An ultra-sensitive T2-weighted MR contrast agent based on Gd3+ ion chelated Fe3O4 nanoparticles

Jing Chen,Hui-Hui Xiang,Zu-Zhi Zhao,Yun-Kai Wu,Meng-Yu Fei,Meng-Meng Song

An ultra-sensitive T2-weighted MR imaging contrast agent was prepared based on Fe3O4 nanoparticles and Gd3+ ions (Fe3O4@Gd). Amino modified Fe3O4 nanoparticles were conjugated to diethylenetriamine pentaacetic acid, and finally coordinated with Gd3+ ions. The nanoparticles had a uniform morphology with a size of 100 nm and a Gd/Fe mass ratio of 1/110. The r2 (transverse relaxivity) of the Fe3O4 nanoparticles increased from 131.89 mM−1 s−1 to 202.06 mM−1 s−1 after coordination with Gd3+ ions. MR measurements showed that the aqueous dispersion of Fe3O4@Gd nanoparticles had an obvious concentration-dependent negative contrast enhancement. Hepatoma cells were selected to test the cytotoxicity and MR imaging effect. The application of Fe3O4@Gd nanoparticles as contrast agents was also exploited in vivo for T2-weighted MR imaging of rat livers. All the results showed the effectiveness of the nanoparticles in MR diagnosis.

Magnetite nanocluster and paclitaxel-loaded charge-switchable nanohybrids for MR imaging and chemotherapy

Lingjie Wu,Ming Wu,Xinyi Lin,Xiaolong Zhang,Xiaolong Liu,Jingfeng Liu

Highly efficient accumulation of nanoscaled theranostic agents in a tumor site is crucial for cancer diagnosis and therapy. In this study, we designed a drug-loaded charge-switchable nanohybrid system (HNPs–DA) triggered by the low pH value of tumor microenvironment (pH 6.5) to enhance the uptake efficiency of NPs in cancer cells; the nanohybrid could exhibit T2-MR imaging enhancement and chemotherapy ability, ascribed to the loaded magnetite nanocluster (MNC) and paclitaxel (PTX), respectively. The HNPs–DA comprises two distinct functional components: (1) a biocompatible amphiphilic polymer (Pluronic F127) to act as a nanovehicle for MNC and PTX after self-assembly in an aqueous solution; and (2) a hydrophilic polymeric shell derived from stearoyl-polyethylenimine-2,3-dimethylmalefic anhydride (SC-g-PEI-DMMA). SC-g-PEI-DMMA switches the surface charge of the HNPs–DA from negative to positive by diminishing the anionic part of DMMA and sequentially recovering the cationic instinct of the PEI part at pH 6.5, which could facilitate the cellular uptake and therefore enhance the theranostic effects. In vitro studies demonstrated a darker T2-MRI image in the HNPs–DA-treated HepG2 cells at pH 6.5 as compared to that at pH 7.4. Moreover, CCK8 assay indicated that HNPs–DA exhibited a much higher cytotoxicity against HepG2 cells at pH 6.5, and flow cytometric analysis suggested that the cell death induced by HNPs–DA occurred via apoptosis, which was detected by Annexin V antibody and propidium iodide staining.

Manganese and dysprosium codoped carbon quantum dots as a potential fluorescent/T1/T2/CT quadri-modal imaging nanoprobe

Chunxing Li,Yun Wang,Huayan Nong,Xiaoxi Hu,Yu Wu,Yanjun Zhang,Caimei Liang,Keguang Chen,Songliang Li

