Solanum nigrum L. (SN) berry is an edible berry containing abundant polyphenols and bioactive compounds, which possess antioxidant and antiinflammatory properties. However, the effects of SN on alcohol-induced biochemical changes in the enterohepatic axis remain unclear. In the current study, a chronic ethanol-fed mice ALD model was used to test the protective mechanisms of SN berries. Microbiota composition was determined via 16S rRNA sequencing, we found that SN berries extract (SNE) improved intestinal imbalance by reducing the Firmicutes to Bacteroides ratio, restoring the abundance of Akkermansia microbiota, and reducing the abundance of Allobaculum and Shigella . SNE restored the intestinal short-chain fatty acids content. In addition, liver transcriptome data analysis revealed that SNE primarily affected the genes involved in lipid metabolism and inflammatory responses. Furthermore, SNE ameliorated hepatic steatosis in alcohol-fed mice by activating AMP-activated protein kinase (AMPK), acetyl-CoA carboxylase (ACC), peroxisome proliferator-activated receptor α (PPAR-α). SNE reduced the expression of toll-like receptor 4 (TLR4), myeloid differentiation factor-88 (MyD88) nuclear factor kappa-B (NF-κB), which can indicate that SNE mainly adjusted LPS/TLR4/MyD88/NF-κB pathway to reduce liver inflammation. SNE enhanced hepatic antioxidant capacity by regulating NRF2-related protein expression. SNE alleviates alcoholic liver injury by regulating of gut microbiota, lipid metabolism, inflammation, and oxidative stress. This study may provide a reference for the development and utilization of SN resources.

Solanum nigrum is a well-documented cadmium (Cd) hyperaccumulator; however, its Cd-induced tolerance capability and detoxification mechanism remain elusive. Hence, a short-term hydroponic experiment was performed in a multiplane glasshouse to determine the influence of Cd toxicity on subcellular distribution, chemical forms, and the physiological responses of cell wall towards Cd stress in a 4-week-old plant. The experiment was conducted following completely randomized design (CRD) with five treatments ( n = 4 replicates). The results showed that Cd stress showed dose-dependent response towards growth inhibition. The subcellular distribution of Cd in S. nigrum was in the order of cell wall > soluble fractions > organelles, and Cd was predominantly extracted by 1 M NaCl (29.87~43.66%). The Cd contents in different plant tissues and cell wall components including pectin, hemicellulose 1 (HC1), hemicellulose 2 (HC2), and cellulose were increased with the increase in Cd concentrations; however, the percentage of Cd concentration decreased in pectin and cellulose. Results of the polysaccharide components such as uronic acid, total sugar contents, and pectin methylesterase (PME) activity showed Cd-induced dose-dependent increase relative to exposure Cd stress. The pectin methylesterase (PME) activity was significantly ( p < 0.05) enhanced by 125.78% at 75 μM Cd in root, 105.78% and 73.63% at 100 μM Cd in stem and leaf, respectively. In addition, the esterification, amidation, and pectinase treatment of cell wall and Fourier transform infrared spectroscopy (FTIR) assay exhibited many functional groups that were involved in cell wall retention Cd, especially on carboxyl and hydroxyl groups of cell wall components that indicated that the –OH and –COOH groups of S. nigrum cell wall play a crucial role in Cd fixation. In summary, results of the current study will add a novel insight to understand mobilization/immobilization as well as detoxification mechanism of cadmium in S. nigrum .

Multiple drug resistance (MDR) often occurs after prolonged chemotherapy, leading to refractory tumors and cancer recurrence. In this study, we demonstrated that the total steroidal saponins from Solanum nigrum L. (SN) had broad-spectrum cytotoxic activity against various human leukemia cancer cell lines, especially in adriamycin (ADR)-sensitive and resistant K562 cell lines. Moreover, SN could effectively inhibit the expression of ABC transporter in K562/ADR cells in vivo and in vitro. In vivo, by establishing K562/ADR xenograft tumor model, we demonstrated that SN might overcome drug resistance and inhibit the proliferation of tumors by regulating autophagy. In vitro, the increased LC3 puncta, the expression of LC3-II and Beclin-1, and the decreased expression of p62/SQSTM1 in SN-treated K562/ADR and K562 cells demonstrated autophagy induced by SN. Moreover, using the autophagy inhibitors or transfecting the ATG5 shRNA, we confirmed that autophagy induced by SN was a key factor in overcoming MDR thereby promoting cell death in K562/ADR cells. More importantly, SN-induced autophagy through the mTOR signaling pathway to overcome drug resistance and ultimately induced autophagy-mediated cell death in K562/ADR cells. Taken together, our findings suggest that SN has the potential to treat multidrug-resistant leukemia.

