In this paper, using Styphnolobium japonicum, Cryptomeria fortunei and Pinus yunnanensis as raw materials, cellulose (CE) was first extracted by chemical method (CM1) and biological enzymatic method 2 (BM2), and then further prepared into nanocellulose fiber (CNF) by biological enzymatic method 1(CPM (CPM stands for cellulase and pectinase)). The CE and CNF were characterized by FTIR, XRD, SEM, DSC and TG. The results showed that the CNF prepared by CM1-CPM was cellulose type II, CNF prepared by BM2-CPM was cellulose type I. The best crystallinity of the CNF prepared by CM1-CPM reached 74.1% (C. fortunei CNF), the best yield was 24.0% (S. japonicum CNF) and the minimum average diameter was 62.6 nm (C. fortunei CNF). However, the best crystallinity of CNF prepared by BM2-CPM reached 80.1% (C. fortunei CNF), the best yield was 27.4% (C. fortunei CNF), and the minimum average diameter was 31.6 nm (C. fortunei CNF). And also, the thermal degradation analysis of the CNF prepared from the three raw materials revealed that the CNF prepared by BM2-CPM had better thermal degradability. Finally, the thermal stability analysis of the C. fortunei CNF was carried out, and it was found that the residual mass of CNF prepared by BM2-CPM was 48%, while the residual mass of C. fortunei CNF prepared by BM1-CPM was 22%, which showed the C. fortunei CNF prepared by BM2-CPM had better thermal stability. Therefore, the properties of nanocellulose prepared by BM2-CPM are optimal.

The flower buds of Styphnolobium japonicum (FBSJ) , a medicinal and food plant, exhibit broad application prospects and diverse therapeutic effects, including cooling the blood, stopping bleeding, lowering blood sugar, and exerting antioxidant and anti-inflammatory activities. To provide a reference for the identification, resource development, and utilization, a widely targeted metabolomics study was conducted on FBSJ samples from four production regions in China: Shandong (SJsd), Anhui (SJah), Hebei (SJhb), and Henan (SJhn). In this research, the metabolomic analysis provided the possibility of identifying metabolites to differentiate plants from different origin to broaden a limited metabolome coverage and unclear origin-metabolite-efficacy correlations. A UPLC-MS/MS-based metabolomics analysis was selected to investigate the metabolites of FBSJ from different geographic origins, utilizing the MWDB database and the multi-reaction monitoring mode of triple-quadrupole mass spectrometry (TQMS). A total of 1,559 metabolites were identified, of which 294 were annotated as key active ingredients in traditional Chinese medicine (TCM), and 193 as active pharmaceutical components against 11 diseases, including cancer and diabetes. Among the 708 differential metabolites identified across the four different origin groups, 176 were annotated by KEGG and distributed across 87 metabolic pathways. The most significantly altered metabolic pathway was isoflavonoid biosynthesis ( p < 0.01), where SJsd samples exhibited an upregulation trend in key differential metabolites compared to the other groups. Among the 193 disease-active metabolites, 13 (e.g., 3’-Methoxydaidzein, Maackiain) were at least twofold more abundant in SJsd compared to any other origin, highlighting their critical role in explaining SJsd’s superior pharmacological potential. This study provides a comprehensive molecular-level analysis of FBSJ metabolites, offering useful insights into its pharmacological analysis across different origins. These findings enhance our understanding of FBSJ metabolites annotated as anti-disease-related components in public databases and offer a valuable reference for investigating their potential in human health applications and the development of novel functional foods.

Introduction This research was to evaluate the beneficial effects of Styphnolobium japonicum (L.) Schott flower extract (SJF) on the adjuvant-induced arthritis rat model. Methods Arthritis was evoked by injection of complete Freund’s adjuvant (CFA) in the hind paw. SJF (150 or 300 mg/kg/day) or Celecoxib (5 mg/kg/day) were administered intragastrically from the 0th day to the 28th day. The arthritis symptoms (paw edema, arthritic scores, mechanical hyperalgesia, and thermal hyperalgesia), inflammation biomarkers (RT and CRP), related enzymes (MMP1 and MMP13), oxidative stress markers (CAT, SOD, GPx, and MDA), and inflammatory cytokines (IL-6, IFN-γ, TNF-α, and IL-1β) of SJF-treated CFA rats were evaluated. Results CFA rats exhibited severe arthritis symptoms, increased oxidative stress, and inflammatory cytokines. Interestingly, SJF treatment relieving arthritis symptoms and restored body weight gain compared with those in the CFA group. SJF treatment decreased the levels of CRP, RF, MMP1, and MMP13 in the CFA group. Besides, SJF treatment increased the activities of antioxidant enzymes and decreased the MDA content and inflammatory cytokines compared with those in the CFA group. Moreover, SJF could increase the mRNA expression of GPx-1 and CAT and inhibit the mRNA expression of IL-6 and TNF-α in the ankle tissue of CFA rats.