It has been challenging to integrate various medical imaging modalities into an ultra-small nanoparticle with good biocompatibility to build highly efficient multimodal imaging nanoprobes. A new manganese and dysprosium codoped carbon quantum dots (Mn,Dy-CQDs) with a mean diameter of 1.77 nm was synthesized for fluorescence imaging, T1/T2-weighted magnetic resonance imaging (T1/T2-weighted MRI), and x-ray computed tomography (CT) imaging using a simple one-step hydrothermal approach. The obtained Mn,Dy-CQDs showed good water solubility, long-term stability, strong stable fluorescence property (fluorescence quantum yield of 31.62%), and excellent biocompatibility. The cell imaging verified that the Mn,Dy-CQDs have high efficiency of fluorescence imaging. The Mn,Dy-CQDs, on the other hand, had a superior x-ray absorption performance (47.344 HU l g−1), a higher longitudinal relaxivity (r1 = 7.47 mM−1 s−1), a higher transverse relaxivity (r2 = 42.686 mM−1 s−1). In vitro T1/T2-weighted MRI and CT imaging showed that Mn,Dy-CQDs can produce a strong contrast enhancement impact. To summarise, the Mn,Dy-CQDs may be used as a T1/T2-weighted MRI/CT/fluorescent quadri-modal imaging nanoprobe, indicating that they have a lot of uses in biomedical multimode imaging and clinics.

Core@shell Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles for triple-modality T1/T2-weighted MRI and NIR-to-NIR upconversion luminescence imaging agents

Yang Luo,Sinan Du,Wei Zhang,Zhengfang Liao,Fang Zuo,Shengtao Yang

Core@shell structures of Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles (NPs) with multifunctional properties were prepared using a hydrothermal route with a seed-growth procedure. The effect of Mn2+ ions on the phase, shape, and upconversion luminescence (UCL) of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was explored using X-ray diffraction, transmission electron microscopy, dynamic light scattering, and photoluminescence spectroscopy. Furthermore, hydrophobic to hydrophilic surface modification of the Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was achieved by coating the NPs with an amphiphilic polymer (poly(maleic anhydride-alt-1-octadecene) (C18PMH)) modified with amine-functionalized methyl ether poly(ethylene glycol). Then, the NIR-to-NIR UCL ascribed to the Tm3+ ions, T1-weighted MRI ascribed to the Mn2+ ions, and T2-weighted MRI ascribed to the Fe3O4 core of the hydrophilic Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs were evaluated. The results indicate that Mn2+ doping is an effective method to control the size/shape and improve the UCL intensity of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs, which are promising as imaging agents for NIR-to-NIR UCL and T1/T2-weighted MRI in biomedical research.

Single Molecular Wells–Dawson-Like Heterometallic Cluster for the In Situ Functionalization of Ordered Mesoporous Carbon: A T 1- and T 2-Weighted Dual-Mode Magnetic Resonance Imaging Agent and Drug Delivery System

Qianqian Zhang,Peiyuan Wang,Yun Ling,Xiaomin Li,Lixue Xia,Yongtai Yang,Xiaofeng Liu,Fan Zhang,Yaming Zhou

Developing a feasible way to prepare highly dispersed heterometallic nanoparticles incorporated in porous carbon composites is of significant importance for multifunctional materials. In this work, heterometallic γ-Fe 2 O 3 and GdPO 4 nanoparticles that are incorporated in ordered mesoporous carbon composites are facilely prepared by a one-pot in situ method using a Wells–Dawson-like cluster of [Fe 6 Gd 6 (μ 3 -O) 2 (CO 3 )(O 3 PPh) 6 (O 2 C t Bu) 18 ] ({Fe 6 Gd 6 P 6 } for short) as the precursor. It is verified that the γ-Fe 2 O 3 and GdPO 4 nanoparticles are highly dispersed and embedded into the carbon matrix with a particle size smaller than 5 nm, even when the carbon matrix is changed from 2D hexagonal P6mm to 3D body-centered cubic Im-3m symmetry. Additionally, a formation mechanism is proposed. Furthermore, dual-mode magnetic resonance (MR) imaging and drug carrier properties are evaluated by in vitro experiments, which show a satisfactory T 1 - and T 2 -weighted MR imaging effect with r 1 and r 2 relaxivity values of 2.7 and 183.7 mM −1 s −1 , respectively, and doxorubicin hydrochloride carrier amount of 102 mg g −1 , identifying a combined function for potential diagnostic and therapy.

