Ethnopharmacological relevance Syringa Pubescens Turcz. (SP), a member of the Oleaceae family, is a species of plant known as Syringa . Flowers, as the medicinal part, are commonly used in the treatment of hepatitis and tonsillitis. Aim of the study: The research was the first to assess the antioxidant and anti-inflammatory potential of different parts of SP flowers (SPF) in vitro . The most promising fraction was ethyl acetate fraction of SP flower (SPFEA). The antioxidant, anti-inflammatory and analgesic activities of SPFEA were further studied, and the chemical components were identified. Methods HPLC was used to identify the major components in various fraction of SPF. DPPH and ABTS + radical scavenging assays as well as FRAP test and β-carotene bleaching test were employed to assess the antioxidant potential of SPF fraction in vitro . The inhibitory effect on NO production in LPS-treated RAW264.7 cells and heat-induced protein denaturation test were used to evaluate the anti-inflammatory potential of SPF fraction. Further analysis of the biological activity of SPFEA was performed. Acute toxicity test was conducted to assess the toxicity of SPFEA. The anti-inflammatory effect was assessed by utilizing xylene induced ear edema model, carrageenan-induced foot edema model and peritonitis model in vivo . The analgesic effect of SPFEA was evaluated using hot plate test, tail immersion test, formaldehyde test as well as acetic acid-induced abdominal writhing pain experiment in vivo . In carrageenan induced foot edema model, ELISA kits were employed to measure levels of inflammation factors (NO, TNF-α, IL-6, COX-2, IL-1β) in foot tissue as well as MDA, CAT, SOD, GSH-PX levels in liver tissue. Results HPLC results showed that there were significant differences in bioactive substances among different fractions of SPF, and SPFEA was rich in bioacitve components. Compared with other fractions of SPF, SPFEA exhibited better antioxidant and anti-inflammatory abilities. The 3000 mg/kg SPFEA group in mice had no obvious side effects. The xylene-induced ear edema model, carrageenan-induced foot edema and peritonitis models demonstrated that the SPFEA had significant anti-inflammatory effect. Moreover, inflammation factors including NO, TNF-α, IL-6, COX-2, IL-1β were significantly reduced in SPFEA groups in foot tissue induced by carrageenan. Additionally, SPFEA effectively decreased liver tissue oxidative stress levels (MDA, SOD, GSH-PX and CAT). The bioactivities of SPFEA demonstrated a clear dose-dependent relationship. The results of the hot plate test, tail immersion test, formaldehyde test and acetic acid-induced abdominal writhing pain experiments indicated the SPFEA possessed an excellent analgesic effect, and this effect was in dose-dependent manner. Conclusion The study provides a scientific foundation for understanding the pharmacological action of SPFEA. It has been indicated that SPFEA has excellent antioxidant, analgesic and anti-inflammatory effects.

In this work, deep eutectic solvents (DESs) were synthesized using ultrasound-assisted and stirring heating methods. DES combined with ultrasonic-assisted extraction (UAE) was proposed for the efficient extraction of glycosides from Syringa pubescens Turcz. A series of DES systems were used to evaluate the extraction efficiency of glycosides in S. pubescens . The DES composed of choline chloride/glycerol extracted higher than those of conventional solvents and other DESs. The extraction procedure of DES-UAE was optimized using response surface methodology. The extraction solvent was choline chloride-glycerol formed a DES with the mole ratio of 1:2 with 20 % of water content. The optimum conditions for echinacoside (ECH) and oleuropein (OLE) were ultrasound power of 200 W, extraction temperature of 68 °C, solvent to solid ratio of 20:1 mL/g and the extraction time of 45 min. Furthermore, the DES with 30 % water content can be used as dilution solvent before HPLC analysis of DES extract. In addition, the enrichment of ECH and OLE from DES extract was achieved using HPD-450 macroporous resin, with the recoveries of 80.04 % and 86.21 %, respectively.

Syringa pubescens Turcz. has been universally employed to make flower tea for the treatment of cough in the western part of Henan Province. Many researchers have confirmed that the phenylethanoid glycosides from S. pubescens were bioactive compounds. In this study, various macroporous resins have been employed to evaluate the potential of adsorption and desorption for phenylethanoid glycosides. The HPD-450 resins were appropriate for the purification of phenylethanoid glycosides, and the optimum purification conditions were established. The adsorption properties were well modeled using the pseudo-second-order kinetics. Meanwhile, the adsorption isotherm could be better represented by the Langmuir equation, suggesting that the process was an exothermic reaction. Moreover, the purified sample possessed a stronger antioxidant capacity. The inhibitory activity of the crude extract against α-amylase and α-glucosidase was stronger than that of purified sample. The findings obtained provided a scientific foundation for further research and high-value exploration of this plant and a potential technique for scale-up purification of phenylethanoid glycosides.

