Effects of sophorolipids and coconut wax incorporation on the physical, structural, and antibacterial properties of cellulose nanofibers-based Pickering emulsion for cherry tomato preservation

Guangshun Li,Tong Wang,Qing Wei,Zhigang Jin,Huipei Han,Huixia Zhu,Xiaojing Ma

A novel acidic sophorolipid-enhanced cellulose nanofiber-based coconut wax Pickering emulsion (ASL-CCPE) using cellulose nanofibers (CNF) as a stabilizer, lactonic sophorolipids (LSL) as an antimicrobial agent, and acidic sophorolipids (ASL) as a co-stabilizer were developed. Through optimization of the oil phase type, oil-to-water ratio, and ASL content, the emulsion exhibited shear-thinning behavior and elastic gel characteristics, making it suitable for coating applications. After 182 days of storage at room temperature, the emulsion's droplet size remained uniform, with no significant flocculation observed. Compared to CCPE, ASL-CCPE exhibited reduced surface tension and formed a more uniform, dense, and washable coating. Cherry tomatoes coated with ASL-CDPE remained fresh for at least 12 days at room temperature. Furthermore, ASL-CCPE treatment resulted in a 5.10 % reduction in weight loss and a 45.83 % increase in vitamin C content even after 15 days of storage. Additionally, the inclusion of LSL significantly inhibited microbial growth, thus reducing decay. This green, safe, and effective emulsion offers a promising, environmentally friendly approach for fruit and vegetable preservation.

Amide Modification of Glycolipid Biosurfactants as Promising Biocompatible Antibacterial Agents

Tianyou Cui,Lianpeng Ge,Mengqian Zhao,Li Luo,Xuwei Long

Discovering new antibacterial agents is crucial to addressing the increasing risk of bacterial infections induced by antimicrobial resistance in food and agricultural industries. Here, biocompatible acidic-type sophorolipids (ASLs) and glucolipids (GLs) prepared via chemical modification of natural sophorolipids from fermentation were functionalized via amide modification for use as potential antibacterial agents. It was found that the arginine methyl ester derivative of GLs (GLs-d-Arg-OMe) showed excellent antibacterial activity, killing more than 99.99% of Escherichia coli at 200 mg/L. The sterilization dosage of the GLs against Bacillus subtilis, Bacillus cereus, and Staphylococcus aureus was 16–64 mg/L, in contrast to 32–64 mg/L for the fungus Candida albicans. In particular, GLs-d-Arg-OMe showed the best biocompatibility with a therapeutic index of up to 18. It was shown that amide modification of glycolipids can effectively improve antibacterial activity while maintaining biocompatibility, which can be exploited for the development of novel antibiotics in food and agricultural fields.

Efficient Preparation of Sophorolipids and Functionalization with Amino Acids to Furnish Potent Preservatives

Yujing Tang,Mingjie Jin,Tianyou Cui,Yang Hu,Xuwei Long

The development of new preservatives is an ongoing investigation in the food industry, especially those which are safe and environmentally friendly. In this study, biosurfactant sophorolipids (SLs) functionalized with amino acids were developed as efficient preservative agents. SLs were first isolated from fermentation broth by Candida bombicola ATCC 22214, hydrolyzed, and purified by extraction. The typical recovery is around 70%, while the extracted material consists of over 90% deacetylated acidic SLs (SL-COOH). Four types of SL derivatives were then synthesized via dicyclohexylcarbodiimide amidation reactions from prepared SL-COOH. Among the derivatives produced, the arginine SL conjugates (SL-d-Arg) displayed the highest activity against Gram-positive bacteria and fungi and even inhibited the cell growth of Gram-negative bacteria and mildew. Furthermore, the arginine conjugates performed the broadest antimicrobial activity among the derivatives evaluated. The sterilization dosage of the arginine conjugates against the food-spoilage pathogen Bacillus spp. was 63–125 mg/L, in contrast to 250 mg/L for the enterotoxin producer Staphylococcus aureus and 500 mg/L for fungi. More importantly, SL-d-Arg displayed excellent biocompatibility, with a therapeutic index of over 7.94. SL-d-Arg has excellent potential as an alternative to traditional chemical preservatives.

