Effects of phosphate on the toxicity and bioaccumulation of arsenate in marine diatom Skeletonema costatum

Qunhuan Ma,Lizhao Chen,Li Zhang

The effects of nutrient phosphate (P) at environmentally relevant levels on the toxicity of arsenic (As) in marine microalgae have been rarely known. In the present study, we explored the toxicity and bioaccumulation of As in a globally distributed diatom species Skeletonema costatum at different ambient P concentrations. The results showed that As toxicity was suppressed at elevated P concentrations. Intracellular As content ([As] intra ) and the molar ratio of intracellular As to P ([As:P]) were negatively correlated with the ambient P concentrations. The trends of As bioaccumulation were substantially different between the relatively low (0, 0.5 and 1.5 μM) and high P concentrations (7.5 and 37.5 μM). Both [As] intra and [As:P] suggested that As bioaccumulation was a better factor to explain the As toxicity comparing to the ambient As concentration. The 4 h As uptake kinetics at different P concentrations followed Michaelis–Menten kinetic pattern. The maximum uptake rates (V max ) decreased with the increase in P concentrations, and the half-saturation constants (K d ) remained constant except for that at extremely high P concentration (37.5 μM-P), suggesting the depression of P on As uptake was mainly due to the non-competitive effect. Overall, these results demonstrate that the P concentration in seawater is an important factor affecting As toxicity and bioaccumulation in the marine diatom. This study therefore helps us better understand the effects of eutrophication on the toxicity and biogeochemistry of As in the marine environment.

Growth inhibition of the microalgae Skeletonema costatum under copper nanoparticles with microplastic exposure

Xiaolin Zhu,Weihong Zhao,Xiaohua Chen,Ting Zhao,Liju Tan,Jiangtao Wang

In order to investigate the combined toxicities of copper nanoparticles (nano-Cu) with microplastic on microalgae Skeletonema costatum , growth inhibition tests were carried out. The toxic effects of copper nanoparticles and microplastic on the microalgae under singleness and coexistence conditions were investigated. Both copper nanoparticles and microplastic inhibited the growth of S. costatum . The growth inhibition ratio (IR) increased with the increasing of particle concentrations and incubation time. The toxicity of copper nanoparticles was reduced with the addition of microplastic. The concentrations of Cu 2+ in the medium with or without addition of microplastic were determined. It was found that adsorption of Cu 2+ on microplastic and aggregation between copper nanoparticles and microplastic are the main reasons for attenuation of toxicity of nano-Cu with adding microplastic. The adhesion and aggregate interactions between microalgae and nanomaterial were also approved by the observations through scanning electron microscopy.

Mitigation effects of CO2-driven ocean acidification on Cd toxicity to the marine diatom Skeletonema costatum

Fang Dong,Pu Wang,Wei Qian,Xing Tang,Xiaoshan Zhu,Zhenyu Wang,Zhonghua Cai,Jiangxin Wang

Ocean acidification (OA) is a global problem to marine ecosystems . Cadmium (Cd) is a typical metal pollutant, which is non-essential but extremely toxic to marine organisms. The combined effects of marine pollution and climate-driven ocean changes should be considered for the effective marine ecosystem management of coastal areas. Previous reports have separately investigated the influences of OA and Cd pollution on marine organisms. However, little is known of the potential combined effects of OA and Cd pollution on marine diatoms. We investigated the sole and combined influences of OA (1500 ppm CO 2 ) and Cd exposure (0.4 and 1.2 mg/L) on the coastal diatom Skeletonema costatum . Our results clearly showed that OA significantly alleviated the toxicity of Cd to S. costatum growth and mitigated the oxidant stress, although the intercellular Cd accumulation still increased. OA partially rescued S. costatum from the inhibition of photosynthesis and pyruvate metabolism caused by Cd exposure. It also upregulated genes involved in gluconeogenesis, glycolysis, the citrate cycle (TCA), Ribonucleic acid (RNA) metabolism, and especially the biosynthesis of non-protein thiol compounds. These changes might contribute to algal growth and Cd resistance. Overall, this study demonstrates that OA can alleviate Cd toxicity to S. costatum and explores the potential underlying mechanisms at both the cellular and molecular levels. These results will ultimately help us understand the impacts of combined stresses of climate change and metal pollution on marine organisms and expand the knowledge of the ecological risks of OA.

Electrochemical biosensor based on strand displacement reaction for on-site detection of Skeletonema costatum

Liu Yaling,Zhang Yibo,Ye Changrui,He Yayi,Wang Shaopeng,Zhang Man,Fu Hao,Lu Shan,Wang Liwei

An electrochemical biosensor was constructed for quantitatively detecting  Skeletonema costatum ( S. costatum ) by combining the electrode modification material (NC-Au) with a strand displacement reaction (SDR). The SDR process addresses the issues of steric hindrance and electrostatic repulsion resulting from the large size of genomic DNA. It enhances the efficiency of the interfacial hybridization reaction and endows the biosensor with remarkable sensitivity. The limit of detection (LOD) and limit of quantification (LOQ) were 33.43 fg/μL (831 cells/L) and 87.21 fg/μL (2112 cells/L), respectively. Additionally, the biosensor has demonstrated excellent accuracy compared to methods such as microscopy and ddPCR ( P  > 0.05). Subsequent assessment of the adjacent waters of the Beibu Gulf using biosensors indicated a low risk of S. costatum red tide outbreaks in the region, which is consistent with the findings of the ecological survey. Therefore, we believe that this biosensor can provide a completely new idea for the dynamic monitoring and early warning of S. costatum red tide. Graphical abstract

