Sulfation of the extracellular polysaccharide from the edible fungus Stropharia rugosoannulata with its antioxidant activity

Hui Hao,Chun Cui,Yuqing Xing,Xuewei Jia,Bingjie Ma,Wenyi Kang,Tianxiao Li,Mingqi Gao,Chunping Xu

ABSTRACT An acidic extracellular polysaccharide (EPS) was gained from Stropharia rugosoannulata fermentation broth by the alcohol precipitate method. Sulfated derivative of EPS (S-EPS) was gained by chlorosulfonic acid-pyridine method and the conditions were as follows: pyridine as the reaction solvent, temperature 80 °C, and reaction time 90 min. The chemical structure and chain conformation of EPS and S-EPS were characterized by FT-IR, NMR and size exclusion chromatography (SEC) coupled with multi-angle laser light scattering (MALLS) detection. FT-IR spectra indicated the sulfated group was linked to the polymer. The EPS mainly consists of mannose, glucose and glucuronic acid, and the glycosidic bond type is mainly (1→4)-linked and (1→6)-linked. C6 and C2 were substituted by sulfate groups. The EPS was degraded into smaller molecular weight ( M w ) polysaccharides with the larger molecular size ( R g ) after sulfation and S-EPS has better hydrophilicity. Moreover, the sulfated derivative S-EPS was found to have a better scavenging ability on hydroxyl radicals than EPS. It proved that the chemical modification of S. rugosoannulata polysaccharides by sulfation effectively enhanced their antioxidant activity.

Protective effect of Stropharia rugosoannulata polysaccharide and its derivatives on dextran sodium sulfate-induced colitis

Ye Hongling,He Yaling,Jin Mingzhi,Ye Ziyang,Huang Qianli,Chen Hui,Ye Ming

Ulcerative colitis is a common inflammatory bowel disease. This study evaluated the protective effect and potential mechanism of Stropharia rugosoannulata polysaccharide (SP-1a) and its carboxymethylated and phosphorylated derivatives (CSP-1a and PSP-1a) against dextran sulfate sodium (DSS)-induced ulcerative colitis. The results showed that treatment with SP-1a and its derivatives alleviated the symptoms of weight loss, colon shortening, and blood in the stool induced by . SP-1a and its derivatives restore the integrity of the intestinal barrier by controlling the expression of tight junction proteins. Furthermore, they play an anti-inflammatory role by reducing oxidative stress and inhibiting the expression of pro-inflammatory factors, pro-inflammatory proteins, and nuclear factor kappa-B (NF-κB) signaling pathways. The anti-inflammatory effects of CSP-1a and PSP-1a were greater than those of SP-1a. These results indicate that SP-1a could be a potential natural medicine to protect against DSS-induced colitis.

Effects of polysaccharides from Stropharia rugosoannulata on cyclophosphamide-induced immunosuppression and hepatic injury in mice

Li Wang,Yupeng Yang,Xuelian Tang,Chunnan Wen,Zhiqiang Zhang,Xia Ma,Bingji Ma

BACKGROUND The immune system is essential for human health. Traditional chemical immunomodulators often have toxic side effects; for example, long-term use of cyclophosphamide (CTX) can lead to severe adverse consequences. Stropharia rugosoannulata , a flavorful edible mushroom native to China, exhibits antioxidant, immunomodulatory, and antibacterial activity, with its polysaccharides being the key active components responsible for its immunomodulatory effects. RESULTS Polysaccharides were extracted from S. rugosoannulata using a three-phase method to investigate their immunomodulatory activity and hepatoprotective effects in cyclophosphamide-induced mice. In vivo experiments showed that these polysaccharides significantly increased body weight, spleen index, and phagocytic index, regulated the CD4+/CD8+ ratio, and promoted secretion of tumor necrosis factor- α (TNF- α ), interleukin-1 β (IL-1 β ), interleukin-6 (IL-6), immunoglobulin A (IgA), and immunoglobulin G (IgG). The mechanism involved activation of the TLR4/MyD88/NF- κ B signaling pathway, effectively enhancing immune function. Regarding hepatic protection, S. rugosoannulata polysaccharides (SRP) increased serum malondialdehyde (MDA) and decreased superoxide dismutase (SOD), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) levels in immunosuppressed mice, reversing CTX-induced liver injury. Histopathological analysis confirmed that SRP ameliorated CTX-induced pathological changes, including single-cell necrosis in the jejunal mucosa and insufficient hematopoietic compensation, and reduced lymphocytes in splenic white pulp. CONCLUSION This study provides a theoretical basis for developing and applying SRP as an immunomodulator or hepatoprotective agent, with potential value in functional food and pharmaceutical applications. © 2025 Society of Chemical Industry.

From White to Reddish-Brown: The Anthocyanin Journey in Stropharia rugosoannulata Driven by Auxin and Genetic Regulators

Mei Wang,Tingting Song,Qunli Jin,Zuofa Zhang,Yingyue Shen,Guoying Lv,Lijun Fan,Weilin Feng,Yingmin Qu,Mengyu Wang,Meng Shen,Heqiang Lou,Weiming Cai

Stropharia rugosoannulata, or wine-cap Stropharia, is a well-known edible mushroom cultivated globally. The pileipellis color is a crucial quality attribute of S. rugosoannulata, exhibiting significant variation throughout its developmental stages. However, the pigment types and regulatory mechanisms behind color variation remain unclear. The metabolome analysis found that the anthocyanin biosynthesis pathway was significantly enriched and anthocyanins accumulated steadily in fruiting bodies during three developmental stages. The pileipellis pigment was extracted, and HPLC-MS confirmed the presence of anthocyanins. Notably, significant differences in anthocyanin content were observed among the various colored varieties. Thus, anthocyanins contribute to the pileipellis color of S. rugosoannulata. Through further investigation, this study elucidated, for the first time, the relationship between the “SrNFYA-SrDRF2” regulatory module and anthocyanin accumulation. Combined multiomics assays and HPLC analysis revealed that auxin functions as a signaling molecule that regulates the accumulation of anthocyanins in the pileipellis. Subsequently, the hub gene of anthranilate synthase for auxin synthesis was identified as SrTRP1, and the transcription factor SrMYB1 was verified as a regulator of SrTRP1, influencing auxin accumulation. These findings provide a valuable resource for the targeted enhancement of the quality of S. rugosoannulata.

