Hydroponic experiments and computational simulations were conducted to investigate the changes in the gene expression, structure, and binding mode of the rice transporter protein OsTIL to OPEs by the presence of Cd 2+ and Zn 2+ . OPEs and Zn 2+ were observed to promote seedling growth and OPEs alleviated the suppressive effect of Cd 2+ on seedlings. Usually, Cd 2+ and Zn 2+ inhibited the accumulations of OPEs in plants accompanied by the decrease of RCF (root concentration factor) and TF (transport coefficient). In particular, Zn 2+ promoted TDCPP accumulation only in roots accompanied by the increase of RCF and the decrease of TF. Furthermore, Cd 2+ and Zn 2+ affected the gene expressions of OPEs transporter-OsTIL response in the accumulation of OPEs in both single and compound pollution. After molecular dynamics simulation analysis, RMSD of the protein backbone, binding pocket, and ligand only in TDCPP-OsTIL complex also were significantly affected by two metal ions. Furthermore, two metal ions can press the interaction of OPEs and OsTIL by reducing the stability of protein structure and the numbers of HB (hydrogen bonds) and enlarging the pocket. However, Zn 2+ extra supports an enlarged entrance for TDCPP, which could facilitate the entry of the coordination complex of Zn 2+ with TDCPP into the pocket and improve TDCPP capacity of OsTIL.

The ubiquity and ecotoxicity of tri (1,3-dichloro-2-propyl) phosphate (TDCPP) in aquatic environments make it essential to develop efficient related treatment techniques. The present research aims to systematically explore the mechanisms and toxicity changes of TDCPP degradation through UV-driven hydrogen peroxide process (UV/H 2 O 2 ). The results revealed degradation efficiency of TDCPP attained 95 % with 0.5 mM H 2 O 2 , corresponding to a reaction rate constant ( k ) of 0.04919 min −1 . Furthermore, three typical chlorinated OPEs (Cl-OPEs) were simultaneously degraded by the UV/H 2 O 2 process, with different degradation efficiencies, according to the following order: TCEP

Tris (1,3-dichloro-2-propyl) phosphate (TDCPP), a halogen-containing phosphorus flame retardant, is widely used and has been shown to possess health risks to humans. The sustained release of artificial nanomaterials into the environment increases the toxicological risks of their coexisting pollutants. Nanomaterials may seriously change the environmental behavior and fate of pollutants. In this study, we investigated this combined toxicity and the potential mechanisms of toxicity of TDCPP and titanium dioxide nanoparticles (TiO 2 NPs) aggregates on human neuroblastoma SH-SY5Y cells. TDCPP and TiO 2 NPs aggregates were exposed in various concentration combinations, revealing that TDCPP (25 μg/mL) reduced cell viability, while synergistic exposure to TiO2 NPs aggregates exacerbated cytotoxicity. This combined exposure also disrupted mitochondrial function, leading to dysregulation in the expression of mitochondrial fission proteins (DRP1 and FIS1) and fusion proteins (OPA1 and MFN1). Consequently, excessive mitochondrial fission occurred, facilitating the translocation of cytochrome C from mitochondria to activate apoptotic signaling pathways. Furthermore, exposure of the combination of TDCPP and TiO 2 NPs aggregates activated upstream mitochondrial autophagy but disrupted downstream Parkin recruitment to damaged mitochondria, preventing autophagosome-lysosome fusion and thereby disrupting mitochondrial autophagy. Altogether, our findings suggest that TDCPP and TiO 2 NPs aggregates may stimulate apoptosis in neuronal SH-SY5Y cells by inducing mitochondrial hyperfission and inhibiting mitochondrial autophagy.

