Gloiopeltis tenax(G. tenax) is widely distributed along the Chinese coastal areas and is commonly used in the treatment of diarrhea and colitis. This study aimed at investigating the bioactivities of the volatile constituents inG. tenax. We extracted the essential constituents ofG. tenaxby supercritical carbon dioxide extraction (CO2-SFE), then identified and analyzed the constituents by gas chromatography-mass spectrometry (GC-MS). In total, 30 components were identified in theG. tenaxextract. The components showed remarkable antioxidant activity (radical scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH)), lipid peroxidation inhibition capacity (in a β-carotene/linoleic acid-coupled oxidation reaction), and hydroxyl radical-scavenging activity (by deoxyribose degradation by iron-dependent hydroxyl radical), compared to butylated hydroxytoluene. In microdilution assays,G. tenaxextracts showed a moderate inhibitory effects onStaphyloccocus aureus(minimum inhibitory concentration (MIC) = 3.9 mg/mL),Enterococcus faecalis(7.8 mg/mL),Pseudomonas aeruginosa(15.6 mg/mL), andEscherichia coli(3.9 mg/mL). Antioxidant and antimicrobial activities ofG. tenaxwere related to the active chemical composition. These results suggest that the CO2-SFE extract fromG. tenaxhas potential to be used as a natural antioxidant and antimicrobial agent in food processing. Keywords:Gloiopeltis tenax;GC-MS;CO2-SFE;antioxidant activity;antimicrobial activity

Physiologically based in vitro methods have been developed to measure bioaccessibility of organic contaminants in soils. However, bioaccessibility of hydrophobic organic contaminants (HOCs) can be underestimated by in vitro tests if gastrointestinal (GI) solution fails to provide sufficient sorption sink for HOCs. To circumvent this drawback, Tenax was included in GI solution as sorption sink to trap mobilized HOCs and maintain the desorption gradient between soil and GI solution. Polycyclic aromatic hydrocarbons (PAHs) were selected as target HOCs, and physiologically based extraction test (PBET) was selected as the in vitro method. Inclusion of Tenax in GI solution increased bioaccessibility of PAHs in five spiked soils from 8.25–20.8% to 55.7–65.9% and the bioaccessibility of PAHs in a field contaminated soil from 3.70–6.92% to 16.3–31.0%. Our results demonstrated the effectiveness of Tenax as sorption sink to enhance PAH mobilization in bioaccessibility measurement in soils.

For hydrophobic organic contaminants , physiologically based in vitro methods may need to include a sorption sink to simulate in vivo intestinal uptake. We compared PAH bioaccessibility in contaminated soils using five in vitro methods including physiologically based extraction test (PBET), in vitro digestion assay (IVD), method from Deutsches Institut für Normung (DIN), unified bioaccessibility method (UBM), and fed organic estimation human simulation test (FOREhST) in the absence and presence of Tenax as a sorption sink. The PAH bioaccessibility without Tenax were pretty low with values ranging from below detection limit to 13.4%, indicating the limited capacity of these in vitro models to accommodate PAHs. With addition of Tenax, bioaccessibility increased to 0.59–75.5% for all PAH congeners. Even with the dominant effect of sorption sink, bioaccessibility values significantly varied among all the five methods with DIN result being the highest at 7.0–34.8%. Based on multiple linear regression, Tenax, incubation time, and bile contents are identified to be the most important factors in controlling bioaccessibility. The understanding of these key factors for bioaccessibility is highly necessary to standardize in vitro methods, which helps to refine the assessment of health risk through exposure to ingested contaminants.