Difference in Volatile Aroma Components of Stropharia rugosoannulata under Two Cultivated Environments Investigated by SPME-GC-MS

Yanbin Wang,Dan Wu,Yingqi Wu,Xiaoqing Tong,Yuchuan Qin,Liling Wang

In order to study the effect of both greenhouse and forest cultivating environments onStropharia rugosoannulata, its volatile aroma compounds were measured by a headspace solid phase micro extractions—gas chromatograph—mass spectrometer (SPME–GC–MS). The optimal adsorption temperature was 75 °C and the optimal adsorption time was 40 min. A total of 36 volatile aroma compounds were identified by GC–MS, including 8 aldehydes, 2 ketones, 4 alcohols, 15 alkenes, and 4 alkanes. Hexanal, 3-Octanone, 2-Undecanone, (E)-Nerolidol, and (Z)-β-Farnesene made great aromatic contributions. Among them, Hexanal, 3-Octanone, 2-Undecanone were the key aroma compounds for which odor activity values (OAVs) were more than 1. (E)-Nerolidol showed odor modification in the forest samples and showed a key aroma effect in greenhouse samples. (Z)-β-Farnesene showed odor modification in greenhouse samples. 3-Octanone was the largest contributing compound for which the OAV was more than 60. The total content of volatile aroma compounds first increased and then decreased with growth time; it reached the highest level at 48 h: 2203.7 ± 115.2 μg/kg for the forest environment and 4516.6 ± 228.5 μg/kg for the greenhouse environment. The aroma was the most abundant at this time. All samples opened their umbrella at 84 h and become inedible. Principal component analysis (PCA), hierarchical cluster analysis (HCA), and orthogonal partial least squares discriminant analysis (OPLS–DA) were combined to analyze the aroma difference ofS. rugosoannulataunder two cultivation modes. PCA and HCA could effectively distinguish the aroma difference in different growth stages. Under different culturing methods, the aroma substances and their changes were different. The samples were divided into two groups for forest cultivation, while the samples were divided into three groups for greenhouse cultivation. At the end of growth, the aroma ofS. rugosoannulatawith the two cultivation modes was very similar. OPLS–DA clearly distinguished the differences between the two cultivation methods; 17 key aroma difference factors with variable importance projection (VIP) > 1 were obtained from SPLS–DA analysis. Keywords:Stropharia rugosoannulata;cultivation mode;volatile aroma compounds;mushroom;gas chromatography—mass spectrometry

Synergistic effect of novel CS/N-TiO₂/NT coating and low temperature in prolonging the storage life of Stropharia rugosoannulata

Xingjun Lu,Kun Qiao,Xinyan Liu,Xiaozhen Peng,Bangzhu Peng

Postharvest Stropharia rugosoannulata deteriorates from fungal infection and active metabolism. A novel chitosan/nitrogen doped titanium dioxide/natamycin (CS/N TiO₂/NT) composite film was developedinhibiting Fusarium pseudoanthophilum and Talaromyces purpureogenus by 71.30% and 75.04%, with 132.20% higher tensile strength and 69.29% lower oxygen permeability. Preservation efficacy was markedly temperature dependent. At 25 °C, only marginal improvements were observed. At 10 °C, the coating significantly reduced total colony counts and modulated enzymatic activities, notably activating peroxidase, retarding the decline of superoxide dismutase (SOD), and inhibiting polyphenol oxidase. Optimal preservation was achieved at 4 °C, where respiratory intensity and microbial proliferation were suppressed while SOD activity was effectively sustained. Cox proportional hazards analysis further confirmed that coated samples stored at 4 °C exhibited a 33.33%–38.46% extension in storage life relative to uncoated controls. These findings establish a temperature responsive preservation strategy and provide a mechanistic framework for extending the shelf life of perishable mushrooms.

Characterization of gelatin-based composite films loaded with polysaccharide and carbon dots from Stropharia rugosoannulata and their application in pork fresh-keeping

Jintao Wu,Zhaolan Yan,Peng Shan,Wei Li,Kun Wang,Hui Li

In this study, bio-based composite films with enhanced barrier, mechanical, antioxidant and antimicrobial properties were prepared by incorporating Stropharia rugosoannulata waste (SP)-derived polysaccharides (SPP) and carbon dots (SPCDs) as an active substance into gelatin (GEL) matrix, which were evaluated for their potential application in pork preservation. The effects of varying concentrations of SPCDs on the morphology, microstructure, and overall performance of the films were systematically investigated. The findings demonstrated that the addition of SPCDs to the GEL/SPP matrix significantly improved the mechanical strength, water vapor permeability, and ultraviolet light blocking ability of the composite films. Specially, the GEL/SPP/SPCDs composite films containing 3 % carbon dots exhibited radical scavenging rates of 53.74 % for DPPH and 73.19 % for ABTS, along with inhibition rates of 58.37 % for E. coli and 70.25 % for S. aureus . Preservation experiments revealed that the prepared GEL/SPP/SPCDs-3 % films provided superior preservation effect on pork, highlighting their potential as eco-friendly active food packaging materials.