Dopamine can alleviate cadmium (Cd) stress in plants and promote plant growth but has differential regulatory effects on Cd accumulation among different plant species. To enhance the phytoremediation capacity of hyperaccumulators, the effects of dopamine on the Cd accumulation in hyperaccumulator Solanum nigrum var. humile were investigated using a pot experiment. The Cd treatment (5 mg/L) induced stress, inhibiting the growth of S. nigrum var. humile by reducing biomass, photosynthetic pigment content, and photosynthetic gas exchange parameters, while increasing antioxidant enzyme activity. Under Cd stress, dopamine treatment (100 μmol/L) increased the root length, tolerance coefficient, and biomass of S. nigrum var. humile, without significantly affecting plant height or root/shoot ratio. Compared to Cd treatment, the root and shoot biomass treated with dopamine treatment increased by 18.97% and 11.57%, respectively. Additionally, dopamine treatment enhanced photosynthetic pigment content, photosynthetic gas exchange parameters, antioxidant enzyme activities, Cd content, and Cd accumulation amount of S. nigrum var. humile. Relative to Cd treatment, the chlorophyll a content, chlorophyll b content, carotenoid content, root Cd content, shoot Cd content, and whole plant Cd accumulation amount treated with dopamine treatment increased by 34.00%, 41.01%, 10.92%, 23.16%, 12.36%, and 35.64%, respectively. Furthermore, correlation and grey relational analyses revealed that root Cd content, superoxide dismutase activity, peroxidase activity, and catalase activity were closely correlated with shoot Cd content. Therefore, dopamine can promote Cd uptake and enhance the phytoremediation capacity of S. nigrum var. humile.

This paper reports a one-step hydrothermal carbonation method for the synthesis of yellow-green carbon dots from Solanum nigrum L, with phosphoric acid as dopant. The phosphorus-doped carbon dots ( P-CDs) were characterized with TEM, XRD, FT-IR, XPS, and UV–Visible. P-CDs provided good fluorescence enhancement of ciprofloxacin with excellent selectivity and sensitivity, while maintaining robust performance against ionic interference and pH variations. The P-CDs demonstrated high sensitivity toward (ciprofloxacin) CIP, with a wide linear detection range of 0.01–0.1 μM and an ultralow detection limit of 6.37 × 10 −3 μM. Moreover, the P-CDs effectively inhibited S. aureus and E. coli growth at a MIC of 0.27 mg/mL. The P-CDs enable rapid antibiotic detection within 5 min while demonstrating potent antibacterial efficacy with a remarkably low propensity for inducing drug resistance, thereby opening new avenues for combating drug-resistant bacterial infections. Notably, the P-CDs offer a simple, rapid, and highly sensitive detection method for CIP analysis in milk samples, making them a promising tool for food safety monitoring. This finding will support the future development of carbon dots for food chemistry applications.

Solanum nigrumL., a widely consumed Asian medicinal edible plant, is a promising source of bioactive saponins for functional food applications. This study optimized the extraction of saponins fromS. nigrumfruits (8.59% total saponin yield), followed by isolation via column chromatography and structural elucidation using spectroscopic analyses (IR, NMR, and MS). Concurrently, the antioxidant properties and antibacterial activity of the purified substances were detected and analyzed. The three saponins (SNL1, SNL2, SNL3) were identified as γ2-Solamargine , Diosgenin, and β-Solanine. Then-butanol -purified fraction demonstrated a remarkable capacity to scavenge DPPH, hydroxyl, and ABTS radicals (DPPH IC50 = 0.0096 mg/mL; hydroxyl radical IC50 = 0.8 mg/mL; ABTS IC50 = 0.061 μg/mL), indicating the inhibition of a multi-pathway oxidative chain reaction. Concurrently, the saponins exhibited selective antimicrobial efficacy against key foodborne pathogens, particularly Escherichia coli. To the best of our knowledge, this work provides the first empirical evidence ofS. nigrumfruit saponins as dual-functional natural preservatives, synergistically suppressing lipid oxidation and microbial growth. These findings highlight their potential as safer, multi-mechanistic alternatives to synthetic additives, aligning with clean-label food industry demands.