Chemical degradation is one of the crucial methods for the remediation of hydrophobic organic compounds (HOCs) in soil/sediment. The sequestration effect of black carbon (BC) can affect the adsorption state of HOCs, thereby affecting their chemical degradability . Our study focused on the chemical degradability of 2-Chlorobiphenyl (PCB1) sequestrated on the typical BC (fly ash (FC), soot (SC), low-temperature biochar (BC400) and high-temperature biochar (BC900)) by iron-nickel bimetallic nanomaterials (nZVI/Ni) based on TENAX desorption technology . The results showed that PCB1 adsorbed in various states were simultaneously dechlorinated by nZVI/Ni. Specifically, rapid-desorption-state PCB1 tended to degrade more easily than resistant-desorption-state PCB1. Moreover, the degradation mechanism varied according to the type of BC. In the case of FC and SC, the degradation rate was lower than the desorption rate for the PCB1 in rapid and slow desorption states, and the degradation rate of PCB1 in the resistant desorption state was negligible. The PCB1 on FC and SC was first desorbed from BC and then degraded. However, in terms of BC400 and BC900, the degradation rate was higher than the desorption rate, and the degradation rate of the resistant-desorption-state PCB1 was 1.4 × 10 −2 h −1 and 4.1 × 10 −2 h −1 , respectively. The graphitized structure of BC900 can directly transfer electrons , so more than 90% of the resistant-desorption-state PCB1 could be degraded. In addition, BC may affect the longevity of nZVI/Ni, thereby affecting its degradability. Therefore, the chemical degradability of BC-adsorbed HOCs should be comprehensively evaluated based on the adsorption state and the properties of BC.

The occurrence of nitrated polycyclic aromatic hydrocarbons (NPAHs) in sediments has been widely reported, but research on NPAH bioavailability is lacking. In this study, a self-made zeolite imidazolate framework-8/hexagonal boron nitride (ZIF-8/h-BN) solid-phase microextraction (SPME) fiber and commercial Tenax are compared as efficient tools to predict the bioavailability of NPAHs in sediments with bioassays using Cipangopaludina chinensis . During the process of SPME, the NPAH concentrations on the ZIF-8/h-BN fibers reached extraction equilibrium after 72 h. The fiber extraction of NPAHs in sediments was well-fitted by the pseudo first-order kinetic model with a rate constant of 2 × 10 −2 h −1 (R 2 > 0.98). The extraction rates ranking of NPAHs in sediments was 2-nitrobiphenyl>1-nitropyrene>5-nitroacenaphthene>2-nitrofluorene. Compared with SPME, NPAH concentrations reached equilibrium after 168 h for the Tenax extraction. The orders of magnitude of fast, slow, and very slow desorption rate constants were 10 −1 , 10 −2 , and 10 −4 , respectively. At extraction equilibrium (168 h), the SPME was close to the bioavailability of the NPAHs in sediments to Cipangopaludina chinensis with a slope statistically approximated to one. In addition, the linear regression for SPME (R 2 = 0.7285) was slightly higher than that of the Tenax extraction (R 2 = 0.7168) over a short time (6 h). This could be because the coating material of ZIF-8/h-BN can rapidly adsorb freely dissolved NPAHs, and the SPME fibers can accurately predict the bioaccumulated concentrations of NPAHs in exposed organisms by measuring the concentration of NPAHs in the pore water of sediment. This study provides a time-saving and easy procedure to predict the bioavailability of NPAHs in sediments.

Bioaccessibility of hydrophobic organic contaminants (HOCs) from unintentional ingestion of soil is increasingly assessed with in vitro gastrointestinal models incorporating a sorption sink. In this study, the bioaccessibility of DDTs in contaminated soils (n = 11) was determined using “unfed” unified bioaccessibility method (UBM) and fed organic estimation human simulation test (FOREhST) with/without Tenax as an absorbent. By adding Tenax, the bioaccessibility of DDTs determined using UBM was significantly increased from 4.9–30.6% to 31.6–86.0%. In contrast, the bioaccessibility of DDTs determined using FOREhST without/with Tenax were similar with values of 20.0–60.9% vs 31.5–47.6%, implying that the influence of food components on the absorption efficiency of the sink should not be overlooked. Much high fraction of DDTs (bioaccessibility: 11.7–24.8%) remained in FOREhST supernatant after Tenax collection, suggesting that prediction of bioavailability through bioaccessibility obtained by absorbent needs to be treated with caution when bioaccessibility is determined using a “fed state” in vitro method.
