Acetaminophen, also known as paracetamol (APAP), is a commonly used over-the-counter medication that is often used to treat headaches, toothaches, joint pain, muscle pain, and to lower body temperature. However, overdose can lead to liver damage, gastrointestinal distress, kidney damage, and cardiovascular disease. Therefore, it is very important to establish a method to quickly detect APAP. A novel “ON-OFF-ON” colorimetric and fluorescence dual-signal sensing system was constructed for the quantitative detection of APAP based on 5,10,15,20-tetrakis(4-sulphonatophenyl) porphyrin (TSPP) dual-signal probe. The absorbance and fluorescence intensity of TSPP respectively were quenched when Fe 3+ was introduced into TSPP solution. At this point, the color of the corresponding solution changed from red to green. The absorbance and fluorescence intensity of TSPP respectively were restored when APAP was added to the TSPP-Fe 3+ system. At this time, the color of the solution changed from green to colorless. Therefore, an “ON-OFF-ON” dual-signal sensing study of APAP were constructed using TSPP as the colorimetric and fluorescent probe. The proposed colorimetric sensing system had a wide linear range in the 13.12 mM ∼ 23.20 mM with 0.11 mM of limit of detection (LOD, S/N = 3). And the proposed fluorescence sensing system had a wide linear range in the 3.45 mM ∼ 12.50 mM and 41.67 mM ∼ 65.22 mM with 0.83 mM of limit of detection (LOD, S/N = 3). The dual-signal sensing system were applied to the APAP detection of real samples.

Histidine (His) plays an important role in human health and life activities. It will harm human health when His intake is insufficient or excessive. Its residue also will pollute the natural environment. Therefore, establishing a reliable and sensitive method for detecting histidine is particularly important. However, most of the existing detection methods of His rely on the single change of single signal, which are susceptible to interference from testing environmental factors and prone to generating false positive results. In contrast, the fluorescence and colorimetric dual-signal sensing system can not only effectively improve the reliability of detection, but also significantly reduce the risk of false positives. Therefore, the dual-signal sensing system has gradually become the research hotspot. Based on this, 5,10,15,20-tetra-(4-sulfophenyl) porphyrin (TSPP) was selected as the fluorescence and colorimetry dual-signal probe for the rapid detection of His in this study. Since TSPP could interact with many substances due to the specificity of molecular structure, it is the best choice to build an “ON-OFF-ON” sensing system in order to improve the specificity of the sensing system. Therefore, citric acid (CA) as an intermediate based on TSPP probe was successfully developed fluorescence and colorimetric dual-sensing system for the quantitative detection of histidine in real samples. The signal of the dual sensing system was “turned on” when the red TSPP solution obtained the fluorescence emission wavelength of 642 nm at the 514 nm optimal excitation wavelength and the UV–Vis absorption at 413 nm, respectively. The fluorescence intensity and absorbance of TSPP gradually decreased with the introduction of CA. At this time, the signal of the dual-sensing system became “turned off”, and the color of the solution changed from light pink to light green. The quenched fluorescence intensity and absorbance, however, was restored upon the introduction of histidine into the system. Simultaneously, the color of the solution changed from light green to light pink, and the dual-sensing system became “turned on”. Therefore, a novel fluorescence and colorimetric dual-signal sensing system based on TSPP was proposed for histidine detection. The results indicated that the linear ranges of the fluorescence sensing system were 8.34 × 10 −6 M − 1.51 × 10 −4 M and 1.85 × 10 −4 M − 1.4 × 10 −3 M with detection limits of 0.282 μM and 10.91 μM (LOD, S/N = 3), respectively. The linear range of the colorimetric sensing system was 2.04 × 10 −4 M–4.35 × 10 −4 M with a detection limit of 11.97 μM (LOD, S/N = 3). Meanwhile, the dual-sensing system provided a promising platform for practical samples sensing applications.

Exploring sensory systems from metal ion receptors enables the continuous development of ion sensing technologies. Herein we report the synthesis of carbon quantum dots (CQDs) from 5,10,15,20-tetrakis(4-sulfophenyl)porphyrin (TSPP) and additional carbon source, and its application in Cu 2+ imaging and quantifying in living cells. The CQDs are synthesized via a hydrothermal method in a successive polymerization and carbonization process. Due to the surface traps introduced by TSPP residue, the CQDs are featured with dual-emissive property of green (505 nm) and red (682 nm) fluorescence. Moreover, the red-to-green fluorescence ratio is readily customizable by tuning the molar fraction of TSPP. Benefited from Cu 2+ complexation induced red fluorescence quenching, the CQDs can be served as the ratiometric fluorescent nanosensor for Cu 2+ detection with ultra-high sensitivity and excellent specificity. The nanosensor is demonstrated to linearly detect Cu 2+ in the range of 0−50 nM, with an extremely low limit of detection (LOD) of 37 pM. More importantly, interference from other biological compositions, especially Fe 3+ , is successfully excluded via the site-selective coordination and specific electron transfer path. Based on the excellent analytical performance of the nanosensor, quantifying Cu 2+ in living cells is realized according to the linear correlation between fluorescence ratio and intracellular Cu 2+ content, which is parallelly detected by inductively coupled plasma mass spectrometry (ICP-MS). To demonstrate its practicality, the nanosensor is challenged to measure Cu 2+ uptake kinetics for cell lines derived from different tissues.

An upconversion luminescence (UCL) probe for the detection of Cu 2+ is constructed by assembling meso-tetra(4-sulfonatophenyl)porphine dihydrochlorid (TSPP) on SiO 2 -encapsulated β-NaYF 4 :Yb,Er,Gd nanorods which were synthesized by hydrothermal reaction and encapsulated with a layer of ultrathin silica through a water-in-oil reverse microemulsion strategy. Under excitation of 980 nm, the nanorods emit upconversion lights at 545 nm and 659 nm, respectively. While in the presence of Cu 2+ , Cu 2+ forms complex with TSPP, which shifts the maximum absorption of TSPP from 515 to 545 nm. As the maximum absorption of TSPP-Cu 2+ is overlapped with the emission at 545 nm of the nanorods, the luminescence energy of the nanorods can be transferred to TSPP–Cu 2+ complex, resulting in a quench of the emission at 545 nm, while the upconversion emission at 659 nm is not affected by Cu 2+ , and can be used as a reference. Cu 2+ was therefore quantified based on the ratio of the upconversion luminescence at 545 to that at 659 nm. Using the ratiometric upconversion luminescence as detection signal, the nanoprobe shows high selectivity and sensitivity towards Cu 2+ with a limit of detection of 2.16 μM, and a quick response time of within 10 s. The analysis of Cu 2+ in Hela cell lysates indicates the potential applications of this nanoprobe in the near-infrared fluorescent imaging of Cu 2+ in cells.
