Polyethylene terephthalate (PET) is one of the most widely used plastics, but its fragmentation into microplastics poses significant environmental challenges. The recycling of PET microplastics is hindered by their low solubility and widespread dispersion in the environment, making microbial in-situ degradation a promising solution. However, existing PET-degrading strains exhibited the limited effectiveness, primarily due to the diffusion of secreted hydrolases away from the PET surface. In this study, Stenotrophomonas pavanii JWG-G1 was engineered to achieve the targeted aggregation of PET hydrolase PETase on the cell surface by fusing it with an endogenous anchor protein. This approach aims to maximise the local concentration of PETase around PET, thereby increasing the overall rate of PET degradation. The PETase surface-aggregated system, S. pavanii /PaL-PETase, demonstrated the highest degradation efficiency, achieving 63.3 % degradation of low-crystallinity PET (lcPET) and 27.3 % degradation of high-crystallinity PET bottles (hcPET) at 30 °C. This represents the highest degradation rate reported for a displayed whole-cell system at ambient temperature. Furthermore, this system exhibited broad-spectrum degradation activity against various polyesters. These findings suggest that this system offers a promising, eco-friendly solution to PET and other polyester pollution, with potential implications for environmental bioremediation strategies.

Although several abiotic processes have been reported that can transform antibiotics, little is known about whether and how microbiological processes may degrade antibiotics in the environment. This work isolated one tetracycline degrading bacterial strain, Stenotrophomonas maltophilia strain DT1, and characterized the biotransformation of tetracycline by DT1 under various environmental conditions. The biotransformation rate was the highest when the initial pH was 9 and the reaction temperature was at 30 °C, and can be described using the Michaelis-Menten model under different initial tetracycline concentrations. When additional substrate was present, the substrate that caused increased biomass resulted in a decreased biotransformation rate of tetracycline. According to disk diffusion tests, the biotransformation products of tetracycline had lower antibiotic potency than the parent compound. Six possible biotransformation products were identified, and a potential biotransformation pathway was proposed that included sequential removal of N-methyl, carbonyl, and amine function groups. Results from this study can lead to better estimation of the fate and transport of antibiotics in the environment and has the potential to be utilized in designing engineering processes to remove tetracycline from water and soil.

A novel prolyl endopeptidase from Stenotrophomonas maltophilia , SmPEP, was discovered and characterized. The specific activity of the recombinant SmPEP expressed by Escherichia coli BL21 (DE3), was 68.3 U/mg at pH 8.0 and 37 °C. In order to improve the substrate specificity for long-chain peptide, rational design was applied based on the structure constructed by homology modeling. Inter-domain sites within the β-propeller domain were chosen for the mutation to weaken the inter-domain interaction and form an open conformation for long-chain substrate entering into the active site. The substrate specificity on a designed long-chain substrate, PQPQLPYPQPQLP, of the mutants F263A and E184 G increased 8.77 and 5.75 times respectively versus wild-type. After the saturated mutation of the both sites, the reactive rate of mutant F263 V on 13-mer peptide was 10.2 times higher than that of the wild-type. Then the mutant F263 V was used in the hydrolysis of casein, and the ACE inhibitory activity of the hydrolysate was significantly improved compared with wild type enzyme, which verified the efficiency of the design strategy.

An abamectin (ABM)-degrading bacterium, Stenotrophomonas maltophilia ZJB-14120, was isolated and identified. This strain is capable of degrading 84.82% of ABM at an initial concentration of 200 mg/L over a 48 h incubation period. This strain showed efficient biodegradation ability (7.81 mg/L/h) to ABM and high tolerance (1000 mg/L) to all macrolides tested. In addition to ABM, emamectin, erythromycin and spiramycin can also be degraded by this strain. Modifications involving either reduction of the double bond between C22–C23 or replacement of the C25-group of ABM with a cyclohexyl group can completely inhibit biodegradation of ABM. The ABM-degrading capability of strain ZJB-14120 is likely to be intrinsic to its metabolism and could be inhibited by incubating with erythromycin, azithromycin, spiramycin or rifampicin. A new and successive degradation pathway was proposed based on metabolite analysis. Although there is evidence for metabolite inhibition, this strain has high ABM degradation activity and reusability. Further investigation showed that activated macrolide efflux pump(s) and an undetermined mechanism for regulating the intracellular ABM concentration are responsible for normal uptake of essential metabolites while pumping out excess harmful compounds. Strain ZJB-14120 may provide efficient treatment of water and soil contaminated by toxic levels of abamectin and emamectin.

