety of flavone synthesis-related metabolites were imaged. Most of these flavone-related metabolites presented stronger ion intensities in root phloem. The biosynthetic network of flavones and their glycosides in S. baicalensis were visualized for the first time. Moreover, we characterized the region-specific activities of four crucial enzymes in flavone synthesis pathway, including l -phenylalanine ammonia-lyase, cinnamate 4-hydroxylase, 4-coumarate coenzyme A ligase, and flavone synthase. In line with the spatial characteristic of flavones, all these four enzymes exhibit higher activity in the root phloem of S. baicalensis . The combination of spatially-resolved metabolites and enzymes information greatly broadens our understanding of flavone biosynthetic network.

Effects of copper treatment on the redox state and secondary metabolism of Scutellaria baicalensis Georgi roots

Simeng Wan,Weiwei Wu,Chunlei Ou,Ying Cao,Xinyun Guo,Hua Liu,Lihua Yao,Hu Su

Copper treatment can lead to the accumulation of reactive oxygen species, alter the cellular redox state in plants, and trigger plant adaptive mechanisms such as changes in gene expression and shifts in secondary metabolism. We investigated the effects of copper treatment on the redox state of Scutellaria baicalensis Georgi, characterized by glutathione (GSH) and oxidized glutathione (GSSG) levels. We also determined the concentrations of baicalin and baicalein, and analyzed the correlation between the redox state and these metabolites. Moreover, we analyzed the activity of glutathione reductase (GR, EC 1.6.4.2) and the expression levels of GR, phenylalanine ammonia lyase (PAL, EC 4.3.1.5) and isochorismate synthase (ICS, EC 5.4.4.2) genes. Results indicated that copper treatment increased GSH concentration at 24 and 48h, and the ratio of GSH:GSSG, and upregulated GR expression. While the baicalin concentration showed a non-significant increase at 24h and 72h, baicalein exhibited a significant decrease at 48h and 72h. The two key genes in the salicylic acid pathway, PAL and ICS, exhibited opposite trends at 24 and 48h after copper treatment, followed by significant decreases in both PAL and ICS at 72h. Our results suggest that plants can mitigate the toxic effects of copper through increasing GSH biosynthesis. Baicalin and baicalein showed varying accumulation patterns in S. baicalensis subjected to different copper treatments.

Adsorption and Purification of Baicalin from Scutellaria baicalensis Georgi Extract by Ionic Liquids (ILs) Grafted Silica

Yunchang Fan,Di Wu,Sheli Zhang

Baicalin which has multiple biological activities is the main active component of the root ofScutellaria baicalensisGeorgi (SBG). Although its isolation and purification by adsorption methods have aroused much interest of the scientific community, it suffered from the poor selectivity of the adsorbents. In this work, an environmentally benign method was developed to prepare ionic liquids (ILs) grafted silica by using IL 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide ([C4mim]NTf2) and ethanol as reaction media. The IL 1-propyl-3-methylimidazolium chloride ([C3mim]Cl) grafted silica ([C3mim]+Cl−@SiO2) was used to adsorb and purify baicalin from the root extract ofScutellaria baicalensisGeorgi (SBG). Experimental results indicated that the adsorption equilibrium can be quickly achieved (within 10 min). The adsorption behavior of [C3mim]+Cl−@SiO2for baicalin was in good agreement with Langmuir and Freundlich models and the adsorption was a physisorption process as suggested by Dubinin–Radushkevich model. Compared with commercial resins, [C3mim]+Cl−@SiO2showed the strongest adsorption ability and highest selectivity. After desorption and crystallization, a purity of baicalin as high as 96.5% could be obtained. These results indicated that the ILs grafted silica materials were promising adsorbents for the adsorption and purification of baicalin and showed huge potential in the purification of other bioactive compounds from natural sources. Keywords:ionic liquids (ILs);grafted silica;adsorption;baicalin;purification

Development of a Quantitative NMR Method for Direct and Simultaneous Determination of Four Flavonoids in Scutellaria baicalensis georgi Extracts

Su Feng,Chen Ji-Peng,Zhu Pei-Xi,Wang Yi-Fan,Liang Xian-Rui,Su Wei-Ke

Introduction: Scutellaria Baicalensis Georgi (SBG), a Traditional Chinese Medicine (TCM) used for the treatment of antiviral therapy, contains many flavonoids. Materials and Methods: A simple, fast and accurate 1H quantitative nuclear magnetic resonance (1HqNMR) method was established for simultaneously determination of four flavonoids (Baicalin, Baicalein, Wogonin and Wogonoside) in SBG extract by using 3,4,5-trichloropyridine as an internal standard (IS). All the NMR determination work was performed at 308 K on an NMR sample tube inserted with a co-axial tube. The NMR sample tube was added with the sample (SBG extract and IS), the co-axial insert tube was added with D2O. Quantification of four flavonoids was carried out by calculating the relative peak area ratio of the selected proton signals from the target compounds and the IS. Results: The validated 1H-qNMR method was successfully used to determine the four flavonoids in 9 batches of SBG from different origins and the results were in good accordance with those obtained from HPLC. Conclusion: The established 1H-qNMR method could be used as a powerful tool for the quality control of SBG from different sources and had potential in the quantification of main components from other TCMs.

