Ethnopharmacological relevance The dried roots of Sophora flavescens Ait. (Leguminosae) is traditionally used as antipyretic medicine to reduce inflammation. It is well known that alkaloids and flavonoids are the main constituents of S. flavescens . However, the clinical researches and applications of S. flavescens is mainly based on its water-extracted alkaloids, its flavonoids may still remain in residues and have been underused. With development and manufacturing of S. flavescens in recent years, more herb residues are being produced. Since they are typically treated as waste and dumped openly in landfill sites, which can cause pollution, there is a great need to explore these wastes as recyclable resources and increase their added value. To date, whether other bioactive components would be found in the residues of S. flavescens is still unknown. If the extraction method of these active ingredients was established, the residues of S. flavescens could be turned from the harm to a benefit and make great sense of the comprehensive utilization of S. flavescens resources. This study aimed to establish an extraction method of the residues of S. flavescens and investigate the anti-inflammatory effect of it both in vivo and in vitro . Materials and methods Dried S. flavescens were decocted with distilled water firstly, then the residues were powdered and extracted with ethyl acetate by using ultrasonic wave. HPLC was utilized to analyze the chemical constituents of the water extracts of S. flavescens (WSF) and the ethyl acetate extracts of residues of S. flavescens (RSF). In vivo , the anti-inflammatory effect of WSF and RSF were evaluated using the xylene-induced auricle edema, acetic acid-induced peritoneal permeability and carrageenan-induced hind paw edema methods. In vitro, the inhibitory activities of WSF and RSF on NO, TNF-α, IL-6 and MCP-1 production of LPS-treated RAW264.7 cells were measured. Results The major ingredients of RSF were flavonoids, while WSF almost had no flavonoids. In vivo , WSF and RSF (200 mg/kg) could significantly inhibit the edema in the xylene-induced mice auricle edema and carrageenan-induced hind paw edema as well as the peritoneal permeability increased by acetic acid. They can also lower production levels of PGE 2 in inflamed paw tissues. In vitro experimental results showed that RSF (25-100 μg/mL) could significantly inhibit the release of pro-inflammatory cytokines NO, TNF-α, IL-6 and MCP-1 on LPS-induced RAW264.7 cells. The in vitro suppress effect of WSF had no dose-response relationship. Conclusions The residues of S. flavescens had obvious flavonoids with anti-inflammatory activity. This study provided evidence for the reuse of residues from S. flavescens in the food additive , medicine and cosmetic industries.

Different endophytes isolated from the seeds of Sophora flavescens were tested for their ability to produce matrine production. Response surface methodology (RSM) was applied to optimize the medium components for the endophytic fungus. Results indicated that endophyte Aspergillus terreus had the ability to produce matrine. The single factor tests demonstrated that potato starch was the best carbon source and the combination of peptone and NH 4 NO 3 was the optimal nitrogen source for A. terreus . The model of RSM predicted to gain the maximal matrine production at 20.67 µg/L, when the potato starch was 160.68 g/L, peptone was 24.96 g/L and NH 4 NO 3 was 2.11 g/L. When cultured in the optimal medium, the matrine yield was an average of 20.63 ± 0.11 µg/L, which was consistent with the model prediction. This study offered an alternative source for the matrine production by endophytic fungus fermentation and may have far-reaching prospect and value.

Carbamazepine (CBZ), an antiepileptic with narrow therapeutic window, is a substrate of CYP 3A4 which metabolizes CBZ to carbamazepine-10,11-epoxide (CBZE). CBZE is an active and toxicity metabolite, and it is a substrate of MRP-2. Using CBZ for a long time can cause hepatic injury. Sophora flavescens (SF) is a medicinal herb used for the protected hepatic injury. This study investigated the acute and chronic effects of SF on the pharmacokinetics of CBZ in rats. The concentrations of CBZ and CBZE in plasma and tissues were determined by HPLC method. The results showed that SF which significantly decreased the AUC 0-t of CBZ, increased CBZE conversely. Tissue analysis showed that the concentrations of CBZ and CBZE in brain and liver were decreased by SF. In addition, the distribution of CBZE in kidney was reduced significantly, which influenced the CBZE excretion and increased the drug toxic potentially. Results in the current study suggest that patients using CBZ might be cautioned in the use of SF extract or Sophora -derived products. Meanwhile, patients receiving drugs which are substrates of CYP 3A4 and/or MRP-2 should be advised of the potential herb–drug interaction to reduce the risk of therapeutic failure or increased toxicity of conventional drug therapy.

