Endoplasmic reticulum chaperone GRP78 participates in fluoride-induced autophagy in LS8 cells by regulating the IRE1-TRAF2-JNK pathway

Lin Zhao,Liyuan Wang,Han Wang,Tao Xi,Sijia Liu,Yang Li,Jianping Ruan,Yongqing Huang

Fluoride induces endoplasmic reticulum (ER) stress in ameloblasts, which is responsible for enamel mineralization disorder. Fluoride induces autophagy in ameloblasts, but the molecular mechanisms through which ameloblasts respond to fluoride-induced cellular stress and autophagy remain unclear. This study investigated ER stress-induced autophagy and the regulatory role of the ER molecular chaperone GRP78 in fluoride-induced autophagy in ameloblast LS8 cells. To explore the relationship between fluoride-induced ER stress and autophagy, we assessed changes in fluoride-induced autophagy in LS8 cells following overexpression and/or silencing of the ER stress molecular chaperone GRP78. We found that autophagy induced by fluoride was further increased after GRP78 overexpression in LS8 cells. Fluoride-induced autophagy was reduced in GRP78-silenced LS8 cells. Furthermore, we found that ER stress can regulate autophagy in fluoride-treated ameloblasts (LS8 cells) and that the GRP78/IRE1/TRAF2/JNK pathway is involved in the underlying regulation. Our study suggests that ER stress plays a role in fluoride-induced damage by inducing ameloblast autophagy.

PKCζ phosphorylates TRAF2 to protect against intestinal ischemia–reperfusion–induced injury

Zhou Wei,Yao Jihong,Wang Guangzhi,Chen Zhao,Li Zhenlu,Feng Dongcheng,Li Yang,Qasim Wasim,Tan Wenzhi,Ning Shili,Tian Xiaofeng

Intestinal ischemia–reperfusion (I/R) is a common clinical problem that occurs during various clinical pathological processes. Excessive apoptosis has an indispensable role in intestinal I/R injury. Tumor necrosis factor receptor-associated factor 2 (TRAF2) and PKC ζ have an essential role in apoptosis. Here, we aimed to investigate the effects of PKC ζ and TRAF2 and to explore the correlation between PKC ζ and TRAF2 in intestinal I/R injury. Mice were subjected to intestinal I/R injury in vivo . In vitro experiments were conducted by treating Caco-2 cells with hypoxia/reoxygenation (H/R) stimulation to simulate intestinal I/R. Intestinal tissue samples and Caco-2 cells were examined using various approaches. Intestinal I/R induced the membrane translocation and phosphorylation of PKC ζ . Pretreatment with the PKC ζ activator phosphatidylcholine remarkably attenuated gut injury by suppressing apoptosis. H/R induced PKC ζ to combine with TRAF2, which was phosphorylated by PKC ζ at Ser 55 , but not at Ser 11 , under intestinal I/R or H/R conditions. In addition, TRAF2 Ser 55 phosphorylation increased cell survival by inhibiting cell apoptosis in the H/R model. Mechanistically, TRAF2 Ser 55 phosphorylation promoted NF- κ B activation but suppressed c-Jun activation in Caco-2 cells under H/R conditions. The results of this study demonstrate that the PKC ζ /TRAF2 pathway represents a novel protective mechanism against intestinal I/R injury. Therefore, the PKC ζ /TRAF2 pathway is a novel target for potential treatments of intestinal I/R injury-related diseases.

Ginsenoside Rb1 reduced ischemic stroke-induced apoptosis through endoplasmic reticulum stress-associated IRE1/TRAF2/JNK pathway

Wei Liangli,Yuan Yuqi,Yang Ziteng,Li Yuqing,Wang Tingting,Hu Shenglin,Cai Biao,Wang Guangyun

