A turn-on fluorescent sensor based on carbon dots from Sophora japonica leaves for the detection of glyphosate

Jingzhou Hou,Xianfeng Wang,Shiyu Lan,Chao Zhang,Changjun Hou,Qiang He,Danqun Huo

Carbon dots (CDs) having low cost and low toxicity and synthesized via a green route were applied to establish a fluorescent nanoprobe for the measurement of glyphosate. The synthesis was realized via a one-pot hydrothermal procedure using Sophora japonica leaves as the carbon source. It was found that electron transfer occurred between Fe3+ and the as-prepared CDs. Therefore, Fe3+ exhibited a specific dynamic-quenching toward CDs. However, the electron transfer process was inhibited by glyphosate. The fluorescence of the quenched CDs/Fe3+ system was recovered by the addition of glyphosate. It resulted from the strong complexation between Fe3+ and the functional groups (like –PO3H2 and –COOH) in the glyphosate molecule. These functional groups captured Fe3+ from the CD/Fe3+ system to reduce the electron transfer. With such a design, the rapid detection of glyphosate could be realized by this turn-on fluorescent sensor based on the CD/Fe3+ system. Under optimal conditions, the CD/Fe3+ system showed a concentration-dependent fluorescent response toward glyphosate in the linear range from 0.1 to 16 ppm. The limit of detection was calculated to be as low as 8.75 ppb (3σ/S). In addition, the successful detection of glyphosate in real samples with satisfactory recoveries exhibited a practical application of the CD/Fe3+ nanoprobe in food safety and environmental monitoring.

Synthesis of a magnetic micelle molecularly imprinted polymers to selective adsorption of rutin from Sophora japonica

Hua Zhang,QianYu Zhao,JunQiang Qiu,ZhanHua Wang,Xin Yang

Rutin is a naturally active compound with biological and medical value. The traditional extraction and separation method not only destroys the structure and activity of rutin, but results in a low extraction rate. In this work, the magnetic micellar molecularly imprinted polymer of rutin with a selective recognition function, i.e., RMMMIP was synthesized from 4 to Vinylphenylboron acid and 4-Vinylpyridine as functional monomer, derivatives of cholic acid as amphiphilic molecules. The internal hydrophobic and external hydrophilic characteristics of micelle was used to weaken the solvation of rutin and strengthen the non-covalent interaction between functional monomer and rutin. Fe 3 O 4 , as the core, endowed the composite materials with good magnetic responsiveness and was easy to separate solid from liquid. Then its structure and adsorption were studied, adsorbing capacity and recognition specific factor of RMMMIP are 11.9 mg·g −1 and 3.55 respectively. RMMMIP was used for the separation of rutin from crude extracts of Sophora japonica Linn and showed a better selective adsorption capacity than quercetin, naringin and cyanidin-3-O-glucose. It indicated that RMMMIP as a specific adsorbent had the potential to be a practical way to purify rutin from rutin crude extracts in the future.

A novel and specific molecular imprinted polymer using cellulose as a carrier for the targeted separation of quercetin from Sophora Japonica

Xiaodan Wu,Yawei Zhu,Sansan Bao,Jingsong Cao,Chunjian Zhao,Xiuhua Zhao,Zhiguo Liu,Xiqing Wang,Yujie Fu

In this study, a quercetin molecular imprinted polymer using cellulose as a carrier (CMMs@MPS@MIPs) with high selectivity for quercetin was prepared. Thermogravimetric Analysis (TG), Fourier Transform Infrared spectroscopy (FTIR), Scanning Electron Microscope (SEM), Transmission electron microscope (TEM) and X-ray diffraction (XRD) were used to analyze the material. The adsorption conditions were optimized, resulting in a CMMs@MPS@MIPs adsorption quantity of 13.19 mg g −1 . The adsorption conditions were optimized and the adsorption amount of CMMs@MPS@MIPs could reach 13.19 mg g −1 . Furthermore, the material could still retain 83.5 % of the adsorption performance after five cycles. CMMs@MPS@MIPs was used as an adsorbent to enrich quercetin in Sophora Japonica extract, and its content was greatly increased from 15.73 % to 68.79 %, which proved that CMMs@MPS@MIPs can efficiently separate and enrich quercetin. This research also provides a scientific basis for the targeted separation of target compounds in the food and pharmaceutical by molecularly imprinted polymers using natural polysaccharides as carriers.