The aim of this work is to study the synergistic effect of Stenotrophomonas sp. N5 and Advenella sp. B9 co-culture (COC) on enhancement of phenol biodegradation. These two strains utilizing phenol as sole carbon and energy source were isolated from phenol-containing coking wastewater. The results of biodegradation experiment showed the COC of N5 and B9 has stronger capability to degrade phenol than either of mono-culture (MOC). Growth kinetics studies indicated inhibitory effect of phenol on COC was reduced by the interaction of N5 and B9 in COC. The RNA-Seq results demonstrated that phenol biodegradation was enhanced by metabolic division of labor (DOL) in COC based on the expression of key genes for phenol degradation. GO enrichment analysis of differentially expressed genes (DEGs) indicated DEGs between COC and MOC degradation systems are mainly concentrated in the synthesis of cell components, microbial growth and metabolism, and catalytic activity. The expression of 3 transcriptional factors (LysR, Two-component system response regulator, and TetR families) which can regulate degradation of aromatic compounds, was identified beneficial to phenol degradation.

C-di-GMP is a bacterial second messenger implicated in the regulation of many key functions including antibiotic tolerance and biofilm formation. Our understanding of how c-di-GMP exerts its action via receptors to modulate different biological functions is still limited. Here we used a c-di-GMP affinity pull-down assay coupled to LC-MS/MS to identify c-di-GMP-binding proteins in the opportunistic pathogen Stenotrophomonas maltophilia. This analysis identified Smlt3238 (SodA), a protein of the superoxide dismutase family, as a c-di-GMP-binding protein. Microscale thermophoresis showed that purified SodA protein bound c-di-GMP with an estimated dissociation constant (Kd) value of 141.5 μM. Using various in vivo and in vitro experiments, we demonstrated that c-di-GMP modulates the enzyme activity of SodA directly. Circular dichroism experiments revealed that SodA protein gradually altered its basic structure with increasing levels of c-di-GMP. Phenotypic experiments conducted in the presence of a range of intracellular c-di-GMP levels showed that SodA function is modulated by c-di-GMP. The findings thus identify a novel c-di-GMP binding protein that governs oxidative stress tolerance in S. maltophilia .

Organophosphorus insecticides have been widely used, which are highly poisonous and cause serious concerns over food safety and environmental pollution. A bacterial strain being capable of degrading O , O -dialkyl phosphorothioate and O , O -dialkyl phosphate insecticides, designated as G1, was isolated from sludge collected at the drain outlet of a chlorpyrifos manufacture plant. Physiological and biochemical characteristics and 16S rDNA gene sequence analysis suggested that strain G1 belongs to the genus Stenotrophomonas . At an initial concentration of 50 mg/L, strain G1 degraded 100% of methyl parathion, methyl paraoxon, diazinon, and phoxim, 95% of parathion, 63% of chlorpyrifos, 38% of profenofos, and 34% of triazophos in 24 h. Orthogonal experiments showed that the optimum conditions were an inoculum volume of 20% (v/v), a substrate concentration of 50 mg/L, and an incubation temperature in 40 °C. p -Nitrophenol was detected as the metabolite of methyl parathion, for which intracellular methyl parathion hydrolase was responsible. Strain G1 can efficiently degrade eight organophosphorus pesticides (OPs) and is a very excellent candidate for applications in OP pollution remediation.

Immobilization technology is a promising way to improve effectiveness and stability of microbial remediation for polycyclic aromatic hydrocarbons (PAHs), in which carrier material is one of key factors restricting removal efficiency. In this study, fulvic acid-wheat straw biochar (FA/WS) composites were applied for immobilization of an efficient PAHs degrading bacterium Stenotrophomonas maltophilia (SPM). FA/WS&SPM showed superior degradation capacity than free bacteria and biochar-immobilized bacteria, with the removal efficiency of pyrene (20 mg L -1 ) reaching 90.5 % (7 days). Transcriptome analysis revealed that FA in the carrier materials can promote transportation and degradation of pyrene, and cell growth, as well as inhibit cell apoptosis. Enzyme activity and degradation products detection showed that SPM utilized both phthalic acid and salicylic acid metabolic pathways to degrade pyrene. Practicality of FA/WS&SPM for different kinds of PAHs remediation had been verified in contaminated soil, demonstrating a great potential in the field of PAHs polluted sites remediation.

Polyethylene terephthalate (PET) is a general plastic that produces a significant amount of waste due to its non-biodagradable properties. We obtained four bacteria ( Stenotrophomonas pavanii JWG-G1, Comamonas thiooxydans CG-1, Comamonas koreensis CG-2 and Fulvimonas soli GM-1) that utilize PET as a sole carbon source through a novel stepwise screening and verification strategy. PET films pretreated with S. pavanii JWG-G1 exhibited weight loss of 91.4% following subsequent degradation by Thermobifida fusca cutinase (TfC). S. pavanii JWG-G1 was able to colonize the PET surface and maintain high cell viability (over 50%) in biofilm, accelerating PET degradation. Compared with PET films with no pretreatment, pretreatment with S. pavanii JWG-G1 caused the PET surface to be significantly rougher with greater hydrophilicity (contact angle of 86.3 ± 2° vs. 96.6 ± 2°), providing better opportunities for TfC to contact and act on PET. Our study indicates that S. pavanii JWG-G1 could be used as a novel pretreatment for efficiently accelerating PET biodegradation by TfC.