The neuroprotective function of ginsenoside Rb1 (GRb1) in cerebral ischemia–reperfusion (I/R) was lately emphasized. However, whether GRb1 plays a regulatory role on endoplasmic reticulum (ER) stress-associated pathway in cerebral I/R damage is still unclear. The aim of this study is to explore the function of GRb1 in cerebral ischemia-induced ER stress and the underlying mechanism related to IRE1/TRAF2/JNK pathway. Longa method, cerebral infarct volume, and HE staining were used to evaluate the efficacy of GRb1 in mice with a mouse model of middle cerebral artery occlusion reperfusion (MCAO/R). We also investigated the effect and mechanism of GRb1 against ischemic stroke using in vitro oxygen–glucose deprivation reperfusion (OGD/R) model. We found that GRb1 could improve neurological scores, infarct volume, and histological injury in ischemic mice. Ischemic attack also activated neuronal apoptosis and ER stress, and this effect was attenuated by GRb1. In addition, GRb1 significantly reduced I/R-induced IRE1-TRAF2 interaction, IRE1, and JNK phosphorylation. The present study also confirmed that GRb1 significantly improved OGD/R-induced PC12 cells injury. GRb1 could decrease ER stress in OGD/R-injured PC12 cells, which was reflected by the decreased expression of GRP78 and CHOP. The ER stress inducer tunicamycin partially prevented the effects of GRb1 on cell viability, ER stress, and apoptosis after OGD/R, whereas the ER stress inhibitor 4-PBA exerted the opposite effect. Moreover, GRb1 markedly decreased IRE1-TRAF2 interaction, IRE1, and JNK phosphorylation in the presence of OGD/R insult. Furthermore, JNK inhibitor SP600125 and IRE1 inhibitor DBSA pretreatment further promoted the inhibition of GRb1 on ER stress induction and cell damage induced by OGD/R. Molecular docking further elucidated that the mechanism by which GRb1 improves cerebral ischemia maybe related to its direct binding to the kinase domain of IRE1, which in turn inhibited the phosphorylation of IRE1. Collectively, these results demonstrated that GRb1 reduced ischemic stroke-induced apoptosis through the ER stress-associated IRE1/TRAF2/JNK pathway and GRb1 has the potential as a protective drug for the treatment of cerebral ischemia.

C1QBP Drives M2 Macrophage Polarization Via TRAF2-CCL2 to Promote Oral Squamous Cell Carcinoma Progression

Xiyuan Qin,Kai Kang,Bowen Zhu,Yujie Shi,Shoushan Bu

Introduction and aims Oral squamous cell carcinoma (OSCC), frequently observed in the head and neck region, possesses mechanisms that have yet to be thoroughly clarified. Poor prognosis in OSCC is closely linked to the polarization of macrophages toward the M2 phenotype. Complement component 1q subcomponent-binding protein (C1QBP), a versatile protein commonly dysregulated in OSCC, has an unclear association with M2 macrophages. Methods Immunohistochemical (IHC) staining was conducted to determine the relationship between C1QBP expression and M2 macrophage infiltration. Flow cytometry (FC) and enzyme-linked immunosorbent assay (ELISA) were employed to explore the functional role of C1QBP in macrophages polarization. Furthermore, RNA sequencing combined with quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting (WB) were utilized to verify the increased expression of TNF receptor-associated factor 2 (TRAF2) and C-C motif chemokine 2 (CCL2). Additional validation of these findings was performed through in vivo experiments. Results A negative correlation was identified between C1QBP expression and CD206, a representative marker of M2 macrophages. Decreased levels of C1QBP in OSCC cells enhanced macrophage polarization toward the M2 phenotype and facilitated tumor progression by activating pivotal elements of the tumor necrosis factor (TNF) signaling pathway, notably TRAF2 and CCL2. Conclusion These findings suggest that C1QBP modulates the polarization of macrophages toward the M2 phenotype via the TRAF2-CCL2 signaling axis, thus contributing to OSCC malignancy progression. Consequently, targeting C1QBP to regulate M2 macrophage polarization may provide a promising therapeutic avenue for OSCC management.