Effects of two typical quinolone antibiotics in the marine environment on Skeletonema costatum

Lin Yuxin,Li Tiejun,Zhang Yurong

This study investigated the effects of levofloxacin (LEV) and norfloxacin (NOR) on Skeletonema costatum, focusing on cell growth, chlorophyll a (Chla) content, maximal quantum yield of PSII (Fv/Fm), protein content, enzyme activities of superoxide dismutase (SOD), glutathione reductase (GR), and glutathione peroxidase (GSH-PX), and the membrane lipid peroxidation product malondialdehyde (MDA) content were conducted to analyze the responses of S. costatum under LEV and NOR exposure. Cell growth, Chla content, Fv/Fm, protein content, enzyme activities, and MDA content were assessed to elucidate physiological changes. Both LEV and NOR inhibited S. costatum growth, except for 10 mg/L NOR, which promoted growth. Algal cells exhibited higher sensitivity to LEV, with 96h-IC50 values of 14.770 mg/L for LEV and 44.250 mg/L for NOR. Low NOR concentration (10 mg/L) increased Chla content, while high antibiotic concentrations (>20 mg/L for LEV, >100 mg/L for NOR) decreased Chla content and Fv/Fm, indicating an impact on photosynthesis. Elevated LEV and NOR levels reduced protein and MDA content but increased GR, SOD, and GSH activities, indicating induced oxidative stress. The study provides a comprehensive analysis of LEV and NOR effects on marine microalgae growth and underlying physiological mechanisms, shedding light on potential ecological risks posed by antibiotics in marine ecosystems.

Behavior of tetracycline and polystyrene nanoparticles in estuaries and their joint toxicity on marine microalgae Skeletonema costatum

Li-Juan Feng,Yi Shi,Xiang-Yu Li,Xiao-Dong Sun,Fu Xiao,Jia-Wen Sun,Yue Wang,Xiao-Yun Liu,Shu-Guang Wang,Xian-Zheng Yuan

Polystyrene nanoplastics (PS NPs), which are newly emerging as particulate pollutants, are one of the most abundant plastic types in marine debris. Although there has been extensive research on microplastics , the sorption behavior of PS NPs in surface waters remains unknown. In addition, in the previous joint toxicity studies, the concentration of organic pollutant in the joint system was based on the EC 50 of this pollutant, rather than the actually amount of this pollutant adsorbed on nanoplastics (NPs). In this study, the sorption behavior of PS NPs with different surface charges in the surface water of estuaries and joint toxicity of that absorbed tetracycline antibiotic in equilibrium were investigated for the first time. Because of the electrostatic repulsion, salting-out effect, and partition function, the sorption capacity of tetracycline antibiotic by differently charged PS NPs was enhanced with increasing salinity . The biological effects of exposure to tetracycline-saturated PS NPs were complicated, which can be attributed to the surface characteristics of mixtures such as hydrophobicity and charges. Thus, the role of NPs in the natural environment as a carrier of antibiotics may provide an alternative for antibiotic inputs from inland water to coastal marine water, which would not only change the environmental fate and ecotoxicology of antibiotics and NPs, but also pose challenges to the safety of coastal aquaculture and marine ecosystem.

Physiological effects of sulfadiazine and sulfamethoxazole on Skeletonema costatum and toxicological evaluation using IBRv2 index

Yurong Zhang,Tiejun Li,Yuxin Lin,Dongdong Xu,Haifeng Jiao

Sulfonamide antibiotics, widely used in human and veterinary medicine as well as agriculture, pose environmental concerns due to their stability and poor biodegradability. This study fills a critical gap in understanding the ecological impact of sulfonamide antibiotics on marine microalgae, particularly Skeletonema costatum , a key primary producer in marine ecosystems. This study investigated the biological responses of the marine microalga Skeletonema costatum to sulfadiazine (SD) and sulfamethoxazole (SMX). Both antibiotics significantly impacted S. costatum , with SD having a more pronounced effect. Growth studies showed a clear dose-response relationship: Low concentrations (0.5 mg/L) of SD and SMX stimulated growth, while higher concentrations (3 mg/L, 5 mg/L, and 10 mg/L) inhibited growth. The 96-hour half-maximal inhibitory concentrations (96h-IC 50 ) were 1.654 mg/L and 1.838 mg/L, respectively, initially indicating that SD has a stronger inhibitory effect on S. costatum than SMX. Photosynthetic activity, measured by chlorophyll a content and the maximum quantum yield of photosystem II ( Fv/Fm ) values, showed that low concentrations (0.5 mg/L) of SD and SMX increased photosynthetic efficiency, while high concentrations (3 mg/L, 5 mg/L, and 10 mg/L) significantly inhibited it. Antioxidants activity analysis revealed that SD and SMX exposure altered superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR), and malondialdehyde (MDA) levels. SOD, GR, and GSH-Px levels initially increased but later decreased, suggesting a synergistic effect, while MDA levels consistently increased, indicating oxidative stress and biochemical disruption in algal cells. The Integrated Biomarker Response Version 2 (IBRv2) index provided a comprehensive evaluation of the ecological risks posed by SD and SMX, demonstrating that these antibiotics can significantly disrupt the physiology of marine microalgae. The IBR v2 index provided a comprehensive evaluation of the ecological risks posed by SD and SMX, demonstrating that these antibiotics can significantly disrupt the physiology of marine microalgae. Higher IBR v2 values for SD exposure indicated more substantial impacts on S. costatum . This study underscores the significant ecological risks of sulfonamide antibiotics in marine environments, highlighting the need for further research and regulation to mitigate their impact.

