Active-target T1-weighted MR Imaging of Tiny Hepatic Tumor via RGD Modified Ultra-small Fe3O4 Nanoprobes

Zhengyang Jia,Lina Song,Fengchao Zang,Jiacheng Song,Wei Zhang,Changzhi Yan,Jun Xie,Zhanlong Ma,Ming Ma,Gaojun Teng,Ning Gu,Yu Zhang

Developing ultrasensitive contrast agents for the early detection of malignant tumors in liver is highly demanded. Constructing hepatic tumors specific targeting probes could provide more sensitive imaging information but still faces great challenges. Here we report a novel approach for the synthesis of ultra-small Fe 3 O 4 nanoparticles conjugated with c(RGDyK) and their applications as active-target T 1 -weighted magnetic resonance imaging (MRI) contrast agent (T 1 -Fe 3 O 4 ) for imaging tiny hepatic tumors in vivo . RGD-modified T 1 -Fe 3 O 4 nanoprobes exhibited high r 1 of 7.74 mM -1 s -1 and ultralow r 2 /r 1 of 2.8 at 3 T, reflecting their excellent T 1 contrast effect at clinically relevant magnetic field. High targeting specificity together with favorable biocompatibility and strong ability to resist against non-specific uptake were evaluated through in vitro studies. Owing to the outstanding properties of tumor angiogenesis targeting with little phagocytosis in liver parenchyma, hepatic tumor as small as 2.2 mm was successfully detected via the T 1 contrast enhancement of RGD-modified T 1 -Fe 3 O 4 . It is emphasized that this is the first report on active-target T 1 imaging of hepatic tumors, which could not only significantly improve diagnostic sensitivity, but also provide post therapeutic assessments for patients with liver cancer.

Biodegradable GdPO4·H2O/PLGA microcarriers for stem cell delivery and non-invasive MRI translocation tracing

Zhu Wenhao,Wu Zhenxu,Wang Peng,Liu Tianyi,Guo Min,Ji Jiansong,He Mingfeng,Wang Haifeng,Zhang Peibiao

Within brain tissue engineering, stem cell implantation assisted by biodegradable injectable scaffolds is a crucial method. However, implanted scaffolds, especially microcarriers, may translocate due to the fluidity of the materials. Therefore, the development of an MRI contrast enhancement microcarrier, which could noninvasively trace the location of the implant in vivo, is necessary to guide the evaluation of treatment prognosis. In this study, GdPO 4 ·H 2 O nanoparticles were utilized as the dispersed phase to endow PLGA microcarriers with T1-weighted MRI contrast ability. By adjusting the dispersed phase of Gd compound nanoparticle in PLGA Matrix, a cross-comparison experiment between MRI contrast enhancement imaging and biocompatibility was conducted, and 0.8% Gd was found to be the most suitable rate for preparing nano-composites. Moreover, 0.8% Gd/PLGA microcarriers were suspension-cultured with stem cells in vitro and implanted into traumatic brain injury rats in vivo. Excellent cytocompatibility and enhancement of the T1 phase of MRI were confirmed. This work created a T1-weighted MRI contrast-enhanced microcarrier, which provides a clinical noninvasive tracing cell scaffold for brain tissue engineering.

Manganese-Doped Carbon Dots with Redshifted Orange Emission for Enhanced Fluorescence and Magnetic Resonance Imaging

Sijia Sun,Lining Zhao,Di Wu,Hongxin Zhang,Haichen Lian,Xiaolong Zhao,Aiguo Wu,Leyong Zeng

Metal-doped carbon dots (CDs) exhibited promising application in fluorescence and magnetic resonance (MR) imaging, but developing manganese-doped CDs (Mn-CDs) with long wavelength emission and enhanced MR performance is a challenge. Herein, using a one-step solvothermal method, Mn-CDs with redshifted orange emission and enhanced longitudinal relaxation were synthesized for fluorescence and MR imaging. The results indicated that the prepared Mn-CDs had a uniform size distribution, and the average size was 5 nm. Moreover, Mn-CDs possessed a stronger fluorescence performance than Mn-free CDs, and simultaneously, the emission wavelength can redshift from 542 nm (green emission) to 578 nm (orange emission), owing to the increasing N-doping. Because of the movement limit of Mn2+, Mn-CDs exhibited high T1-weighted MR imaging performance with a longitudinal relaxation rate of 12.69 mM–1·s–1. The in vivo experiments demonstrated their excellent fluorescence and MR imaging with safety and reliability. Therefore, the prepared Mn-CDs with orange emission can be an excellent candidate as a dual-modal nanoprobe for fluorescence and MR imaging.