M2 Macrophages Attenuate AQP2⁺ Collecting Duct Cell Apoptosis via the TRAF1–TRAF2 Complex to Suppress Randall's Plaque Formation

Liang Tang,Zhangcheng Liao,Meng Gao,Minghui Liu,Yongchao Li,Jian Wu,Hao Yu,Cheng He,Feng Zeng,Jinbo Chen,Hequn Chen,Zewu Zhu

Calcium oxalate (CaOx) kidney stones are common and recur frequently, yet effective pharmacological prevention is limited. Randall’s plaques (RPs), calcium deposits on renal papillae, act as anchoring sites for CaOx crystal growth, but the cellular origin and immunoregulatory mechanism underlying early calcium deposition remain unclear. Here, we found that early calcium deposition was partially localized to the renal interstitium surrounding aquaporin 2 (AQP2)-labeled collecting duct cells with apoptotic phenotype, and inhibiting high calcium-induced apoptosis of AQP2+ cells markedly reduced cell layer calcium deposition. Given the regulatory role of macrophages in renal stone formation, we revealed that M2 macrophages protected AQP2+ cells by delivering TRAF2 via exosomes. Mechanistically, exosomal TRAF2 interacted with intracellular TRAF1 to form a stable complex, which mutually inhibited their ubiquitin-mediated degradation by excluding the shared E3 ubiquitin ligase CBLC. This complex activated downstream nuclear factor κB1 (NF-κB1) and NF-κB2 signaling. NF-κB1 enhanced TRAF1 transcription, whereas NF-κB2 increased BCL2 expression while suppressing BAX and cleaved-PARP1, thereby limiting apoptosis and subsequent calcium deposition. Based on these findings, we developed M2 exosome-loaded, AQP2+ cell membrane-coated poly (lactic-co-glycolic acid) nanoparticles (Exo@A-P) for renal-targeted delivery. Exo@A-P treatment reduced collecting duct apoptosis and calcium deposition in hypercalciuria and renal calcified mice (Umod−/−) without detectable toxicity, potentially supporting a targeted nanotherapeutic strategy to prevent early RP formation by utilizing TRAF2-rich exosome from M2 macrophages.

Ketamine enhances autophagy and endoplasmic reticulum stress in rats and SV-HUC-1 cells via activating IRE1-TRAF2-ASK1-JNK pathway

Yanming Yu,Daoxu Wu,Yongwei Li,Hui Qiao,Zhengfei Shan

Background Ketamine-related cystitis (KC) has been researched in many clinical studies, but its exact mechanism is ambiguous and needs further research. Methods We established a KC rat model and analyzed physiological, biochemical, and urodynamic parameters of ketamine (KET)-related bladder injury. Bladder histologic feature, reactive oxygen species (ROS), autophagy-, apoptosis-, and endoplasmic reticulum stress (ERS)-related markers were examined by hematoxylin and eosin staining, Masson staining, ROS kit, quantitative real-time polymerase chain reaction, and western blot. In vitro, effects of 0.01, 0.1, and 1 mM KET on cell vitality, apoptosis, ROS level, autophagy-, apoptosis-, and ERS-related markers were examined again. Effects of KET-1 and salubrinal on complex formation, autophagy-, apoptosis-, and ERS-related markers were examined by Co-Immunoprecipitation and western blot. After transfection with shIRE1, complex formation, cell biological behaviors, ROS level, autophagy-, apoptosis-, and ERS-related markers were examined again. Results KET induced bladder hyperactivity and injury. KET facilitated urinary frequency, ROS production, and induced bladder histologic injury by activating autophagy-, apoptosis-, and ERS-related markers in rats. In vitro, KET (0.01, 0.1, and 1 mM) restrained cell vitality and elevated apoptosis and ROS level via activating autophagy-, apoptosis-, and ERS-related markers. Moreover, salubrinal reversed the promotion of KET-1 on complex formation, autophagy-, apoptosis-, and ERS-related marker expressions. After transfection with shIRE1, shIRE1 weakened complex formation induced by KET-1, and the effects of KET-1 on cells were offset by shIRE1. Conclusion KET enhanced autophagy and ERS in vivo and in vitro via restraining IRE1-TRAF2-ASK1-JNK pathway.