Inhibition of α-glucosidase is an attractive strategy for the control of hyperglycaemia. Naturally occurring bioactive compounds are potential sources of α-glucosidase inhibitors. The Syringa pubescens Turcz (SP) is commonly used as Chinese folk medicine for treatment of hepatitis and cirrhosis. However, the anti-hyperglycemic activity of SP was lack of study and the report on compounds of α-glucosidase inhibitory activity of SP was also insufficient. In this study, the α-glucosidase inhibitory activity of SP extract was firstly assessed in vitro . The quantification analysis of bioactive glycosides in SP collected at different geographical origins and its correlation with α-glucosidase inhibitory activities were investigated. As the results, six glycosides including salidroside, echinacoisde, forsythoside B, verbascoside, isoacteoside and oleuropein were simultaneously determined using HPLC-DAD method. Moreover, SP collected from different geographical origins exhibited different concentrations of bioactive glycosides and had potential α-glucosidase inhibitory activity. The results of correlation analysis indicated that the amounts of glycosides were positively correlated with the α-glucosidase inhibitory activities of SP. Then, the evaluation of α-glucosidase inhibitory activity of six glycosides was performed. And docking study was conducted to predict the possible binding sites of forsythoside B. Forsythoside B might be the most possible α-glucosidase inhibitor in SP.

Introduction Syringa pubescens Turcz. was reported to be abundant in the Funiu Mountains of Henan Province and can be used to treat hepatitis and cirrhosis. In order to develop and utilise the resource, a fast and simple technique to extract bioactive compounds is needed. Objectives Our aims were to provide an extraction technique of glycosides from S. pubescens and study the antioxidant activity of this material. Methods Box–Behnken design (BBD) was employed with three factors at three levels. The process parameters such as ethanol concentration ( X 1 ), temperature ( X 2 ), and solvent–solid ratio ( X 3 ) could significantly influence efficiency and yield of target compounds. High-performance liquid chromatography (HPLC) was used to determine the content of glycosides. DPPH (α,α-diphenyl-β-picrylhydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) and reducing power were used to evaluate the antioxidant activity of S. pubescens extract. Results The optimal parameters for the maximal extraction yield were obtained with ethanol concentration of 68%, temperature of 89°C, solvent–solid ratio of 46 mL/g, and time of 20 min. The S. pubescens extract showed strong antioxidant properties in vitro . Conclusion The findings indicated the potential application of solvothermal extraction method to extract bioactive compounds from S. pubescens Turcz. Furthermore, the S. pubescens extract could be used as an important resource of antioxidant activity.

A high-performance liquid chromatograph with diode array detector was established for the simultaneous determination of five phenylethanoid glycosides in Syringa pubescens Turcz. The optimal chromatographic conditions were achieved on a Zorbax C18 column using gradient elution with 0.5% aqueous acetic acid and acetonitrile as the mobile phase at the flow rate of 1.0 mL/min. The detection wavelength was developed as follows: 0–10 min, 276 nm; 10–45 min, 332 nm. The validation of the method including linearity, precision, stability, accuracy, repeatability and recovery was tested. The chemometric analysis including hierarchical cluster analysis and principal component analysis was employed to investigate the similarity and difference of samples from different geographical origin. The results revealed that S. pubescens samples were divided into four clusters based on the phenylethanoid glycosides contents. Antioxidant activity of extract was measured using three different methods including α,α-diphenyl-β-picrylhydrazyl and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) radical scavenging assays, and ferric reducing antioxidant power assay. Furthermore, different phenylethanoid glycosides exhibited different contribution to antioxidant capacities. This study provides a foundation for the quality evaluation and offers scientific data for the utilization of S. pubescens resources.

