This study addresses the challenge of transitioning Siniperca chuatsi (mandarin fish) from live bait (LF) to compound feed (CF) in aquaculture by examining their stress responses during short-distance transportation. The aim was to compare the physiological and biochemical differences between CF-fed and LF-fed fish under transport stress. The results showed that CF-fed fish exhibited higher levels of lysozyme (LZM) activity, malondialdehyde (MDA), triglycerides (TGs), and glucose in multiple tissues (brain, liver, kidneys, muscles, stomach, pyloric caecum, intestines, and blood) compared to LF-fed fish ( p < 0.05), indicating greater energy retention and microbial resistance but weaker antioxidant capacity. The intestine was particularly affected by dietary differences. This study concludes that while CF-fed S. chuatsi demonstrate benefits in energy metabolism and microbial defense, their reduced antioxidant capacity highlights the need for future research to improve CF formulations to enhance oxidative stress resistance. These findings are valuable for optimizing aquaculture practices, ensuring healthier fish production, and supporting sustainable feed development in the industry. The transition from live bait (LF) feeding to compound feed (CF) feeding in aquaculture is of great production significance. In recent decades, cultivation with CF has become a focus for practitioners and researchers dealing with Siniperca chuatsi . This study focused on experimental subjects of S. chuatsi fed with CF and LF, using short-distance transportation as a stimulating factor. For the first time, the differences between S. chuatsi fed with CF and LF were analyzed from the perspective of stress response during transportation. This study found that after transportation stimulation, the activities of LZM and the contents of MDA, TGs, and glucose in the brain, liver, kidneys, muscles, stomach, pyloric caecum, intestines, and blood of S. chuatsi fed with CF were higher compared to S. chuatsi fed with LF ( p < 0.05). Significant differences were observed in the impacts of various diets on the gastrointestinal tract, particularly in the intestine. In summary, this study found that S. chuatsi fed with CF could retain more energy after transport stimulation and exhibited stronger resistance to microbial stress, but they had a weaker antioxidant capacity. Therefore, in future research on CF for S. chuatsi , we need to focus on its ability to enhance antioxidant capacity.

Tandem Mass Tag (TMT)-based proteomics was employed to elucidate the pivotal role of endogenous enzymes in the quality deterioration of Mandarin fish ( Siniperca chuatsi ) during refrigerated storage. Proclean 300 was used to inhibit the growth of psychrophilic microorganisms, emphasizing the contribution of endogenous enzymes. Quality characteristics, including pH, water-holding capacity, color, texture, and myofibrillar fragmentation index (MFI), were compared between the Proclean 300-treated group (P-group) and the control group (C-group). Correlation analysis underscored the collaborative role of calpain and cathepsin B in postmortem degradation. Pearson correlation analysis showed that texture characteristics were significantly negatively correlated with MFI and significantly positively correlated with whiteness and L* in the P-group. Differentially expressed proteins (DEPs) analysis identified 66 DEPs, with 40 exhibiting low abundance, including cytoskeleton proteins and hemoglobin subunits, and 26 showing high abundance, primarily metabolic enzymes. Bioinformatics analysis revealed significant enrichment of DEPs within the glycolysis/gluconeogenesis pathway. Furthermore, protein-protein interaction analysis highlighted key proteins linked to the quality deterioration of Mandarin fish, including beta-enolase, phosphoglycerate kinase 1, and triosephosphate isomerase B. These findings offer valuable insights into the mechanisms behind the quality deterioration of refrigerated Mandarin fish and the intricate relationship with endogenous enzymes.

