Based on the properties of photodynamic therapy (PDT) and magnetic resonance imaging (MRI), combining them to construct multifunctional nanotheranostics can leverage strengths and avoid weaknesses for tumor diagnosis and treatment. However, certain problems remain unsolved, notably the short observation window caused by insufficient retention time. In this study, a GSH-responsive shape-transformable nanotheranostics Gd-Ce6-FFVLGGGC-SS-PEG are designed (abbreviated as GdCFS) by combining a single metal Gd with Ce6, and peptide (Phe-Phe-Val-Leu-Gly-Gly-Gly-Cys) disulfide-conjugated with polyethylene glycol (PEG) to perform dual-modal T 1 /T 2 MRI specifically at the tumor site. Due to its amphiphilic features, GdCFS can self-assemble to spherical nanoparticles, while transforming to nanofibers (NFs) in the presence of intracellular overexpressed GSH stimuli. After NFs formation, the hydrophobic core of GdCFS is exposed, resulting in T 1 positive contrast enhancement, whereas NFs can simultaneously achieve T 2 negative contrast enhancement. Furthermore, GSH depletion can cause imbalance in intracellular redox state, ultimately improving PDT efficacy. GdCFS benefits from the shape-transformation in terms of sufficient retention time, thus positively minimizing the toxicity risk. On the tumor model of nasopharyngeal carcinoma, in vivo and in vitro outcomes confirm that GdCFS can be a promising candidate for early diagnosis, real-time monitoring, and precise treatment of tumors with great biocompatibility.

This study reports a multifunctional core/shell nanoparticle (NP) that can be used for amplified magnetic resonance image (MRI), enhanced photothermal therapy (PTT) and magnetic hyperthermia therapy (MHT) due to its surface coating with a porous shell. Importantly, by means of introducing the surface coating of a porous shell, it helps entrap large quantities of water around NPs and allow more efficient water exchange, leading to greatly improved MR contrast signals. Besides, the porous shell helps the near-infrared (NIR) absorbance of the core, and then the extremely enhanced thermal effect can be obtained under synergistic combination of PTT and MHT. By synthesizing multifunctional porous MnFe2O4/PB as an example, we found that the transversal relaxivity (r2) of MnFe2O4 NPs might improve from 112.11 to 123.46 mM−1 s−1, and the specific absorption rate (SAR) of MnFe2O4/PB nanoparticles reached unprecedented levels of up to 4800 W g−1 compared with the SAR 1182 W g−1 of PTT under an 808 nm laser and 180 W g−1 of MHT under an external AC magnetic field. Meanwhile, when MnFe2O4 was decorated on PB nanoparticles, the magnetic properties became lower slightly, but the synergistic photothermal/magnetic hyperthermia conversion was enhanced greatly. Subsequently, in vitro T1–T2 dual-modal MRI, PTT and MHT results verified that MnFe2O4/PB could serve as an excellent MRI/PTT/MHT theranostic agent. Furthermore, the MnFe2O4/PB NPs were applied as a T1–T2 dual-modal MRI, PTT and MHT theranostic agent for in vivo MRI-guided photothermal and magnetic hyperthermia ablation of tumors by intratumoral injection in 4T1 tumor-bearing mice. The T1–T2 dual-modal MR imaging result shows a significantly contrast in the tumor site. The MPB-mediated PTT and MHT result shows high therapeutic efficiency as a result of high photothermal and magnetic hyperthermia conversion efficiency. The multifunctional NPs have a great potential application for future clinical tumorous diagnosis and treatment.

Emerging nanotechnologies in cancer therapy increasingly highlight the importance of integrating therapeutic and diagnostic functions into a single platform for enhanced efficacy and real-time monitoring. Here, we present a vortex‐shaped nanoring Fe 3 O 4 @MnO 2 @PDA (polydopamine) engineered for dual‐mode T 1 – T 2 magnetic resonance imaging (MRI)‐guided, dual‐thermal, and Fenton‐like chemodynamic therapy (CDT). The ring‐shaped nano Fe 3 O 4 core provides magnetothermal hyperthermia, while the MnO 2 layer, acting as a responsive nanozyme, catalyzes endogenous H 2 O 2 to generate oxygen and bolster CDT. Meanwhile, the PDA coating efficiently absorbs near‐infrared light, amplifying photothermal effects for tumor ablation. Under dual‐heating conditions, the temperature rise rate (2.05–2.92 times) and final equilibrium temperatures (1.57–2.27 times) exceed those of single‐mode heating, underscoring a robust synergistic effect. Additionally, due to the shell’s shielding effect weakening T 2 signals and the tumor microenvironment enhancing T 1 relaxation, the T 1 (5.75 Mm −1 s −1 ) to T 2 (91.56 mM −1 s −1 ) ratio is optimized specifically at tumor sites, thus achieving effective dual‐modal MRI contrast. Moreover, the smart MnO 2 design not only furnishes dynamic imaging enhancements but also enables environment‐triggered Mn 2+ release, offering further diagnostic and therapeutic benefits. In vitro and in vivo evaluations confirm significant tumor suppression with minimal systemic toxicity. These findings position nanoring Fe 3 O 4 @MnO 2 @PDA as a promising dual‐mode MRI‐guided, dual‐thermal, and chemodynamic platform for advanced cancer therapy.

Biodegradable Two-dimensional (2D) nanomaterials have attracted tremendous interest in biomedical application. Herein, a biodegradable ferrous sulfide nanosheet with ultrathin structure and uniform size was developed. Following its development, the application of the self-enhanced photothermal nanoplatform (FMPN) in vivo by combining Mn-DOTA and dimeric PD-L1 affibody for targeted dual-mode magnetic resonance imaging (MRI) guided synergistic tumor therapy is explored. Dual-contrast enhanced subtraction imaging (DESI) was used to evaluate the excellent T1-T2 MRI ability of FMPN. FMPN displayed excellent photothermal conversion efficiency (η = 40.33 %) and biodegradability, thus had good tumor photothermal therapy effects with good biosafety. In addition, FMPN with pH-responsive Fe 2+ release can effectively induce tumor cell ferroptosis, further intensifying PTT and subsequent immunologic cell death. PD-L1 affibody could endow this nanoplatform with efficient tumor targeting, terminating T cells’ immune suppression by effectively blocking the interaction between PD-1 and PD-L1. Systemic delivery of FMPN significantly inhibited tumor progression in primary, metastatic and abscopal tumors, and showed obvious dual-mode MRI enhancement. Therefore, this nanoplatform exhibited promising potential for MRI-guided synergistic ferroptosis/photothermal/immunotherapy.