Atopic dermatitis (AD) is a chronic inflammatory skin disorder with limited effective treatment. Sophora flavescens Aiton (Kushen) exhibits anti-inflammatory properties, but its key active components and mechanisms against AD remain unclear. This study aimed to identify its therapeutic compounds and the underlying molecular mechanisms. Network pharmacology and ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry were applied to screen for the potential bioactive compounds from Kushen, which were validated in AD mouse models. Formononetin (FMN) was topically administered to evaluate its curative efficacy through histopathological and immunofluorescent staining of skin lesions. Transcriptomic profiling explored the molecular mechanisms, with subsequent validation of neutrophil extracellular traps (NETs)-associated markers by immunofluorescent staining and western blotting. Co-treatment with PAD4 inhibitor GSK484 and molecular docking were used to study the underlying mechanisms. The study identified FMN as an important active component in Kushen, which showed therapeutic effects comparable to dexamethasone in AD models. FMN normalized cutaneous hyperplasia, reduced infiltration of CD3 + T cells, mast cells, and neutrophils, and suppressed inflammatory cytokine expression. Mechanistically, FMN inhibited NETs formation evidenced by decreased levels of citrullinated histone H3 (citH3), myeloperoxidase (MPO), and peptidylarginine deiminase 4 (PAD4). GSK484 co-treatment showed non-synergistic effects, suggesting that FMN suppresses NETs formation primarily through PAD4/MPO inhibition, which was confirmed by molecular docking. These findings highlight FMN as a primary anti-AD constituent of Kushen, therapeutically suppressing NETs-driven crosstalk of innate and adaptive immunity. FMN's multitarget mechanisms provide mechanistic insights and position it as a candidate for AD treatment.

A simple and sensitive electrochemical sensor fabricated with graphene nanosheets (GNs) and a hydroxyapatite (HA) nanocomposite–modified glassy carbon electrode (GCE) was developed for the determination of matrine (MT). The as-prepared electrode (GNs/HA/GCE) was verified to outperform bare a GCE and GNs-modified electrode with increased oxidation peak currents and the decreased over-potential in the redox process of MT, indicating the great enhancement of electrocatalytic activity toward the oxidation of MT by the composite of GNs and HA. Under the optimized conditions, the oxidation peak currents were related linearly with the concentration of MT, ranging from 2 μM to 3 mM, and the detection limit (S/N = 3) was 1.2 μM. In addition, the proposed electrochemical sensor can be successfully applied in the quantitative determination of MT inSophora flavescensextract. Keywords:matrine;voltammetric sensors;Sophora flavescens;electrochemistry;hydroxyapatite

Streptococcosis disease caused by Streptococcus agalactiae (Group B Streptococcus, GBS) results in a huge economic loss of tilapia culture. It is urgent to find new antimicrobial agents against streptococcosis. In this study, 20 medicinal plants were evaluated in vitro and in vivo to obtain medicinal plants and potential bioactive compounds against GBS infection. The results showed that the ethanol extracts of 20 medicinal plants had low or no antibacterial properties in vitro, with a minimal inhibitory concentration ≥256 mg/L. Interestingly, in vivo tests showed that 7 medicinal plants could significantly inhibit GBS infection in tilapia, and Sophora flavescens (SF) had the strongest anti-GBS activity in tilapia, reaching 92.68%. SF could significantly reduce the bacterial loads of GBS in different tissues (liver, spleen and brain) of tilapia after treated with different tested concentrations (12.5, 25.0, 50.0 and 100.0 mg/kg) for 24 h. Moreover, 50 mg/kg SF could significantly improve the survival rate of GBS-infected tilapia by inhibiting GBS replication. Furthermore, the expression of antioxidant gene cat , immune-related gene c-type lysozyme and anti-inflammatory cytokine il-10 in liver tissue of GBS-infected tilapia significantly increased after treated with SF for 24 h. Meanwhile, SF significantly reduced the expression of immune-related gene myd88 and pro-inflammatory cytokines il-8 and il-1β in liver tissue of GBS-infected tilapia. The negative and positive models of UPLC-QE-MS, respectively, identified 27 and 57 components of SF. The major components of SF extract in the negative model were α, α-trehalose, DL-malic acid, D- (−)-fructose and xanthohumol, while in the positive model were oxymatrine, formononetin, (−)-maackiain and xanthohumol. Interestingly, oxymatrine and xanthohumol could significantly inhibit GBS infection in tilapia. Taken together, these results suggest that SF can inhibit GBS infection in tilapia, and it has potential for the development of anti-GBS agents.