Annual global plastic waste exceeds 287 million tonnes, devastating ecosystems and health. As a promising solution, yellow mealworm larvae have shown the ability to degrade plastics. In this study, we explored the regional characteristics of the yellow mealworm and isolated a novel enteric bacterium, S . maltophilia ZSL2493, from the intestinal tract of yellow mealworm larvae, which achieved a biodegradation efficiency of 14.7% of polystyrene (PS) within 30 days. The two most promising candidate enzymes, kynurenine 3-monooxygenase (Kmo) and 4-hydroxyphenylpyruvate dioxygenase (Hpd), were identified by transcriptome mining and molecular docking simulations. Their depolymerisation ability was also confirmed by heterologous expression experiments. Recombinant Kmo and Hpd reduced the mass of PS by 6.14% and 3.96%, respectively, within 30 days. Quantitative analysis by liquid chromatography showed that the yield of styrene monomer was 1.37 μg/mL (Kmo-treated group) and 0.95 μg/mL (Hpd-treated group), respectively, after 30 days of incubation, which is a direct evidence of the enzyme's cleavage of polymer chains. This study elucidates the key enzymatic drivers in the biodegradation of PS and promotes the development of enzymatic recycling strategies for difficult-to-degrade plastics.

Bioremediation of highly chlorinated polychlorinated biphenyls (PCBs) typically requires sequential anaerobic-aerobic treatment, highlighting the need for robust facultative anaerobic degraders. In this study, a novel facultative anaerobic strain, Stenotrophomonas sp. FS3, was resuscitated with resuscitation-promoting factor (Rpf) under alternating anaerobic-aerobic conditions. Whole-genome sequencing revealed a comprehensive set of genes for biphenyl, benzoate, catechol, and protocatechuate catabolism, indicating strong intrinsic potential for PCB biodegradation. FS3 achieved 61.2 % removal of Aroclor 1260 at 10 mg/L after 30 days, and Rpf supplementation significantly enhanced both anaerobic dechlorination and aerobic degradation, resulting in a 1.66-fold increase in removal efficiency and greater chloride ion release relative to controls. Viability and morphophysiological analyses revealed that Rpf preserved cell integrity and metabolic activity, preventing the transition into a viable but non-culturable (VBNC) state. Transcriptomic analysis revealed that Rpf supplementation significantly upregulated bph cluster genes and downstream catabolic genes (e.g., pobA , ligB , pcaGH ) without altering the overall degradation pathway, which proceeded via the HOPDA-benzoate pathway followed by catechol- and protocatechuate-mediated mineralization. Collectively, this work establishes FS3 as a robust degrader suitable for PCB bioremediation under fluctuating redox conditions and underscore Rpf supplementation as a viable strategy to enhance the resilience and metabolic capacity of PCB-degrading bacteria.

Polyethylene terephthalate (PET) microplastics pose significant environmental and human health risks due to their resistance to degradation and accumulation in ecosystems. In this study, we engineered Stenotrophomonas pavanii JWG-G1, a robust biofilm-forming bacterium, to overexpress the PET hydrolase (DuraPETase) for PET microplastics degradation at ambient temperature. Nine endogenous PET hydrolases were identified through genome sequencing of S. pavanii , and were successfully expressed in Escherichia coli BL21(DE3). Among them, hydrolase Est_B achieved 100% degradation of bis(2-hydroxyethyl) terephthalate (BHET) at an initial concentration of 0.23 mg/mL at 30 °C within 4 h, identifying it as a novel BHETase. However, the PET degradation performance of all endogenous PET hydrolases was inferior to that of DuraPETase. The engineered strain overexpressing DuraPETase demonstrated a significant enhancement in PET degradation, achieving a 38.04 μM total product release of high-crystallinity PET microplastics after 30 days at 30 °C. The degradation extent was greater than that of low biofilm-forming engineered strains, attributing to the aggregation of DuraPETase on the PET surface in the presence of biofilm. Additionally, this engineered strain also maintained PET degradation activity across various water environments and demonstrated effectiveness in degrading other polyester plastics. This is the first report demonstrating that an engineered strain of Stenotrophomonas species is capable of simultaneously secreting exogenous hydrolase and degrading polyester microplastics, representing a novel approach in the development of engineered bacteria with potential applications in bioreactor systems and environmental remediation.