Bio-Surfactant (Sophorolipids) Pre-Coating for Thin-Film Composite Membranes With Enhanced Dye Separation Performance

Xin-Xi Ding,Cheng Jiang,Yong-Feng Yang,Shu-Tian Du,Ming-Guang Li,Dan-Dan Shao

With increasing concerns over resource depletion and environmental pollution, resource recycling has emerged as a key strategy for promoting sustainable development. Nanofiltration (NF) membranes, characterized by their well-defined pore size and surface charge, offer significant potential for small-molecule separation and resource recovery. However, conventional polyamide (PA) NF membranes prepared via interfacial polymerization often exhibit dense selective layers, resulting in limited permeability and constrained practical application. In this study, a cost-effective and biodegradable biosurfactant, sophorolipids, was employed as a pre-coating agent on porous polyethersulfone (PES) substrates to modify the interfacial polymerization process. The resulting NF membranes demonstrated a nearly fivefold increase in water permeability (from 5 to 25 L m −2 h −1 bar −1 ) while maintaining excellent selectivity—achieving high rejection of large molecular dyes and low rejection of small molecular dyes. The membranes effectively separated dye mixtures containing Alcian blue (MW: 1298.86 g·mol −1 ) and crystal violet (MW: 407.98 g·mol −1 ), as well as dye/salt mixtures with sodium chloride, exhibiting outstanding dye/salt selectivity. These results highlight the strong potential of sophorolipids-modified NF membranes for the sustainable treatment and resource recovery of textile dyeing wastewater.

Formulation and Physicochemical and Biological Characterization of Etoposide-Loaded Submicron Emulsions with Biosurfactant of Sophorolipids

Ma Xiaojing,Wang Tong,Yu Zequan,Shao Junqian,Chu Jun,Zhu Huixia,Yao Risheng

Etoposide (ETO), a traditional anticancer chemotherapeutic agent, is commercialized in oral soft gelatin capsules and non-aqueous parenteral solutions form. Novel formulation application and new excipients exploration are needed to improve the water-solubility and comfort of the drug administration. In the present study, novel etoposide-loaded submicron emulsions (ESE) with the biosurfactants of acidic sophorolipid (ASL) and lactonic sophorolipid (LSL) instead of the chemical surfactant of Tween-80 were prepared and characterized. Firstly, parameters of medium-chain triglyceride: long-chain triglyceride (MCT:LCT), lecithin concentration, homogenization pressure and cycle, and type and concentration of surfactants were investigated to optimize the formation of ESEs. Then the physicochemical properties, antitumor activity, stability, and security of ESEs were compared. The results showed that ASL performed the best properties and activities than Tween-80 and LSL in ESE formation. ASL-ESE showed higher drug loading capacity, slower release rate, and significantly increased antitumor activity against ovarian cancer cell line A2780 via apoptosis than Tween-ESE and commercial ETO injection. Besides, both ASL-ESE and Tween-ESE caused no hemolysis, and the safe dose of ASL was 2.14-fold that of Tween-80 in the hemolysis test, making ASL more reliable for drug delivery applications. Furthermore, ASL-ESE exhibited equivalent long-term and autoclaving stability to Tween-ESE. These results thus suggested the excellent competences of ASL in ESE formation, efficacy enhancement, and safety improvement. Graphical abstract

Efficient purification of sophorolipids via chemical modifications coupled with extractions and their potential applications as antibacterial agents

Yujing Tang,Qianqian Ma,Yulu Du,Lu Ren,Leonardo J. Van Zyl,Xuwei Long

Efficient synthesis and purification of sophorolipids produced by fermentation into ester and free acid derivatives has attracted significant interest as these compounds have many potential specific applications. In this study, the ester and free acid derivatives of sophorolipids were prepared via chemical modifications, and the purification via a series of stepwise extractions was optimized. Acidic type sophorolipids (SL-COOH) was readily obtained by hydrolysis of sophorolipids isolated from fermentation broth, while the lactonic sophorolipids (SL-ester) can be prepared by the esterification of SL-COOH. Then, a series of newly developed stepwise extractions using optimized solvent systems recovered more than 50% of the SL-COOH and about 80% of the SL-ester with high purity, respectively. Both modified SLs have great surface activities, can reduce the surface tension to 40–43 mN/m with critical micellar concentrations ranged from 50 to 120 mg/L. More importantly, SL-ester presented excellent antibacterial activity against B. subtilis and Moesziomyces sp ., and the inhibition efficiency reached over 96% and 83% at concentration of 1 g/L, was much better than that by natural SLs and SL-COOH. Such chemical modifications followed by the newly developed purification would be conducive to the practical applications of sophorolipids in several fields, especially as antibacterial agents.

