Effects of Astaxanthin on oxidative stress of sows and piglet growth performances

Xue Qin,Menghao Yang,Xiaolin Wang,Haoqi Xiao,Chaoyue Cao,Jiawen Chai,Weijun Pang

Sows commonly suffered ROS due to increased energy demands and metabolic burdens during late gestation (LG) and lactation. Astaxanthin (Asta) is a potent antioxidant, which can enhance livestock health and productivity. Therefore, this study aimed to evaluate the effects of Asta on antioxidant status, reproductive outcomes in sows, and the growth performance of piglets. A total of 80 large white sows were randomly divided equally into dietary treatments supplemented with 0, 4, 8 and 12 mg/kg Asta from 85 days of gestation to 21 days of lactation. The results demonstrated that with increasing levels of dietary Asta, the concentrations of Asta in placenta, colostrum, and piglet liver significantly increased ( P < 0.05), which notably reduced the levels of reactive oxygen species (ROS) and malondialdehyde (MDA), while significantly increasing total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) activity in sow serum, placenta, colostrum, and piglet serum ( P < 0.05). In addition, Asta supplementation significantly increased the number of live births (NLB) in sow and average daily gain (ADG) of piglet with a significant improvement in placental angiogenesis and duodenal development status of piglets ( P < 0.05). Overall, dietary Asta in sows can be transferred to piglets through the placenta or milk, which can improve sow reproductive outcomes and piglet growth by reducing oxidative stress during late gestation (LG) and lactation.

Bifidobacterium breve CCFM1310 enhances immunity in immunosuppressed mice via modulating immune response and gut microbiota

Huiting Fang,Huizhen Li,Yang Chen,Xiaoming Liu,Jianxin Zhao,Paul Ross,Catherine Stanton,Wei Chen,Bo Yang

There has been a rising interest in studying how Bifidobacterium breve can affect the immune system over the years. The aim of this study was to evaluate the effect of B. breve on the immune system and to explore the mechanism in a mouse model of cyclophosphamide (CTX)-induced immunosuppression. The results of the study showed that B. breve CCFM1310 significantly increased the thymus index (p < 0.05), promoting the secretion of immunoglobulins and the number of immune cells in the blood. Moreover, B. breve CCFM1310 could repair the spleen injury. It increased the mRNA expression level of T-bet (p < 0.05) and decreased the mRNA expression level of TGF-β and FOXP3 (p < 0.05). B. breve CCFM1310 alleviated oxidative stress by increasing enzymatic activities of SOD, CAT and T-AOC and reducing the concentration of MDA. Moreover, it repaired intestinal damage by increasing the number of goblet cells, upregulating mRNA expression of tight junction proteins, and promoting the production of short-chain fatty acids (SCFA). B. breve CCFM1310 could regulate the gut microbiota by increasing the relative abundance of Lactobacillus (p < 0.05) and decreasing the relative abudance of Lachnospiraceae NK4A136 group . Overall, our study suggests that B. breve CCFM1310 could enhance immunity in CTX-induced immunosuppressed mice by affecting immune responses, oxidative stress and gut microbiota.

Monotropein alleviates septic acute liver injury by restricting oxidative stress, inflammation, and apoptosis via the AKT (Ser473)/GSK3β (Ser9)/Fyn/NRF2 pathway

Kunmei Xie,Feibiao Wang,Yue Yang,Shoujie Pan,Junyao Wang,Nan Xiao,Xinyan Wang,Zhihao Ma,Xiaolong Xu,Zibo Dong

Sepsis-associated acute liver injury (ALI) is a deadly condition resulting from a systemic inflammatory response to liver cell damage and malfunction. Monotropein (MON) belongs to the iris group of compounds extracted from the natural product Mollen dae officinalis radix, which has strong anti-inflammatory and antioxidant pharmacological effects. The purpose of this study was to elucidate the underlying mechanism of MON in the treatment of sepsis ALI. In this study, an in vivo caecal ligation puncture (CLP)-induced ALI model and in vitro LPS-stimulated AML12 cells and RAW264.7 cells model were established. Additionally, a variety of experimental techniques, including CCK8, H&E staining, DHE probe labelling, biochemical, QPCR, and Western blotting and blocking tests, were used to explore the role of MON in ALI. The results showed that MON improved liver morphological abnormalities, oedema, histopathological injury, and elevated ALT and AST, providing a protective effect against ALI. MON reduced CYP2E1 expression, alleviated oxidative stress (downregulation of MDA levels and upregulation of GSH, CAT, and T-AOC levels) and ROS accumulation with the involvement of the NRF2-Keap-1 pathway. MON inhibited inflammation via the TLR4/NF-κB/NLRP3 inflammasome pathway. In addition, it activated the Akt (Ser473)/GSK3β (Ser9)/Fyn pathway and accelerated NRF2 nuclear accumulation; MK-2206 blockade reversed the NRF2 nuclear accumulation and anti-inflammatory function of MON. MON also restricted the mitochondrial apoptosis pathway, a process specifically blocked by MK-2206. In summary, we concluded that MON alleviated septic ALI by restricting oxidative stress, inflammation, and apoptosis via the AKT (Ser473)/GSK3β (Ser9)/Fyn/NRF2 pathway.

Hypoglycemic and H2O2-induced oxidative injury protective effects and the phytochemical profiles of the ethyl acetate fraction from Radix Paeoniae Alba

Lu Zhang,Chun-yan Peng,Pei-xin Wang,Linju Xu,Jia-hui Liu,Xing Xie,Ling Lu,Zong-cai Tu

Radix Paeonia Alba (RPA) is often used as food and medicine. This study aimed to enrich and identify the antioxidant and hypoglycemic bioactive compounds from RPA. The results indicated that the ethyl acetate fraction (EAF) showed the highest total phenolic content, DPPH, ABTS + scavenging ability, and α-glucosidase inhibition ability (IC 50 = 7.27 μg/ml). The EAF could alleviate H 2 O 2 -induced oxidative stress in HepG2 cells by decreasing the MDA and ROS levels, improving cell apoptosis, increasing the enzyme activity of GPX-Px, CAT, SOD, Na + /K + -ATP, and Ca 2+ /Mg 2+ -ATP, and stimulating T-AOC expression, which also enhanced the glucose uptake of insulin-resistant HepG2 cells. In addition, the EAF significantly reduced the fasting blood glucose level and improved glucose tolerance in diabetic mice. An HPLC-QTOF-MS/MS analysis displayed that procyanidin, digallic acid isomer, methyl gallate, tetragalloylglucose isomer, dimethyl gallic acid, and paeoniflorin were the major compounds in the EAF. These findings are meaningful for the application of the EAF in the medicinal or food industry to prevent and treat oxidative stress and diabetes mellitus.
