A novel phosphorus (P) modified biochar (PLBC) was produced by pyrolyzing biomass of the dietic herb Taraxacum mongolicum Hand-Mazz (TMHM) and treating it with monopotassium phosphate (KH 2 PO 4 ). This phosphorous loaded biochar was then assessed as adsorbent for As(III) removal from contaminated water . In the current research, the adsorbent was characterized before and after P loading by means of SEM-EDX, TEM , FTIR and XRD techniques . It was evidenced that the presence of P on the surface of the biochar (BC) could improve its efficiency to remove As(III) from contaminated environments. Adsorption kinetics were evaluated by performing batch-type experiments at varied times and pH values (5, 7 and 9). The kinetic study revealed that a contact time of 24 h was required to attain equilibrium and the experimental data were best fitted to the pseudo-second-order kinetic model (q e = 17.1 mg g −1 ). In addition, several batch experiments were conducted with varied arsenic concentrations. During the adsorption tests, the maximum adsorption of As(III) was found at pH 5. The adsorption study further showed that compared to BC, PLBC depicted increased removal of As(III) from contaminated solutions. The adsorption experimental data showed the best fit to the Langmuir isotherm model (with R 2 = 0.84), with maximum As(III) adsorption capacity reaching 30.76 mg g −1 for PLBC.

Ethnopharmacological relevance Triple-negative breast cancer (TNBC) has a poor clinical prognosis and shows resistance to traditional treatments. Ferroptosis is a potential therapeutic strategy for TNBC. Although dandelion extract has demonstrated anti-tumor effects against TNBC, its role in regulating ferroptosis has not been elucidated. Aim of the study This study aimed to investigate the role and mechanisms of dandelion extract in inhibiting TNBC via the induction of ferroptosis. Materials and methods Cell viability and cell death were detected by CCK-8 assay and PI/Hoechst 33342 dual staining, respectively. Proteomic profiling was used to identify the mechanisms of dandelion extract-induced cell death. Ferroptosis was assessed by measuring key hallmarks, including Fe 2+ accumulation, lipid peroxidation, mitochondrial dysfunction, and ultrastructural changes. Mechanistic validation was achieved through target-specific interference and subsequent biomolecular analysis. Results Dandelion extract selectively induced cell death in TNBC cells without affecting normal MCF-10A cells. Proteomic and experimental evidence indicates that dandelion extract induces ferroptosis in TNBC cells, characterized by elevated Fe 2+ , ROS, lipid peroxidation, MDA levels, and mitochondrial dysfunction. Crucially, ferroptosis inhibitors attenuated the inhibitory effect of dandelion extract on TNBC cell viability, confirming ferroptosis as the primary cell death mechanism. Mechanistic studies have demonstrated that dandelion extract promoted ferroptosis in TNBC cells both in vitro and in vivo through NCOA4-mediated ferritinophagy. Conclusions This study demonstrates that dandelion extract induces ferroptosis in TNBC cells both in vitro and i n vivo by regulating ferritinophagy mediated by the NCOA4-FTH1 axis, thus suggesting novel therapeutic strategies for TNBC treatment.

Due to the interest in the potential pharmacological application of dandelion, the chemical constituents and activities of Taraxacum mongolicum Hand.-Mazz were studied. Box–Behnken response surface methodology was employed to optimize the protocol for extraction of flavonoid from dandelion. The molecular structures of different flavonoid compounds were acquired and analyzed by liquid chromatography-mass spectrometry (LC–MS) and nuclear magnetic resonance (NMR) spectroscopy. Several major flavonoid compounds were isolated and purified, namely, hesperetin-5′-O-β-rhamnoglucoside, hesperetin-7-glucuronide, kaempferol-3-glucoside, baicalein, hyperseroside, which were extracted for the first time from dandelion. Hesperetin-5′-O-β-rhamnoglucoside was identified as a new type of flavonoid that had never reported in the literature. This new flavonoid has outstanding antioxidant activity, as shown by its IC 50 value (8.72 mg/L) for scavenging DPPH free radicals. The determination of the structure-related antioxidant activities could be interpreted based on DFT calculations. As such, we have not only illustrated the rich flavonoid contents in Taraxacum mongolicum Hand.-Mazz , but also revealed new types of flavonoid compounds in dandelion in terms of structure and antioxidant properties.