Ethanol extract of Sophora japonica flower bud, an effective potential dietary supplement for the treatment of hyperuricemia

Danni Song,Honghui Zhao,Lingling Wang,Fuqi Wang,Lina Fang,Xu Zhao

Hyperuricemia (HUA) is a common metabolic condition associated with a variety of diseases, mainly manifested by the increase in uric acid (UA) levels. As xanthine oxidase (XOD) is the key enzyme in UA production, it has become a target for treating HUA. In this study, an extract named EESJ was extracted from Sophora japonica using 80% ethanol solution and administered to HUA mice to verify its UA-lowering effect. The results confirmed that administering EESJ could reduce the UA levels in vivo by inhibiting XOD activity, and ameliorating renal injury in a dose-dependent manner. To further study the active components in EESJ, 243 chemical components and 32 blood components were identified by the UHPLC-Q-TOF-MS/MS technology, of which flavonoids were the main constituents. The molecular docking results showed that the following six compounds, namely hydroxygenkwanin, genistein , tectorigenin, kaempferol , sophoricoside, and quercetin , had strong binding properties with XOD; among which hydroxygenkwanin had the best binding capacity. In conclusion, this study confirmed that EESJ could effectively reduce UA levels, and speculated the active compounds playing a role in lowering UA levels, which has certain guiding significance for Sophora japonica and natural compounds as potentially effective treatment options for HUA.

Simultaneous determination of phytochemicals and low-weight organic acids in Sophora japonica bud and premium noni juice through column-switching liquid chromatography connected with ion chromatography system

Qiqi Wu,Zhiwei Lin,Haibao Zhu,Chengzhu Ni,Meilan Chen

In this work, a liquid chromatography connected with an ion chromatography by the column-switching technique was developed for the simultaneous determination of three phytochemicals (scopoletin, rutin, and quercetin), three anions (chloride, sulfate, and phosphoric), and seven organic acids (lactic, acetic, propionic, formic, malic, oxalic, and citric acid) in Chinese traditional herbs, S. japonica bud and premium noni juice. A concentration column was used to collect organic acids and anions, and then the concentrated analytes were eluted to an analytical column by column-switching technique in which target analytes could be separated successfully and detected simultaneously within 70 min. This method showed good linearity with the linear correlation coefficients in the range of 0.9971–1.0000. The relative standard deviations of the peak areas were all less than 5.63% (n = 6). The limits of detection (LOD, S/N = 3) of anions and organic acids were in the range of 0.0006–0.0857 mg/L, whereas phytochemicals were in the range of 0.0123–0.0197 mg/L. Recoveries were in the range of 86.53–117.01% for the spiked sample. This method was successfully applied to the determination of the low-weight organic acids and phytochemicals in Chinese traditional herbal medicines, S. japonica bud and premium noni juice.

One-pot preparation of quercetin using natural deep eutectic solvents

Yuan-Yuan Zang,Xi Yang,Zhi-Gang Chen,Tao Wu

In this study, we have established a green and efficient preparation method of quercetin. Rutin was first extracted from Sophora japonica using natural deep eutectic solvents (NADESs), then hydrolyzed into quercetin by rutin degrading enzyme (RDE) obtained from germinated tartary buckwheat in situ. Rutin solubility tests showed that most of the 11 NADESs increased the solubility of rutin by 67-3116 times compared to water. Thus, NADESs could be prior candidate to extract rutin. Extraction efficiency of rutin varied with different NADESs, and a maximum of 291.57 mg g −1 was achieved in NADES ChGly, which was prepared by mixing choline chloride and glycerol at a molar ratio of 1:1. After that hydrolysis was performed directly in extraction system by adding RDE with degradation rate of up to 8.36 mg min -1 ·L -1 . Our findings suggest that preparation of quercetin using NADESs was simple and feasible to operate, environmentally friendly, efficient, and inspired the preparation method of bioactive components from a new perspective.

