Molecular mechanisms of Tetrastigma hemsleyanum Diels&Gilg against lung squamous cell carcinoma: From computational biology and experimental validation

Ping Li,Changchang Wang,Gun Chen,Yixiao Han,Hanyu Lu,Nan Li,Yangbin Lv,Chu Chu,Xin Peng

Ethnopharmacological relevance Tetrastigma hemsleyanum ( T. hemsleyanum ), valued in traditional medicine for its potential to boost immunity and combat tumors, contains uncharacterized active compounds and mechanisms. This represents a significant gap in our understanding of its ethnopharmacological relevance. Aim of the study To involve the mechanism of anti-lung cancer effect of T. hemsleyanum by means of experiment and bioinformatics analysis. Materials and methods The anticancer mechanism of T. hemsleyanum against lung squamous carcinoma (LUSC) in zebrafish was investigated. The LUSC model was established by injecting NCI–H2170 cells in the zebrafish and evaluating its anti-tumor efficacy. Next, component targets and key genes were obtained by molecular complex detection (MCODE) analysis and protein-protein interaction (PPI) network analysis. Component analysis of T. hemsleyanum was performed by UPLC-Q-TOF-MS. Molecular docking was used to simulate the binding activities of key potential active components to core targets were simulated using. Prognostic and pan-cancer analyses were then performed to validate the signaling pathways involved in the prognostic genes using gene set enrichment analysis (GSEA). Subsequently, Molecular dynamics simulations were then performed for key active components and core targets. Finally, cellular experiments were used to verify the expression of glutamate metabotropic receptor 3 (GRM3) and glutamate metabotropic receptor 7 (GRM7) in the anticancer effect exerted of T. hemsleyanum . Results We experimentally confirmed the inhibitory effect of T. hemsleyanum on LUSC by transplantation of NCI–H2170 cells into zebrafish. There are 20 main compounds in T. hemsleyanum , such as procyanidin B1, catechin , quercetin , and kaempferol , etc. A total of 186 component targets of T. hemsleyanum and sixteen hub genes were screened by PPI network and MCODE analyses. Molecular docking and molecular dynamics simulation results showed that Gingerglycolipid B and Rutin had higher affinity with GRM3 and GRM7 , respectively. Prognostic analysis, Pan-cancer analysis and verification experiment also confirmed that GRM3 and GRM7 were targets for T. hemsleyanum to exert anti-tumor effects and to participate in immune and mutation processes. In vitro experiments suggested that the inhibitory effect of T. hemsleyanum on cancer cells was correlated with GRM3 and GRM7. Conclusion In vivo, in vitro and in silico results confirmed the potential anticancer effects against LUSC of T. hemsleyanum , which further consolidated the claim of its traditional uses.

Shoot Organogenesis from Tetrastigma hemsleyanum Leaf and Petiole Explants, and Subsequent Plant Regeneration and Acclimatization

Pang Jinhui,Xiong Yuping,Zeng Yujie,Chen Xiaohong,Li Jianrong,Zhang Xinhua,Li Yuan,Wu Kunlin,Zeng Songjun,da Silva Jaime A. Teixeira,Ma Guohua

Tetrastigma hemsleyanum is a perennial evergreen vine of the Vitaceae. The entire herb is used in traditional Chinese medicine as a broad-spectrum plant-based antibiotic, so it has high economic and social value. Wild T. hemsleyanum resources are scarce, so it has been declared an endangered and rare medicinal plant. Seed yield is low and vegetative propagation by cuttings results in limited plant production, so development of the T. hemsleyanum industry requires optimized propagation protocols and the development of new biotechnologies to proliferate this plant in commercial quantities. In this study, shoot organogenesis was successfully induced from leaves and petioles. Two plant growth regulators, 6-benzyladenine (BA) and thidiazuron, induced callus and adventitious shoots, but the ideal adventitious shoot induction medium was Murashige and Skoog (MS) medium containing 1.0 mg L −1 BA and 0.1 mg L −1 α-naphthaleneacetic acid (NAA). This resulted in a shoot proliferation coefficient (SPC) of 6.73 within 30 d at a light intensity of 100 µmol m −2  s −1 . When light intensity was increased from 50 to 200 µmol m −2  s −1 , SPC (7.35), chlorophyll a (Chl a ), Chl b , and total Chl ( a  +  b ) content increased. On MS medium containing 0.1–2.0 mg L −1 NAA or indole-3-butyric acid, 100% of adventitious shoots formed adventitious roots. Plantlets showed no obvious morphological variation, and their survival exceeded 98% on a substrate of peat and river sand ( v:v  = 2:1). This study’s protocols allow for the mass production of adventitious shoots for conservation purposes, and potentially for the commercial propagation of T. hemsleyanum .

