Structural characterization, antioxidant and hepatoprotective activities of polysaccharides from Sophorae tonkinensis Radix

Liangliang Cai,Shanshan Zou,Dengpan Liang,Libiao Luan

In current study we present two polysaccharides, STRP1 and STRP2, purified from Sophorae tonkinensis Radix via column chromatography. Structural analyses indicated that STRP1 and STRP2 were consisted of mannose, rhamnose, glucuronic acid, glucose, galactose and arabinose in a similar molar ratio with main backbones of (1 → 3)-linked-α- d -Gal and (1 → 4)-linked-α- d -Glc, while average molecular weights were 1.30 × 10 4 and 1.98 × 10 5  Da, respectively. We observed a strong chelating ability on ferrous ions; substantial radical scavenging activities on DPPH, hydroxyl and superoxide anion radicals in vitro; and significant attenuation on acetaminophen-induced hepatic oxidative damage in mice for STRP1 and STRP2. The promising data on these polysaccharides showcase the need to further develop novel natural antioxidant and liver-protecting drugs.

Multi-omics and chemical profiling approaches to understand the material foundation and pharmacological mechanism of sophorae tonkinensis radix et rhizome-induced liver injury in mice

Si-Wei Rao,Cheng-Jun Liu,Dong Liang,Yuan-Yuan Duan,Zi-Hao Chen,Jin-Jin Li,Han-qing Pang,Feng-Xiang Zhang,Shi Wei

Ethnopharmacological relevance Sophorae tonkinensis Radix et Rhizoma (STR) is an extensively applied traditional Chinese medicine (TCM) in southwest China. However, its clinical application is relatively limited due to its hepatotoxicity effects. Aim of the study To understand the material foundation and liver injury mechanism of STR. Materials and methods Chemical compositions in STR and its prototypes in mice were profiled by ultra-performance liquid chromatography coupled quadrupole-time of flight mass spectrometry (UPLC-Q/TOF MS). STR-induced liver injury (SILI) was comprehensively evaluated by STR-treated mice mode. The histopathologic and biochemical analyses were performed to evaluate liver injury levels. Subsequently, network pharmacology and multi-omics were used to analyze the potential mechanism of SILI in vivo . And the target genes were further verified by Western blot. Results A total of 152 compounds were identified or tentatively characterized in STR, including 29 alkaloids, 21 organic acids, 75 flavonoids, 1 quinone, and 26 other types. Among them, 19 components were presented in STR-medicated serum. The histopathologic and biochemical analysis revealed that hepatic injury occurred after 4 weeks of intragastric administration of STR. Network pharmacology analysis revealed that IL6, TNF, STAT3, etc. were the main core targets, and the bile secretion might play a key role in SILI. The metabolic pathways such as taurine and hypotaurine metabolism, purine metabolism, and vitamin B6 metabolism were identified in the STR exposed groups. Among them, taurine, hypotaurine, hypoxanthine, pyridoxal, and 4-pyridoxate were selected based on their high impact value and potential biological function in the process of liver injury post STR treatment. Conclusions The mechanism and material foundation of SILI were revealed and profiled by a multi-omics strategy combined with network pharmacology and chemical profiling. Meanwhile, new insights were taken into understand the pathological mechanism of SILI.

Efficient and green ultrasonic-assisted extraction of flavonoids from Sophorae Tonkinensis Radix Et Rhizoma using deep eutectic solvent: Optimization and extraction mechanism

Jianwei Luo,Zhengmin Yang,Xinghai Huang,Jinyun Cai,Haiyi Zhong

For the advancement of an environmentally friendly and sustainable plant industry, an excellent high-efficiency extraction procedure was exploited for total flavonoids from Sophorae Tonkinensis Radix Et Rhizoma (STRR) based on deep eutectic solvents (DESs) integrated with ultrasonic-assisted extraction (UAE). A comparative study assessed the extraction performance of traditional organic solvents and 17 DESs, with the choline chloride and acrylic acid combination yielding the best results. Optimization through response surface methodology (RSM) achieved a maximum flavonoid yield of 3.785 ± 0.026 mg/g under optimal conditions: 61 min, 65 ℃, solid/liquid ratio 1:148 g/mL, molar ratio 1:4, and 20 % water content. HPLC-MS was employed to quantify the three flavonoids (quercetin, rutin, and formononetin) in the extract, revealing their extraction yield in the DES to be 3.039 ± 0.226 mg/g, 41.274 ± 2.545 mg/g, and 73.926 ± 6.603 mg/g, respectively. These levels are 2–4 times higher than those in the methanol and ethanol extracts. Furthermore, the extraction mechanism was further elaborated with density functional theory (DFT) analysis showing the presence of non-covalent interactions (hydrogen bonding and van der Waals forces) between DES and the samples. The analysis highlighted the excellent extraction performance of DES from multiple aspects and perspectives, including electrostatic potential (ESP), mutual penetration distance (MPD), interaction region indicator (IRI), binding energy, and electrostatic potential quantitative distributions. Thus, the findings of this study demonstrate that DES-UAE represents a highly efficient and environmentally sustainable approach for extracting bioactive compounds from plants, thereby providing valuable support for the development and application of this green extraction technique in the plant industry.

Biolabel-led research pattern positions the effects and mechanisms of Sophorae Tonkinensis radix et rhizome on lung diseases: A novel strategy for computer-aided herbal medicine research based on omics and bioinformatics

Shuai-nan Zhang,Ya-feng He,Xu-zhao Li,Wu-de Yang,Ying Zhou

Previous studies have shown that Sophorae Tonkinensis radix et rhizome (ST) can be used to treat some lung diseases . However, the therapeutic potentials, therapeutic advantages, mechanism of action, and material basis of ST treatment of lung diseases remain unclear. Thus, the aim of this study was to carry out an integrated analysis based on the biolabel-led research pattern. Proteomics and metabonomics were applied to explore the biolabels responsible for the effect of ST on lung tissue. Based on the biolabels, a bioinformatics database was used to topologically analyze the therapeutic potentials, therapeutic advantages, mechanism of action, and material basis of ST in treating lung diseases. Four human lung-cancer cell models were used to validate the results of the biolabel analysis. In total, 45 proteins and 3 metabolites were significantly enriched in 13 pathways and were considered as biolabels. Bioinformatics revealed that the therapeutic potentials of ST involved a variety of lung diseases, especially lung neoplasms. Under the mediation of 40 biolabels, 29 compounds may be the material basis of ST in treating lung diseases. In a verification experiment, ST had a significant inhibitory effect on the H226 cell line (lung squamous cell carcinoma), which ranks first in morbidity and mortality among lung cancers in China. Additionally, five biolabels (CPS1, CKM, CPT1B, COX5B, and COX4I1) were involved in the anti-lung cancer mechanism of ST and 3 compounds (gallic acid, betulinic acid , and caffeic acid). These findings indicate that the biolabel-led research pattern was helpful in achieving the objectives of this study.