Artificial diet slowed early evacuation yet hastened overall gastric emptying. • The artificial diet group had markedly elevated digestive enzyme activities. • Fish fed artificial diet showed early immune responses, then metabolic shifts. Postprandial digestion and metabolism are fundamental to understanding how mandarin fish ( Siniperca chuatsi ) respond to artificial diets. However, whether artificial diets alter the temporal patterns of postprandial digestion and metabolism within a single feeding cycle remains unclear. In this study, fish fed live prey (L) or an artificial diet (A) were sampled at 0, 3, 6, 9, 12, 18, and 24 h postprandially to evaluate gastric evacuation, digestive activity, and blood biochemical parameters, while hepatic transcriptomic responses were analyzed at 0, 3, 12, and 24 h. Gastric evacuation in both groups followed a square-root model, but exhibited markedly different patterns. During the early postprandial phase, the L group showed a progressive reduction in stomach contents, whereas the A group exhibited a transient increase in stomach wet weight; thereafter, stomach contents declined in both groups, with model-estimated complete evacuation occurring earlier in the A group. The A group showed elevated digestive enzyme activity, particularly α-amylase, and a faster and sustained increase in blood glucose. During the early postprandial phase, energy metabolism pathways, including oxidative phosphorylation, glycolysis/gluconeogenesis, pyruvate metabolism, and fatty acid metabolism, were more prominent in the L group, whereas immune-related pathways were more prominent in the A group; at later stages, glucose and lipid metabolism, PPAR signaling, and protein turnover became prominent in the A group. Collectively, artificial diets reshape postprandial physiology in mandarin fish. These findings provide a dynamic physiological perspective on the adaptation of mandarin fish to formulated feed. Lipids are the key aroma contributors and nutrients in fermented fish products. A total of 376 lipid molecules were identified in mandarin fish during fermentation by untargeted lipidomics, including glycerolipids, glycerophospholipids, lysoglycerophospholipid, sphingolipids, fatty acids (FAs), and sterol lipids. Both lipid composition and content changed dynamically during fermentation. Triglyceride (TAG, 30.05%) and phosphatidylcholine (PC, 14.87%) were the two major lipids, with especially 39.36% saturated FAs in PCs and 35.34% polyunsaturated FAs in TAGs. The content of TAGs and PCs reached a peak point at 0 and 6 days, respectively. Fermented mandarin fish expressed a high nutritional value, and the ratio of total linoleic acid/total linolenic acid was about 5:1. Glycerophospholipid metabolism was a potential metabolic pathway, and the oxidation of derived FAs contributed to flavor. These data progress in understanding lipid dynamic variation during fermentation and provide thoughts on controlling the flavor quality and safety of fermented fish products.

There are unique hypothalamic fatty acid (FA) sensing systems in fish that are different from those in mammals. In order to explore the effect of medium-chain FA (MCFA) on the FA sensing systems and appetite regulation, Chinese perch Siniperca chuatsi were subjected to intracerebroventricular (ICV) injection with saline solution alone or saline containing octanoic acid. Food intake of these fish was computed at 2, 4, 6, 12 and 24 h post-treatment, respectively. The transcript levels of genes related to the FA sensing mechanism ( cpt1c , cd36 , pparα and srebp1c ) and hypothalamic neuropeptide ( agrp , npy , pomc and cart ) in the hypothalamus were estimated at 4 h post-treatment. Results showed that at 4 h post-ICV injection with octanoic acid, the food intake of Chinese perch was significantly reduced with the decreasing mRNA level of agrp and the increasing mRNA levels of cd36 and pparα in the hypothalamus. The data indicated that the hypothalamic FA sensing systems of Chinese perch could be activated by octanoic acid, and the underlying mechanism might be that the enhancement of binding ability of FAT/CD36 upregulates the mRNA level of pparα . In addition, with the decrease in the gene expression of agrp , the food intake of Chinese perch was significantly reduced as a result of ICV injection with octanoic acid. These results demonstrated that MCFA might play a key role in central FA sensing and appetite regulation of Chinese perch.