Plain Language Summary Background: Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is a widely used organophosphorus flame retardant that has raised growing concern because it is persistent, can bioaccumulate, and is toxic. However, its possible role in chronic kidney disease (CKD) is still not well understood. Methods: We used network toxicology, molecular docking, transcriptomic validation, and mouse exposure experiments to uncover the mechanisms linking TDCPP exposure to kidney injury. Results: We found 1270 overlapping targets between predicted TDCPP-binding proteins and CKD-related genes. Enrichment analyses showed strong links to inflammatory and apoptotic processes, as well as key signaling pathways including PI3K–Akt, MAPK, Ras, and cAMP. Machine learning methods (LASSO, SVM-RFE, RF) identified two hub genes, CTRB1 and HSPA1A , which were both significantly downregulated in CKD transcriptomes and showed perfect diagnostic performance (AUC = 1.0). Immune cell analysis showed that CKD tissues had increased regulatory T cells, monocytes, M2 macrophages, and neutrophils, and CTRB1 / HSPA1A expression was correlated with specific immune cell subsets. Molecular docking predicted favorable binding of TDCPP to both proteins, with the strongest affinity for CTRB1 (–7.2 kcal/mol). In vivo , TDCPP exposure caused dose-dependent tubular degeneration, inflammation, and increased serum BUN and creatinine, along with marked downregulation of CTRB1 and HSPA1A . Conclusion: Taken together, these findings suggest that TDCPP may contribute to CKD by disrupting CTRB1 / HSPA1A and activating PI3K–Akt/MAPK signaling, which leads to immune dysregulation and progressive kidney injury. We propose a new adverse outcome pathway (AOP) framework linking TDCPP exposure to CKD, and highlight CTRB1 and HSPA1A as potential biomarkers and mechanistic targets for environmental nephrotoxicity. Plain Language Summary This study explores how the flame retardant TDCPP might contribute to chronic kidney disease (CKD). Researchers used various methods, including network toxicology and mouse experiments, to identify 1270 targets shared between TDCPP-binding proteins and CKD-related genes. They found that TDCPP affects inflammatory and apoptotic processes and key signaling pathways. Two genes, CTRB1 and HSPA1A, were identified as crucial, showing strong links to immune cell changes in CKD. TDCPP binds well to these proteins, especially CTRB1, and exposure in mice led to kidney damage and inflammation. The study suggests TDCPP disrupts these genes, contributing to CKD, and proposes them as biomarkers for environmental nephrotoxicity. Text is machine generated and may contain inaccuracies. FAQ

Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) has been frequently detected in environmental media and biological samples. However, knowledge of its adverse health consequences is limited. In the current study, Caenorhabditis elegans ( C. elegans , L1 larvae) were exposed to TDCPP at environmentally relevant concentrations (control, 0.1, 1, 100 and 1000 μg L −1 ) for 72 h to explore any association between TDCPP and the aging process. Some of the degenerative age-related indicators were observed, including locomotion behaviors and lifespan. As crucial biomarkers of aging, the accumulation of lipofuscin, and lipid peroxidation (LPO) products exemplified by 4-hydroxynon-2-enal (4-HNE) were detected. This product forms as a result of oxidative stress , as confirmed by an N-acetyl-L-cysteine (NAC) pharmacological assay. Moreover, a significant increase in reactive oxide species (ROS) production in a dose-dependent manner using a fluorescent probe was observed. For the underlying molecular mechanism of the above aging phenotypes, significantly upregulated transcription of genes related to antioxidant systems, especially a subset of glutathione S-transferase ( gst-5, gst-6, gst-9, gst-10, gst-19, gst-24, gst-26, gst-29, gst-33, and gst-38 ), was found by RNA-Seq and further confirmed by RT-qPCR. The elevated glutathione S-transferase (GST) was attributed to the significant increase in 4-HNE because mutations in gst-5 and gst-24 inhibited the conjugation of GSTs with 4-HNE. Therefore, GST play an indispensable role in the detoxification process of TDCPP exposure and further confirmed LPO accumulation at the molecular mechanism level. In conclusion, TDCPP accelerated the aging process induced by the LPO products, 4-HNE, response to reactive oxidative species in C. elegans .

Flame retardants , such as Tetrabromobisphenol A (TBBPA), Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) and tributyl phosphate (TBP), are frequently detected in surface water. However, the effects of FRs exposure on aquatic organisms especially freshwater microalgae are still unclear. In this study, the toxicities of TBBPA, TDCPP and TBP to microalgae Chlorella sorokiniana , in terms of growth inhibition, photosynthetic activity inhibition and oxidative damage, were investigated, and according ecological risks were assessed. The results showed that TBBPA, TDCPP and TBP had inhibitory effects on C. sorokiniana , with 96 h EC 50 (concentration for 50% of maximal effect) values of 7.606, 41.794 and 49.996 mg/L, respectively. Fv / Fm decreased as the increase of exposure time under 15 mg/L TBBPA. Under 50 mg/L TDCPP and 80 mg/L TBP exposure, Fv / Fm decreased significantly after 24 h. However, Fv / Fm rose after 96 h, indicating that the damaged photosynthetic activity was reversible. The content of chlorophyll a decreased, as the increase of TBBPA concentration from 3 to 15 mg/L. However, chlorophyll a increased first and then decreased, as the increase of TDCPP and TBP concentrations from 0 to 50 mg/L and 0–80 mg/L, respectively. Results indicated that C. sorokiniana could use the phosphorus of TDCPP and TBP to ensure the production of chlorophyll a . The risen content of reactive oxygen species , malondialdehyde as well as superoxide dismutase activity indicated that exposure to FRs induced oxidative stress . Additionally, the risk quotients showed that tested FRs had ecological risks in natural waters or wastewaters. This study provides insights into the toxicological mechanisms of different FRs toward freshwater microalgae for better understanding of according environmental risks.

Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is an emerging environmental contaminant of concern due to its widespread presence and potential toxicity. Its metabolite, bis(1,3-dichloro-2-propyl) phosphate (BDCPP), plays a critical role in mediating TDCPP-induced toxicities and may exacerbate aging-related processes. In this study, we quantitatively compared the bioconcentration and toxicity of TDCPP and BDCPP in Caenorhabditis elegans. BDCPP exhibited higher environmental persistence and bioaccumulation potential than TDCPP. Exposure to 0.1–1000 μg/L TDCPP and BDCPP increased Oil red O staining (7.88%–55.44%, 6.42%–33.92%), lipofuscin levels (5.44%–35.98%, 3.54%–28.66%), and reduced the mean lifespan (3.09%–10.31%, 1.84%–8.74%). Overall, TDCPP caused greater triglyceride (TG) accumulation, lipofuscin formation, and lifespan reduction compared to BDCPP. TDCPP reduced brood size by 5.30%–18.95%, while BDCPP exposure increased brood size by up to 13.90%. Mechanism investigations revealed that TDCPP and BDCPP induce aging through distinct pathways. The adverse outcome pathways revealed that BDCPP disrupted germline stem cells (GSCs) apoptosis, transmitting reproductive signals through the kri-1-daf-16-lipl-4 signaling cascade, impairing TG hydrolysis and lifespan, while TDCPP acted through a GSC-independent route. These findings underscore the central role of GSCs in BDCPP-induced aging, enhance our understanding of BDCPP as a secondary pollutant, and provide critical data for assessing the ecological and health risks of chlorinated organophosphate esters.

Alkyl phosphate esters (APEs) are mass-produced substances characterized by a wide range of physicochemical properties and applications. Their endocrine-disrupting effects via inhibition of placental 3β-hydroxysteroid dehydrogenase (3β-HSD) remain unclear. The objective of this study was to examine the inhibitory effects of 22 APEs on human 3β-HSD1 (h3β-HSD1) its rat homolog r3β-HSD4, critical enzymes in progesterone synthesis, using placental microsomes and computational analysis. The results demonstrated that h3β-HSD1 was more sensitive to inhibition by 100 μM APEs, exhibiting a V-shaped potency trend. Among the tested APEs, triheptyl phosphate ester showed the strongest inhibition of h3β-HSD1 (IC 50 = 3.33 μM), followed by trihexyl (IC 50 = 5.53 μM), triamyl (IC 50 = 22.67 μM), triethylhexyl (IC 50 = 22.85 μM), and tributyl phosphate esters (IC 50 = 102.53 μM). In contrast, r3β-HSD4 exhibited lower sensitivity, with significant inhibition observed for only three APEs: diethyl, triamyl, and trihexyl phosphate esters, with IC 50 values of 38.94, 109.76, and 51.49 μM, respectively. All tested APEs functioned as mixed-type inhibitors, and they suppressed progesterone production in human JAr cells at 10 and/or 100 μM (noncytotoxic concentrations). Lipophilicity and molecular size were identified as key factors influencing inhibitory potency against h3β-HSD1, as supported by 3D-QSAR modeling and molecular docking. These findings suggest that the mode of inhibition involves interactions at the NAD + /steroid interface, including hydrogen bonds and hydrophobic forces. Network toxicology analysis highlighted the association of h3β-HSD1 with endometrial cancer and preeclampsia. This research provides mechanistic insights into the potential reproductive risks of APEs and emphasizes the need for further studies on their environmental and health impacts.

Chlorinated organophosphate flame retardants (Cl-OPFRs) make up a class of highly refractory pollutants that have raised wide environmental concerns. Conventional AOPs, e.g., the benchmark O3/H2O2 (peroxone), are not efficient in removing them from water/wastewater, because of their relatively low reaction rates with hydroxyl radical (•OH). We report here that Cl-OPFRs (i.e., TCEP, TCPP, and TDCPP) can be efficiently removed by catalytic ozonation with zinc oxide (O3/ZnO) through a novel reaction pathway. Based on in situ characterization with confocal Raman spectrum and fluorescence microscopy imaging, we confirmed that the ozone–ZnO interaction resulted in the formation of surface-bound •OH, with yields 50% higher than those of the peroxone system. Using TCEP as a model Cl-OPFR and ATR-FTIR characterization, we recognized the coordination between the phosphoryl oxygen and the surface Zn(II) of ZnO. The surface coordination improved the reactivity of TCEP toward the surface-bound •OH, which was confirmed with a competitive reaction on ZnO. The O3/ZnO was most effective at neutral pH for TCEP removal and can achieve partial mineralization and detoxification of the compound. This work highlights the importance of surface reaction in removing the highly refractory Cl-OPFRs.