Styphnolobium japonicum (L.) S chott is widely cultivated in China, and its flowers and flower buds (FFB-SJ) are commonly used as traditional Chinese medicine. This work aimed to assess variations in the chemical components and antioxidant and tyrosinase inhibitory activities of S. japonicum extract during five flower maturity stages (ES1–ES5). The results showed that the contents of total flavonoids, rutin, and narcissin were highest at ES1, whereas the contents of quercetin and isorhamnetin were highest at ES3. ES1 presented considerable antioxidant activities in terms of reducing power (RP) and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH . ) and hydroxyl radical ( . OH) scavenging capacity, whereas ES3 showed excellent tyrosinase inhibitory activity and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical (ABTS .+ )- and O 2 .− -scavenging capacity. Rutin and quercetin are the main bioactive components of FFB-SJ with antioxidant and tyrosinase inhibition, and the immature flower buds of S. japonicum (S2 and S3) with excellent biological activities and relatively high extract yields were the best for product development.

HighlightsWhat are the main findings? We identified 1550 metabolites (flavonoid-dominant), 152 key active ingredients of traditional Chinese medicine (TCM-KAIs), and 204 pharmaceutical disease-resistant ingredients (PDRIs) in the flower bud ofStyphnolobium japonicumcv. Jinhuai (FBSJvJ).Geographic origin drove metabolic divergence. Guangxi samples were enriched in lipids and nucleotides, while Sichuan samples were enriched in flavonoids and phenolic acids. In addition, three core biomarkers were identified. What are the implications of the main findings?Biomarkers enable origin authentication and precision quality control compared to a single marker (rutin), supporting traceability and regulatory compliance for food and medicinal products.Regional metabolic signatures can help identify functional ingredients and guide product formulation, processing optimization, and cultivar selection for food and medicinal development.Background/Objectives:Styphnolobium japonicumcv. Jinhuai (SJvJ) represents a medicinal and edible plant whose metabolite composition is strongly shaped by its growing location. Current quality control methods mainly rely on rutin quantification, lacking comprehensive metabolic markers for origin discrimination. Therefore, this study aimed to profile interregional metabolic differences between Guangxi and Sichuan SJvJ flower buds, identify characteristic differential markers, and clarify relevant metabolic pathways, thereby guiding quality control, germplasm evaluation, and functional food development. Methods: Ultra-high performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) was employed to identify metabolites. The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and Cancer HSP were applied to screen the key active ingredients of traditional Chinese medicine (TCM-KAIs) and disease-related pharmaceutical ingredients (PDRIs). Specifically, we targeted six highly prevalent human diseases and another five disorders based on therapeutic indications documented in the Chinese Pharmacopoeia. Multivariate analyses, such as principal component analysis, hierarchical clustering analysis, and other statistical methods, were applied to investigate differential metabolites. The Kyoto Encyclopedia of Genes and Genomes (KEGG) database was utilized for pathway enrichment analysis of marker metabolites. Results: In total, 1550 metabolites were identified across 12 categories, predominantly flavonoids. Additionally, 152 TCM-KAIs and 204 PDRIs against 11 diseases were screened. Multivariate analyses indicated that geographical origin was closely associated with observed metabolic variation among the tested samples: Guangxi samples accumulated higher lipids and nucleotides, whereas Sichuan samples showed higher levels of flavonoids and phenolic acids. Vanilloloside, protocatechuic acid-4-O-glucoside, and gallic acid-4-O-glucoside were identified as key inter-group biomarkers. KEGG enrichment analysis revealed enhanced metabolism of nucleotide/pyrimidine in Guangxi, whereas zeatin biosynthesis was upregulated in Sichuan, consistent with discrepancies in regional climatic patterns. Conclusions: This study established a more comprehensive metabolomic dataset for FBSJvJ. It also clarified the correlations between origin and quality and unraveled the underlying mechanisms. These findings facilitate origin authentication, standardized quality control, and rational exploitation of FBSJvJ as raw materials of functional foods.