Three polysaccharides, namely SHC, SFH, and SFA, were extracted from Sophora sophora under various conditions. A single-factor experimental design was employed to investigate the influence of reaction solvent, sulfating reagent ratio, reaction time, and temperature on the yield of sulfated products, the degree of sulfate group substitution, and their inhibitory effects on HepG2 cell proliferation. Three sulfated derivatives, SHCS, SFHS, and SFAS, were synthesized and confirmed through Fourier Transform Infrared Spectroscopy, Ultraviolet Spectroscopy, and Nuclear Magnetic Resonance Spectroscopy. Among these, SFHS exhibited the highest degree of substitution (DS) at 1.22, followed by SFAS at 0.97 and SFCS at 0.56. In vitro analyses revealed that sulfated modification significantly enhanced free radical scavenging activity, tumor cell growth inhibition, and immune response enhancement. Specifically, SFHS exhibited scavenging activities against DPPH, hydroxyl, and superoxide anion free radicals with rates of 69.8 % ± 6.1 %, 50.4 % ± 4.2 %, and 58.5 % ± 5.1 %, respectively, at 5 mg/mL. Furthermore, it inhibited the proliferation of HepG2 cells by 51.9 % ± 5.6 % at 400 μg/mL and HeLa cells by 43.7 % ± 1.8 % at 800 μg/mL. At 800 μg/mL, SFHS treatment resulted in a phagocytosis rate of neutral red by RAW264.7 macrophages, which was 246.6 % ± 13.9 % relative to the control group, while nitric oxide production was measured at 237.6 % ± 6.2 %. These results suggest that sulfation can enhance the biological functions of polysaccharides derived from S. flavescens , indicating potential applications in functional foods and pharmaceuticals.

A simple, rapid, and sensitive nonaqueous capillary electrophoresis-electrospray ionization-ion trap-mass spectrometry (NACE-ESI-IT-MS) method was developed for determination of matrine and oxymatrine in Sophora Flavescens and medicinal preparations. The conditions for NACE separation and MS detection were systematically optimized. The optimum NACE buffer contained 30 mM ammonium acetate, 1% acetic acid, and 15% acetonitrile in methanol and the applied voltage on separation capillary was set at 25 kV. Berberine was selected as internal standard. In order to generate a stable electrospray, a sheath liquid (isopropanol/H2O, 2/1, v/v) was used, which could also boost the flow through the ESI needle. The matrine and oxymatrine solutions were introduced into MS detection by a syringe pump for collecting the MSn spectra to investigate the main fragment ions and its possible cleavage pathways. Both matrine and oxymatrine showed good linearity in the concentration ranges from 0.5 to 400 µg/mL, with linear correlation coefficient R > 0.99 and the limit of detections were 37.5 ng/mL for matrine and 50.0 ng/mL for oxymatrine, respectively. The recoveries at different content of Sophora Flavescens were 98.3%–102.9% for MT and 95.3%–100.6% for OMT, which indicates the reliability of this method.

The extraction and determination of cytisine , sophocarpine, matrine , sophoridine and oxymatrine in Sophora flavescens Ait. were performed using subcritical water extraction and capillary electrophoresis with field-amplified sample stacking. The effects of extraction temperature, pressure, time and cycle number on the extraction yields were investigated systematically for accelerated solvent extraction with ethanol (ASE) and accelerated solvent extraction with water (subcritical water extraction, SWE). The extraction yields obtained using SWE, ASE, water ultrasonic extraction and chloroform soaking extraction methods were compared. The electrophoresis separation buffer was monosodium phosphate (pH 3.0; 110 mM)-isopropanol (85:15, v/v). The effect of phosphoric acid added to the sample matrix on the reproducibility of the peak heights of the analytes was also examined. Cytisine, sophoridine and oxymatrine showed good linearity ( R 2 > 0.999) within 0.125–4.0 μg/mL, and sophocarpine and matrine exhibited good linearity ( R 2 > 0.998) within 0.0625–2.0 μg/mL, with the detection limits in the range of 0.004–0.0013 μg/mL. The five alkaloid contents in medicinal plants from different sources and Sophora instant granule were determined and compared.

