Accumulation of microplastics and Tcep pollutants in agricultural soil: Exploring the links between metabolites and gut microbiota in earthworm homeostasis

Jing Cao,Qian Wang,Yumeng Lei,Xiaofeng Jiang,Mei Li

Agricultural soil contamination with plastic film has become a critical global environmental problem, requiring greater research on the possible occurrence and biological risk of microplastics (MPs) and their additives in soil ecosystems. The presence of MPs and tris (2-chloroethyl) phosphate (Tcep) in agricultural soil was investigated at nine sites in the present study. Polyethylene MPs (PE-MPs) and Tcep were found at all nine sites. To study co-exposure effects on soil microbiota and earthworms, and to mimic a realistic exposure scenario, 0.05 % (w/w) PE-MPs with three particle size ranges were combined with Tcep (1.0 mg/kg). After 28 days of exposure, there was no indication that added PE-MPs and/or Tcep significantly affected the soil microbial community structure. In earthworms, size-selective intake, digestion and egestion of PE-MPs may occur, with Tcep co-exposure affecting the residual Tcep concentration in earthworm intestines (3.52–9.31 μg/g dw). Long-term earthworm PE-MPs intake caused intestinal damage, and Tcep co-exposure increased oxidative stress, thereby influencing their feeding behavior and growth, resulting in weight loss (3.42 %–14.96 %), especially for the most common PE-MPs sizes (0–300 μm). High performance liquid chromatography-mass spectrometry (LC-MS) was used for metabolomic analysis, revealing the significant up-regulation of citrate ( p  < 0.001) and down-regulation of l -glutamate ( p  < 0.05) in co-exposure groups. Co-exposure resulted in the alteration of most metabolic pathways, thereby impairing nervous, digestive and excretory systems in the earthworm, with an associated decrease in amino acid metabolism and changes in tricarboxylic acid (TCA) cycle intermediates. Gut microbiota, such as Proteobacteria ( Verminephrobacter and Bradyrhizobium ) and Firmicutes ( Bacillus ), are critically important in maintaining earthworm metabolic homeostasis, particularly for the TCA cycle and amino acid metabolism. Overall, MPs and Tcep co-exposure in agricultural soil enhanced their toxicity to earthworms and may potentially endanger the development of agricultural sustainability.

Effects of TCPP and TCEP exposure on human corneal epithelial cells: Oxidative damage, cell cycle arrest, and pyroptosis

Zhen-Ning Zhang,Dan-Lei Yang,Hai Liu,Jue Bi,Ya-Bo Bao,Jiao-Yang Ma,Qin-Xiang Zheng,Dao-Lei Cui,Wei Chen,Ping Xiang

Tris(2-chloroisopropyl) phosphate (TCPP) and Tris(2-chloroethyl) phosphate (TCEP) are the widely used organophosphorus flame retardants indoors and easily accessible to the eyes as the common adhesive components of dust and particle matter, however, hardly any evidence has demonstrated their corneal toxicity. In this study, the adverse effects of TCPP, TCEP, and TCPP + TCEP exposure on human corneal epithelial cells (HCECs) were investigated. The cell viability and morphology, intracellular reactive oxygen species (ROS), cell cycle, and the expressions of cell cycle and pyroptosis-related genes were assessed to explain the underlying mechanisms. Compared to individual exposure, co-exposure to TCPP 20 +TCEP 20 showed higher cytotoxicity with a sharp decrease of >30% in viability and more serious oxidative damage by increasing ROS production to 110.92% compared to the control group. Furthermore, the cell cycle arrested at the S phase (36.20%) was observed after combined treatment, evidenced by the upregulation of cyclin D1 , CDK2 , CDK4 , CDK6 , p21, and p27 . Interestingly, pyroptosis-related genes GSDMD , Caspase-1 , NLRP3 , IL-1β , IL-18 , NLRP1, and NLRC4 expressions were promoted with cell swelling and glowing morphology. Oxidative stress and cell cycle arrest probably acted as a key role in TCPP 20 +TCEP 20 -induced cytotoxicity and pyroptosis in HCECs. Our results suggested that TCPP 20 +TCEP 20 co-exposure induced severer corneal damage, further illustrating its significance in estimating indoor health hazards to humans.