A Contrast-Enhanced Tri-Modal MRI Technique for High-Performance Hypoxia Imaging of Breast Cancer

Quanliang Mao,Mengyin Gu,Chengyuan Hong,Huiying Wang,Xinzhong Ruan,Zhusheng Liu,Bo Yuan,Mengting Xu,Chen Dong,Lei Mou,Xiang Gao,Guangyu Tang,Tianxiang Chen,Aiguo Wu,Yuning Pan

Personalized radiotherapy strategies enabled by the construction of hypoxia-guided biological target volumes (BTVs) can overcome hypoxia-induced radioresistance by delivering high-dose radiotherapy to targeted hypoxic areas of the tumor. However, the construction of hypoxia-guided BTVs is difficult owing to lack of precise visualization of hypoxic areas. This study synthesizes a hypoxia-responsive T 1 , T 2 , T 2 mapping tri-modal MRI molecular nanoprobe (SPION@ND) and provides precise imaging of hypoxic tumor areas by utilizing the advantageous features of tri-modal magnetic resonance imaging (MRI). SPION@ND exhibits hypoxia-triggered dispersion-aggregation structural transformation. Dispersed SPION@ND can be used for routine clinical BTV construction using T 1 -contrast MRI. Conversely, aggregated SPION@ND can be used for tumor hypoxia imaging assessment using T 2 -contrast MRI. Moreover, by introducing T 2 mapping, this work designs a novel method (adjustable threshold-based hypoxia assessment) for the precise assessment of tumor hypoxia confidence area and hypoxia level. Eventually this work successfully obtains hypoxia tumor target and accurates hypoxia tumor target, and achieves a one-stop hypoxia-guided BTV construction. Compared to the positron emission tomography-based hypoxia assessment, SPION@ND provides a new method that allows safe and convenient imaging of hypoxic tumor areas in clinical settings.

Structural and Molecular Fusion MRI Nanoprobe for Differential Diagnosis of Malignant Tumors and Follow-Up Chemodynamic Therapy

Weitao Yang,Cuijun Deng,Xiudong Shi,Yan Xu,Chenyu Dai,Hui Wang,Kexin Bian,Tianming Cui,Bingbo Zhang

Enhanced imaging techniques using contrast agents enable high-resolution structural imaging to reveal space-occupying lesions but rarely provide detailed molecular information. To this end, we report a structural and molecular fusion magnetic resonance imaging (MRI) nanoprobe for differential diagnosis between benign and malignant tumors. This fusion nanoprobe, termed FFT NPs, follows a working mechanism involving a T1-/T2-weighted magnetic resonance tuning effect (MRET) between a magnetic Fe3O4 core and a paramagnetic Fe-tannic acid (Fe-TA) shell. The FFT NPs with an “always-on” inert T2 signal provide structural MRI (sMRI) contrast of tumors while affording an activated T1 signal in the presence of ATP, which is overproduced during the rapid growth of malignant tumors to enable molecular MRI (mMRI) of tumor lesions. We propose the use of the ratiometric mMRI:sMRI intensity to assist in the differential diagnosis of malignant 4T1 tumors from benign L929 fibroblast tumors. Furthermore, the dissociated FFT NPs were found to be able to catalyze H2O2 conversion in 4T1 tumors to generate excess reactive oxygen species (ROS) for chemodynamic therapy. The described fusion nanoprobe strategy enables the differential diagnosis of tumors from a combined spatial and molecular perspective with one-stop MRI imaging with potential applications in precision intervention.

Ultrasmall Fe3O4 nanoparticles self-assembly induced dual-mode T1/T2-weighted magnetic resonance imaging and enhanced tumor synergetic theranostics.

Qinghua Xie,Xuemei Wang,Gaorui Zhang,Dawei Zhou,Yuxuan Zhao,Hong Liu,Jiazhi Duan,Dexin Yu,Yuanhua Sang