Enzyme reaction-guided identification of active components from the flowers of Sophora japonica var. violacea

Yunci Zhang,Dongyu Gu,Shuai He,Jing Meng,Jifeng Wang,Yunxiao Wang,Yi Wang,Jing Tian,Yi Yang

The flower of S. japonica is a favorite food and used as traditional medicine. In the present study, a facile and effective method based on the changes in the composition before and after the enzyme reaction was established to screen the active compounds from complex natural products. The separation of an active compound from the ethanolic extracts of Sophora japonica var. violacea, which exhibited the α-amylase inhibitory activity is presented as an example. The analysis of HPLC showed that one component was reduced by 25% after the enzyme reaction. The potential active compound was isolated via LH-20 gel permeation chromatography and identified as kaempferol 3-O-α-L-rhamnopyranosyl-(1 → 6)-β-D-galactopyranosyl-7-O-α-L-rhamnopyranoside by 1H and 13C NMR. The in vitro test indicated that the compound had the α-amylase inhibitory activity, and the IC50 was 88.56 ± 0.60 μg mL−1. The molecular docking study of this compound showed that the compound enfolded in the active sites of α-amylase completely and interacted with the amino acid residues through hydrogen bonds, van der Waals force and hydrophobic interactions.

Sophora japonica Extract Inhibits Ulcerative Colitis and Colitis-Associated Colon Cancer via Regulation of Lipid Metabolism

Ying Zheng,Tianjiao Li,Xi Luo,Mengnan Jia,Liying Han,Hao Yu,Yongrui Bao,Shuai Wang,Xiansheng Meng

Inflammation is a well-known risk factor for tumor development. Intervening in chronic inflammation may be an effective approach to inhibit the progression of ulcerative colitis (UC) to colitis-associated colon cancer (CAC). Sophora japonica is a medicinal and food plant commonly used to treat hemorrhoids, bleeding, and inflammation, symptoms that coincide with those of patients with CAC. However, studies on using Sophora japonica to treat CAC are scarce. Therefore, this study aimed to investigate the anti-inflammatory and antitumor effects of Sophora japonica extract (SJE) using dextran sodium sulfate (DSS)-induced UC and azoxymethane (AOM)/DSS-induced CAC models. Furthermore, we employed ultraperformance liquid chromatography–quadrupole time-of-flight–tandem mass spectrometry (UPLC–QTOF–MS/MS) technology to explore the metabolomics mechanism through which SJE inhibited the progression of UC to CAC. The results demonstrated that SJE significantly inhibited UC and CAC and alleviated symptoms such as bloody stools, colon shortening, inflammation, and intestinal tissue damage in model mice. Further, 57 differential metabolites were identified by UPLC–QTOF–MS/MS, which were composed mainly of lipids (fatty acids, glycerophospholipids, sterol lipids, pregnenolone lipids, etc.) and peptides (amino acids). Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis suggested that SJE might inhibit the progression of UC to CAC by regulating lipid metabolic pathways such as arachidonic acid metabolism, primary bile acid biosynthesis, and linoleic acid metabolism. This study, for the first time, substantiated the inhibitory effect of SJE on CAC. Employing metabolomics techniques, the research delved into the potential pathways through which SJE may exert its inhibitory effects on CAC by suppressing the progression of UC. These findings contribute scientific evidence to the application of Sophora japonica in functional foods and the development of natural drugs for the prevention and treatment of UC and CAC.

Functional Food Potential of Chrysanthemum morifolium, Perilla frutescens, and Sophora japonica in Managing Hyperuricemia through Dual Enzyme Inhibition

Kai-lin Chen,Dong-dong Xie,Man-ping Luo,Bing Liu,Yang Li,Yu-jie Zhao,Xiao-xiao Zhao,Jia-mei Pei,Yong-gang Ding,Zai-Ping Feng,Bei Wang,Xin-guo Zhang

Amid growing concerns regarding gout and hyperuricemia associated with high-protein and purine-rich diets, the need for effective prevention and management strategies with minimal side effects has become increasingly critical. This study evaluates the potential of three commonly consumed plant-based functional foods, Chrysanthemum morifolium, Perilla frutescens, and Sophora japonica, inhibiting xanthine oxidase (XO) and adenosine deaminase (ADA), key enzymes in uric acid metabolism. Results from hyperuricemia model mice indicate that this blend significantly reduces serum uric acid levels, mirroring the efficacy of conventional prevention and management strategies such as allopurinol but with fewer adverse effects. Liquid chromatography–mass spectrometry (LC-MS) analysis confirms that flavonoids are the primary bioactive agents, exhibiting a strong affinity for XO. These findings highlight the viability of integrating plant-based functional foods into comprehensive gout management strategies, underscoring their role in enhancing patient health through dietary innovation.