In order to evaluate fatty acid (FA) sensing systems based on binding to FAT/CD36 in hypothalamus of Chinese perch ( Siniperca chuatsi ) and its sensitivity to FAs with the same chain length and different unsaturation levels. The effects of Stearate (SA; C18:0), oleate (OA; C18:1 n-9), linoleic acid (LA; C18:2 n-6), and α-linolenic acid (ALA; C18:3 n-3) on hypothalamic FA sensing were evaluated by intracerebroventricular (ICV) administration. Food intake was assessed after 2, 4, 6, 8 and 12 h. Gene expression associated with FA sensing mechanism such as cd36 , pparα and srebp1c , and neuropeptides controlling appetite such as pomca , cart , agrp2 and npy were assessed after 6 h. The ICV treatment of OA, LA and ALA activated FAT/CD36 and PPARα, rather than SA, and modulated gene expression levels of hypothalamic neuropeptides associated with appetite. And then, OA, LA and ALA inhibited food intake, which was consistent with the activation of hypothalamus FA sensing. Our data indicated some mechanisms of the hypothalamic FA sensing systems also existed in Chinese perch. It's worth noting that polyunsaturated fatty acids (PUFA) could also activate hypothalamic FA sensing mechanisms in Chinese perch. The unsaturation of FA appears to be extremely important for FA sensing mechanisms, since no major influences in Chinese perch after SA treatment. Our findings will contribute to the study of long-chain FAs sensing mechanisms in fish hypothalamus and highlight the importance of PUFAs in fish species.

Summary Identification of aroma compounds in Stinky Mandarin fish ( Siniperca chuatsi ) and comparison of volatiles during fermentation (with spices and salt) and storage were analysed by electronic nose (e-nose) combining with gas chromatography–mass spectrometry (GC-MS). Among the sixty-one detected volatiles, thirteen aroma-active compounds, especially linalool, were identified in stinky mandarin fish according to thresholds and concentrations. Totally, twenty-four aroma compounds correlated well with the periods of fermentation and storage. Trimethylamine, indole, sulphur-containing compounds, acetic acid, esters and phenols increased continually, while aldehydes decreased. According to these quality indicators, e-nose data using principal component analysis showed a clear discrimination of the fermented fish and were in good agreement with the results of GC-MS. In conclusion, fermentation favoured to retard spoilage and provided new aroma compounds. The technique employing an e-nose in combination with GC-MS could compare and identify the aroma and quality of stinky mandarin fish.

To investigate the impact of betaine on high-carbohydrate-diet-induced oxidative stress and endoplasmic reticulum (ER) stress, mandarin fish (Siniperca chuatsi) (23.73 ± 0.05 g) were fed with control (NC), betaine (BET), high carbohydrate (HC), and high carbohydrate + betaine (HC + BET) diets for 8 weeks. The results showed that betaine significantly promoted the growth of mandarin fish irrespective of the dietary carbohydrate levels. The HC diet induced oxidative stress, as evidenced by significantly elevated MDA levels. The HC diet significantly stimulated the mRNA levels of genes involved in ER stress (ire1,perk,atf6,xbp1,eif2α,atf4,chop), autophagy (ulk1,becn1,lc3b), and apoptosis (bax). However, betaine mitigated HC-diet-induced oxidative stress by modulating antioxidant enzymes and alleviated ER stress by regulating the mRNA of genes in the PERK-eIF2a-ATF4 pathway. Additionally, betaine significantly reduced the mRNA levels ofbecn1andbax, along with the apoptosis rate, indicating a mitigating effect on autophagy and apoptosis. Overall, dietary betaine improved growth, attenuated HC-diet-induced oxidative stress and ER stress, and ultimately alleviated apoptosis in mandarin fish. These findings provide evidence for the use of betaine in aquafeeds to counter disruptive effects due to diets containing high carbohydrate levels. Keywords:mandarin fish;high carbohydrate diet;betaine;oxidative stress;ER stress