Manganese and dysprosium codoped carbon quantum dots as a potential fluorescent/T1/T2/CT quadri-modal imaging nanoprobe

Chunxing Li,Yun Wang,Huayan Nong,Xiaoxi Hu,Yu Wu,Yanjun Zhang,Caimei Liang,Keguang Chen,Songliang Li

It has been challenging to integrate various medical imaging modalities into an ultra-small nanoparticle with good biocompatibility to build highly efficient multimodal imaging nanoprobes. A new manganese and dysprosium codoped carbon quantum dots (Mn,Dy-CQDs) with a mean diameter of 1.77 nm was synthesized for fluorescence imaging, T1/T2-weighted magnetic resonance imaging (T1/T2-weighted MRI), and x-ray computed tomography (CT) imaging using a simple one-step hydrothermal approach. The obtained Mn,Dy-CQDs showed good water solubility, long-term stability, strong stable fluorescence property (fluorescence quantum yield of 31.62%), and excellent biocompatibility. The cell imaging verified that the Mn,Dy-CQDs have high efficiency of fluorescence imaging. The Mn,Dy-CQDs, on the other hand, had a superior x-ray absorption performance (47.344 HU l g−1), a higher longitudinal relaxivity (r1 = 7.47 mM−1 s−1), a higher transverse relaxivity (r2 = 42.686 mM−1 s−1). In vitro T1/T2-weighted MRI and CT imaging showed that Mn,Dy-CQDs can produce a strong contrast enhancement impact. To summarise, the Mn,Dy-CQDs may be used as a T1/T2-weighted MRI/CT/fluorescent quadri-modal imaging nanoprobe, indicating that they have a lot of uses in biomedical multimode imaging and clinics.

Core@shell Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles for triple-modality T1/T2-weighted MRI and NIR-to-NIR upconversion luminescence imaging agents

Yang Luo,Sinan Du,Wei Zhang,Zhengfang Liao,Fang Zuo,Shengtao Yang

Core@shell structures of Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles (NPs) with multifunctional properties were prepared using a hydrothermal route with a seed-growth procedure. The effect of Mn2+ ions on the phase, shape, and upconversion luminescence (UCL) of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was explored using X-ray diffraction, transmission electron microscopy, dynamic light scattering, and photoluminescence spectroscopy. Furthermore, hydrophobic to hydrophilic surface modification of the Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was achieved by coating the NPs with an amphiphilic polymer (poly(maleic anhydride-alt-1-octadecene) (C18PMH)) modified with amine-functionalized methyl ether poly(ethylene glycol). Then, the NIR-to-NIR UCL ascribed to the Tm3+ ions, T1-weighted MRI ascribed to the Mn2+ ions, and T2-weighted MRI ascribed to the Fe3O4 core of the hydrophilic Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs were evaluated. The results indicate that Mn2+ doping is an effective method to control the size/shape and improve the UCL intensity of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs, which are promising as imaging agents for NIR-to-NIR UCL and T1/T2-weighted MRI in biomedical research.

Achieving Ultrasmall Prussian Blue Nanoparticles as High-Performance Biomedical Agents with Multifunctions

Zhiguo Qin,Bo Chen,Yu Mao,Chu Shi,Yan Li,Xiao Huang,Fang Yang,Ning Gu

Prussian blue nanoparticles (PBNPs), which belong to the iron-based metal–organic frameworks, are important biomedical agents. Reducing the size of PBNPs can bring improved functional properties, but unfortunately, has been a long-standing challenge. Herein, sub-5 nm ultrasmall PBNPs (USPBNPs) were successfully synthesized by using ethanol/water mixture as the solvent and polyvinyl pyrrolidone (PVP) as the surface capping agent. Adjusting the ethanol/water ratio is not only able to control the nucleation time and size of PBNPs but also tune the conformation of PVP molecules so as to prevent interparticle attachment and enlargement. At an ethanol/water ratio of 3:1, highly stable USPBNPs with a size of ∼3.4 nm were synthesized. Due to their large specific surface area, they demonstrated high peroxidase-like and catalase-like activities, which outperform PBNPs synthesized by a conventional method. In addition, they also showed a high longitudinal relaxation rate (r1) of 1.3 mM–1 S–1, suggesting their potential to be used as T1 MRI agent.

