Simple SummaryThe experiment was designed to investigate the effect of gestational and lactational exposure to the T-2 toxin and its effects on the puberty of female mice offspring. T-2 toxin contaminates feed participates in the maternal diet. It directly interferes with the normal physiological function of offspring because the maternal diet is the most important during pregnancy and lactation. Female mice are more sensitive to the maternal diet during the lactation stage. Thus, their maturity stage is very complex and regulated by the hypothalamus, which controls Gonadotropin-releasing hormone (GnRH), and it also secretes reproductive hormones. Current findings report that T-2 toxins during late gestation and lactation period directly affect offspring, disturb the vaginal environment, delayed normal estrus cycle, and puberty age induces oxidative stress-caused ovarian damage. Based on findings, we think it is necessary to raise people’s attention to T-2 toxin-contaminated feed offered to animals. Current results reveal information about the health risks associated with a maternal diet, which affects the next generation.AbstractAmong foodborne toxicities, the T-2 toxin is the most toxic member of trichothecenes mycotoxins, which has been shown to impair the development and reproductive efficiency of animals. Pups are particularly more quickly prone to programming the effects of the maternal diet during the gestational and lactation periods. Few studies have reported the maternal toxic effect on the next generation. Dams were served the T-2 toxin at a dose of 0.005 and 0.05 mg/kg body weight/day and control group 0 mg/kg from gestation day 14 to lactation day 21. Female mice offspring were selected at the weaning age. Our observations indicate that age during the vaginal opening and di-estrus stage increased and the length of the estrus cycle, first di-estrus, and regular estrus cycling were delayed with prolonged di-estrus in the 0.05 mg/kg group compared to the 0.005 mg/kg and control group. Transcription level analysis showed that mice at a dose of 0.05 mg/kg exhibited a decrease in hypothalamic mRNA expression ofGnrhandGnrhr, Lhb,andFshbin the pituitary gland, with a significant decrease ofFshrandLhrin the ovaries. Present findings report that postnatal exposure to the T-2 toxin delayed puberty age in female mice and induced oxidative stress, ovarian damage, and reduced vaginal epithelium wall majorly in the 0.05 mg/kg group, and showed fewer effects in the 0.005 mg/kg group. Keywords:estrus cycle;mice;puberty;T-2 toxin

T-2 toxin is a kind of group A trichothecenes mycotoxins , frequently detected in various foods and feeds, having high toxic effects on both humans and animals. The present study aims to investigate the toxic effects of T-2 toxin exposure to ICR mice during pregnancy and lactation on liver glycolipid metabolism of young mice. The pregnant mice were given 0, 0.005 and 0.05 mg of T-2 toxin/kg bw daily through oral gavage from late gestation (GD 14) to the lactation (LD 21). Liver and serum samples of the young mice were collected on postnatal day 21 (PND 21), PND 28 and PND 56. The results showed that T-2 toxin increased the contents of triglycerides (TG), total cholesterol (T-CHO) and glucose in serum of young mice on PND 21 and PND 28. In addition, obvious lipid droplets of liver in T-2 toxin treatment groups were observed, especially in 0.05 mg group of PND 21and PND 28. Compared with the control group, T-2 treatment also increased the expressions of genes associated with liver glycolipid metabolism, such as PEPCK , Glut2 , Fas , Acox1 , Hmgcr , PPARα , Srebp1 and CD36 . These results demonstrated T-2 toxin exposure to pregnant mice could cause liver glycolipid metabolism disruption in the young mice and the toxic effects weakened on PND 56.