Abiotic stress-induced changes in Tetrastigma hemsleyanum: insights from secondary metabolite biosynthesis and enhancement of plant defense mechanisms

Minmin Chen,Xiaoqun Wang,Xiawen Zhou,Baiyu Huang,Yujie Zhao,Haiying Liu,Qiuling He

Tetrastigma hemsleyanum , a traditional Chinese medicinal plant with anti-inflammatory, anti-cancer, and anti-tumor properties, faces increasing abiotic stress due to climate change, agricultural chemicals, and industrialization. This study investigated how three abiotic stress factors influence antioxidant enzyme activity, MDA levels, DPPH free radical scavenging capacity, chlorophyll, carotenoids, active compounds, and gene expression in different T. hemsleyanum strains. The comprehensive evaluation indicates that the ZJWZ strain holds potential as a preferred parental material for future resistance breeding. Furthermore, PAL gene expression was strongly positively correlated with flavonoid and phenol contents, highlighting its role in the stress response through the phenylpropanoid-flavonoid pathway. This study contributes to the standardization of the production and breeding of superior strains of T. hemsleyanum . It also lays the foundation for investigating how plants react to environmental stressors.

Purified Tetrastigma hemsleyanum vines polysaccharide attenuates EC-induced toxicity in Caco-2 cells and Caenorhabditis elegans via DAF-16/FOXO pathway

Qiang Chu,Ruoyi Jia,Wen Chen,Yangyang Liu,Yonglu Li,Xiang Ye,Yong Jiang,Xiaodong Zheng

Ethyl Carbamate (EC), as a carcinogen widely found in fermented foods, was verified that its cytotoxicity was associated with oxidative stress. Polysaccharides from natural sources due to their antioxidative capacity have attracted great attention in the past time. In this study, purified polysaccharide from Tetrastigma hemsleyanum vines (TVP) with 64.89 kDA was extracted and conducted multiple analysis to identify its structural information. It could be discovered that TVP was composed of mannose, rhamnose, glucuronic acid, glucose, galactose, and arabinose. In vitro , TVP could inhibit cytotoxicity and genotoxicity, attenuate oxidative damage and mitochondrial dysfunction induced by EC in Caco-2 cells. Meanwhile, TVP could suppress apoptosis by mTOR and Bcl-2 signaling pathways, ameliorate oxidative via Sirt1-FoxO1 and Nrf2-Keap1 signaling pathways. In vivo , EC as well triggered the decline of survival and athletic ability in Caenorhabditis elegans ( C. elegans ) and TVP could reverse the decline. In the meantime, TVP could ameliorate oxidative damage in N2 and daf-2 (-) mutant but fail in daf-16 (-) mutant, which suggested that DAF-16 (FOXO) might affect the antioxidative protection of TVP in C. elegans . In brief, our results manifested that TVP could attenuate EC-induced cytotoxicity both in vitro and in vivo .

The stereoscopic planting mode improved the quality and yield of Tetrastigma hemsleyanum

Wanying Hu,Mengdan Jiang,Zongsuo Liang,Pengguo Xia

Tetrastigma hemsleyanum Diels et Gilg, a plant of the Vitaceae family, is often used as a medicine. Modern pharmacological studies have found that its root tubers have a particular anticancer effect. Polydatin, piceatannol, resveratrol, and kaempferol could be detected stably in this herbal plant, which are vital indicators for controlling and evaluating its quality. At present, its planting mode is chaotic and complicated. In order to explore the optimal cultivation mode with excellent quality and yield, the agronomic indicators, the indicators of conventional medical materials and the content of active ingredients were determined in this study. Then, their discrepancy under different planting bases and various planting modes was analyzed. The T. hemsleyanum cultivated in the greenhouse had the thickest stems and the longest leaves, as well as the largest aerial and underground biomass. Compared with understory planting, the root tubers have higher total flavonoid content in the greenhouse planting mode. In addition, the index component content of the stereoscopic planting was higher than that of the crawl planting. In summary, greenhouse planting combined with stereoscopic planting mode was conducive to improving the yield and quality of T. hemsleyanum simultaneously. These results provided a more comprehensive reference basis for the artificial planting of T. hemsleyanum . Simultaneously, it provided a theoretical basis for strengthening the sustainable development and utilization of medicinal materials from T. hemsleyanum and protection of resources.