Adsorption behavior of hierarchical porous biochar from shrimp shell for tris(2-chloroethyl) phosphate (TCEP): Sorption experiments and DFT calculations

Siyuan Chen,Haiming Cai,Xiaodong Du,Peiwen Wu,Xueqin Tao,Jiangmin Zhou,Zhi Dang,Guining Lu

Tris(2-chloroethyl) phosphate (TCEP) as a new type of flame retardant exists in various water environments, causing great risks to humans and the environment. In this study, shrimp shell was used to prepare an economical and environmental-friendly adsorbent for the efficient removal of TCEP. The systematic studies including characterization, removal performance, and adsorption mechanism of shrimp shell biochar toward TCEP were carried out. Adsorption kinetics and thermodynamics showed that fast equilibrium reached within 30 min, the maximum adsorption capacity q m was 108 μmol g −1 at 298 K, and the adsorption process is spontaneous and exothermic. The environmental factor, such as temperature, pH, inorganic anions and organic matter hardly affected the adsorption performance. Structural characterization indicated that the hierarchical porous structure of shrimp shell biochar is the key to excellent adsorption performance. The adsorption mechanisms were further revealed using density functional theory (DFT) calculations, and the hydrogen bond , van der Waals interactions, Cl–H interactions, and pi-H interactions were identified as potential interaction mechanisms between TCEP and specific biochar structures. The calculated binding energy between TCEP and simplified biochar structure suggested that oxygen-containing groups especially carboxyl, hydroxyl and aldehyde facilitate the adsorption. Our work not only provides a novel strategy for the quick remediation of organophosphate-contaminated water environments but also offers new opportunities for crustacean waste biomass valorization.

The isolated Ca-Nx sites in biochar boosting Fe catalyzed Fenton-like oxidation of Tris(2-chloroethyl) phosphate: Properties, mechanisms, and applications

Chenyu Yang,Qian Yao,Liangzhong Li,Xian Xiao,Lun Lu,Chang Liu,Chunyou Zhu,Sihui Zhan,Haoran Yuan

The biochar with single Ca atom (1.7 wt%) loaded nanoscale zero-valent iron composites (Fe@Ca-N-C) were prepared and effectively degraded tris(2-chloroethyl) phosphate (TCEP) via Fenton-like oxidation. Systematic experiments and theoretical calculations indicated that the enhanced removal of TCEP was attributed to the synergistic effect of adsorption (Ca-N x sites) and degradation (Fe sites). Remarkably, the optimal system of 0.1 g L −1 Fe@Ca-N-C, 1 mM peroxydisulfate (PDS) achieved 99.5 % TCEP degradation (2 mg L −1 ) in 30 min and maintained 100 % TCEP removal for 72 h in continuous-flow reaction column. The Ca-N x sites exhibited high adsorption energy for TCEP (especially the Ca-N 1 sites), and the Fe sites (Fe(0) and Fe(Ⅱ)) activated PDS into HO•, which quickly degrades the adsorbed TCEP through C-O bond breaking and C-Cl bond hydroxylation. This study provided insights into single Ca atom utilization and TCEP pollution remediation.

Reduction Modification of Macromolecules in Wool Fabric for Improving the Adhesion with UV-Curable MXene/PUA Conductive Coating

Yao Jiale,Xu Jiang,Cui Ao,Wang Rui,Hao Longyun

In this study, the molecules in wool fabric were modified to convert their disulfide bonds into sulfhydryl groups (–SH) by using tris (2-carboxyethyl) phosphine hydrochloride (TCEP) as a reducing agent. The XPS, SEM, and UV–visible absorption spectrum were utilized to track the modification process. MXene and UV-curable PUA resin were blended and printed on the modified wool substrates by screen printing way, followed by a sustainable UV irradiation to effectively fix the printed coating on them. The effects of TCEP on the wettability, electrical resistance of conductive coatings were investigated. Especially, the durability of the conductive coatings to mechanical operations, wet and temperature treatment were systematically evaluated by measuring the resistance change rate on unmodified and TCEP modified wool fabrics, respectively. The results indicated that TCEP modification of wool fabrics could significantly improve the durability and stability of conductive coatings. This was because the active –SH groups in modified wool could participate in the polymerization of PUA molecules into a film under the excitation of UV photo-initiators, greatly improving the integrity and toughness of its structure and ultimately resulting in better adhesion between conductive coatings and wool fabrics.

