Metal-organic frameworks (MOFs) are crystalline porous materials with tunable structures, where metal ions or clusters serves as magnetic centers and organic ligands offer spatial separation. These characteristics, combined with their diverse topologies, make MOFs promising candidates for contrast agents (CAs) in magnetic resonance imaging (MRI). Herein we synthesized four MOFs based on the same triangular Fe3O clusters with different topologies: MIL-101(Fe) (mtn net), MIL-100(Fe) (moo net), MIL-59(Fe) (pcu net), and MIL-88B(Fe) (acs net). To clarify the relationship between topologies and T2 relaxivities, the MOFs were tailored into uniform, nanoscale spherical morphologies. Notably, the value of T2 relaxivity for MIL-88B(Fe) with acs topology is nearly three times of that for MIL-101(Fe) with mtn topology at 7.0 T. By comparing with the magnetic properties of Fe3O molecular clusters and Ga-doped MIL-88B(Fe), our analysis demonstrated the significant advantage of MOFs with fixed arrays, adjustable component and diverse topologies in enhancing magnetic relaxation. Cellular MRI experiments further revealed that MIL-88B(Fe) could differentiate between M1 and M2 macrophages, highlighting its potential for monitoring tumor progression. These findings offer valuable insights into how MOF topology can be strategically utilized to enhance T2 relaxivities for MRI applications.

Improving the relaxivity of magnetic resonance imaging (MRI) contrast agents is an important challenge for cancer theranostics. Herein we report the design, synthesis, characterization, theoretical analysis and in vivo tests of a superparamagnetic polymersome as a new MRI contrast agent with extremely high T 2 relaxivity (611.6 mM −1 s −1 ). First, a noncytotoxic cancer-targeting polymersome is synthesized based on a biodegradable diblock copolymer , folic acid-poly( l -glutamic acid)- block -poly( ε -caprolactone) [FA-PGA- b -PCL]. Then, ultra-small superparamagnetic iron oxide nanoparticles (SPIONs) are in situ generated in the hydrophilic PGA coronas of polymersomes to afford magnetic polymersomes. The in vivo MRI assay revealed prominent negative contrast enhancement of magnetic polymersomes at a very low Fe dose of 0.011 mmol/kg. Moreover, this cancer-targeting magnetic polymersome can effectively encapsulate and deliver anticancer drug to inhibit the tumor growth, demonstrating promising theranostic applications in biomedicine .

Superparamagnetic iron oxide nanoparticles (SPIONs) have a history of clinical use as contrast agents in T2 weighted MRI, though relatively low T2 relaxivity has caused them to fall out of favor as new faster MRI techniques have gained prominence. We demonstrate that SPIONs coated with a monolayer of succinylated heparin (Su-HP-SPIONs) exhibit over four-fold increased T2 relaxivity (460 mM−1 s−1) as compared to the clinically approved SPION-based contrast agent Feridex (98.3 mM−1 s−1) due to greatly increased water interaction from increased hydrophilicity and thinner coating as supported by our proposed parametric model. In vivo, the performance increase of the Su-HP-SPIONs in T2 MRI imaging of xenograft tumors is ten-fold that of our in-house synthesized Feridex analogue, due to better tumor localization from the smaller size imparted by the thinner coating. In addition to these significantly improved magnetic properties, the succinylated heparin coating also exhibits favorable synthetic reproducibility, solution stability, and biocompatibility. These findings demonstrate the untapped potential of SPIONs as possible high performance clinical T2 contrast agents.

Magnetosonoporation (MSP) is a relatively safe and efficient approach for instant MR stem cell labeling. In this study, the physical and magnetic properties of different formulations of synthesized superparamagnetic iron oxide nanoparticles (SPION) were characterized. Then, a “closed”
MSP apparatus using focused ultrasound was designed and the feasibility of MSP stem cell labelling using focused ultrasound was validated by evaluating the proliferation, migration and differentiation of the magnetically labeled cells. Subsequently, MSP/SPION labeled neural stem cells (NSCs)
were transplanted into the contralateral striatum of glioma-bearing nude mice, and their migration was monitored using magnetic resonance imaging (MRI) in vivo. The results indicated that SPION-1 with the largest size (28.43±9.55 nm) had the highest T
2 relaxivity
(136.62 Fe mM−1 S−1) and the best MRI contrast effect. Without additional transfection reagents, NSCs were labeled with SPION using focused ultrasound in vitro and the safety of MSP stem cell labeling was validated with the optimized MSP technique. Finally,
confirmed by histological evaluation, pronounced signal attenuation on T
2-weighted images demonstrated the intracranial tumor tropism of NSCs could be monitored non-invasively by MRI. In conclusion, MSP cell labeling using focused ultrasound is a promising technique and the
“closed” device is feasible, convenient and safe for instant magnetic stem cell labeling and MRI cell tracking.

Highly stable and multifunctional fluorescent quantum dots are particularly attractive in practical applications. Here, a new kind of ultra-small-sized silicon quantum dot-gadolinium (SiQD-Gd) was successfully fabricated by a newly-designed facile hydrothermal growth and chelating method. The obtained SiQD-Gd exhibited outstanding water dispersibility, stability and good fluorescent property with the quantum yield of 11.6%. SiQD-Gd displayed a low cytotoxicity in normal cell lines (HELF, HEK293F) and tumor cell lines (H1299, A549). Meanwhile, SiQD-Gd showed excellent magnetic resonance response with r 1 relaxation rate of 10.5 mmol L −1 ·s −1 and r 2 relaxation rate of 47.5 mmol L −1 ·s −1 , which are 2.5 and 7.4 times enhanced comparing to that of the commercial MR agent Magnevist. In vivo studies showed significant contrast enhancement effect of its T 1 - and T 2 -weighted MR imaging. In addition, in vivo fluorescent imaging for mice and zebrafish indicated its potential applications in fluorescent tracking. Thus, the excellent multimodal imaging capacity and biocompatibility of SiQD-Gd make it a potential imaging agent for clinic applications.
