In the water system, antibiotic pollution significantly impacts the human body and the environment. Therefore, it is essential to quickly identify the types of antibiotics in the system and detect their concentration. It has been reported that many metal ions interact with antibiotics, and some of them can also change the enzyme-like catalytic properties of gold clusters (AuNCs). In the experiments, we found significant differences in the experimental results when different antibiotics and metal ions were placed in a TMB-H2O2system with AuNCs as catalysts. Based on this result, we devised a simple and sensitive colorimetric method for the simultaneous detection of multiple antibiotics using AuNCs-metal ions as the sensor, a multifunctional microplate detector as the detection instrument, and LDA as the analytical method. This method was successfully applied for the identification of antibiotics and the detection of their concentrations in river water. Keywords:gold nanocluster;antibiotic;colorimetric detection;linear discriminant analysis;multiplex detection

Utilizing the peroxidase (POD) activity of nanozymes to construct the colorimetric sensors is a popular strategy for assessing the antioxidant capacity of biological molecules, but the detection sensitivity is often compromised by their low enzymatic activity. Pt-based nanozymes, favored by their regulated nanostructures, well provide numerous binding sites for H 2 O 2 , and exhibit enhanced POD performance. Here, we utilized a simple hydrothermal method to synthesize PtTeNi nanomaterials featured with the ultrathin sheets for detecting biological antioxidant (ascorbic acid (AA) as the model). With the TMB-H 2 O 2 system as the colorimetric substrate, PtTeNi nanomaterial exhibited superior POD-like activity with a higher Vmax (5.02 × 10 −4 mM s −1 ) and lower Km (3.14 mM)) in contrast to natural HRP. AA significantly inhibited the colorimetric reaction of PtTeNi-TMB-H 2 O 2 system via competitively binding OH generated by PtTeNi-mediated H 2 O 2 catalysis and further blocking oxTMB formation. Inspired by this fact, the PtTeNi-TMB-H 2 O 2 colorimetric sensor successfully realized highly sensitive and selective detection of AA with a good linear range from 5 μM to 500 μM and the detection limit of 2.94 μM. For analyzing the total antioxidant capability and AA contents in actual samples (commercial drinks and vitamin C tablets), the PtTeNi-based colormetric method displayed comparable results with the commercial total antioxidant capacity assay kit, with the advantages of easy operation, convenient storage and high stability. This work fully affirms the excellent enzyme-like performance of Pt-based nanomaterial and confirms the feasibility of nanozymes-based colorimetric sensing strategy in detecting antioxidant capability of biological molecules.

The successful synthesis of ruthenium oxide/reduced graphene oxide (RuO 2 /rGO) heterostructures by one-pot hydrothermal method using graphene oxides and RuCl 3 as precursors is reported. The heterostructures had high peroxidase-like (POD-like) activities, which catalyzes the oxidation of classical peroxidase substrate 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of H 2 O 2 to create a blue colored reaction product. The catalytic activity was significantly enhanced by the synergistic effect between RuO 2 nanoparticles and rGO. RuO 2 /rGO had a low K m of 0.068 mM and a high v max of 1.228 × 10 −7 M·s −1 towards TMB in the TMB-H 2 O 2 catalytic oxidation system. In addition, the POD-like activity originating from the electron transfer mechanism was confirmed by cytochrome C (Cyt C) oxidation experiment. A colorimetric method based on RuO 2 /rGO heterostructures was developed with good sensitivity and selectivity for glucose detection with a limit of detection of 3.34 μM and a linear range of 0–1500 μM. The RuO 2 /rGO heterostructures have potential applications in the biomedical areas, such as biosensor and diagnostics. Graphical abstract

A novel colorimetric aptasensor has been developed for highly sensitive tetracycline (TC) detection based on the peroxidase-like activity of Fe 3 O 4 @Cu nanoparticles and “sandwich” oligonucleotide hybridization. The Fe 3 O 4 @Cu nanoparticles with high peroxidase-like activity were successfully synthesized under mild conditions. Then, a “sandwich” oligonucleotide hybridization probe (a short amino-modified complementary sequence of a portion of the TC aptamer (cDNA1), TC aptamers, and a long complementary to 5′ terminal TC aptamer sequence (cDNA2)) was created in 96-wells plates via DNA hybridization in the absence of TC from the detection system. The unique “sandwich” oligonucleotide hybridization probe adsorbed large numbers of Fe 3 O 4 @Cu nanozymes while further enhancing its peroxidase-like activity. Based on the 3,3′,5,5′-tetramethylbenzidine (TMB)-hydrogen peroxide (H 2 O 2 ) reporting system, the blue color of the solution decreased linearly with the increase of TC concentration, ranging from 10 −3 to 10 3 μg/L with an ultralow limit of detection (LOD) of 0.91 ng/L (2 pM). The proposed method was successfully applied to detect TC in spiked milk samples, with recoveries of 81.8 to 112%, demonstrating the excellent potential for highly sensitive TC detection in milk. In this work, a novel and ultrasensitive electrochemical biosensor was constructed for DNA detection based on functionalized gold clusters/graphene nanohybrids (AuNCs/GR nanobybrids) and exonuclease III (Exo III)-aided cascade target recycling. By utilizing the capacity of GR as universal template, different metal nanoclusters including AuNCs/GR nanobybrids and PtNCs/GR nanohybrids were synthesized through convenient ultrasonic method. Exo III-aided cascade recycling was initiated by target DNA, generating the final cleavage product (S2), which acted as a linkage between capture probe and the functionalized metal nanoclusters/GR conjugates in the construction of the biosensor. The AuNCs/GR-DNA-enzyme conjugates acted as interfaces of enzyme-catalyzed silver deposition reaction, achieving DNA detection ranging from 0.02 fM to 20 pM with a detection limit of 0.057 fM. In addition, PtNCs/GR-DNA conjugates presented peroxidase-like activity and the functionalized PtNCs/GR nanohybrids-based electrochemical biosensor also realized DNA detection by catalyzing the 3,3′,5,5′-tetramethylbenzidine-hydrogen peroxide (TMB-H 2 O 2 ) system to produce electrochemical signal. This metal clusters/GR-based multiple-amplified electrochemical biosensor provided an universal method for DNA detection.