Matrine is an active ingredient in traditional Chinese medicine that has been shown to be effective in treating bone disorders. The anti-osteoarthritis (OA) effects of matrine were assessed using both in in vitro and in vivo systems, and the mechanisms underlying the effects were investigated by focusing on the activity of miR-29b-3p/PGRN axis. The miR was chosen as potential target for matrine after chondrocytes were treated with both IL-1β and matrine. Changes in cell viability, cell apoptosis, inflammation, and miR-29b-3p/PGRN axis were detected. In vitro assays results were validated using collagen-induced arthritis (CIA) rat models. Incubation with IL-1β reduced cell viability, induced cell apoptosis, and inhibited production of cytokines in chondrocytes, which was associated with the up-regulation of miR-29b-3p and down-regulation of PGRN. In CIA rats, matrine reduced bone destruction and weight loss in a dose-dependent manner. Matrine also reduced the systemic levels of cytokines. At the molecular level, matrine inhibited the expression of miR-29b-3p while increasing the expression of PGRN. The findings outlined in the current study showed that matrine exerted its anti-OA effects by modulating the miR-29b-3p/PGRN axis.

Sophora flavescenshas anti-inflammatory, analgesic, antibacterial, antiviral, and anti-tumor properties due to its active compounds, including alkaloids and flavonoids. Exploring the biosynthetic regulation mechanism of these compounds is crucial. Here, we identified 227 flavonoids and 55 alkaloids among five tissues (roots, stems, leaves, flowers, and pods) using wide-targeted metabolomics. Flavonoids were predominantly in roots, whereas alkaloids were primarily in roots and flowers. RNA sequencing revealed 18,488 differentially expressed genes (DEGs) in non-root tissues compared to roots. These DEGs were significantly enriched in pathways such as plant hormone signal transduction, carbon metabolism, flavonoid biosynthesis, and isoflavonoid biosynthesis. Utilizing K-means clustering and WGCNA, we identified ten transcription factors (TFs) potentially regulating the biosynthesis and accumulation of flavonoids (e.g., SfWRKY50 and SfbHLH078) and seven TFs involved in alkaloid biosynthesis (e.g., SfbHLH109 and SfbHLH162) inS. flavescens. These TFs can serve as candidate genes for studying the biosynthetic mechanisms of flavonoids and alkaloids, providing insights into the production of key active compounds and supporting the rational use ofS. flavescenstissues.

In this study, an eco-friendly extraction method that used tailor-made deep eutectic solvents (DESs) for the extraction of oxymatrine and matrine from Sophora flavescens Aiton root was implemented for the first time. The extraction parameters, including the DES type, DES water content, solid-to-liquid ratio, extraction time, and extraction temperature, which affected the extraction efficiencies of oxymatrine and matrine were optimised. High-performance liquid chromatography with a diode-array detector and quantitative analysis of the DES components was conducted. The highest oxymatrine and matrine extraction yields (21.04 and 1.53 mg·g −1 , respectively) were obtained using DES-2 (comprising choline chloride and malonic acid; molar ratio of 1:2) with 50% water and DES-8 (comprising choline chloride and ethylene glycol; molar ratio of 1:2) with 30% water as the extraction solvents, respectively. Furthermore, magnetic molecularly imprinted polymers (MMIPs) were successfully synthesised using oxymatrine, acrylic acid, ethylene glycol dimethylacrylate, azobisisobutyronitrile, and acetonitrile–methylbenzene (3:1, v/v) as the template, functional monomer, cross-linker, initiator, and porogen, respectively. The MMIPs were characterised using transmission electron microscopy, Fourier-transform infrared spectroscopy, and X-ray diffraction analyses, and the results demonstrated the successful preparation of MMIPs. The adsorption properties of MMIPs were further analysed using static and dynamic adsorption experiments, and the results indicated that MMIPs presented good adsorption capacities for oxymatrine and matrine (110.8 and 63.4 mg·g −1 , respectively). Lastly, MMIPs were used as sorbents for the extraction and enrichment of oxymatrine and matrine from DES extracts of S. flavescens roots. The extraction efficiencies of oxymatrine and matrine (80.21–89.15% and 85.33–95.28%, respectively) were satisfactory. The proposed DES-based extraction followed by MMIPs secondary enrichment appeared to be an effective method for the selective extraction of specific components from complex samples.
