CDs–MnO2–TPPS Ternary System for Ratiometric Fluorescence Detection of Ascorbic Acid and Alkaline Phosphatase

Hongzhi Lu,Shoufang Xu

Manganese dioxide (MnO2) nanosheet-based fluorescence sensors often use oxidase-like activity or wide absorption spectrum for detection of antioxidants. In those strategies, MnO2 nanosheets were reduced to Mn2+ by antioxidants. However, few strategies emphasize the role of Mn2+ obtained from MnO2 reduction in the design of the fluorescence sensor. Herein, we expanded the application of a MnO2 nanosheet-based fluorescence sensor by involving Mn2+ in the detection process using ascorbic acid (AA) as a model target. In this strategy, carbon dots (CDs), MnO2 nanosheets, and tetraphenylporphyrin tetrasulfonic acid (TPPS) comprise a ternary system for ratiometric fluorescence detection of AA. Initially, CDs were quenched by MnO2 nanosheets based on the inner filter effect, while TPPS maintained its fluorescence intensity. After the addition of AA, MnO2 nanosheets were reduced to Mn2+ so that the fluorescence intensity of CDs was recovered and TTPS was quenched by coordination with Mn2+. Overall, AA triggered an emission intensity increase at 440 nm for CDs and a decrease at 640 nm for TPPS. The ratio intensity of CDs to TPPS (F440/F640) showed a good linear relationship from 0.5 to 40 μM, with a low detection limit of 0.13 μM for AA detection. By means of the alkaline phosphatase (ALP)-triggered generation of AA, this strategy can be applied for the detection of ALP in the range of 0.1–100 mU/mL, with a detection limit of 0.04 mU/mL. Furthermore, this sensor was applied to detect AA and ALP in real, complex samples with ideal recovery. This novel platform extended the application of MnO2 nanosheet-based fluorescence sensors.

Hollow Polymeric Capsules from POSS-Based Block Copolymer for Photodynamic Therapy

Zhenghe Zhang,Yudong Xue,Pengcheng Zhang,Axel H. E. Müller,Weian Zhang

A novel amphiphilic diblock copolymer, PHEMAPOSS-b-P(DMAEMA-co-CMA), was prepared via reversible addition–fragmentation chain transfer (RAFT) polymerization, where PHEMAPOSS block was first synthesized using a methacrylate monomer based on polyhedral oligomeric silsesquioxane (HEMAPOSS), and PHEMAPOSS was further utilized to prepare the block copolymer via RAFT copolymerization of 2-(dimethylamino)ethyl methacrylate (DMAEMA) and reduction-cleavable coumarin methacrylate (CMA) monomer. PHEMAPOSS-b-P(DMAEMA-co-CMA) could self-assemble in water to form spherical micelles with POSS core and stimuli-responsive shell. The micelles were cross-linked by photodimerization of coumarin, and then hollow polymeric capsules could be finally obtained via etching the POSS core in the solution of hydrofluoric acid (HF). The morphologies of the micelles and hollow polymeric capsules were well characterized by TEM, SEM, and DLS. The hollow polymeric capsules are responsive to typical physiological stimuli such as pH, and redox potential, and could be further utilized in the encapsulation and release of tetraphenylporphyrin tetrasulfonic acid hydrate (TPPS) for photodynamic therapy (PDT). The in vitro release of TPPS-loaded polymeric capsules allowed a relatively low TPPS release at pH = 7.4. However, a burst release of TPPS was observed in the presence of 10 mM glutathione (GSH) at pH = 5.5. Confocal laser scanning microscopy (CLSM) confirmed that TPPS-loaded polymeric capsules could well improve the internalization rate in MCF-7 cells. According to the result of MTT assay, TPPS-loaded polymeric capsules demonstrated efficient PDT efficacy and low dark toxicity toward MCF-7 cells. Thus, TPPS-loaded polymeric capsules have presented potential application in PDT.

Analyzing urinary hippuric acid as a metabolic health biomarker through a supramolecular architecture

Wenhui Li,Hua Dong,Kejing Niu,Huan-Yu Wang,Wenqian Cheng,Hualong Song,An-Kang Ying,Xiaobing Zhai,Kefeng Li,Huijuan Yu,Dong-Sheng Guo,Yuefei Wang

