Obesity is a major risk factor for type 2 diabetes. Some traditional Chinese medicine (TCM) compounds can improve obesity by increasing energy consumption. This study aimed to identify and verify TCM compounds that promote adipose thermogenesis to alleviate obesity via transcriptomic analysis, molecular docking, network pharmacology, and Connectivity Map (CMap) analysis. Thirty-six TCMs related to adipose tissue thermogenesis were first collected to generate transcriptomic data. Then a ranking method based on integrated transcriptomic data was used to select emodin from the TCM Da Huang for network pharmacology to explore its potential mechanisms and further experimental validation. CMap analysis of thermogenesis signature genes from ProFAT and GEO datasets identified triptolide as another compound for improving obesity through enhanced thermogenesis. In vitro experiments with 3T3-L1 cells and in vivo experiments with zebrafish were conducted. The results showed that both emodin and triptolide could upregulate the NAD + /NADH ratio, increase AMPK and SIRT1 expression, suppress lipid accumulation, and promote lipolysis, as verified by both in vitro and in vivo experiments. They also upregulated thermogenesis-related genes (e.g., UCP1, PGC1α, and PRDM16), lipolysis-related genes (e.g., PKA, ATGL, and HSL), mitochondrial biogenesis-related genes (e.g., NRF1, NRF2, and TFAM), and β-oxidation-related genes (e.g., CPT1α and CPT1β). Meanwhile, they downregulated lipogenesis-related genes such as FASN, SREBP1, and ACC. These results provide a basis for understanding the effects of emodin and triptolide on obesity. The findings highlight the effectiveness of combined screening approaches in exploring compounds with beneficial metabolic effects.

s In this paper, a hybrid system of ultrafiltration and membrane distillation was used to study the resourceful reuse of active compounds and water resources in traditional Chinese medicine wastewater. Pre-treatment of turbid wastewater by ultrafiltration (UF) was found to maximise the retention of macromolecular organic matter in the wastewater using a 10 kDa ultrafiltration membrane at a trans-membrane pressure of 0.15 MPa. The UF filtrate was subjected to vacuum membrane distillation (VMD) at 60 °C, 95 kPa, 36 L·h −1 , and pH = 3, which ultimately recovered about 85 % of chlorogenic acid as well as about 74 % of baicalin. The UF-MD system can effectively separate the large organic molecules, small molecules of active compounds and water in the turbid wastewater of traditional Chinese medicine, so as to realise the recovery and reuse of valuable resources in the turbid wastewater of traditional Chinese medicine.

Marine biofouling and corrosion have become the main problems affecting the development of the marine industry. Silicone-based coatings have been widely used for antifouling and anticorrosion due to their low surface energy. However, the poor adhesion and low mechanical stability of these materials limit their application in complex marine environments. In this work, we presented a marine antifouling and anticorrosion coating named POSS-DMA@PDMS-TCM through photoinitiated thiol–ene click reaction combined with (mercaptopropyl) methylsiloxane dimethylsiloxane (PDMS-SH), dopamine methacrylamide (DMA), sulfhydryl-functionalized organosiloxanes (POSS-(SH)8), and N-(2,4,6-trichlorophenyl) maleimide (TCM). The POSS-DMA@PDMS-TCM coating exhibited strong stability and bonding ability both in air (2.17 MPa) and underwater (2.11 MPa) when the DMA content was 3 wt %. The high antibacterial (98.1% for Staphylococcus aureus and 99.5% for Escherichia coli) and antidiatom (94.5%) properties of the POSS-DMA@PDMS-TCM coatings have also been confirmed. Moreover, the POSS-DMA@PDMS-TCM coatings show excellent antifouling abilities in 120-day marine field tests, reducing fouling by 65.5% in comparison to the blank group. The coating also displayed superior anticorrosion performance with Ecorr values of −0.055 V, Icorr values of 7.67 × 10–6 , and Rp values of 3.10 × 105 Ω for Cu, which benefited from excellent chelating effect and liquid repellency. This study provides a novel strategy for the development of high-quality marine antifouling and anticorrosion coatings.

Ethanol precipitation serves an important separation technology used for the manufacturing of more than 200 traditional Chinese medicines (TCM). In the present study, a membrane dispersion micromixer was employed for the ethanol precipitation of Codonopsis Radix concentrates to reduce the coating loss of active components. As membrane pore size decreased, mixing chamber width decreased, or mixing chamber depth decreased, lobetyolin recovery increased and dry matter removal decreased. Higher dry matter contents of concentrates (DM C ) or lower flow rates of two phases could lead to reduced lobetyolin recovery and increased dry matter removal. The mass ratio of ethanol solution to concentrate (ECR, g/g) showed little effect on lobetyolin recovery. Higher dry matter removal was obtained when ECR was higher. Higher continuous phase Reynolds number ( Re c ) or dispersed phase Reynolds number ( Re d ) resulted in higher lobetyolin recovery and lower dry matter removal. Lobetyolin recovery could be higher than 95% when using micromixing technology, which was about 10% higher than that of stirring. Membrane dispersion technology is promising to reduce the coating loss of active compounds in TCM ethanol precipitation.