Physiologically based toxicokinetic modelling of Tri(2-chloroethyl) phosphate (TCEP) in mice accounting for multiple exposure routes

Jiaqi Ding,Wanyu He,Wanxiao Sha,Guoqiang Shan,Lingyan Zhu,Lin Zhu,Jianfeng Feng

Exposure routes are important for health risk assessment of chemical risks. The application of physiologically based toxicokinetic (PBTK) models to predict concentrations in vivo can determine the effects of harmful substances and tissue accumulation on the premise of saving experimental costs. In this study, Tri(2-chloroethyl) phosphate (TCEP), an organophosphate ester (OPE), was used as an example to study the PBTK model of mice exposed to different exposure doses by multiple routes. Different routes of exposure (gavage and intradermal injection) can cause differences in the concentration of chemicals in the organs. TCEP that enters the body through the mouth is mainly concentrated in the gastrointestinal tract and liver. However, the concentrations of chemicals that enter the skin into the mice are higher in skin, rest of body, and blood. In addition, TCEP was absorbed and accumulated very rapidly in mice, within half an hour after a single exposure. We have successfully established a mouse PBTK model of the TCEP accounting for multiple exposure Routes and obtained a series of kinetic parameters. The model includes blood, liver, kidney, stomach, intestine, skin, and rest of body compartments. Oral and dermal exposure route was considered for PBTK model. The PBTK model established in this study has a good predictive ability. More than 70% of the predicted values deviated from the measured values by less than 5-fold. In addition, we extrapolated the model to humans. A human PBTK model is built. We performed a health risk assessment for world populations based on human PBTK model. The risk of TCEP in dust is greater through mouth than through skin. The risk of TCEP in food of Chinese population is greater than dust.

Degradation of tris(2-chloroethyl) phosphate (TCEP) by thermally activated persulfate: Combination of experimental and theoretical study

He Liu,Jiahao Liang,Xiaodong Du,Rui Wang,Ting Tang,Xueqin Tao,Hua Yin,Zhi Dang,Guining Lu

Organophosphorus esters (OPEs), one kind of the emerging contaminants with high frequency of detection, is rather refractory in natural environment, thus posing great threat to human health. This study investigated the feasibility and mechanism of tris(2-chloroethyl) phosphate (TCEP) degradation in thermally activated persulfate (TAP) system. Influence of impact factors, such as PDS dosage, temperature, initial pH, and presence of natural water matrix (Cl − , NO 3 − , H 2 PO 4 − , NH 4 + , humic acid), were evaluated. Results showed that 100% degradation of TCEP can be achieved in TAP system in 40 min at 60 °C. SO 4 ·− as the dominant oxidant for TCEP degradation was proved by quenching experiment and verified by EPR analysis. Alkaline condition exerted great inhibitory effect by affecting the constituents of oxidative radicals. It is suggested that Cl − and H 2 PO 4 − at lower dosages promoted the degradation by stimulating ·OH production and forming oxidative radicals with better selectivity . Intermediates identified by high resolution mass spectrometer was suggested less toxic than TCEP by ECOSAR program. Meanwhile, the illustrated oxidation mechanism mainly involved radical attack at C Cl bond and cleavage of C O bond, as further confirmed by frontier electron density calculation and wavefunction analysis. Moreover, cyclic degradation of TCEP indicated the constant release of SO 4 ·− in 450 min, suggesting high efficiency and stability of PDS in TAP system. Four selected OPEs achieved complete removal in TAP system and their degradation discrepancy was further discussed based on the distinctive structures. Altogether, TAP technology can be used as an efficient method in TCEP removal with great potential for application.