The influences of dissolved inorganic and organic phosphorus on arsenate toxicity in marine diatom Skeletonema costatum and dinoflagellate Amphidinium carterae

Qunhuan Ma,Li Zhang

In this study, arsenate (As(V)) uptake, bioaccumulation, subcellular distribution and biotransformation were assessed in the marine diatom Skeletonema costatum and dinoflagellate Amphidinium carterae cultured in dissolved inorganic phosphorus (DIP) and dissolved organic phosphorus (DOP). The results of 3-days As(V) exposure showed that As(V) was more toxic in DOP cultures than in DIP counterparts. The higher As accumulation contributed to more severe As(V) toxicity. The 4-h As(V) uptake kinetics followed Michaelis–Menten kinetics. The maximum uptake rates were higher in DOP cultures than those in DIP counterparts. After P addition, the half-saturation constants remained constant in S. costatum (2.42–3.07 μM) but decreased in A. carterae (from 10.9 to 3.8 μM) compared with that in the respective P-depleted counterparts. During long-term As(V) exposure, A. carterae accumulated more As than S. costatum . Simultaneously, As(V) was reduced and transformed into organic As species in DIP-cultured S. costatum , which was severely inhibited in their DOP counterparts. Only As(V) reduction occurred in A. carterae . Overall, this study demonstrated species-specific effects of DOP on As(V) toxicity, and thus provide a new insight into the relationship between As contamination and eutrophication on the basis of marine microalgae.

Toxic effects of nano-ZnO on marine microalgae Skeletonema costatum: Attention to the accumulation of intracellular Zn

Cai Zhang,Jiangtao Wang,Liju Tan,Xiaohua Chen

To explore toxic mechanisms of nano-ZnO on marine microalgae, algal growth inhibition test was carried out, and total Zn in the cell and total dissolved Zn in f/2 medium were determined. It was found that nano-ZnO was more toxic than bulk-ZnO on marine microalgae Skeletonema costatum . No matter under nano-ZnO or bulk-ZnO treatment, accumulation of intracellular Zn had a good linear correlation with growth inhibition ratio (IR). The total Zn content in the cell of microalgae was the dominant toxic mechanism of ZnO. Intracellular total Zn could be an indispensable parameter to indicate the toxic effects of nano-ZnO. Higher intracellular total Zn under nano-ZnO treatment than bulk-ZnO resulted in more toxicity of nano-ZnO on microalgae. Compared with Zn 2+ released by nano-ZnO into medium, lipid peroxidation (MDA level) injury, aggregation of nano-ZnO and transmembrane process of nano-ZnO also contributed to toxicity of nano-ZnO on Skeletonema costatum . The accumulation of intracellular Zn provides a new insight into toxic mechanisms of nano-ZnO.

The effect of polystyrene plastics on the toxicity of triphenyltin to the marine diatom Skeletonema costatum—influence of plastic particle size

Yi Xianliang,Wang Jianfeng,Li Zhaochuan,Zhang Zherui,Chi Tongtong,Guo Miao,Li Wentao,Zhou Hao

The effect of polystyrene (PS) particles on the toxicity of triphenyltin (TPT) to the marine diatom Skeletonema costatum was investigated. The 0.1-μm PS particles attached to the cell walls of S. costatum but did not cause adverse effects on the growth of the diatom. The adsorption of TPT to PS particles was negligible in seawater systems, but the presence of 0.1-μm PS significantly reduced the bioavailable concentrations of TPT in f/2-Si medium, indicating a potential three-way interaction between TPT, PS particles, and components of f/2-Si medium. The adsorption of TPT to PS of smaller size (i.e., 0.1 μm) was stronger than that of PS of larger size (i.e., 5 μm), which was probably attributed to larger surface areas of smaller PS particles. The presence of PS could reduce the toxicity of TPT. IC 50 values of TPT increased from 0.56 to 0.85 and 0.71 μg/L at the presence of 20 mg/L 0.1-μm PS and 5-μm PS, respectively. The overall results of this study profiled the combined toxic effects of PS and TPT on marine phytoplankton species and highlighted the difference in adsorption of organic pollutants by microplastics in different ambient mediums.
