BACKGROUND Indoxacarb, a type of chiral pesticide, is used to control Lepidoptera insects. Most studies had focused on the environmental behavior and selective toxicity of indoxacarb enantiomers, and the mechanism behind its selective biological activity against target organisms is not well understood. RESULTS Spodoptera frugiperda was selected as the target insect. The lethal concentrations (96 h) of indoxacarb enantiomers on S. frugiperda were 2.61 mg/kg ( S -indoxacarb) and 463.52 mg/kg ( R -indoxacarb). S -Indoxacarb resulted in stronger oxidative damage to S. frugiperda than R -indoxacarb, and catalase and malondialdehyde were upregulated by 40.46% and 68.64% respectively after treatment with S -indoxacarb. Furthermore, cytochrome P450 and carboxylesterase were activated by S -indoxacarb, increasing by 39.62% and 63.68% respectively. Decarbomethoxyllated JW062 (DCJW), a metabolite of indoxacarb, has insecticidal activity. The concentration of DCJW in the S -indoxacarb treatment group was 2.73 times that in the R -indoxacarb treatment group. Molecular docking results demonstrated that S -indoxacarb could spontaneously bind to metabolic enzymes and be metabolized. CONCLUSIONS Enantiomeric bioactivity of indoxacarb enantiomers against on S. frugiperda was observed. S -Indoxacarb demonstrated remarkable insecticidal efficacy. Upon ingestion by S. frugiperda , it induced oxidative damage. Furthermore, S -indoxacarb was metabolized to DCJW, which has a significant role in its insecticidal properties. The selective bioactivity of indoxacarb enantiomers in S. frugiperda might be attributed to the enantiomeric metabolites. These findings offer a new perspective on the selective mechanisms of chiral pesticides. © 2025 Society of Chemical Industry.

BACKGROUND The fall armyworm, Spodoptera frugiperda (J. E. Smith), is a polyphagous moth species that is spreading all around the globe. It uses ( Z )-9-tetradecenyl acetate (Z9-14:Ac) and ( Z )-7-dodecenyl acetate ( Z 7-12:Ac) (100:3.9) as essential sex pheromone components. However, our understanding of the molecular basis of pheromone detection of S. frugiperda is still incomplete. RESULTS Herein, we identified six PRs , i.e. SfruOR6 , 11 , 13 , 16 , 56 , and 62 , by transcriptome sequencing. Subsequently, we heterologously expressed them in Drosophila OR67d neurons and determined their response spectra with a large panel of sex pheromones and analogs. Among them, SfruOR13-expressing neurons strongly respond to the major sex pheromone component Z9-14:Ac, but also comparably to ( Z , E )-9,12-tetradecadienyl acetate (Z9,E12-14:Ac) and weakly to ( Z )-9-dodecenyl acetate (Z9-12:Ac). Both SfruOR56 and SfruOR62 are specifically tuned to the minor sex pheromone component Z7-12:Ac with varying intensities and sensitivities. In addition, SfruOR6 is activated only by Z9,E12-14:Ac, and SfruOR16 by both ( Z )-9-tetradecenol (Z9-14:OH) and ( Z )-9-tetradecenal (Z9-14:Ald). However, the OR67d neurons expressing SfruOR11 remain silent to all compounds tested, a phenomenon commonly found in the OR11 clade of Noctuidae species. Next, using single sensillum recording, we characterized four sensilla types on the antennae of males, namely A, B, C and D types that are tuned to the ligands of PRs, thereby confirming that S. frugiperda uses both SfruOR56 and SfruOR62 to detect Z7-12:Ac. Finally, using wind tunnel assay, we demonstrate that both Z9,E12-14:Ac and Z9-14:OH act as antagonists to the sex pheromone. CONCLUSION We have deorphanized five PRs and characterized four types of sensilla responsible for the detection of pheromone compounds, providing insights into the peripheral encoding of sex pheromones in S. frugiperda .

