Biodegradation mechanism of 1H-1,2,4-triazole by a newly isolated strain Shinella sp. NJUST26

Wu Haobo,Shen Jinyou,Wu Ruiqin,Sun Xiuyun,Li Jiansheng,Han Weiqing,Wang Lianjun

The highly recalcitrant 1H-1,2,4-triazole (TZ) is widely used in the synthesis of agricultural pesticide and considered to be an environmental pollutant. In this study, a novel strain NJUST26 capable of utilizing TZ as the sole carbon and nitrogen source, was isolated from TZ-contaminated soil, and identified as Shinella sp. The biodegradation assays suggested that optimal temperature and pH for TZ degradation by NJUST26 were 30 °C and 6–7, respectively. With the increase of initial TZ concentration from 100 to 320 mg L −1 , the maximum volumetric degradation rate increased from 29.06 to 82.96 mg L −1 d −1 , indicating high tolerance of NJUST26 towards TZ. TZ biodegradation could be accelerated through the addition of glucose, sucrose and yeast extract at relatively low dosage. The main metabolites, including 1,2-dihydro-3H-1,2,4-triazol-3-one (DHTO), semicarbazide and urea were identified. Based on these results, biodegradation pathway of TZ by NJUST26 was proposed, i.e., TZ was firstly oxidized to DHTO, and then the cleavage of DHTO ring occurred to generate N-hydrazonomethyl-formamide, which could be further degraded to biodegradable semicarbazide and urea.

lA multiple PAHs-degrading Shinella sp. strain and its potential bioremediation in wastewater

Zan Wang,Haiyang Hu,Zhan Zhang,Yongming Xu,Ping Xu,Hongzhi Tang

Polycyclic aromatic hydrocarbons (PAHs) and heterocyclic derivatives are organic pollutants which threaten ecosystems and human beings. In this study, a new strain, Shinella sp. FLN 14, was isolated and characterized. It can utilize fluorene as its sole carbon source and effectively co-metabolize multiple PAHs and heterocyclic derivatives, including phenanthrene , acenaphthene, and fluoranthene . Two possible metabolic pathways are proposed (i.e., salicylic acid pathway and phthalic acid pathway). Whole-genome sequencing revealed that strain FLN14 possesses a chromosome and four plasmids. However, when combined with ensemble genetic information, novel fluorene-degrading functional gene clusters were not located within the genome of FLN 14, except for some new dioxygenases and electron transport chains, which typically initiate the oxidation of aromatic compounds. In wastewater bioremediation, strain FLN14 removed nearly 95 % of PAHs within 5 days and maintained high degrading activity during the 18-day reaction compared to the control. Overall, our study provides a promising candidate to achieve bioremediation of PAHs-contaminated environments.

Reduction of tellurite in Shinella sp. WSJ-2 and adsorption removal of multiple dyes and metals by biogenic tellurium nanorods

Shijuan Wu,Tengfei Li,Xian Xia,Zijie Zhou,Shixue Zheng,Gejiao Wang

Te(IV) reduction to Te(0) is a detoxification process in bacteria. Elemental Te nanostructures are new materials, however, the functions of them, such as adsorption ability, have not been fully explored. Shinella sp. WSJ-2 is a highly Te(IV)-resistant bacterium with a MIC of 8 mmol/L. It was able to rapidly reduce 0.3 mmol/L Te(IV) to Te nanorods (TeNRs) within 28 h, and its Te(IV) reduction ability was relatively stable after 14 cycles repeatedly. The size of TeNRs ranged from 50 to 120 nm. The surface of TeNRs was mainly enveloped with proteins and lipids. Moreover, the biogenic TeNRs were capable of removing multiple dyes and metals and were effectively recycled with 100 mmol/L NaCl. Adsorption equilibrium isotherms showed that the maximum adsorption amounts were 618.8, 1076.8, 496.3, 179.3, 58.7 and 275.3 mg/g for Methylene Blue , Azure B, Malachite Green, Cd(II), Cu(II) and Pb(II), respectively. In conclusion, this is the first evidence that the genus Shinella is involved in Te(IV) reduction. The strong sorption of cationic dyes and metals onto TeNRs is probably due to the electrostatic interaction. This study showed the potential of Shinella sp. WSJ-2 in the bioremediation of toxic Te(IV) and adsorption removal of multiple dyes and metals by biogenic TeNRs.

Tetracycline hydrochloride degradation in polarity inverted microbial fuel cells: Performance, mechanisms and microbiology

Yongkang Li,Guangyi Zhang,Danxin Liang,Xiaoqin Wang,Haifeng Guo

Tetracycline antibiotics are widely used in veterinary medicine, human therapy and agriculture, and their presence in natural water raises environmental concerns. In this study, more than 94% of tetracycline hydrochloride (TCH) could be rapidly degraded within 48 h in polarity-inverted microbial fuel cells. The electrochemically active bacteria had the best electrochemical performance at 1 mg/L of TCH with the minimum internal resistance of 77.38 Ω. The electron-rich functional groups of TCH were continuously attacked and finally degradated into small molecules in three possible degradation pathways. Microbial community structure analysis showed that Comamonas and Shinella were enriched at the electrode as polarity-inverted bacteria. Genomic analysis showed that both direct and indirect electron transfer participated in the degradation of TCH in polarity-inverted microbial fuel cell ( MFC) and the functional genes related to electrical conductivity in polarity-inverted MFC were more enriched on the electrode surface than non-polarity-inverted MFC. This study can facilitate further investigations about the biodegradation of TCH in polarity-inverted microbial fuel cell.