This study aimed to investigate the combined ecotoxicological effects of Cd, Pb, Zn, Hg and regulation mechanisms in Solanum nigrum L. In this work, the co-exposure of these four heavy metals hindered the transformation of Cd, Zn, and Hg (except Pb) from available to non-available chemical forms. Individual Cd, Pb, Zn and Hg induced the oxidative damages to S. nigrum L., while their combination further enhanced this ecological toxicity. By internal regulation, the ecological toxicity of metals to S. nigrum L. could be alleviated to a certain extent. Specifically, S. nigrum L. was a hyperaccumulator of Cd with BCF >1. Moreover, since BCFroot of Pb, Zn and Hg were all greater than BCFshoot, S. nigrum L. could accumulate Pb, Hg and Zn mainly in plant roots, which was beneficial for the detoxification of plants. Meanwhile, the immobilization by cell wall (the proportions of Cd, Pb, Zn and Hg in the cell wall were 54.46–84.92%, 38.33–49.25%, 48.38–56.19% and 45.97–63.47% in low metal concentration treatments) and the sequestration in vacuole (the proportions of Cd, Pb, Zn, and Hg in the soluble fractions are 50.99–59.00%, 41.05–45.46%, 37.54–61.04% and 33.47–61.35% in high metal concentration treatments) also act as important detoxification pathways. The external regulation was mainly the changes of soil microbial communities influenced by plants. Specifically, the richness and diversity of bacteria in rhizosphere soil were increased, and roots of S. nigrum L. recruited some potentially beneficial microbials. This study provided a theoretical basis and guidance for S. nigrum L. as a phytoremediation plant under combined heavy metal pollution.

In this research, two novel polysaccharides (S1 and S2) from Solanum nigrum L were extracted and purified. Then homogeneity, molecular weights, major chemical contents and monosaccharide compositions of S1 and S2 were determined. Then, the effects of S1 and S2 on human faecal microbial community and short-chain fatty acid production were investigated using an in vitro fermentation model. Results showed that S1 and S2 have different impacts on human gut microbiota in vitro . S1 selectively promoted the abundance of 9 genera and the production of propionic acid, butyric acid, isobutyric acid, valeric acid and isovaleric acid; while S2 selectively promoted the abundance of 8 genera and the production of acetic acid, propionic acid, butyric acid, isobutyric acid, valeric acid, isovaleric acid and succinic acid. Also, S1 group had higher abundance of genera Butyricimonas and Megamonas and higher levels of lactic acid than S2; while S2 group had higher abundance of Megaphaera and higher levels of butyric acid, valeric acid, isobutyric acid, isovaleric acid and succinic acid comparably. We concluded that S1 and S2 may have potential prebiotic functions.

This study employs Fourier Transform Infrared Spectroscopy (FTIR) to quantitatively analyze the distribution patterns and metabolic roles of α-solanine across different organs (roots, stems, leaves, flowers, and fruits) of Solanum nigrum . By extracting absorbance values from key characteristic peaks (2923 cm −1 , 2853 cm −1 , 1639 cm −1 , 1152 cm −1 , and 1072 cm −1 ) and applying a normalization ratio method for quantitative assessment, our results reveal that fruits serve as the primary accumulation site of α-solanine, followed by flowers and leaves, whereas the lowest concentrations are detected in roots and stems. Further analysis suggests that the high accumulation in fruits is likely associated with defensive functions and seed protection, while leaves and flowers function as the primary sites for α-solanine biosynthesis and signaling regulation, respectively. In contrast, roots and stems mainly facilitate transport and storage. This study highlights FTIR as a rapid, non-destructive technique for the quantitative analysis of plant secondary metabolites, demonstrating its potential in botanical research and biochemical profiling. Moreover, these findings provide valuable insights into the ecological roles of Solanaceae secondary metabolites and their potential applications in agriculture and pharmaceutical sciences.