Wastewater discharge of traditional Chinese medicine production is a serious threat to the environment in China. This study investigated the effectiveness of the UF-FO-MD hybrid system for resources recycle from the TCM wastewater. Real TCM wastewater was treated with the self-assemble UF-FO-MD hybrid system. Moreover, the natural surfactant rejection and the FO fouling impact were evaluated by separate FO operation with synthetic TCM wastewater, and MD wetting was also assessed. Results show that the UF-FO-MD hybrid system successfully isolated the macromolecular substances, bioactive compounds and water in the TCM wastewater for recovering useful resources. Natural surfactants in the TCM wastewater could be effectively rejected by FO, avoiding the subsequent MD wetting problem which has occurred in our previous study on the resource recovering with the UF-MD hybrid system. However, FO membrane fouling might be an issue affecting the recovery efficiency of bioactive compounds from TCM wastewater. The bioactive compounds could not be completely concentrated due to potential adsorption on the fouling layer .

Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality, with high postoperative recurrence rates due to occult micrometastases or minimal residual disease, markedly worsening the prognosis for HCC patients. Current therapies lack effective strategies to prevent recurrence, while traditional Chinese medicine (TCM) shows potential in delaying HCC progression. Combining a hemostatic hydrogel with nanoparticle-based delivery of active TCM components provides a strategy to enhance tumor recurrence prevention. Herein, we develop a nanocomposite hydrogel (HPS@ZCJ) by encapsulating Jianpi-Huayu essential oils (JEO) into zein-based nanoparticles (zein@chondroitin sulfate@JEO, ZCJ) and embedding them in a hydroxymethyl cellulose/Pluronic F-127/sodium alginate (HPS) hydrogel matrix. HPS@ZCJ hydrogel enhances cytotoxic T-lymphocyte infiltration, inhibits the polarization of tumor-associated macrophages to M2 phenotype, induces tumor cell death, reverses immunosuppression, and inhibits angiogenesis within the tumor. The antitumor mechanism involves dual downregulation of GPNMB and DHCR7, key genes in HCC progression and immune evasion. In vitro and in vivo experiments demonstrate that HPS@ZCJ hydrogel-mediated targeted comprehensive therapy simultaneously achieves intraoperative hemostasis, impedes primary tumor growth and prevents HCC postoperative recurrence. This study provides a promising postoperative HCC treatment strategy, leveraging TCM's therapeutic potential with significant clinical translation prospects.

The separation of active ingredients in traditional Chinese medicine (TCM) is of great significance in determining the efficacy of TCM as well as the pharmacological study of active ingredients, improving the scope of application of TCM and the efficiency of disease prevention and treatment. In the current study, we have successfully synthesized a core–shell structure magnetic microsphere Fe 3 O 4 -COF for the separation of α-asarone and β-asarone in the combined decoction of Acorous Tatarinowii Rhizoma (ATR) and Polygalae Radix (PR). The successful synthesis of Fe 3 O 4 -COF was demonstrated chemically and morphologically by using five characterizations such as Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM). Extraction conditions were investigated and optimized, including adsorbent dosage, adsorption time, sample pH value, elution solvent, elution time and volume of elution solution. Under the optimal experimental conditions, we got a low limit of detection (LOD) of α-asarone and β-asarone were 0.0379 μg/mL and 0.0432 μg/mL, and the relative standard deviations (RSD) were less than 2.3%. And the number and intensity of impurity peaks in the chromatogram after extraction of complex samples were significantly reduced and the good recoveries were still maintained after multiple extractions of complex samples, which showed satisfied specific adsorption and excellent resistance to matrix interference. The successful establishment of this method will provide a new direction for the separation and analysis of active ingredients in traditional Chinese medicine.

TCM wastewater treatment is a significant environmental issue in China. This study investigated the effectiveness of the UF-MD hybrid system on recycling valuable resources in TCM wastewater. Synthetic TCM wastewater was prepared and separated with the self-assemble UF-MD hybrid system. Moreover, the feed temperature (45 °C, 55 °C, 65 °C) and membrane materials' influence on MD performance were also evaluated. Results show that the UF-MD hybrid system is efficient to achieve a complete separation and resources recycle from the TCM wastewater. The MD flux production increased with the increment of feed temperature. However, some bioactive compounds could not be concentrated in MD due to their potential degradation at high temperatures, and membrane wetting might be an issue in long-term operation. Although PP membranes exhibited the highest flux among PP , PTFE and PVDF membranes, considering the wetting and fouling resistance , PTFE membranes would be a better option for long-term MD operation at 65 °C.