The prevalence of metabolic disorders has been found to increase concomitantly with alternations in habitual diet and lifestyle, indicating the importance of metabolic health monitoring for early warning of high-risk status and suggest effective intervention strategies. Hippuric acid (HA), as one of the most abundant metabolites from the gut microbiota, holds potential as a regulator of metabolic health. Accordingly, it is imperative to establish an efficient, sensitive, and affordable method for large-scale population monitoring, revealing the association between HA level and metabolic disorders. Upon systematic screening of macrocycle•dye reporter pair, a supramolecular architecture (guanidinomethyl-modified calix [5] arene, GMC5A) was employed to sense urinary HA by employing fluorescein (Fl), whose complexation behavior was demonstrated by theoretical calculations, accomplishing quantification of HA in urine from 249 volunteers in the range of 0.10 mM and 10.93 mM. Excitedly, by restricted cubic spline, urinary HA concentration was found to have a significantly negative correlation with the risk of metabolic disorders when it exceeded 0.76 mM, suggesting the importance of dietary habits, especially the consumption of fruits, coffee, and tea, which was unveiled from a simple questionnaire survey. In this study, we accomplished a high throughput and sensitive detection of urinary HA based on supramolecular sensing with the GMC5A•Fl reporter pair, which sheds light on the rapid quantification of urinary HA as an indicator of metabolic health status and early intervention by balancing the daily diet.

Bioinspired electron carrier mediated transmembrane photocatalytic hydrogen evolution in silica colloidosomes

Chengkun Bai,Bingdi Wang,Zhengshun Jiang,Chunying Lv,Zhenning Liu,Shiyu Wang,Song Liang,Hongying Zang

Inspired by the ordered processes of chloroplasts in photosynthesis involving coordinative linkages and efficient compartmentalization, a bioinspired compartmentalized photocatalytic system (BCPS) with an inorganic compartment is designed for photocatalytic hydrogen evolution with light driven electron carrier mediated cross-membrane energy transport. A typical BCPS comprises silica colloidosomes containing co-catalysts as microreactors, where positively charged N-methyl-4-cyanopyridinium (MCP+) serves as the electron carrier. The MCP+ molecules harvest energy from photoexcited g-C3N4, and transit to electrically neutral species MCP0, which can pass the colloidosome. Driven by light, the MCP0 diffuse across the membrane and transport electrons to H+ for hydrogen evolution, in which Pt particles are employed as co-catalysts. Analogous to natural photosynthesis, hydrogen evolution occurs within compartments, separating light and dark reactions, protecting the dark reaction from interference during light harvesting and minimizing potential side effects. The system enables sustained evolution of hydrogen with a higher yield than that with the control catalysis in a bulk catalyst suspension. The dramatic influence of SDS on the hydrogen evolution was revealed, and a possible mechanism of electron carrier amount buffering was proposed.

Peroxynitrite scavenger FeTPPS binds with hCT to effectively inhibit its amyloid aggregation

Bin Xiao,Junhao Xiao,Sisi Liu,Xiaoying Xiao,Shengping Dai,Yan Sui,Jinming Wu,Huixian Ye

Human calcitonin (hCT) is an endogenous polypeptide commonly employed in treating bone resorption-related illnesses, but its clinical application is limited due to its high aggregation tendency. Metalloporphyrins are effective in suppressing amyloid fibrillation, positioning them as potential drug candidates for amyloidogenic disorders like Alzheimer's and type 2 diabetes. In this work, we investigated the effects of Fe(III) meso-tetra(4-sulfonatophenyl)porphine chloride (FeTPPS), a highly efficient ONOO− decomposition catalyst, on hCT aggregation. Our findings reveal that FeTPPS effectively precludes hCT fibrillation by stabilizing the monomers and delaying the structural transition from α-helix bundles to β-sheet-rich aggregates. The macrocyclic ring of FeTPPS plays a significant role in disrupting hCT self-associations. Among various porphyrin analogs, those with an iron center and negatively charged peripheral substituents exhibit a stronger inhibitory effect on hCT aggregation. Spectroscopic analyses and computational simulations indicate that FeTPPS binds to hCT's core aggregation region via complexation with His20 in a 1 : 1 molar ratio. Hydrophobic interaction, hydrogen bonding, and π–π stacking with the residues involving Tyr12, Phe19, and Ala26 also contribute to the interactions. Collectively, our study provides a promising approach for developing novel hCT drug formulations and offers theoretical guidance for designing metalloporphyrin-based inhibitors for various amyloidosis conditions.

Interfacial Chirality Transfer Induced by Vortices inside Microdroplets

Tianshun Ding,Shi Feng,Hao Li,Zelong Zhang,Shengyang Tao

Chiral materials have broad applications in optics. Precise regulation of the chiral signal, however, remains highly challenging. Herein, a microfluidic strategy is proposed in which the cooperative action of dual driving forces regulates vortical flow inside droplets. The flow rate and interfacial tension are adjusted to induce ordered Marangoni convection and shear flow within microdroplets. Under these conditions, chiral signals are transmitted both within droplets and across the liquid-liquid interface. Tetrakis(4-sulfonatophenyl)porphyrin and phenethylamine are employed as a supramolecular co-assembly pair to probe the relation between vortex strength/distribution and chiral-signal transfer. Assemblies obtained under microdroplet vortex fields exhibit markedly enhanced and nearly mirror-image circular dichroism (CD) responses in comparison with those prepared under conventional magnetic stirring. These observations confirm that vortical flow efficiently promotes chiral amplification and enantioselective supramolecular assembly. Flow-field visualization by micro-particle image velocimetry (μ-PIV), combined with hydrodynamic analysis, reveals the formation mechanism and evolution of microvortices under the dual driving forces. Overall, this study demonstrates a practical approach to modulating supramolecular chiroptical signals via vortexes in confined environments.

