Quantitative fluorescence turn-on and colorimetric detection of glutathione (GSH) with rapid speed, low cost have attained much attention. Herein, we developed a sensitive fluorescence turn-on and colorimetric sensor for GSH based on the inner-filter effect (IFE), which is the first time to select oxTMB and TMPyP as the IFE absorber and fluorophore pair, respectively. The absorption band of oxTMB matches well with the emission band of TMPyP in the IFE-based fluorescent assay. In the absence of GSH, the absorption peak of oxTMB at 652 nm significantly overlaps with the emission of TMPyP, resulting in the efficient IFE and inhibition of the fluorescence of TMPyP. In the presence of GSH, the absorption intensity at 652 nm decreases, generating the recovery of the fluorescence of TMPyP. Therefore, this approach is demonstrated to be a novel candidate for detection of GSH, with high sensitivity and selectivity. The linear dynamic range for the concentrations of GSH is between 0.1 μM to 20 μM along with a limit of detection (LOD) of about 30 nM (calculated LOD as 3 σ /slope). Finally, this novel sensor was successfully applied for GSH detection in fetal calf serum , and satisfactory recovery was achieved.

Blood lead poisoning is a universal and severe health problem that greatly threatens human health in various industries. Elimination of blood lead relying on chelating agents and combination with hemoperfusion adsorbents has achieved considerable progress, but it is still suffering from the compromised selectivity of adsorbents as well as in absence of real time monitoring during treatment. Herein, we proposed a selective blood lead adsorbent integrated with real-time visualized/electrochemical bi-modal monitoring based on TMPyP-bound guanosine-borate (GB) supramolecular hydrogel as potential smart hemoperfusion device. The GB hydrogel possessed stability in physiological environment, self-healing ability resistant to fluid shear, blood compatibility, selective adsorption of lead ions superior to conventional adsorbents, anti-fouling performance to blood components and renewability. Benefiting from binding with TMPyP and the intrinsic conductivity, GB hydrogel was endowed with the ability to qualitatively diagnose the presence of blood lead via simple color change and quantitatively reflect the amount of adsorbed lead from blood accurately through electrochemical technique . This work puts forward an integrated treatment/monitoring hemoperfusion device with high selectivity, simple fabrication and low-cost, providing a paradigm for next generation design of intelligent, monitorable theranostic hemopurification system, which is also an extensible platform for the other research fields such as environmental monitoring and remediation.

We have developed a new fluorescent probe of thioglycolic acid (TGA)-capped CdTe quantum dots (QDs) complexed with a model drug, meso -tetrakis( N -methylpyridinium-4-yl)porphyrin (TMPyP) for detecting deoxyribonucleic acids (DNAs). This probe operates with an “Off–On” mode: TMPyP quenches the photoluminescence (PL) of QDs via a photo induced electron-transfer (PIET) process; the presence of DNA can break the QD/TMPyP complexation, interrupting the PIET process, and switch on the PL of QDs. Sensitive detection of DNA with the detection limit of 0.16 nM and a linear detection range of 0.25–6.0 nM are achieved. Importantly, this probe can be used to distinguish the binding modes of DNA–TMPyP interactions, exhibiting the DNA sequence-dependent PL recovery behaviors. The obtained binding constant for poly(dA)·poly(dT) is ∼3.30 × 10 7 L mol −1 , which is approximately one order of magnitude larger than those for native DNAs and poly(dG)·poly(dC). Furthermore, the thymine bases preferential of the TMPyP–DNA interaction is proved by this probe.