Simple SummaryThis study investigated the effects of maternal T-2 toxin exposure on the development of testis in the mice offspring. The detrimental effects were assessed by testicular weight, antioxidant capacity, and testosterone synthesis and secretion. Studies have shown that the toxin carried by the mother has bad effects on the testicular development of offspring at puberty, affecting the antioxidant system and testosterone synthesis in the testis, but the maternal exposure of T-2 toxin had no significant impact on the testes of offspring after sexual maturity, suggesting the recovery of reproductive function.AbstractT-2 toxin, the most toxic member of trichothecene mycotoxin, is widely distributed in cereals, and has been extensively studied, but few studies focus on the toxicity of maternal exposure to offspring. This study focused on the effects of maternal exposure to T-2 toxin (during gestation and lactation) on the testicular development of mice offspring. Dams were orally administered with T-2 toxin at 0, 0.005, or 0.05 mg/kg body weight from the late stage of gestation to the end of lactation. Testicular samples of the mice offspring were collected on the postnatal day 21, 28, and 56. The results showed significant decreases in body weight and testicular weight on the postnatal day 28. Moreover, significant inhibition of antioxidant system and testosterone synthesis was detected on the postnatal day 28. Furthermore, there were significant decreases in the gene expression levels ofStARand3β-HSD, which are involved in testosterone synthesis. In general, present results demonstrated that maternal exposure to T-2 toxin during gestation and lactation led to bad effects on the capacity of antioxidant system and inhibited testosterone synthesis in testes during pre-puberty with no significant effects on post-puberty.Keywords:maternal exposure;oxidative stress;T-2 toxin;testicular development;testosterone synthesis

The T-2 and HT-2 toxins, the main metabolites of Fusarium poae , induce toxicity in broilers and accumulate in tissues. Consequently, during the breeding process of broilers, diets are frequently supplemented with physical adsorbents to protect birds against the toxicity induced by mycotoxins. In the present research, T-2 and HT-2 were produced in maize inoculated with F. poae . Mont, the strongest adsorbent based on in vitro adsorption ratios, was added to the contaminated diet. One-day-old chickens were randomly and equally divided into the following four groups: control diet group, Mont supplemented diet group, contaminated diet group and detoxification diet group. The experiment lasted for 42 days. Compared to the control group, the contaminated group showed significant decrease in body weight, feed intake and TP ( P < 0.05), and marked increase in FCR, ALP, AST and ALT activity, T-2/HT-2 residues in the tissues and the relative expressions of apoptosis-related mRNAs ( P < 0.05). Mont supplementation provided protection for the treated broilers in terms of performance, blood biochemistry, hepatic function, T-2/HT-2 residue of tissues and apoptosis. Therefore, Mont may be suitable as a detoxification agent for T-2/HT-2 in feed for broilers.

The “turn-off” mode lateral flow immunoassay (LFA) based on the conventional competitive format and “turn-on” mode LFA mainly determined by inner filter effect (IFE-LFA) were objectively compared using T-2 toxin as a model molecule and T-2-BSA and anti-T-2 monoclonal antibody as immunocomplex pairs to improve the sensitivity of the LFA. First, Au nanoparticles (Au NPs), amorphous carbon nanoparticles (ACNPs), quantum dots nanospheres (QDs), and time-resolved fluorescent microspheres (TRFMs) were selected as labels for developing the “turn-off” mode LFAs (Au NPs-LFA, ACNPs-LFA, QDs-LFA, and TRFMs-LFA, respectively). Thereafter, Au NPs and ACNPs were used as absorbers, and QDs and TRFMs were selected as fluorescers, for preparing the “turn-on” mode IFE-LFAs (IFE-LFA based on Au NPs as absorbers and QDs as fluorescers [Au NPs-mAb-QDs-BSA], IFE-LFA based on Au NPs as absorbers and TRFMs as fluorescers [Au NPs-mAb-TRFMs-BSA], IFE-LFA based on ACNPs as absorbers and QDs as fluorescers [ACNPs-mAb-QDs-BSA], and IFE-LFA based on ACNPs as absorbers and TRFMs as fluorescers [ACNPs-mAb-TRFMs-BSA]). Under optimized conditions, the naked-eye observation cut-off values for the aforementioned eight LFAs were 4, 2, 2, 2, 3, 2, 1.5, and 1 ng/mL, with quantitative limit of detection values of 0.50, 0.23, 0.24, 0.23, 0.47, 0.45, 0.27, and 0.22 ng/mL, respectively. Among all LFAs, ACNPs-mAb-TRFMs-BSA showed the highest sensitivity. The results of this study provide a perspective for improving the sensitivity of the LFAs to meet the requirements of on-site screening for trace substances.