Hand-Held Electrostatic Spray Ionization Mass Spectrometry Enables On-Site Metabolomic Analysis of Sophora japonica and Robinia pseudoacacia

Song Mengyang,Luo Haiyan,Ji Zhenyang,Ye Zhihao,Yang Qinqin,Liu Xiaoyi,Shi Buyin,Li Yuze,Nie Zongxiu

Mass spectrometry plays a key role in agricultural and food chemistry, yet direct ionization of plant samples is complicated by endogenous salts and other matrix components that destabilize conventional direct-current nanoelectrospray ionization (DC-nanoESI). To address this limitation in point-of-need analysis, we developed hand-held electrostatic spray ionization (HESSI), which produces stable ionization from nanoliter sample volumes without direct electrical contact with the solution. A compact pulsed-discharge module and dielectric barrier limit electrochemical reactions and reduce the risk of discharge at the emitter while remaining compatible with salt-containing plant extracts. Approximately 50 nL of the sample sustained MS signals for up to 5 min. In a proof-of-concept application, 208 compounds were tentatively annotated from Sophora japonica and Robinia pseudoacacia extracts in negative-ion mode, and a neural network model distinguished the two groups with an accuracy of 83.3%. These results support HESSI as a compact and sample-efficient platform for the rapid mass-spectrometric characterization of botanical materials.

Novel bayberry-and-honeycomb-like magnetic surface molecularly imprinted polymers for the selective enrichment of rutin from Sophora japonica

Huijia Song,Haipin Zhang,Yulian He,Ruixia Gao,Yue Wang,Wenting Wang,Lisa D. Pfefferle,Xiaoshuang Tang,Yuhai Tang

Rutin (RT), a widely distributed natural flavonoid compound, has been generally utilized as an important active ingredient owing to its considerable biomedical and economic value. Inspired by the structure features of densely-packed bayberry and well-orientated honeycomb, a novel type of magnetic molecularly imprinted polymers (HB-TI-MMIPs) with abundant high-affinity and uniformly-distributed binding sites was rationally constructed for the selective enrichment of RT from Sophora japonica. The polymerization conditions, physicochemical properties, and adsorption performance of the imprinted nanomaterials were systematically investigated. The optimized HB-TI-MMIPs display a high adsorption capacity, fast adsorption rate, and satisfactory selectivity towards RT. Meanwhile, the proposed analytical methodology using HPLC, with HB-TI-MMIPs as adsorbents, successfully applied to enrich and detect RT from Sophora japonica with high recoveries (87.2–94.6%) and good RSDs (lower than 4.3%). Therefore, the fabricated HB-TI-MMIPs with a fast magnetic responsivity and desirable adsorption performance would be attractive in plant active ingredients extraction fields.

Extraction and Application of Natural Rutin From Sophora japonica to Prepare the Novel Fluorescent Sensor for Detection of Copper Ions

Yang Shilong,Sun Lu,Song Zhiwen,Xu Li

Rutin (R), a representative flavonoid found in various biomasses, can be used to prepare different fluorescent sensors for environmental, biological and medical fields. In this work, the natural R in Sophora japonica was extracted and purified to prepare fluorescent-responding sensor systems intended to recognize copper ions with both strong selectivity as well as appropriate sensitivity. Results showed that neat R had no obvious fluorescent emission peak in PBS buffer solution. However, when R and (2-hydroxypropyl)-β-cyclodextrin (CD) were introduced within buffer solution, fluorescent emission intensity was significantly increased due to the resultant R-CD inclusion complex. In addition, the formed R-CD inclusion complex was shown to behave as the aforementioned fluorescent sensor for copper ions through a mechanism of quenched fluorescent emission intensity when R-CD became bound with copper ions. The binding constant value for R-CD with copper ions was 1.33 × 106, allowing for quantification of copper ions between the concentration range of 1.0 × 10–7–4.2 × 10–6mol⋅L–1. Furthermore, the minimum detection limit was found to be 3.5 × 10–8mol⋅L–1. This work showed the prepared R-CD inclusion complex was both highly selective and strongly sensitive toward copper ions, indicating that this system could be applied into various fields where copper ions are of concern.