Target discovery of natural products is a key step in the development of new drugs, and it is also a difficult speed-limiting step. In this study, a traditional Chinese medicine microspheres (TCM-MPs) target fishing strategy was developed to discover the key drug targets from complex system. The microspheres are composed of Fe 3 O 4 magnetic nanolayer, oleic acid modified layer, the photoaffinity group (4- [3-(Trifluoromethyl)-3H-diazirin-3-yl] benzoic acid, TAD) layer and active small molecule layer from inside to outside. TAD produces highly reactive carbene under ultraviolet light, which can realize the self-assembly and fixation of drug active small molecules with non-selective properties. Here, taking Shenqi Jiangtang Granules (SJG) as an example, the constructed TCM-MPs was used to fish the related proteins of human glomerular mesangial cells (HMCs) lysate. 28 differential proteins were screened. According to the target analysis based on bioinformatics, GNAS was selected as the key target, which participated in insulin secretion and cAMP signaling pathway. To further verify the interaction effect of GNAS and small molecules, a reverse fishing technique was established based on bio-layer interferometry (BLI) coupled with UHPLC-Q/TOF-MS/MS. The results displayed that 26 small molecules may potentially interact with GNAS, and 7 of them were found to have strong binding activity. In vitro experiments for HMCs have shown that 7 active compounds can significantly activate the cAMP pathway by binding to GNAS. The developed TCM-MPs target fishing strategy combined with BLI reverse fishing technology to screen out key proteins that directly interact with active ingredients from complex target protein systems is significant for the discovery of drug targets for complex systems of TCM.

Reliable, non-invasive, continuous monitoring of pulse and blood pressure is essential for the prevention and diagnosis of cardiovascular diseases. However, the pulse wave varies drastically among individuals or even over time in the same individual, presenting significant challenges for the existing pulse sensing systems. Inspired by pulse diagnosis methods in traditional Chinese medicine (TCM), this work reports a self-adaptive pressure sensing platform (PSP) that combines the fully printed flexible pressure sensor array with an adaptive wristband-style pressure system can identify the optimal pulse signal. Besides the detected pulse rate/width/length, “Cun, Guan, Chi” position, and “floating, moderate, sinking” pulse features, the PSP combined with a machine learning-based linear regression model can also accurately predict blood pressure such as systolic, diastolic, and mean arterial pressure values. The developed diagnostic platform is demonstrated for highly reliable long-term monitoring and analysis of pulse and blood pressure across multiple human subjects over time. The design concept and proof-of-the-concept demonstrations also pave the way for the future developments of flexible sensing devices/systems for adaptive individualized monitoring in the complex practical environments for personalized medicine, along with the support for the development of digital TCM.

The global COVID-19 pandemic caused by SARS-CoV-2 has underscored the urgent need for the development of new broad-spectrum antivirals to combat its high mutation rate and the emerging variants. Host ER α-glucosidases I/II are promising host-targeted therapeutic targets for the development of broad-spectrum antivirals against viral strains that depend on ERQC for infectivity. Herein, we designed and synthesized a series of TCM theory-inspired DNJ-flavonoid conjugates as novel α-glucosidase inhibitors, which were screened against their in vitro antiviral activities in non-replicative SARS-CoV-2 pseudovirus (PsV) assay. Remarkably, DNJ-20 not only demonstrated remarkable inhibition activity against α-glucosidase and viral entry process, but also exerted potent and broad-spectrum anti-coronaviral activity against SARS-CoV-2 pseudovirus (PsV), several SARS-CoV-2 variants, as well as HCoV-229E and HCoV-OC43, with EC 50 values up to 1.49 μM, which is more potent than the benchmark α-glucosidase inhibitor UV-4 ( DNJ-3 ). Besides, it had no observable cytotoxicity in HeLa-ACE2, HEK-293T and Beas-2B cells. Therefore, TCM theory-inspired DNJ-flavonoid conjugates can be served as promising drug leads for pan-coronavirus therapeutic development to combat current and future coronavirus pandemics.

Patulin (PAT) is a kind of mycotoxin which must be monitored for the sake of quality and safety in traditional Chinese medicine (TCM) owing to its harm to human health. On this account, a rationally designed ratiometric fluorescent aptasensor was developed based on the study of the interaction mechanism between PAT and its aptamer (PAT-APT). Firstly, CD spectroscopy, molecular docking, and molecular dynamic simulation were applied to investigate the details on how PAT-APT bind with its target molecule. The results indicated that the structure of PAT-APT changed to a certain extent and was stabilized after binding with PAT. C-11, C-37 and C-38 were the key sites for the recognition and interaction between PAT-APT and its target. Secondly, based on these results, a ratiometric aptasensor made use of fluorescence resonance energy transfer (FRET) and synchronous fluorescence spectroscopy was designed. The complementary sequence (cDNA) to the aptamer with appropriate length and hybridization position was obtained through rationally design and optimization. Both PAT-APT and cDNA were labeled by a pair of fluorophores, which could generate FRET when the two single-stranded oligonucleotides hybridized. The accurate detection of PAT could be realized according to the change ratio of fluorescence intensity at the corresponding wavelength of the two fluorophores before and after assay. The aptasensor achieved an ultralow limit of detection 0.16 nM, perfect selectivity, and satisfactory practibility in complex TCM sample. To our knowledge, this is the first aptasensor for PAT designed through the interaction mechanism between aptamer and target molecule. Moreover, the assay for PAT is cost-effective, does not need complicated pretreatment and only takes less than an hour. Summarily, this study makes contribution to safety control of TCM, and provides a thinking mode from mechanism to rational design to conquer the problem of sensitive aptasensing of one component in a complex system.