Anti-fouling loose polyamide nanofiltration membrane preparation by biodegradable sophorolipids for precise separation of simulated dyeing wastewater

Xin-Xi Ding,Cheng Jiang,Mingguang Li,Xianghong Li,Dan-Dan Shao

Background Strengthening the recycling and utilization of resources is deeply significant in achieving carbon neutrality. The nanofiltration membranes possess separation ability within the nanoscale by utilizing special pore size ranges and surface charge properties, showing great application prospects in resource recycling. The commercial nanofiltration membranes are mainly prepared by interfacial polymerization with fast reaction speed and easy scaling-up advantages. However, the obtained polyamide nanofiltration membranes possess dense selective layers with low permeance and separation efficiency to the molecules. Herein, the biodegradable surfactant sophorolipid was added to the aqueous solution to improve the performance of the polyamide nanofiltration membranes. Results Compared with the traditional nanofiltration membranes, the permeance of the sophorolipid-modified nanofiltration membrane was improved from 3 to 18 L·m −2 ·h −1 ·bar −1 . The modified membrane showed high rejection of the dyes with large molecular structures and low rejection of those with small molecular structures. Conclusion The modified membrane may separate the mixed dye solution of congo red (molecular weight 696.66 g/mol) and indigo carmine (molecular weight 407.98 g/mol) and the mixed congo red/NaCl solution precisely. Besides, the modified membrane showed good anti-fouling properties in the long-term stability test. This study may offer research significance for the resource treatment of printing and dyeing wastewater. © 2024 Society of Chemical Industry (SCI).

Preparing Biosurfactant Glucolipids from Crude Sophorolipids via Chemical Modifications and Their Potential Application in the Food Industry

Tianyou Cui,Yujing Tang,Mengqian Zhao,Yang Hu,Mingjie Jin,Xuwei Long

This investigation developed a novel strategy for efficiently preparing glucolipids (GLs) by chemically modifying crude sophorolipids. Running this strategy, crude sophorolipids were effectively transformed into GLs through deglycosylation and de-esterification, with a yield of 54.1%. The acquired GLs were then purified via stepwise extractions, and 66.2% of GLs with 95% purity was recovered. GLs are more hydrophobic and present a stronger surface activity than acidic sophorolipids (ASLs). More importantly, these GLs displayed a superior antimicrobial activity to that of ASLs against the tested Gram-positive food pathogens, with a minimum inhibitory concentration of 32–64 mg/L, except against E. coli . This activity of GLs is pH-dependent and especially more powerful under acidic conditions. The mechanism involved is possibly associated with the more efficient adsorption of GLs, as demonstrated by the hydrophobicity of the cell membrane. These GLs could be used as antimicrobial agents for food preservation and health in the food industry.

Efficient preparation of diacetylated lactonic sophorolipids and their potential for use as antibacterial agents in the agricultural industry

Qianqian Ma,Mingda Guo,Mengqian Zhao,Deyu Zhang,Bin Yang,Xuwei Long

A novel strategy prepares highly pure Di-Ac LSLs with a yield over 77%. • Di-Ac LSLs exhibit potent antibacterial activity against Gram-positive bacteria. • Di-Ac LSLs show significant inhibitory activity against fungal mycelia. • The mechanism involves disruption of cell membranes and inhibition of biofilm formation. Diacetylated lactonic sophorolipids (di-Ac LSLs), the most hydrophobic of the sophorolipids congeners, show great promise as antibacterial agents in a variety of fields. However, their efficient production is challenging. This study proposes a novel strategy for efficiently preparing highly pure di-Ac LSLs from fermented SLs, using oleic acid as a hydrophobic carbon source for the fermentation and isolation processes with crystallization. More than 77% of the di-Ac-LSLs with purity over 95% were obtained through crystallization. The resulting di-Ac-LSLs exhibited significantly superior antibacterial activity against Gram-positive bacteria than crude SLs or ASLs. The minimum inhibitory concentrations (MIC) of di-Ac-LSLs against B. cereus and B. subtilis were as low as in the 8–16 mg/L range. The di-Ac-LSLs acted by disrupting cell membranes and inhibiting the formation of biofilm. They also exhibited remarkable inhibitory activity against fungal mycelium, demonstrating their great potential for use as green and efficient antibacterial agents in agriculture. Efficient preparation and isolation of sophorolipids with excellent pH-regulated water solubility via environmentally friendly acid-induced chemical modifications

Yang Hu,Tianyou Cui,Zhihong Yang,Lu Ren,Li Luo,Mengqian Zhao,Mingjie Jin,Xuwei Long

Sophorolipids (SLs) show great potential application in many fields as an alternative to chemically synthesized surfactants. However, their poor water solubility limits industrial applications in many fields. In this study, a new environmentally friendly strategy based on acid-induced chemical modifications has been developed to prepare and isolate water-soluble SLs from natural SLs that not dissolve or precipitate in fermentation broth. Over 70 % of SLs were obtained as sediments from fermentation broth after chemical modification at a pH of 1 at 120 °C for 5 min. The obtained SLs demonstrated excellent pH-regulated water solubility, superior foaming ability, and comparable surface activities with natural SLs. The involved mechanism of the present process was attributed to the generation of glucolipids with excellent water solubility. The present findings are conducive to preparing water-soluble SLs, addressing the problem that SLs cannot be recovered from fermentation broths via acid-induced precipitation in certain case.