Effects of tris (2-chloroethyl) phosphate (TCEP) on growth, reproduction and gene transcription in the protozoan Tetrahymena thermophila

Hui Hao,Siliang Yuan,Shiyang Cheng,Qian Sun,John P. Giesy,Chunsheng Liu

As a typical organophosphorus flame retardant, tris (2-chloroethyl) phosphate (TCEP) has been widely detected in various environmental media. Toxicity of TCEP to vertebrates have been investigated, but potential effects on lower trophic level species were unknown to date. In this study, toxic effects and molecular mechanisms of toxic actions of TCEP on the aquatic protozoan Tetrahymena thermophila were evaluated by use of phenotypic observations, transcriptome sequencing analysis and real-time quantitative PCR detection. Exposure to 0.044, 0.411 or 4.26 mg/L TCEP for 5 days decreased the theoretical population, cell viability, number of cilia and cell size of Tetrahymena thermophila in a time- and dose-dependent manner. Meanwhile, RNA-Seq analysis indicated that exposure to 4.26 mg/L TCEP significantly changed expression of 2932 genes (up-regulation: 1228; down-regulation: 1704). Of these, expressions of 9, 10 and 17 genes that were enriched in soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) interaction in vesicular transport, proteasome and endocytosis pathway respectively were down-regulated. Data collected during this study suggested that exposure to high concentrations of TCEP might affect growth and reproduction of Tetrahymena thermophila through down-regulating transcriptional levels of genes encoding proteins associated with vesicle trafficking, proteasome and endocytosis.

Lucigenin-Tris(2-carboxyethyl)phosphine Chemiluminescence for Selective and Sensitive Detection of TCEP, Superoxide Dismutase, Mercury(II), and Dopamine

Muhammad Saqib,Shahida Bashir,Haijuan Li,ShanShan Wang,Yongdong Jin

Development of simple and effective chemiluminescence (CL) systems for multiple sensing applications is significantly important but still a challenge. Until now, the majority of CL systems primarily utilized hydrogen peroxide (H2O2) as coreactant, which is limited in its stability and selectivity due to the easy decomposition of H2O2 in the presence of several ions. In this study, we develop a new and intense CL system by combined use of tris(2-carboxyethyl)phosphine (TCEP), a highly solution stable and pH-tolerant tertiary phosphine, with lucigenin for the first time. The effective pairing leads to a significant ∼23 times CL enhancement over classic the lucigenin–H2O2 system without employing additional catalysts. By virtue of this fascinating platform, a sensitive CL method has been developed for the multiple detection of TCEP (LOD = 70 nM), lucigenin (LOD = 4.0 nM), superoxide dismutase (LOD = 0.8 ng/mL), Hg2+(LOD = 0.3 nM), and dopamine (LOD = 3.0 nM), with a linear range of 0.1–320 μM, 0.01–55 μM, 0.005–0.5 μg/mL, 1.0–600 nM, and 0.01–0.8 μM, respectively. Remarkably, this CL method exhibited superior selectivity over several potential interferents. Moreover, the proposed method achieved excellent recoveries in the range of 94.0–102.3% for both Hg2+ detection in lake water and dopamine detection in human serum real samples. We envision that broad applications of TCEP may lead to construct new CL systems, pushing forward for efficient detection of various analytes.

Enhancing the peroxidase activity and decreasing the protease activity of ficin with rational modification and its application to one-step colorimetric detection of glucose

Yijuan Long,Wen Zheng,Danyang Yi,Yadi Pan,Huzhi Zheng

Ficin has dual enzyme activity , i.e., protease and peroxidase-like activity. In some respects, its application is limited by the protease activity of ficin. Herein, we used tris (2-carboxyethyl) phosphine hydrochloride (TCEP) to break the three pairs of disulfide bonds of ficin, and then blocked the free thiol groups with N-ethylmaleimide (NEM) to synthesize ficin-TN. The results showed that ficin-TN had increased peroxidase-like activity and reduced protease activity. According to this phenomenon, we have exploited a colorimetric method with high sensitivity and selectivity for the one-step detection of glucose. Comparing with ficin, ficin-TN has wider detection range (0.1–300 μM) and lower detection limit (88 nM), and our method is simpler and more timesaving than other two-step methods. Furthermore, the actual appliances of ficin-TN for glucose detection in human serum have been illustrated with satisfied result, suggesting that its promising utilization in various fields.