SERS and Chiral Properties of Cinnamic Acid Derivative Langmuir-Blodgett Films Complexed with Dyes

Xingdi Zhao,Xinyu Li,Pengfei Bian,Qingrui Zhang,Yuqing Qiao,Mingli Wang,Tifeng Jiao

Chiral molecules are crucial in the field of optical devices, molecular recognition, and other novel functional materials due to their unique spatially asymmetric configuration and optical activity. In this study, a chiral molecule, Cholest-3-yl (E)-3-(4-carbamoylphenyl)acrylate (CCA), was combined with dyes containing large conjugated structures, tetramethylporphyrin tetrasulfonic acid (TPPS), and Nickel(II) phthalocyanine-tetrasulfonic acid tetrasodium salt (TsNiPc), and composite LB films of CCA/TPPS and CCA/TsNiPc were successfully prepared by using Langmuir-Blodgett (LB) technology. The circular dichroism (CD) test proved that the CCA/TPPS composite film had a strong CD signal at 300–400 nm, and the composite film showed chirality. This significant optical activity provides a new idea and option for the application of LB films in chiral sensors. In the Surface Enhanced Raman Spectroscopy (SERS) test, the CCA/TPPS composite film was sensitive to signal sensing, in which the enhancement factorEF= 2.28 × 105, indicating that a large number of effective signal response regions were formed on the surface of the film, and the relative standard deviation (RSD) = 12.08%, which demonstrated that the film had excellent uniformity and reproducibility. The high sensitivity and low signal fluctuation make the CCA/TPPS composite LB film a promising SERS substrate material.

A Redox Stimulation-Activated Amphiphile for Enhanced Photodynamic Therapy

Yudong Xue,Jia Tian,Zhiyong Liu,Jianbo Chen,Mengsi Wu,Yongjia Shen,Weian Zhang

The development of more efficient photosensitizers with minimal damage to surrounding normal tissues has been a valuable and challenging subject during photodynamic therapy (PDT). Herein, a stimuli-activated porphyrinic photosensitizer (PEG-TPP-DNB; PEG = poly(ethylene glycol); TPP = 5,10,15,20-tetraphenylporphyrin; DNB = 2,4-dinitrobenzene) with capabilities of fluorescence and, remarkably, singlet oxygen quenching was prepared successfully for photodynamic therapy with high efficiency and biosecurity. The amphiphilic PEG-TPP-DNB could be self-assembled into nanomicelles in aqueous media and dissociated in response to reductive thiol such as glutathione. Meanwhile, the fluorescence and singlet oxygen generation of porphyrinic photosensitizer would be activated to regenerate. Moreover, the intracellular uptake and localization effectively confirmed the redox-responsive and activated behavior of PEG-TPP-DNB micelles. The cytotoxicity in vitro revealed that the micelles had low dark toxicity and great phototoxicity, and in vivo bioimaging and antitumor evaluation further indicated that the micelles possessed selective tumor imaging and targeted PDT antitumor effect as well as low systemic toxicity. Overall, this tumor microenvironment-activated photosensitizer system may provide a useful strategy for precise photodynamic therapy.

Interfacial aggregation behavior of novel carbazole-based composite Langmuir-Blodgett films for photoelectric conversion and catalytic performance

Pengfei Bian,Na Li,Guohua Li,Senlin Zhang,Xiujuan Liu,Jianmin Gu,Bo Liu,Tifeng Jiao

The development of multifunctional film materials from novel organic small molecules has attracted the attention of researchers. In this study, novel carbazole organic molecules (CS-14, CS-38 and CS-39) were successfully composited with TsNiPc and TPPS subphases into multilayer films by LB technology. The nanostructure and morphology of the composite films were successfully assembled and analyzed by scanning electron microscopy (SEM) and atomic force microscopy (AFM). In addition, the stability of the composite film was proved by UV–vis spectroscopy and Fourier transform infrared spectroscopy . The results demonstrate that Langmuir films are transferred to the surface of substrates in different aggregated morphologies. In addition, the self-assembled LB film electrodes exhibit excellent photoelectrochemical performance. The photoelectric conversion of the multilayer films transferred to ITO (indium tin oxide) was tested, and CS-38/TsNiPc Langmuir film exhibits a stronger photoelectric conversion effect. The electrolysis reaction of LB films to water was also tested. Further analysis shows that the composite films with TsNiPc as the subphase all showed better catalytic performance than the composite films with TPPS as the subphase. Therefore, the prepared LB film has the potential to become an optoelectronic functional ultrathin film device.