Rational utilization of the competition among internal functional groups in chemical adsorbents for heavy metal ions can significantly enhance adsorption efficiency, while also driving the development of advanced adsorbents. This study proposes an innovative strategy employing a group competition mechanism to develop a dual-function platform for Cd(II) detection and adsorption, along with field detection applications. Nitrogen-doped graphene quantum dots (NGQDs) anchor the Cd(II) detection agent, 5,10,15,20-tetrakis(1-methyl-4-pyridinio)porphyrin tetra( p -toluenesulfonate) (TMPyP), onto sodium alginate chains to form an aerogel (T-NGQDs-CAA). FT-IR and XPS characterizations confirm that NGQDs facilitate a rapid Cd(II) transfer pathway through electrostatic interactions and π-π stacking. The Cd(II) detection speed of T-NGQDs-CAA is enhanced fivefold, while density functional theory analysis reveals that TMPyP exhibits a significantly stronger binding affinity toward Cd(II) compared to nitrogen- and oxygen-containing groups. Combined experimental and theoretical analyses demonstrate that this rapid ion transfer pathway intensifies TMPyP’s competition for Cd(II), thereby promoting the coordination reaction between TMPyP and Cd(II). Additionally, the Cd(II) adsorption by T-NGQDs-CAA follows the pseudo-second-order kinetic and Langmuir isotherm models, exhibiting a 148.7% increase in saturation adsorption capacity. This strategy, through constructing rapid ion channels with NGQDs and leveraging the group competition mechanism, enables in-situ Cd(II) detection and removal, enhancing the understanding and application of interactions between functional groups and metal ions.

Pathogenic bacterial infections associated with wound healing progress usually result in serious complications. Herein, biocompatible and antimicrobial electrospun nanofibrous mats with photodynamic therapy (PDT) effect were fabricated to accelerate the infected wound healing. The nanofibrous mats were fabricated by co-electrospining of polyanionic poly(γ-glutamic acid) (γ-PGA) and cationic photosensitizer 5,10,15,20-tetrakis(1-methylpyridinium-4-yl)porphyrin tetra (p-toluenesulfonate) (TMPyP) in aqueous solution and stabilized by the chemical crosslinking. The as-prepared nanofibrous mats can not only confer the moist microenvironment to the wound bed, but also provide potent bactericidal activity upon visible light irradiation by releasing the cytotoxic reactive oxygen species (ROS). The antibacterial assay in vitro showed that they can effectively eradicate the board-spectrum bacteria at a relatively low loading dose of TMPyP ( e.g. , 0.1 wt%). Meanwhile, those nanofibrous mats showed good biocompatibility with no obvious adverse effects on mammalian cells and red blood cells (RBCs). The animal test in vivo suggested that the restrained inflammatory reaction and better wound healing could be achieved upon timely and effective antibacterial treatment with negligible local toxicities. This biocompatible and antibacterial γ-PGA-TMPyP nanofibrous mat may show great potential in practical infection-resistant applications, particularly for wound dressing applications.

ABSTRACT Bacterial infections are among the most critical global health challenges that seriously threaten the security of human. To address this issue, a biocompatible engineered living hydrogel patch was developed by co-embedding engineered photothermal bacteria (E M ), photosensitizer (porphyrin) and reactive oxygen species amplifier (laccase) in a protein hydrogel. Remarkably, the genetice engineered bacteria can express melanin granules in vivo and this allows them to exhibit photothermal response upon being exposed to NIR-II laser (1064 nm) irradiation. Besides, electrostatically adhered tetramethylpyridinium porphyrin (TMPyP) on the bacterial surface and encapsulated laccase (Lac) in protein gel can generate highly toxic singlet oxygen ( 1 O 2 ) and hydroxyl radical (·OH) in the presence of visible light and lignin, respectively. Interestingly, the engineered bacteria hydrogel patch (E M TL@Gel) was successfully applied in synergistic photothermal, photodynamic and chemodynamic therapy, in which it was able to efficiently treat bacterial infection in mouse wounds and enhance wound healing. This work demonstrates the concept of “fighting bacteria with bacteria” combining bacterial engineering and material engineering into an engineered living hydrogel path that can synergistically boost the therapeutic outcome. Statement of Significance Genetically engineered bacteria produce melanin granules in vivo, exhibiting remarkable photothermal properties. These bacteria, along with a photosensitizer (TMPyP) and a reactive oxygen species amplifier (laccase), are incorporated into a biocompatible protein hydrogel patch. Under visible light, the patch generates toxic singlet oxygen ( 1 O 2 ) and hydroxyl radical (·OH), demonstrates outstanding synergistic effects in photothermal, photodynamic, and chemodynamic therapy, effectively treating bacterial infections and promoting wound healing in mice.