Toxicity of Tris(2-chloroethyl) Phosphate (TCEP) to Alfalfa’s Root System: An Insight into TCEP’s Damage to Morphology, Respiration, and Antioxidant Systems

Meijun Liu,Liangzhu Gong,An Yan,Wenjing Liu,Haojie Li,Peiyi Guo

Tris(2-chloroethyl) phosphate (TCEP), as an organophosphate contaminant, poses a significant threat to the growth and development of plants, especially roots. This study aimed to clarify the mechanisms of TCEP’s toxicity and damage to root systems, as well as the mechanisms of its damage to the respiration and energy metabolism of alfalfa root cells. The results showed that TCEP obviously affected the root length, root surface area, root volume, and root diameter of alfalfa. With increasing stress intensity, the total mitochondrial respiration rate and Cytochrome C Oxidase (COX) pathway respiration rate progressively declined, while the Alternative Oxidase (AOX) pathway respiration rate and its proportion of total respiration gradually rose. In addition, adenosine triphosphate (ATP) content and root vigor were significantly reduced. Moreover, with an increase in TCEP concentration, root superoxide anion radical content in alfalfa root cells was significantly elevated, while superoxide dismutase (SOD) and catalase (CAT) activities were significantly lowered, and ascorbate peroxidase (APX) and peroxidase (POD) activities were significantly enhanced. The present study indicated that respiration was disrupted, causing a lack of ATP in root cells under TCEP. Both the overproduction of reactive oxygen species (ROS) from the mitochondrial respiratory electron transport chain (mECT) and the deficiency of ROS-scavenging enzymes caused ROS accumulation, which led to the destruction of the cell membrane structure and exacerbated the disruption of the respiratory metabolism. The disruption of the conversion and reuse of energy by TCEP affected root growth and development.

Efficient removal of organophosphate esters by ligand functionalized MIL-101 (Fe): Modulated adsorption and DFT calculations

Sheng Fan,Xinru Lu,Haoliang Li,Xiaodong Du,Xiaoyu Huang,Yao Ma,Juan Wang,Xueqin Tao,Zhi Dang,Guining Lu

Organophosphate esters (OPEs) are a new class of flame retardants present in diverse waters. The study focused on the removal of aqueous OPEs using functionalized MIL-101(Fe), which was a representative of the metal-organic frameworks (MOFs). Adsorption kinetics of tris(2-chloroethyl) phosphate (TCEP), tributyl phosphate (TnBP), and triphenyl phosphate (TPhP) were investigated. Additionally, TCEP was selected as the model contaminant to study the adsorption isotherms, thermodynamics, and effect of solution matrix properties. Adsorption mechanisms obtained from the experiments were confirmed using density functional theory (DFT) calculations. Adsorption kinetics indicated that functionalized MOFs provided a significant enhancement to the removal of TCEP. The maximum adsorption capacities q m of MIL-101(Fe), MIL-101(Fe)–NH 2 , MIL-101(Fe)–OH, and MIL-101(Fe)–CH 3 at 298 K for TCEP were 76.040, 282.940, 119.680, and 181.274 μmol/g, respectively. By comparing the adsorption behavior of functionalized MOFs, MIL-101(Fe)–NH 2 was proved to be most efficient for TCEP removal . Based on the adsorption experiments and DFT calculations, TCEP removal was dominated by physical adsorption. The van der Waals (vdW) interactions and hydrogen bonding were assumed to be involved in the adsorption. This work proves that appropriate ligand functionalization is promising for the removal of aqueous OPEs, which also provides a new insight for the control of OPEs pollution.
