Background Based on the prodrug principle, aspirin and eugenol, as starting precursors, were esterified to synthesize aspirin eugenol ester (AEE). The aim of the present study was to evaluate the antithrombotic effect of AEE in an animal disease model. In order to compare the therapeutic effects of AEE and its precursors, aspirin, eugenol and a combination of aspirin and eugenol were designed at the same molar quantities as the AEE medium dose in the control group. Methods After oral administration of AEE (dosed at 18, 36 and 72 mg/kg) for seven days, rats were treated with k-carrageenan to induce tail thrombosis. Following the same method, aspirin (20 mg/kg), eugenol (18 mg/kg) and 0.5 % CMC-Na (30 mg/kg) were administered as control drug. Different drug effects on platelet aggregation, hemorheology, TXB2/6-keto-PGF1α ratio and blood biochemistry were studied. Results AEE significantly inhibited ADP and AA-induced platelet aggregation in vivo. AEE also significantly reduced blood and plasma viscosity. Moreover, AEE down-regulated TXB2 and up-regulated 6-keto-PGF1α, normalizing the TXB2/6-keto-PGF1α ratio and blood biochemical profile. In comparison with aspirin and eugenol, AEE produced more positive therapeutic effects than its precursors under the same molar quantity. Conclusion It may be concluded that AEE was a good candidate for new antithrombotic and antiplatelet medicine. Additionally, this study may help to understand how AEE works on antithrombosis in different ways.

Oxidative stress biomarkers have been associated with both acute and chronic health outcomes. However, traditional methods analyze different biomarkers separately, resulting in complex sample preparation, high sample consumption, lengthy processing time, and limited comparability. In this study, we presented a newly developed and validated method for the simultaneous determination of oxidative stress from multiple perspectives (DNA, lipids, and antioxidants). Using a one-step solid-phase extraction and ultra-high-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UHPLC-QqQ-MS/MS), we measured four oxidative stress related biomarkers in urine simultaneously, within a run time of only 12 min. These biomarkers included 8-hydroxy-2′-deoxyguanosine (8-OHdG), 6-sulfatoxymelatonin (aMT6s), 8-isoprostaglandin-F 2α (8-isoPGF 2α ), and 11-dehydro thromboxane B 2 (11-DH-TXB 2 ). The calibration curves showed wide linear ranges (0.4–800 ng/mL for 8-OHdG, 0.2–600 ng/mL for aMT6s, 0.4–600 ng/mL for 8-isoPGF 2α , and 0.4–600 ng/mL for 11-DH-TXB 2 ), with r 2 values above 0.9932 for all analytes. The method demonstrated excellent sensitivity, with detection limits below 0.12 ng/mL, and good precision, with intra- and inter-day coefficients of variation ranging from 1.2 % to 14.4 %. We applied this method to urine samples from two populations living at different altitudes and found significantly higher levels of both 8-OHdG and 11-DH-TXB 2 in the high-altitude group, likely due to hypobaric hypoxia. In the future, this new method could be applied in large-scale epidemiological studies to investigate biological mechanisms of oxidative stress in health risks or for clinical diagnosis.

Background Laminaria japonica polysaccharide (LJP), a fucose enriched sulfated polysaccharide has been demonstrated to have excellent anticoagulant and antithrombotic activities. However, the antithrombotic effect of low molecular weight polysaccharide from enzymatically modified of LJP (LMWEP) remains unknown. Material/Methods LMWEP was prepared by fucoidanase enzymatic hydrolysis, and the antithrombotic and anticoagulant activities, and the underlying mechanism were investigated thoroughly. Rats were randomly divided into 6 groups (8 rats in each group): the blank control group, the blank control group treated with LMWEP (20 mg/kg), the model group, the model group treated with heparin (2 mg/kg), the model group treated with LJP (20 mg/kg), and the model group treated with LMWEP (20 mg/kg). After 7 days of intravenous administration, blood was collected for biochemical parameters examinations. Results LMWEP increased the activated partial thromboplastin time (APTT), thrombin time (TT), prothrombin time (PT), 6-keto prostaglandin F1α (6-Keto-PGF1α), and endothelial nitric oxide synthase (eNOS). In addition, LMWEP decreased fibrinogen (FIB), endothelin-1 (ET-1), thromboxane B2 (TXB2), erythrocyte sedimentation rate (ESR), and hematocrit (HCT). Conclusions LMWEP, an enzymatically modified fragment with a molecular weight of 25.8 kDa, is a potential antithrombotic candidate for treatment of thrombosis related diseases.