Combined magnetic molecularly imprinted polymers with a ternary deep eutectic solvent to purify baicalein from the Scutellaria baicalensis Georgi by magnetic separation

Dandan Wang,Xinxin Luo,Yike Huang,Min Wang,Zhining Xia

A ternary deep eutectic solvent (DES) was applied as a novel functional monomer for synthesis of molecularly imprinted polymers (MIPs) to separate baicalein from the Scutellaria baicalensis Georgi ( S. baicalensis ). For rapid separation from the complex systems, modified Fe 3 O 4 particles were used as the magnetic cores to prepare magnetic deep eutectic solvent molecularly imprinted polymers (DES-MMIPs). The prepared DES-MMIPs were characterized by scanning electron microscopy, transmission electron microscopy, dynamic light scattering, Fourier transform infrared spectra, x-ray diffraction, thermal gravimetric analysis, vibrating sample magnetometer and BET analysis. Moreover, the adsorption capability and selectivity of DES-MMIPs to baicalein were investigated systematically. The results of adsorption experiments demonstrated that DES-MMIPs showed excellent binding capacity to baicalein and the selectivity of DES-MMIPs to baicalein was much higher than that of structure analogues (quercetin, hesperetin and keampferol). Finally, DES-MMIPs were applied in selective extraction of baicalein in the S. baicalensis by magnetic separation. The analysis was conducted by high performance liquid chromatography. The recoveries of baicalein in the S. baicalensis ranged from 91.6% to 99.3%. It had been proven that the DES-MMIPs were an innovative and effective adsorbent for selective recognition and separation of baicalein in complex systems.

Selective separation of the homologues of baicalin and baicalein from Scutellaria baicalensis Georgi using a recyclable ionic liquid-based liquid-liquid extraction system

Lingqi Wang,Changyong Cai,Jiajia Liu,Zhijian Tan

To explore convenient and efficient techniques for the extraction and separation of the active ingredients in medicinal plants is still ongoing. As a new class of molten salt, ionic liquids (ILs) are widely used in the extraction of bioactive compounds from plant resources. In this work, the IL-based biphasic system was developed for the extraction and separation of the homologues of baicalin and baicalein from Scutellaria baicalensis Georgi. The IL-water biphasic system is superior to the other organic solvents-water systems for the extraction and separation of baicalin and baicalein. The phase volume ratio of the IL-water biphasic system was studied, and the other influencing parameters of solid–liquid ratio, extraction temperature, extraction time, and ultrasonic time were optimized through the single factor experiments. The results revealed that the extraction yields were 43.3 mg/g and 14.3 mg/g for baicalin (aqueous phase) and baicalein (IL phase), respectively; and the distribution coefficients were 23.8 and 13.7 for baicalin and baicalein, respectively under the optimized conditions. Finally, the IL can be easily recycled and reused by means of its temperature-responsive characteristic. This study provides a green and efficient method for the selective separation of the homologues in natural plant resources.

Improvement of Baicalein and Wogonin Yields from Scutellaria baicalensis Georgi Roots Fermented by a Novel Endophytic Fungus Capable of Producing Cellulase and β-Glucosidase

Fu Jin-Xian,Jiao Jiao,Gai Qing-Yan,He Xiao-Jia,Fu Yu-Jie,Feng Xue,Gao Jie

Baicalein and wogonin, two health-promoting flavonoid aglycones in Scutellaria baicalensis Georgi roots, have better bioactivity and bioavailability than their glycoside precursors (baicalin and wogonoside). The search for novel microorganisms for the fermentation of S. baicalensis roots to improve the yields of baicalein and wogonin is of great interest in the food and nutraceutical industries. In this study, a novel endophytic fungus ( Penicillium rubens ) capable of producing cellulase (0.82 ± 0.12 U/mL) and β-glucosidase (4.55 ± 0.97 U/mL) was used to ferment S. baicalensis roots to obtain high yields of baicalein and wogonin. Semi-solid-state fermentation with homogeneous fungal cultures was found to be the appropriate strategy. Under the optimal fermentation parameters (temperature 30 °C, inoculation dose of fungal cultures 4 mL/g, and time 60 h), the yields of baicalein and wogonin (46.95 ± 4.98 mg/g DW and 31.41 ± 9.36 mg/g DW) from S. baicalensis roots were tremendously increased by 16.13-fold and 15.47-fold over control, respectively. Results of industrial enzyme bioprocessing and scanning electron microscope indicated that the remarkable increase in aglycone product yields could be due to the synergistic effects of cellulase and β-glucosidase produced by P. rubens on cell wall hydrolysis and glycoside deglycosylation. In addition, extracts of S. baicalensis roots fermented by P. rubens showed a significant increase in antioxidant and antimicrobial activities. Overall, the semi-solid-state fermentation of S. baicalensis roots using the novel P. rubens was an effective strategy that could yield high quantities of baicalein and wogonin, which had promising potential in the food and nutraceutical industries.