T-2 is a trichthecenes mycotoxin responsible for severe food poisoning and toxic influences in humans and animals. Therefore, a rapid and sensitive detection of T-2 is an urgent need to protect the public health mainly caused by the consumption of contaminated agricultural products. Herein, we have prepared one pot synthesis of aptamer functionalized silver nanoclusters (apt-AgNCs) for fluorescent detection of T-2 that relies on novel fluorescence resonance energy transfer (FRET). Apt-AgNCs, act as energy donor, were applied as fluorescent sensing probe whose fluorescence was quenched by MoS 2 employed as fluorescent acceptor. Introduction of target in proposed bioassay leads to desorption of Apt-AgNCs from MoS 2 , and recovery of fluorescence in a toxin concentration dependent-manner. A dynamic quantification range was 0.005–500 ng mL −1 while the LOD was 0.93 pg mL −1 . Spiking analysis reveals the efficiency and robustness of the developed assembly, with recovery percentages ranged from 89.46% to 102.08% and 90.41% to 107.75% in maize and wheat samples, respectively. The results were cross-verified using standard commercial T-2 EISA kit. In conclusion, developed bioassay may find good utility in testing for risk assessment of T-2 toxin.

T-2 toxin, a hazardous mycotoxin often present in cereals and products based on cereals, poses a substantial risk to humans and animals due to its high toxicity. The development of uncomplicated, quick and highly sensitive methods for detecting T-2 toxin is imperative. In this work, a portable sensing system was constructed using water column height as a readout device in combination with a controlled release system, which allows for an accurate quantitative analysis of T-2 toxin without the need for expensive instrumentation or skilled technicians. Hyaluronic acid (HA) hydrogel was constructed by double cross-linked DNA/aptamer hybrids with polyethyleneimine (PEI) and embedded with platinum nanoparticles (Pt NPs). The aptamer specifically bound to T-2 toxin in its presence, resulting in the disruption of the hydrogel and subsequent release of the Pt NPs. These Pt NPs were later mixed with a solution of H 2 O 2 in a confined reaction flask, leading to the decomposition of H 2 O 2 into O 2 . A glass capillary tube containing a column of red water had been inserted into the cap of the reaction flask, and the low solubility of O 2 led to an increase in pressure within the reaction unit, causing the red water column to rise. There is a good linear correlation between the height of the capillary liquid level and the T-2 toxin concentration in the range of 20 ng/mL to 6 μg/mL. The system has been successfully used to detect T-2 toxin in samples of barley tea and corn.

The excellent detection sensitivity in various matrices of T-2 toxin (T-2), which has cytotoxic and immunosuppressive effects in DNA and RNA synthesis, is a highly desirable characteristic. A sensitive molecularly imprinted electrochemical sensor was constructed for the selective detection of T-2. In this study, iron ions (Fe 3+ ) were introduced to increase the chelation of the metal ions and templates for preparing molecularly imprinted polymers (MIPs). With the increased chelation of the metal ions and templates, the selectivity and sensitivity of the MIPs were effectively improved. The imprinted sensor was successfully employed to detect T-2 in cereals and human serum samples.

T-2 toxin belongs to the trichothecenes group A compound, mainly produced by Fusarium fungi. It has been shown that T-2 toxin could cross the placental barrier and breast milk, thus endangering the health of offspring. The present study aimed to explore the effects of maternal T-2 toxin exposure on the integrity of the intestinal barrier and the intestinal microflora of young mice. From late pregnancy (GD 14) to lactation (LD 21), pregnant mice were given T-2 toxin daily at 0, 0.005, or 0.05 mg T-2 toxin/kg BW. Postnatal day 21 (PND21), PND28, and PND56 young mice were chosen as objects to detect the influences of maternal T-2 toxin exposure to mice on the offspring. The results showed that maternal exposure to T-2 toxin disturbed the balance of the intestinal microbial flora of the young mice. Villous adhesions and fusion of ileum were observed in T-2-treated groups. In addition, supplementation of T-2 toxin significantly decreased the gene expressions of Claudin 1 , Occludin , Tjp1 , Il10 , Il6, and Tnf in PND 21. However, in PND 28, the expressions of Tnf were significantly increased . T he expressions of Claudin 1 , Occludin , Tjp1 , Il10 , Il6 and Tnf were significantly increased after T-2 toxin treatment in PND 56. These results suggested that maternal exposure to T-2 toxin has negative influences on the intestine of young mice, which may be due to the alterations of microbial composition.