Cadmium contamination in soil threatens ecological safety and human health. Phytoremediation has gained attention due to its cost-effectiveness and environmental sustainability. Studies show that plant growth-promoting rhizobacteria can enhance the ability of hyperaccumulator plants to remove heavy metals. This research aimed to isolate and identify plant-growth-promoting rhizobacteria under Cd stress and assess their impact on the growth and Cd accumulation ofSolanum nigrumL. Six bacterial strains were isolated from the rhizosphere ofS. nigrum, all showing high Cd tolerance. Among them, LKT25 exhibited multiple growth-promoting traits, including indole-3-acetic acid production, nitrogen fixation, 1-aminocyclopropane-1-carboxylate deaminase, and siderophore synthesis. Under varying Cd concentrations (5, 25, and 50 mg/kg), theBacillus thuringiensisstrain LKT25 significantly improved Cd removal byS. nigrum. At 5 mg/kg Cd, the removal efficiency reached 45.13%. LKT25 also enhanced plant growth, photosynthesis, and antioxidant activity, contributing to improved Cd remediation. This study provides new microbial resources and technical support for using rhizobacteria in remediating heavy metal-contaminated soils.

Introduction Solanum nigrum L. is a traditional medicinal herb and edible plant. Many studies provide evidence that S. nigrum L. is a nutritious vegetable. Polyphenols and steroidal glycoalkaloids are the main components. Objectives This study aimed to systemically evaluate the phytochemical profile, quantification, and bioactivities of polyphenolics and glycoalkaloids in different parts of S. nigrum L. Results Total polyphenols (TPC) and total glycoalkaloids (TGK) were determined using the Folin–Ciocalteu and acid dye colorimetric methods, respectively. A total of 55 polyphenolic constituents (including 22 phenolic acids and 33 flavonoids) and 24 steroidal glycoalkaloids were identified from different parts using ultrahigh-performance liquid chromatography Q-exactive high-resolution mass spectrometry (UHPLC-QE-HRMS), of which 40 polyphenols (including 15 phenolic acids and 25 flavonoids) and one steroidal glycoalkaloid were characterised for the first time in S. nigrum L. Moreover, typical polyphenols and glycoalkaloids were determined using HPLC-UV and HPLC–evaporative light-scattering detector (ELSD), respectively. In addition, the TPC and TGK and their typical constituents were compared in different anatomical parts. Finally, the antioxidant capacities of polyphenolic extracts from different parts of S. nigrum L. were evaluated by ·OH, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ferric-reducing antioxidant power (FRAP) assay in vitro . In addition, the antitumour effects of TGK from different parts of S. nigrum L. on the proliferation of PC-3 cells were investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Polyphenolic and glycoalkaloid extracts from different parts of S. nigrum L. showed different antioxidant and cytotoxic capacities in vitro . Conclusion This is the first study to systematically differentiate between polyphenolic and glycoalkaloid profiles from different parts of S. nigrum L.

Lead (Pb) can possess its solubility, bioavailability and toxicity modified with solid phase transformation, but the potential for such processes in heterogeneous soil-plant system remains unclear. Herein, we investigated the biogeochemical pathways controlling Pb partitioning mediated by strengite (Fe III PO 4 ) and oxalated zero-valent iron (Ox-nZVI). Batch tests verified that Fe III PO 4 and Ox-nZVI achieved the dissolution of stable P and Fe while fixing Pb. Pot experiments with Solanum nigrum L. further showed the simultaneous addition of Fe III PO 4 and Ox-nZVI to soil reduced porewater Pb and boosted the utilization efficiency of P and Fe as a result of favoring Pb retention in the soild phase as Pb 5 (PO 4 ) 3 OH and PbFe 12 O 19 . Characterization results revealed the liquid-solid transfer mass fluctuation depends on precipitation-dissolution and adsorption-desorption reactions of Pb-containing solids driven by Fe III PO 4 reductive dissolution and Ox-nZVI oxidative corrosion. Both Fe III PO 4 and Ox-nZVI promoted plant uptake of Pb in root cell wall as chloropyromorphite crystals but suppressed its upward translocation due to iron plaque barrier. Both Fe III PO 4 and Ox-nZVI recruited more Fe(II)-oxidizing genera and thus facilitated electron transfer processes in near-rhizosphere soil. These findings provide new insights into the role of Fe III PO 4 and Ox-nZVI in microscale Pb stabilization coupled with Fe and P cycling for contaminated media.