Ultra-large-scale production of ultrasmall superparamagnetic iron oxide nanoparticles for T1-weighted MRI

Yuan-Peng Rui,Bo Liang,Fenglin Hu,Jie Xu,Yi-Feng Peng,Pei-Hao Yin,Yourong Duan,Chunfu Zhang,Hongchen Gu

Ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles with high stability in physiological conditions have great potential for T1-weighted MRI. In the current study, we developed a procedure for ultra-large-scale production of USPIOs (1.5 kg per batch) for T1-weighted MRI. The USPIOs were coated with poly(acrylic acid) (PAA@USPIOs). They were uniform in size (∼4.5 nm) and very stable in physiological conditions even after being sterilized in an autoclave. Due to their small size, the PAA@USPIOs demonstrated good T1 MRI contrast effect and could be used for both first-pass and steady-state magnetic resonance angiography (MRA). After being injected with PAA@USPIOs at the dose of 135 μmol Fe per kg, the head–neck vasculatures and abdominal artery of rabbits and the coronary artery of pigs could be clearly visualized by T1-weighted MRI. Moreover, PAA@USPIOs were highly tolerable by animals. Our study indicated that the procedure developed in this study has great promise for large-scale production of USPIOs for T1-weighted MRI for future clinical applications.

Bismuth- and gadolinium-codoped carbon quantum dots with red/green dual emission for fluorescence/CT/T1-MRI mode imaging

Qin Meng,Yun Wang,Chunxing Li,Xiaoxi Hu

Carbon quantum dots (CQDs) with good biosafety can be doped with metals to construct multimodal imaging probes with a wide range of applications in fluorescence and molecular imaging. However, developing CQDs with both long-wavelength emission and a variety of imaging modes remains a challenge. In this article, novel bismuth- and gadolinium-codoped carbon quantum dots (Bi,Gd-CQDs) were developed using a hydrothermal method for green and red fluorescence imaging, X-ray computed tomography (CT) imaging, and T1-weighted magnetic resonance imaging (T1-weighted MRI). In addition to exceptional water solubility, strong steady fluorescence characteristics, and great biocompatibility, the obtained Bi,Gd-CQDs have a high X-ray attenuation coefficient and a short magnetic resonance T1 relaxation time. Additionally, cell imaging involved red fluorescence imaging with deep penetration. Therefore, the prepared Bi,Gd-CQDs with green and red dual-emissions can be used as nanoprobes for fluorescence, CT, and T1-weighted MR imaging, expanding their application prospects in the field of biomedicine.

Black TiO2-based nanoprobes for T1-weighted MRI-guided photothermal therapy in CD133 high expressed pancreatic cancer stem-like cells

Siqi Wang,Wenzhi Ren,Jianhua Wang,Zhenqi Jiang,Madiha Saeed,Lili Zhang,Aiguo Li,Aiguo Wu

At present, transmembrane glycoprotein CD133 highly expressed pancreatic cancer stem cells (PCSCs), with the features of chemotherapeutic/radiotherapeutic resistance and exclusive tumorigenic potential, are considered as the primary cause of metastasis and recurrence in pancreatic cancer, and therefore are an effective target in the disease treatment. Furthermore, with the launch of precision medicine, multifunctional nanoprobes have been applied as an efficient strategy for the magnetic resonance imaging (MRI)-guided photothermal therapy (PTT) of pancreatic cancer. In this research, with the aim of achieving precise MRI-guided PTT in CD133 highly expressed PCSCs, novel bTiO2-Gd-CD133mAb nanoprobes were designed and successfully prepared by loading Gd-DOTA and CD133 monoclonal antibodies on black TiO2 nanoparticles. It was very interesting to find that the r1 relaxivity value of the nanoprobes was 34.394 mM−1 s−1, about 7.5 times that of commercial Magnevist (4.5624 mM−1 s−1), which indicates that the nanoprobes have good potential as MRI T1 contrast agents with excellent performance. Herein, CD133 highly expressed PANC-1 cells were selected and verified as PCSCs model. In vitro experiments demonstrated that the nanoprobes exhibited active-targeting ability in PANC-1 cells, and consequently could specially enhance T1-weighted MR imaging and 808 nm near-infrared (NIR)-triggered PTT efficiency in the PCSCs model. Our study not only provides a new strategy for the effective treatment of pancreatic cancer and its’ stem cells, but also further broadens the application of black TiO2 in the field of cancer theranostics.

Single Molecular Wells–Dawson-Like Heterometallic Cluster for the In Situ Functionalization of Ordered Mesoporous Carbon: A T 1- and T 2-Weighted Dual-Mode Magnetic Resonance Imaging Agent and Drug Delivery System

Qianqian Zhang,Peiyuan Wang,Yun Ling,Xiaomin Li,Lixue Xia,Yongtai Yang,Xiaofeng Liu,Fan Zhang,Yaming Zhou