T-2 and HT-2 toxins belong to mycotoxins which are found in human foods and animal chow. We investigated the toxicity and oxidative stress induced by T-2/HT-2 in broilers and chicken hepatocytes. Maize cultures of Fusarium poae was fed to broilers for 42 d, and the physiological index, biochemical index and expression of mRNAs related to oxidative stress were analyzed. Chicken hepatocytes were treated with different levels of T-2/HT-2, and the following parameters were detected: cell viability, GSH and MDA concentration, LDH leakage , activities of ALT/AST, ROS , GSH-PX, SOD and CAT , and expression of mRNA related to oxidative stress. In vivo, high levels of mycotoxins (4 mg/kg T-2 and 0.667 mg/kg HT-2) in feed caused significant reductions in body weight, weight gain, and serum total protein, and significant increases in feed conversion ratio, ALP , ALT/AST activities, and expression of mRNA related to oxidative stress. In vitro, cells treated with T-2/HT-2 showed reductions of GSH concentration and significant increases in LDH leakage, ALT/AST ROS, GSH-PX, SOD and CAT activities, MDA concentration, and expression of mRNA related to oxidative stress. Consequently, F. poae culture material and T-2/HT-2 induced toxicity and oxidative stress in vivo and in vitro, respectively.

A new highly specific and sensitive monoclonal antibody (MAb) to T-2 toxin (T-2) was produced, providing an IC50value of 1.02 ng/mL and negligible cross-reactivity (CR) to other related mycotoxins. Based on the new MAb, a lateral-flow immunochromatographic assay (LFIA) using colloidal gold (CG) and fluorescent microspheres (FMs) as labels was proposed for T-2. Under the optimized conditions, in rapid qualitative assay, the cut-off values of the CG-LFIA were 400 μg/kg in rice and 50 μg/L in fresh milk, and the cut-off values of the FMs-LFIA were 100 μg/kg in both rice and chicken feed. For the quantitative assay with the FMs-LFIA, the limit of detection (LOD) were 0.23 μg/kg and 0.41 μg/kg in rice and chicken feed, respectively, and the average recoveries ranged from 80.2% to 100.8% with the coefficient of variation (CV) below 10.8%. In addition, we found that the CG-LFIA could tolerate the matrix effect of fresh milk better than the FMs-LFIA, while the FMs-LFIA could tolerate the matrix effect of chicken feed better than CG-LFIA under the same experimental conditions. These results provide a certain reference for the selection of appropriate labels to establish a rapid LFIA in various biological samples. Keywords:monoclonal antibody;colloidal gold;fluorescent microsphere;lateral-flow immuno-chromatographic assay;T-2 toxin

A stable and sensitive method has been developed for use in food and livestock product safety for the detection of mycotoxins. This newly developed method allows for the determination of T-2 toxin, HT-2 toxin and diacetoxyscirpenol (DAS) in heart, liver, spleen, lung, kidney, Glandular stomach, muscular stomach, small intestine, muscle, bone and brain samples from broilers using liquid chromatography coupled to tandem mass spectrometry (LC–MS/MS). The samples were initially extracted with ethyl acetate before being filtered through a 0.22 μm nylon syringe filter and subjected to chromatographic separation on a reversed-phase C18 (50 × 2.1 mm, 3 μm) column. A mobile phase composed of 0.1% acetic acid and 10 mM ammonium acetate in methanol and water was used in an assay of the levels of T-2 toxin, HT-2 toxin and DAS. For the analysis of the target compounds, the mass spectrometer was operated under positive electrospray ionization conditions in the selected reaction monitoring mode. The limit of detection was in the range of 0.02–0.05 ng/g, whereas the limit of quantification was in the range of 0.08–0.15 ng/g. The extraction recoveries of spiked samples from the high, intermediate and low levels ranged from 58.5% to 110.5%, and the relative standard deviation (RSD (%)) values were less than 17.0%. The results of inter- and intra-day precision (RSD (%)) were within 14.7%. The results revealed that the present method could be successfully applied to the analysis of T-2 toxin, HT-2 toxin and DAS in the real samples
