We established a method of determining 13 trace elements (TEs) in whole blood, determined the levels of the TEs in patients with colorectal cancer (CRC), and examined the correlations between the TEs and circulating tumor cells (CTCs). A total of 93 CRC patients and 48 control subjects in Guangzhou, China, were included. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure the blood levels of 13 TEs. CTCs of 13 CRC patients were examined using CanPatrol™. The levels of Mg, Fe, Zn, Se, Sr, Mo, and Ba in CRC patients were lower than those in control subjects (all, P < 0.05). The levels of Mg, Fe, Zn, Se, and Ba decreased with increasing TNM stage. The reductions of Zn and Se levels were higher in early-stage than late-stage CRC patients. The levels of Cu and Ni were higher in CRC patients than in control subjects (both, P < 0.05). The Cu level increased with increased TNM stage, and Ni level was higher in early-stage CRC patients as compared with late-stage CRC patients. Consequently, the TE levels showed differences as the process of TNM stage of CRC patients. There were correlations between the amount of CTCs and epithelial-CTCs (E-CTCs) and interstitial-CTCs (M-CTCs) ( P < 0.05). The levels of Mg, Cr, Fe, Cu, Zn, Se, Sr, and Ba were correlated with the number of CTCs ( P < 0.05). Specific TE patterns in whole blood may assist in the early diagnosis of CRC. The combined determination of TE levels and CTCs may be useful as an adjunct to staging and guiding treatments.

Background Cholangiocarcinoma is a common malignant tumor of digestive system. LncRNA metallothionein 1 J, pseudogene (MT1JP) has been reported to play tumor-suppressing roles in multiple cancers. However, its effect on cholangiocarcinoma has not been evaluated. Methods The expression of MT1JP in intrahepatic cholangiocarcinoma specimens and paired para-carcinoma tissues were detected by real-time PCR. The overexpression plasmid and siRNA of MT1JP were transfected into intrahepatic cholangiocarcinoma cells to change the expression levels of MT1JP. CCK-8, flow cytometry and transwell assays were performed to measure proliferation, cell cycle transition, apoptosis, migration and invasion. Dual-luciferase reporter assay, real-time PCR and western blot were carried out to screen the miRNA bound by MT1JP. In addition, xenograft experiment was used to determine the tumorigenesis of cholangiocarcinoma cells in nude mice. Results MT1JP was downregulated in intrahepatic cholangiocarcinoma specimens, and its expression was related with TNM stage and lymph node metastasis. Overexpression of MT1JP inhibited proliferation, cell cycle transition, migration and invasion, and induced apoptosis in intrahepatic cholangiocarcinoma cells. The knockdown of MT1JP led to opposite results. MT1JP bound to miR-18a-5p to facilitate the expression of fructose-1,6-bisphosphatase 1 (FBP1). MiR-18a-5p was increased in intrahepatic cholangiocarcinoma samples, and its expression was negatively correlated with that of MT1JP. In addition, MT1JP also suppressed tumorigenesis in nude mice. Conclusions MT1JP alleviated proliferation, migration and invasion, and induced apoptosis in cholangiocarcinoma cells by regulating miR-18a-5p/FBP1 axis. These findings may provide novel insights for clinical diagnosis and treatment of cholangiocarcinoma.

SLC16A3/monocarboxylate transporter 4 (MCT4) regulates intracellular lactate transport and is highly expressed in many tumors, indicating poor prognosis. It may be related to inducing hypoxia, apoptosis and other mechanisms, but the study of MCT4 in HCC is far from complete. In this study, we first analyzed the expression of SLC16A3 in HCC tumor and non-tumor tissue samples based on TCGA data and immunohistochemistry. Subsequently, the effects of SLC16A3 expression on cell proliferation and invasion were analyzed using hepatocellular carcinoma (HCC) lines, and Western blot (WB) analysis was performed to explore the changes in pathway proteins and ferroptosis proteins. Finally, the drug sensitivity was tested by CCK8 kit. We found that SLC16A3 was significantly upregulated in tumor tissues, and was significantly correlated with TNM stage, histological grade, and macrovascular invasion. TCGA data and WB analysis showed that the high expression of SLC16A3 induced hypoxia, and knockdown could reverse hypoxia and inhibit ERK phosphorylation, thus limiting the malignant behavior of HCC cells. Moreover, knockdown of SLC16A3 significantly increased the level of lipid peroxidation and reactive oxygen species (ROS), while the expressions of GPX4, DHODH and SLC7A11 were inhibited. The expression of SLC16A3 affected the sensitivity of HCC cells to chemotherapy and targeted drugs, and RNA sequencing data suggested that the expression level influenced tumor microenvironment and response to immunotherapy. So, we draw a conclude that SLC16A3 is associated with poor prognosis of HCC. Inhibition of SLC16A3 expression is a potential therapeutic target for HCC.