The fall armyworm, Spodoptera frugiperda (J. E. Smith) is a notorious insect pest of poaceae and causes severe economic damage to major cereal crop. It has evolved with different levels of insecticide resistance. Insect cytosolic GSTs are a major class of detoxifying enzymes and have been implicated in metabolic resistance and protection against oxidative stress. In this study, an epsilon class GST ( SfGSTe9 ) was identified from S. frugiperda and its transcript levels were highest in the fat bodies, second instar and pupae. Furthermore, SfGSTe9 is upregulated after exposure to beta- cypermethrin, lambda -cyhalothrin, chlorpyrifos, and malathion. The kinetic analysis suggests the purified recombinant proteins exhibit CDNB conjugating activity, with the V max of 11.87 ± 0.25 μM·min −1 ·mg −1 protein and K m of 0.22 ± 0.01 mM. The peroxidase activity assays indicate that SfGSTe9 has antioxidant activity against both CHP and H 2 O 2. The inhibition assays infer that four types of insecticides have strong inhibition on the GST activity. In vitro metabolism assays with HPLC further suggest that SfGSTe9 could be able to deplete 11.2 %, 21.3 % and 19.3 % of beta- cypermethrin, lambda -cyhalothrin and chlorpyrifos within 2 h, respectively, with specific activity varying between 30 and 40 μM·min −1 ·mg −1 protein. However, no metabolites were identified in this case, indicating that SfGSTe9 probably involves in detoxification via binding and sequestration. 3D modeling and molecular docking analysis indicate that above three types of insecticide compounds fit nicely into the hydrophobic pocket in the active site of SfGSTe9 . Our results definitely demonstrate that SfGSTe9 of S. frugiperda plays pivotal role in insecticide detoxification and serves in defence against oxidative stress .

Insects mount robust immune responses to reduce the efficacy of biopesticides. Identification of critical immune targets for biocontrol strategies is thus essential. However, immune targets in the fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), a major pest of corn, remain largely unexplored. Here, we investigated the immune response of S. frugiperda to the entomopathogen Beauveria bassiana (Hypocreales: Cordycipitaceae). Time-series RNA sequencing revealed 863, 717, and 165 differentially expressed genes (DEGs) at 24, 48, and 60 h post-infection, respectively. GO and KEGG enrichment analyses revealed that DEGs were significantly enriched in immune-related terms and pathways. Among the DEGs, the gene encoding immulectin SfIML-1 , a C-type lectin with dual carbohydrate recognition domains, was significantly upregulated during the early and intermediate stages of infection. Genome-wide analysis identified nine IML genes, with SfIML-1 as the only one highly responsive to B. bassiana . Recombinant SfIML-1 binds pathogen-associated molecular patterns and pathogens, enhancing cellular responses. RNAi of SfIML-1 decreased survival after fungal infection, while injection of recombinant SfIML-1 increased survival. Suppression of the NF-κB and JAK-STAT pathways diminished SfIML-1 expression in both fat body and Sf9 cells. These results identify SfIML-1 as a key pattern recognition receptor and a promising target for developing novel biocontrol strategies.

Insects, as the most diverse and numerous group in the animal kingdom, are at least partly dependent on the reproduction process, which is strictly regulated by the ‘classic’ insect hormones: juvenile hormone (JH), and 20-hydroxyecdysone (20E). However, the regulatory mechanism governing the reproduction of JH and 20E inSpodoptera frugiperdaremains unclear. In this study, ovarian development and ovulation in femaleS. frugiperdawere assessed through dissection of the ovaries following treatment with JH analog (JHA) and 20E. Moreover, the expression patterns of the JH-signal and 20E-signal-related genes were determined by quantitative PCR (qPCR), and RNA interference (RNAi) was used to investigate the role of JH and 20E-induced genes. Ovarian development was observed by microdissection, and JH and 20E titers were determined by ELISA.Kr-h1,Vg, andUSPexpression were determined by qPCR. Dissection and qPCR results showed that JHA and 20E promoted ovarian development, egg maturation, and egg laying by upregulatingMethoprene-Tolerant(Met) andEcdysone Receptor(EcR)expression. Additionally, the RNAi results showed that the injection of dsMetand dsEcRmarkedly delayed ovarian development, inhibited egg maturation, and halted egg production. Knockdown ofMetandEcRsignificantly reduced JH and 20E content and inhibited the transcription ofKr-h1andUSP. These results indicate that JH and 20E facilitate adult reproduction through the methoprene-tolerant gene and ecdysone receptor gene in femaleS. frugiperda.

