Bioceramics play an important role in bone regeneration. However, it is challenging to design bioceramic scaffolds with suitable ionic components and beneficial osteo/angio-stimulation ability for enhanced bone regeneration. In this study, we successfully synthesized a pure-phase Sr5(PO4)2SiO4 (SPS) bioactive ceramic through a solid-state reaction method and further prepared highly uniform SPS bioceramic scaffolds with controlled macropore sizes and mechanical strength by a 3D-plotting technique, and the biological responses of rabbit bone marrow stromal cells (rBMSCs) and human umbilical vein endothelial cells (HUVECs) after culturing with different concentrations of SPS extracts and porous scaffolds were systematically studied. The results showed that the ionic products from SPS bioceramics significantly stimulated the proliferation, alkaline phosphate (ALP) activity and osteogenesis-related gene expression (Runx2, ALP, OCN, OPN) of rBMSCs as well as the proliferation and angiogenesis-related gene expression (VEGF, KDR, eNOS, HIF 1α) of HUVECs. 3D-plotted SPS scaffolds could effectively support the attachment and proliferation of both rBMSCs and HUVECs, and the proliferation rates of the two kinds of cells in SPS scaffolds were distinctively higher than those in β-tricalcium phosphate (β-TCP) scaffolds prepared by the same method. In addition, the compressive strength of SPS scaffolds could be well controlled in the range 8–30 MPa when their pore size varied from 100 to 300 μm, which was significantly higher than those of β-TCP scaffolds with similar pore sizes (∼1.5 times). Our results demonstrated that 3D-plotted SPS bioceramic scaffolds with such a specific ionic combination and high mechanical strength as well as good degradability possessed the ability to stimulate both osteogenic and angiogenic differentiation of tissue cells, indicating that they might be promising biomaterials for bone tissue engineering.

Bioactive glass ceramics are widely used for hard tissue repair and bone regeneration due to their most attractive properties such as biocompatibility, bioactivity and non toxicity. In this study, for the first time, sol-gel method was used to synthesize nanosized strontium phosphosilicate (SPS: Sr 5 (PO 4 ) 2 SiO 4 ) with rod shaped morphology by using CTAB as a surfactant . The obtained SPS powder was characterized by FTIR, XRD , Raman, SEM-EDS and TEM analysis. In vitro bioactivity study confirms that SPS possesses bone bonding ability by formation of amorphous apatite layer on its surface. In vitro osteogenesis and angiogenesis studies were performed on SPS powder to determine its biological responses using mBMSCs and HUVECs, respectively. The co-culture of BMSCs with different concentrations of SPS powder (50, 100, 150, 200 and 250 µg/ml) reveals that there's no any cytotoxicity effect on mBMSCs and observed good cell proliferation , alkaline phosphatase activity, calcium nodules formation, osteogenesis-related gene expressions ( alp , runx2 , col1a2 and ocn ). In addition, the angiogenesis study demonstrated that SPS powder can stimulate the cellular response of HUVECs and its gene expressions ( vegfr2 and hif1α-3 ). Therefore, these results confirm that nanosized SPS could act as promising bioceramic material for bone tissue engineering applications.

This work mainly focused on the development of novel nanocomposites with combination of strontium phosphosilicates (SPS: Sr 5 (PO 4 ) 2 SiO 4 ) and graphene (G) for bone regeneration applications. Firstly, we have adopted the sol-gel method to synthesize the rod shaped nanosized SPS powder using CTAB as a surface morphology directing agent. Secondly, we have attempted a new approach to grow the graphene on the nanosized SPS by chemical vapour deposition technique, whereas SPS acts as self catalyst for the generation of graphene. The Raman analysis proved the existence of graphene with a few layers in [email protected] nanocomposites. The XPS confirms the presence of sp2 hybridized carbon atoms in graphene by detecting peak position at 284.4 eV. In vitro apatite study demonstrates the deposition of needle like shaped amorphous apatite layer on [email protected] The 50–250 µg/ml of [email protected] were assessed to examine its in vitro cytocompatibility behaviour using mBMSCs and found to stimulation of cell proliferation. The [email protected] possesses relative alkaline phosphatase activity, calcium deposition and osteogenic differentiation ability. Also, [email protected] shown no toxicity on HUVECs and significantly encouraged the cell proliferation. Therefore, this finding provides the development of [email protected] nanocomposites with improved biological properties can acts potential candidate for future bone tissue engineering applications.

The natural polysaccharide-based hydrogels have gained significant interest to use as 3D scaffolds for bone repair and reconstruction owing to their excellent biocompatibility, biosafety, and biodegradability. Yet, these hydrogels have major obstacles in terms of insufficient properties such as mechanical, osteogenic, angiogenic, and biomineralization . Therefore, this study mainly aims to develop an innovative water-soluble phosphate functionalized chitosan (CSMAP) through consequent modification with methacrylic anhydride (MA), then alteration by phosphonopropionic acid (P). Further, a novel photocrosslinked composite hydrogel was fabricated with a combination of CSMAP and synthesized strontium phosphosilicate (SPS, Sr 5 (PO 4 ) 2 SiO 4 ) bioceramic nanoparticles with a photoinitiator under UV irradiation. The various concentrations of nanosized SPS particles (0.5, 2.5, 5, and 10 mg/mL) were incorporated into the CSMAP matrix and the swelling, mechanical, morphological, physiochemical, osteogenic, angiogenic properties were investigated. The CSMAP-SPS composite hydrogels possessed a well-arranged porous network structure. The composite hydrogel was restricted to swell in distilled water with the increased concentration of SPS particles. The compressive strength and modulus of CSMAP hydrogel were improved by the inclusion of SPS particles. The bioactive Sr, P, and Si ions were released from CSMAP-SPS hydrogels in a sustained and controlled manner at a non-toxic level. The composite hydrogels promoted in vitro amorphous apatite deposition, revealing a superior biomineralization activity. In vitro results demonstrated that the composites hydrogels showed no toxicity to preosteoblast MC3T3-E1 cells, induced MC3T3-E1 osteogenic differentiation with detection of bsp , ocn and opn osteogenic genes, and also promoted the endothelial tube formation with increased capillary length , branch points, and angiogenic gene vegf expression. Thus, the obtained results proved that the fabricated novel CSMAP-SPS hydrogel could act as an attractive candidate for bone regeneration.