The flower ofSyringa pubescensTurcz. is used in Chinese folk medicine and also as a flower tea for healthcare. The effects of five drying methods on the active compound contents, the antioxidant abilities, anti-inflammatory properties and enzyme inhibitory activities were evaluated. The plant materials were treated using shade-drying, microwave-drying, sun-drying, infrared-drying and oven-drying. The seven active compounds were simultaneously determined using an HPLC method. Furthermore, the chemical profile was assessed using scanning electron microscopy, ultraviolet spectroscopy and infrared spectroscopy. The antioxidant capacities and protective effects on L02 cells induced with hydrogen peroxide were measured. The anti-inflammatory effects on lipopolysaccharide-induced RAW264.7 cells were investigated. The enzyme inhibitory activities were determined against α-amylase, α-glucosidase cholinesterases and tyrosinase. The results indicated that drying methods had significant influences on the active compound contents and biological properties. Compared with other samples, the OD samples possessed low IC50values with 0.118 ± 0.004 mg/mL for DPPH radical, 1.538 ± 0.0972 for hydroxyl radical and 0.886 ± 0.199 mg/mL for superoxide radical, while the SHD samples had stronger reducing power compared with other samples. The SHD samples could be effective against H2O2-induced injury on L02 cells by the promoting of T-AOC, GSH-PX, SOD and CAT activities and the reducing of MDA content compared with other samples. Furthermore, SPF samples, especially the SHD sample, could evidently ameliorate inflammation through the inhibition of IL-6, IL-1β and TNF-α expression. All the studied SPF samples exhibited evidently inhibitory effects on the four enzymes. The IC50values of inhibitory activity on α-glucosidase and α-amylase from SHD sample were 2.516 ± 0.024 and 0.734 ± 0.034 mg/mL, respectively. SD samples had potential inhibitory effects on cholinesterases and tyrosinase with IC50values of 3.443 ± 0.060 and 1.732 ± 0.058 mg/mL. In consideration of active compound contents and biological activities, it was recommended that SHD and SD be applied for drying SPF at an industrial scale. Keywords:Syringa pubescensTurcz.;drying method;bioactive component;biological activity

An efficient ultrasound-assisted aqueous two-phase extraction method was developed for the extraction of verbascoside from the leaves of Syringa pubescens Turcz. The ethanol/ammonium sulfate system was used as the extraction solvent. Based on the results of single-factor experiments, the key parameters, including ethanol concentration, ammonium sulfate concentration, and sample mass, were further optimized using response surface methodology. The optimal extraction conditions were obtained as follows: ethanol concentration of 30% (w/w), ammonium sulfate concentration of 19% (w/w), sample mass of 0.63 g, ultrasonic power of 150 W, extraction time of 30 min, and temperature of 40°C. Under these conditions, the experimental verbascoside yield (5.23 ± 0.29 mg/g) was in good agreement with the model-predicted value (5.33 mg/g), confirming the reliability of the established model. Furthermore, the verbascoside extract exhibited potent radical scavenging activities with half‑maximal inhibitory concentration values of 0.039 mg/mL against α, α-diphenyl-β-picrylhydrazyl and 0.058 mg/mL against 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid), as well as a ferric-reducing antioxidant power of 2.78 mg Fe 2 + /g at 2.5 mg/mL. This study demonstrates that the ultrasound-assisted aqueous two-phase extraction method is an efficient, green, and reliable approach for the extraction of verbascoside from S. pubescens leaves, and the obtained verbascoside extract shows promise as a natural antioxidant agent for potential applications in the food and pharmaceutical industries.

The flower of Syringa pubescens Turcz., frequently prepared as herbal tea, exhibits various biological properties, including anti-inflammatory, antioxidant, and hepatoprotective activities. Herein, we identify the chemical constituents from ethanol extract of S. pubescens flower (SPFE) and explore its antioxidant, anti-xanthine oxidase (anti-XOD), and 5-lipoxygenase (5-LOX) enzyme activities, and anti-inflammatory activities and mechanisms. SPFE contained a variety of glycosides and showed good antioxidant capacity and effectively inhibited the activity of 5-LOX and XOD enzymes in vitro. In the model of RAW264.7 cell inflammation generated by lipopolysaccharide, SPFE not only impeded the expression of inflammatory mediators and the accumulation of malondialdehyde, but also restored the capacity of antioxidant enzymes. Moreover, SPFE could notably relieve or treat inflammation by suppressing the expression of inducible nitric oxide synthase and cyclooxygenase-2 proteins. In vivo inflammation models, oral SPFE remarkably reduced the swelling of the ear, paw tissues in mice and had a good anti-inflammatory activity, reducing cell infiltration in the claw tissue, and restoring the degree of oxidative damage in the organism. Our findings demonstrate that SPFE exhibits promising potential as a novel therapeutic agent or nutraceutical for anti-inflammatory applications. Graphical