Individual theranostic agents with dual-mode MRI responses and therapeutic efficacy have attracted extensive interest due to the real-time monitor and high effective treatment, which endow the providential treatment and avoid the repeated medication with side effects. However, it is difficult to achieve the integrated strategy of MRI and therapeutic drug due to complicated synthesis route, low efficiency and potential biosafety issues. In this study, novel self-assembled ultrasmall Fe 3 O 4 nanoclusters were developed for tumor-targeted dual-mode T 1 /T 2 -weighted magnetic resonance imaging (MRI) guided synergetic chemodynamic therapy (CDT) and chemotherapy. The self-assembled ultrasmall Fe 3 O 4 nanoclusters synthesized by facilely modifying ultrasmall Fe 3 O 4 nanoparticles with 2,3-dimercaptosuccinic acid (DMSA) molecule possess long-term stability and mass production ability. The proposed ultrasmall Fe 3 O 4 nanoclusters shows excellent dual-mode T 1 and T 2 MRI capacities as well as favorable CDT ability due to the appropriate size effect and the abundant Fe ion on the surface of ultrasmall Fe 3 O 4 nanoclusters. After conjugation with the tumor targeting ligand Arg-Gly-Asp (RGD) and chemotherapy drug doxorubicin (Dox), the functionalized Fe 3 O 4 nanoclusters achieve enhanced tumor accumulation and retention effects and synergetic CDT and chemotherapy function, which serve as a powerful integrated theranostic platform for cancer treatment.

Plasmid-loadable magnetic/ultrasound-responsive nanodroplets with a SPIO-NP dispersed perfluoropentane core and lipid shell for tumor-targeted intracellular plasmid delivery

Wei Dong,Anqi Huang,Jixiu Huang,Pengying Wu,Shifang Guo,Huasheng Liu,Mengfan Qin,Xinxing Yang,Bo Zhang,Mingxi Wan,Yujin Zong

Using ultrasound activating contrast agents to induce sonoporation is a potential strategy for effective lesion-targeted gene delivery. Previous reports have proven that submicron nanodroplets have a better advantage than microbubbles in that they can pass through tumor vasculature endothelial gaps by passive targeting; however, they cannot achieve an adequate dose in tumors to facilitate ultrasound-enhanced gene delivery. Additionally, a few studies focused on delivering macromolecular genetic materials (i.e. overexpression plasmid and CRISPR plasmid) have presented more unique advantages than small-molecular genetic materials (i.e. miRNA mimics, siRNA and shRNA etc.), such as enhancing the expression of target genes with long-term effectiveness. Thereby, we constructed novel plasmid-loadable magnetic/ultrasound-responsive nanodroplets, where superparamagnetic iron oxide nanoparticle dispersed perfluoropentane was encapsulated with lipids to which plasmids could be adhered, and branched polyethylenimine was used to protect the plasmids from enzymolysis. Furthermore, in vitro and in vivo studies were performed to verify the magnetic tumor-targeting ability of the plasmid-loadable magnetic/ultrasound-responsive nanodroplets and focused ultrasound enhanced intracellular plasmid delivery. The plasmid-loadable magnetic/ultrasound-responsive nanodroplets, carrying 16–19 plasmids per droplet, had desirable diameters less than 300 nm, and integrated the merits of excellent magnetic targeting capabilities and phase transition sensitivity to focused ultrasound. Under programmable focused ultrasound exposure, the plasmid-loadable magnetic/ultrasound-responsive nanodroplets underwent a phase-transition into echogenic microbubbles and the subsequent inertial cavitation of the microbubbles achieved an ∼40% in vitro plasmid delivery efficiency. Following intravenous administration, T2-weighted magnet resonance imaging, scanning electron microscopy and inductively coupled plasma optical emission spectrometry of the tumors showed significantly enhanced intratumoral accumulation of the plasmid-loadable magnetic/ultrasound-responsive nanodroplets under an external magnetic field. And a GFP ELISA assay and immunofluorescence staining indicated that focused ultrasound-induced inertial cavitation of the plasmid-loadable magnetic/ultrasound-responsive nanodroplets significantly enhanced the intracellular delivery of plasmids within the tumor after magnet-assisted accumulation, while only lower GFP levels were observed in the tumors on applying focused ultrasound or an external magnet alone. Taken together, utilizing the excellent plasmid-loadable magnetic/ultrasound-responsive nanodroplets combined with magnetism and ultrasound could efficiently deliver plasmids to cancer cells, which could be a potential strategy for macromolecular genetic material delivery in the clinic to treat cancer.