BACKGROUND Spodoptera frugiperda is an important invasive agricultural pest that causes huge economic losses worldwide. Gut microorganisms play a vital role in host feeding, digestion, nutrition, immunity, growth and insecticide resistance. Illumina high-throughput sequencing was used to study the gut microbial community dynamics across the life cycle (egg, 1st to 6th instar larvae, pupae, and male and female adults) of S. frugiperda fed on maize leaves. Furthermore, the gut microbial community and food intake of the 5th instar S. frugiperda larvae were studied after feeding them antibiotics. RESULTS Enterobacteriaceae and Enterococcaceae dominated the gut during growth and feeding of the larvae. The relative abundance of Enterobacteriaceae was higher in the 4th and 6th instar larvae. With the increase in larval feeding, the relative abundance of Enterococcaceae gradually increased. In addition, principal coordinate analysis and linear discriminant effect size analysis confirmed differences in the structure of gut microbiota at different developmental stages. After antibiotic treatment, the relative abundance of Firmicutes, Proteobacteria and Fusobacteriota decreased. The relative abundance of Enterococcus and Klebsiella decreased significantly. Antibiotic treatment inhibited the gut flora of S. frugiperda , which decreased larval food intake and body weight gain, and prolonged the larval stage. CONCLUSION The composition of the gut bacterial community plays an important role in the growth, development, and feeding of S. frugiperda . The results have a certain theoretical value for the development of bio-pesticides targeting intestinal flora. © 2022 Society of Chemical Industry.

Antimicrobial peptides (AMPs) are critical components of innate immunity in diverse organisms, including plants, vertebrates, and insects. This study identified and characterized a novel Lepidoptera-specific AMP, named lepidoptin, from the invasive pest Spodoptera frugiperda (Lepidoptera: Noctuidae). Lepidoptin is a 116-amino acid protein containing a signal peptide and a novel β -sandwich domain that is distinct from previously reported AMPs. Temporal and spatial expression analyses revealed a significant upregulation of the lepidoptin gene in vivo and in cultured SF9 cells in response to pathogens. Molecular docking analysis identified a specific binding cavity. Enzyme-linked immunosorbent assay and binding assays confirmed that lepidoptin can bind to pathogen-associated molecular patterns, bacteria, and fungi. Recombinant lepidoptin exhibited potent antibacterial activity by inducing bacterial agglutination, inhibiting bacterial growth, increasing bacterial membrane permeability, and preventing biofilm formation. Lepidoptin also showed antifungal activity against the entomopathogenic fungus Beauveria bassiana by inhibiting spore germination, increasing fungal cell permeability, and increasing reactive oxygen species. Injection of recombinant lepidoptin into S. frugiperda larvae increased survival after B. bassiana infection, whereas knockdown of lepidoptin by RNA interference decreased larval survival. In addition, lepidoptin showed antimicrobial activity against the plant pathogen Fusarium graminearum by inhibiting spore germination and alleviating disease symptoms in wheat seedlings and cherry tomatoes. This study demonstrates the remarkable dual functionality of lepidoptin in enhancing S. frugiperda immunity and controlling plant pathogens, making it a promising candidate for biocontrol strategies in both pest management and plant disease prevention.

RNA interference is an eco-friendly alternative to chemical pesticides, yet its efficacy in lepidopterans like Spodoptera frugiperda ( S. frugiperda ) is limited by poor uptake. Here, we report on ZIF-8 polydopamine nanoparticles that protect dsRNA against enzymatic degradation and active the endocytic/phagosome pathways for increased uptake. Furthermore, the uptake of nano-enabled dsRNA induces the overgrowth of Serratia marcescens , this reduces the S. frugiperda reactive oxygen species (ROS) immune response, increasing the effects of plant’s natural defenses, further inhibiting Enteroccous mundtii growth. This work shows the synergistic potential of nanoparticles for influencing the gut bacteria to prevent resistance mechanisms and for RNAi delivery for pest management.

The Fall Armyworm (FAW), Spodoptera frugiperda, is a globally significant invasive lepidopteran pest responsible for severe crop damage, particularly to maize (Zea mays). In this study, we investigated the preference of FAW larvae and adults for regular corn (RC) versus sweet corn (SC) using a two-choice behavioral assay. Potential semiochemicals underlying this preference were isolated and identified through headspace volatile collection coupled with Solid-Phase Microextraction–Gas Chromatography–Mass Spectrometry (SPME-GC-MS). The behavioral activity of identified differential volatile compounds was then comprehensively evaluated using electroantennography (EAG), a Y-tube olfactometer, and cage-based oviposition preference tests. Our results demonstrate that FAW larvae exhibit a significant feeding preference for SC leaves, while adult females show a marked oviposition preference for SC plants. Volatile profiling revealed four compounds consistently expressed at higher levels in SC compared to RC: hexanal, trans-2-hexen-1-ol, linalool, and β-caryophyllene. EAG recordings indicated that trans-2-hexen-1-ol elicited the strongest antennal response in FAW adults. In behavioral assays, FAW larvae showed significant attraction to all four volatiles at concentrations of 10 and 100 mg/mL. Similarly, adults exhibited significant preference for most compound-concentration combinations, with the exception of linalool at 100 mg/mL. The volatile compounds identified in SC present promising candidates for use in behavior-based disruption techniques, offering a more environmentally benign alternative to conventional insecticides for FAW control. These findings provide a scientific foundation for developing volatile-mediated strategies as part of integrated pest management programs against this invasive pest.