Developing a feasible way to prepare highly dispersed heterometallic nanoparticles incorporated in porous carbon composites is of significant importance for multifunctional materials. In this work, heterometallic γ-Fe 2 O 3 and GdPO 4 nanoparticles that are incorporated in ordered mesoporous carbon composites are facilely prepared by a one-pot in situ method using a Wells–Dawson-like cluster of [Fe 6 Gd 6 (μ 3 -O) 2 (CO 3 )(O 3 PPh) 6 (O 2 C t Bu) 18 ] ({Fe 6 Gd 6 P 6 } for short) as the precursor. It is verified that the γ-Fe 2 O 3 and GdPO 4 nanoparticles are highly dispersed and embedded into the carbon matrix with a particle size smaller than 5 nm, even when the carbon matrix is changed from 2D hexagonal P6mm to 3D body-centered cubic Im-3m symmetry. Additionally, a formation mechanism is proposed. Furthermore, dual-mode magnetic resonance (MR) imaging and drug carrier properties are evaluated by in vitro experiments, which show a satisfactory T 1 - and T 2 -weighted MR imaging effect with r 1 and r 2 relaxivity values of 2.7 and 183.7 mM −1 s −1 , respectively, and doxorubicin hydrochloride carrier amount of 102 mg g −1 , identifying a combined function for potential diagnostic and therapy.

Gadolinium-Based MRI Contrast Agent for Targeted Imaging of Vascular Endothelial Growth Factor Receptor-2-Expressing Tumors

Tu Rong,You Xiaoguang,Ding Zhichao,Wu Huanyu,Zhao Linlu,Qiu Zhi,Wei Junjie,Yang Xiaofeng

Development of imaging methods that can detect target molecules will be of great significance for non-invasive molecular diagnosis. Vascular endothelial growth factor receptor-2 (VEGFR-2) is a broadly expressed target, and VEGFR-2-specific agents have been used in biological therapy. In this study, amine-reactive coupling was used to label the polypeptide VEGF125–136 with a Gd(DOTA) complex to create a magnetic resonance contrast agent, VEGF125–136-Gd, which binds to VEGFR-2. Using T1-weighted magnetic resonance imaging, we explored the tumor cell-targeting ability of VEGF125–136-Gd and its enhancement of T1-weighted image intensity using human hepatoma (HepG2) cells and found that the signal-to-noise ratio (SNR) increased commensurately with the concentration of VEGF125–136-Gd. At a concentration of 2 mM, the SNR produced by VEGF125–136-Gd was 5.4-fold higher than that produced by a Gd-labelled non-targeting polypeptide (NTPP-Gd) control. We also evaluated the tumor-targeting efficiency of VEGF125–136-Gd in nude mice injected with human hepatoma (HepG2) cells. Preliminary in vivo imaging of VEGFR-2 in tumor tissue revealed a targeting peak 60 min post administration of VEGF125–136-Gd, the intensity of which was 23% higher than the SNR of NTPP-Gd at the same time point. Our findings may help lay the foundation for clinical applications of VEGF125–136-Gd in targeted imaging.

A Contrast-Enhanced Tri-Modal MRI Technique for High-Performance Hypoxia Imaging of Breast Cancer

Quanliang Mao,Mengyin Gu,Chengyuan Hong,Huiying Wang,Xinzhong Ruan,Zhusheng Liu,Bo Yuan,Mengting Xu,Chen Dong,Lei Mou,Xiang Gao,Guangyu Tang,Tianxiang Chen,Aiguo Wu,Yuning Pan

Personalized radiotherapy strategies enabled by the construction of hypoxia-guided biological target volumes (BTVs) can overcome hypoxia-induced radioresistance by delivering high-dose radiotherapy to targeted hypoxic areas of the tumor. However, the construction of hypoxia-guided BTVs is difficult owing to lack of precise visualization of hypoxic areas. This study synthesizes a hypoxia-responsive T 1 , T 2 , T 2 mapping tri-modal MRI molecular nanoprobe (SPION@ND) and provides precise imaging of hypoxic tumor areas by utilizing the advantageous features of tri-modal magnetic resonance imaging (MRI). SPION@ND exhibits hypoxia-triggered dispersion-aggregation structural transformation. Dispersed SPION@ND can be used for routine clinical BTV construction using T 1 -contrast MRI. Conversely, aggregated SPION@ND can be used for tumor hypoxia imaging assessment using T 2 -contrast MRI. Moreover, by introducing T 2 mapping, this work designs a novel method (adjustable threshold-based hypoxia assessment) for the precise assessment of tumor hypoxia confidence area and hypoxia level. Eventually this work successfully obtains hypoxia tumor target and accurates hypoxia tumor target, and achieves a one-stop hypoxia-guided BTV construction. Compared to the positron emission tomography-based hypoxia assessment, SPION@ND provides a new method that allows safe and convenient imaging of hypoxic tumor areas in clinical settings.