Organophosphate esters (OPEs) are a serious and pervasive indoor environmental pollutant, and their extensive use and frequent detection in indoor environments have raised growing concerns globally about their impact on the respiratory system. Simultaneously, increasing attention has been directed toward interfacial hypochlorous acid (HOCl) chemistry, particularly in relation to indoor air quality and public health. However, the molecular mechanisms underlying HOCl-mediated oxidation of phospholipids in the presence of OPEs remain unclear. This study utilized an online air-water interface reaction monitoring technique to examine the effects of ten OPEs, including alkyl, chlorinated, and aryl OPEs, on the oxidation of a model lung phospholipid 1-Palmitoyl-2-oleoyl-sn-glycero-3-phospho-(1′-rac-glycerol) (POPG) exposed to HOCl at the air-water interface. The results indicated that OPEs inhibit the oxidation of phospholipids by HOCl at the air-water interface, and this inhibitory effect is correlated with the concentration of OPEs and their water-oil partition coefficient (LogP). Mechanistic studies demonstrated that the hydrophilicity of OPEs enables their incorporation into the phospholipid monolayer, resulting in a compact arrangement of POPG. This arrangement partially shields the C=C double bonds from exposure to HOCl, thereby inhibiting the oxidation of POPG at the air-water interface. These findings elucidate the molecular mechanisms by which OPEs modulate HOCl-induced phospholipid oxidation, providing a theoretical foundation for the development of specific drugs targeting related diseases.

Herein, we engineered a novel pH-responsive ratiometric assay by virtue of the composite system of Ce 3+ -enhanced glutathione-encapsulated copper nanoclusters (GSH-CuNCs-Ce 3+ ) and N-doped carbon quantum dots (N-CQDs). The GSH-CuNCs-Ce 3+ and N-CQDs were fabricated using one-pot chemical reduction and hydrothermal approaches, respectively, and their morphologies as well as physical and chemical properties were testified in detail by a series of characterization techniques. At 350 nm excitation wavelength, the GSH-CuNCs-Ce 3+ /N-CQDs system (GCC-NC) featured the 440/650 nm dual-emitting property. The ratios of fluorescence intensities (F 650 /F 440 ) demonstrated a strong pH-dependence from 3.0 to 5.0 with a coefficient of determination of 0.9982. Consequently, the GCC-NC system was feasible for constructing a pH-responsive dual-emitting fluoroprobe. Based on the catalytic effect of acetylcholinesterase on acetylcholine to yield acetic acid and further trigger varying solution pH, the as-constructed GCC-NC fluoroprobe was satisfactorily applied for detecting malathion . Under optimized conditions, the newly developed GCC-NC fluoroprobe supplied a wider linear range (0.25–200 μM), lower detection limit (0.075 μM), satisfactory recoveries (96.3–106.2%) and higher precision for malathion in several kinds of fruit juice samples. The fortified experiments by malathion’s structural analogues and metal ions provided compelling evidence that this fluoroprobe had strong anti-interference capacity and high specificity for malathion. These findings render us to believe that the as-constructed pH-responsive dual-emitting fluoroprobe holds great promise in trace malathion assay in food matrices.

Background Organophosphate flame retardants (OPFRs) are notorious emerging contaminants threatening the environment and human health. Triphenyl phosphate (TPHP), which has an extremely serious biotoxicity, is a typical harmful OPFR. Due to its wide use, TPHP has been discovered in various environmental mediums. Moreover, it is pretty recalcitrant to the removal process, resulting in the need for a technique to understand it better. Hence, accurate and quick discrimination of TPHP in the environment is critical to further evaluate its potential effect on ecosystems and human health. Results An ingenious oxidant-free chemiluminescence (CL) sensor based on the oxidant-free UV/MIL-100(Fe) gel system was established for TPHP detection. The oxidation of luminol in the UV-activated MIL-100(Fe) gel has resulted in remarkable CL emission, which is contributed by reactive oxygen species (ROS) generated by it. Notably, the CL intensity was inhibited significantly after introducing TPHP. An investigation into the mechanism underlying the effect of CL suppression demonstrated that TPHP competed with luminol to consume ROS from UV-activated MIL-100(Fe) gel, contributing to CL inhibition. The subsequent sensing performance experiments demonstrated the advantages of environmentally friendly, economic efficiency, user-friendly operation, rapid determination, potential for compact size, high selectivity, and sensitivity. Additionally, these investigations confirmed the low limit of detection (210 ng L −1 ) and wide linear range (10–1000 μg L −1 ). Significance In this paper, a green, economical, and oxidant-free CL sensing strategy for TPHP has been established. It has the advantage of being rapid, having the potential for compact size, high selectivity, and sensitivity. This ingenious method has promising applications in real-time and online environmental monitoring, and it paves the way for the rapid and environmentally friendly identification of emerging contaminants that are structurally stable and recalcitrant to remove.