Spodoptera frugiperda (J. E. Smith) is a globally invasive pest that poses significant threats to a variety of crops. It has a wide host range of more than 353 recorded plants from 76 families. However, the herbivorous insects could have adaptation mechanisms through physiological modifications in response to constitutive defenses of different host plants. The results revealed that feeding different host plants could significantly affect LC 50 s of broflanilide, chlorantraniliprole, tetraniliprole, indoxacarb, metaflumizone, lufenuron, methoxyfenozide, and spinetoram to S. frugiperda compared to rearing on Zea mays (L.). Detoxification enzyme activity assays and synergist tests indicated that changes in detoxification enzyme activity cause alterations in the susceptibility of S. frugiperda to insecticides. Knockdown of UGT40F3 using RNA interference (RNAi) significantly increased susceptibility to tetraniliprole in all host-fed strains. Microscale thermophoresis (MST) and molecular docking revealed a robust tetraniliprole-UGT40F3 interaction, suggesting that UGT40F3 plays a pivotal role in tetraniliprole metabolism. These results showed that different host plants can influence the detoxification enzyme activity of S. frugiperda , leading to variations in its susceptibility to insecticides. These findings provide a scientific foundation for developing effective pest management strategies.

Camptothecin (CPT) is derived from Camptotheca acuminate and its main action is the inhibition of the DNA topoisomerase I (Top I) complex. Therefore, CPT and its derivatives are always used as antitumor and antimicrobial agents and also have some insecticidal effects. However, the mode of action underlying the toxicity of CPT to the virulence of pests is still not fully understood. In this study, the effects of CPT on Spodoptera frugiperda ( S. frugiperda ) were determined by performing toxicity assessments, metabolomics, and mechanism analysis. The results showed that CPT increased the efficiency of inhibition of larval weight in a dose and time-dependent manner. In addition, the metabolomics of 5th-instar larval hemolymph was determined by UPLC-MS/MS and 1 H NMR analysis. UPLC-MS/MS analysis showed that the energy metabolic pathway was more important than the amino acid pathway for the resistance of S. frugiperda to CPT. Furthermore, metabolites involved in the energy metabolic pathway were analyzed using 1 H NMR, and fumarate and pyruvate were identified as interesting metabolites. Bioassays confirmed that they play a key role in enhancing the resistance of S. frugiperda larvae to CPT. Finally, the underlying mechanisms were found to be related to the restorative effects of the two metabolites on metabolic pathways associated with the TCA cycle. In a word, CPT is a potential biological insecticide that exhibits a strong toxicological effect against S. frugiperda by inhibiting the TCA cycle, which provides theoretical support for applying CPT in the control of pests.

Fluorescent gold nanoclusters (Au NCs) have attracted considerable interest in biological application. Here, we reported a novel Au NCs with blue fluorescence, conjugated with glycine (Gly) and rotenone (R). The structure and properties of Au–BSA–Gly–R conjugates were characterized by transmission electron microscopy (TEM), UV–visible, fluorescence spectroscopy, Fourier transform infrared spectroscopy (FTIR) and Zeta potential measurement. The results of fluorescent microscopic imaging revealed that the Au NCs with different ligands were capable of being selectively uptaken by Spodoptera frugiperda (Sf9) cells. Furthermore, the gold content of Au–BSA–Gly–R in Sf9 cells was 2.2 and 2.6 times higher than that of Au–BSA and Au–BSA–R, respectively. It illustrated that nanoparticles after conjugated with Gly could gain specificity to intracellular transportation. This study indicated that the Au NCs after mediated by Gly not only had great potential for applications in insect cell imaging, but also could play an important role of amino acid for agrochemicals delivery in agricultural fields.