The mandarin fish ( Siniperca chuatsi ) is a significant freshwater carnivorous species in Chinese aquaculture industry, and its farming scale is continuously expanding. The use of feed in aquaculture has become an increasingly common practice. However, the impacts of substituting artificial feed for live bait on fish's biochemical and immune responses are poorly understood. In this study, two hundred forty mandarin fish (weight: 5.60 ± 0.41 g) were divided into two groups and fed live bait or artificial feed (LB and AF groups) over a 63-day aquaculture experiment. We compared the differences between the two groups in terms of histology, biochemical indicators, gene expression, and bacterial resistance. The results showed that artificial feed promoted enhanced growth, evidenced by higher weight ( p < 0.05). The AF group exhibited higher liver and intestinal somatic indices ( p < 0.05), and histological examination revealed denser cytoplasmic content in liver cells, less fragmentation of renal tubular epithelial cells, and less detachment of intestinal epithelial cells in the AF group. Regarding biochemical indicators and gene expression, the AF group showed better performance in glucose regulation and lipid metabolism. The AF group maintained glucose balance ( p < 0.05) and effectively regulated cholesterol transport ( p < 0.05), promoting lipolysis ( p < 0.05) while inhibiting lipogenesis ( p < 0.05). In contrast, live bait consumption resulted in reduced lipolysis ( p < 0.05), increased lipogenesis ( p < 0.05), impaired endoplasmic reticulum function ( p < 0.05), heightened inflammation ( p < 0.05), and diminished antioxidant capacity ( p < 0.05). Additionally, the LB group exhibited lower survival rates and lysozyme levels during bacterial challenges. Overall, artificial feed was more beneficial for the growth, regulate physiology and enhance disease resistance of S. chuatsi , highlighting its potential to improve fish health and increase aquaculture yield.

The edible and sensory quality of fermented fish products, particularly the formation of garlic clove-structured muscle flakes (GCMF), play critical roles in consumer acceptance and consumption. Herein, aiming to obtain the optimal technical process, this study systematically explored the generation and dynamic evolution of GCMF structure of fermented mandarin fish, especially the integrity and peeling properties of GCMF, which would profoundly determine the textural properties of fish flesh. Meanwhile, flavor profiles were also concentrated during the formation of GCMF. Specifically, our results showed that the optimal fermentation conditions were 3% salt concentration and 7 days of fermentation at 7 °C. Under these conditions, the physicochemical indicators (moisture, pH, TVB-N) of the fermented fish remained within reasonable ranges and the sensory score; peeling integrity of GCMF and the texture properties reached the highest values. In addition, with the increase in fermentation time, the content of undesirable flavor compounds, especially nonanal and 1-octen-3-ol, gradually decreased. Overall, these findings provide a theoretical framework for the evaluation of GCMF structure and for understanding flavor development in fermented mandarin fish, thereby laying a foundation for improved quality control of fermented fish products.

The potential role of prickly ash extracts (PAEs) in enhancing garlic-clove shaped protein gel (GSPG)-forming capacity of mandarin fish ( Siniperca chuatsi ) myofibrillar protein (MP) was studied. PAEs with 0%, 20%, 60%, and 100% ethanol were utilized to systematically analyze changes in the physicochemical properties, water retention, chemical forces, and protein structure of GSPG. Results demonstrated that PAE60 (38.09 mg gallic acid equivalent/g) significantly improved gel strength and water-holding capacity compared to the control ( P < 0.05). The immobilized water (23.04% decrease of relaxation time), hydrogen bonds (56.74% increase), disulfide bonds (174.31% increase), and β -sheet (49.42% increase) became significantly better than the control ( P < 0.05). The gel microstructure had more orderly and compact networks with relatively increases of fractal dimension (1.83%), porosity (22.32%), and number of pores (23.66%) with statistical significances ( P < 0.05). The findings could lay a theoretical groundwork for improving garlic-clove shaped fish protein with plant polyphenols.

Endogenous proteases accelerate myofibrillar protein (MP) degradation and quality loss in fish. This study investigated allicin's protective effects on refrigerated mandarin fish ( Siniperca chuatsi ). Allicin treatment significantly inhibited cathepsin B, cathepsin L, and calpain activities and reduced MP degradation, leading to lower MFI values and slower accumulation of TCA-soluble peptides. Molecular docking and enzyme assays demonstrated that allicin binds to key catalytic residues of these endogenous proteases, suggesting a multi-target inhibitory mechanism. SDS-PAGE and histological staining further confirmed slower degradation of MPs and collagen, along with improved maintenance of muscle fiber integrity. CD and intrinsic fluorescence confirmed allicin attenuated α-helix loss, delayed tryptophan exposure, and stabilized sulfhydryl groups, indicating protection against oxidative and conformational damage. Overall, allicin inhibits endogenous proteases and preserves protein conformation, highlighting its potential as a natural preservative to improve fish storage quality.