A glucuronogalactomannan isolated from Tetrastigma hemsleyanum Diels et Gilg: Structure and immunomodulatory activity

Zian Mao,Liu Yang,Yishan Lv,Yuchi Chen,Mingyuan Zhou,Chengnan Fang,Bingqi Zhu,Fangmei Zhou,Zhishan Ding

A novel acidic glucuronogalactomannan (STHP-5) was isolated from the aboveground part of Tetrastigma hemsleyanum Diels et Gilg with a molecular weight of 3.225 × 10 5  kDa. Analysis of chain conformation showed STHP-5 was approximately a random coil chain. STHP-5 was composed mainly of galactose, mannose, and glucuronic acid. Linkages of glycosides were measured via methylation analysis and verified by NMR. Based on the structural characterization, the repeat unit of STHP-5 was proposed. In vitro, STHP-5 induced the production of nitric oxide (NO) and secretion of IL-6, MCP-1, and TNF-α in RAW264.7 cells, indicating STHP-5 had stimulatory activity on macrophages. STHP-5 was proven to function as a TLR4 agonist by inducing the secretion of secreted embryonic alkaline phosphatase (SEAP) in HEK-Blue™-hTLR4 cells. The TLR4 activation capacity was quantitatively measured via EC50, and it showed purified polysaccharides had stronger effects (lower EC50) on activating TLR4 compared with crude polysaccharides. In conclusion, our findings suggest STHP-5 may be a novel immunomodulator.

Tetrastigma hemsleyanum tubers polysaccharide ameliorates LPS-induced inflammation in macrophages and Caenorhabditis elegans

Qiang Chu,Ruoyi Jia,Meng Chen,Yonglu Li,Xin Yu,Yaxuan Wang,Wen Chen,Xiang Ye,Yangyang Liu,Yong Jiang,Xiaodong Zheng

Tetrastigma hemsleyanum Diels et Gilg, a rare herbal plant native to China, possesses high medicinal value. More researches have focused on the flavones in the tubers of Tetrastigma hemsleyanum whereas polysaccharides attract few attentions. In this study, a purified polysaccharide (TTP-1) with the average molecular weights of 478.33 kDa was extracted from Tetrastigma hemsleyanum and conducted multiple analysis to identify its structural information. In vitro , TTP-1 could suppress the inflammation, cytotoxicity, genotoxicity, mitochondrial dysfunction and oxidative damage induced by LPS in RAW264.7 cells. Meanwhile, TTP-1 could attenuate inflammation via COX-2, iNOS, MAPKs pathways and ameliorate oxidative damage through Nrf2-Keap1, Sirt1-FoxO1 pathways in RAW264.7 cells. In vivo , the inflammation induced by LPS triggered the suppression of survival ratio and growing development, as well as the restrain of athletic ability in Caenorhabditis elegans . The treatment of TTP-1 reversed the situation induced by LPS. In a word, TTP-1 could ameliorate LPS-induced inflammation both in vitro and in vivo .

Tetrastigma hemsleyanum leaves extract against acrylamide-induced toxicity in HepG2 cells and Caenorhabditis elegans

Qiang Chu,Wen Chen,Ruoyi Jia,Xiang Ye,Yonglu Li,Yangyang Liu,Yong Jiang,Xiaodong Zheng

Acrylamide (ACR), as a raw material of polyacrylamide that used in water purification, was verified to possess various toxicity. Tetrastigma hemsleyanum (TH) is a medicinal plant widely used to anti-inflammation and anti-tumor in Chinese folks. However, more researches focused on the biological activities in tubers and the leaves were ignored. Thus, the protective effect of Tetrastigma hemsleyanum leaves extract (THLE) against ACR-induced toxicity in HepG2 cells and Caenorhabditis elegans ( C. elegans ) was explored in this study. In vitro , we observed that THLE attenuated ACR-induced toxicity in HepG2 cell via regulating Akt/mTOR/FOXO1/MAPK signaling pathway. Further research proved that 5-caffeoylquinic acid (5-CA) plays a major role in THLE’s amelioration effect of ACR toxicity. In vivo , it was found that THLE possesses the same protective effect in ACR-treated wild-type N2 C. elegans and daf-2 (-) (deficit in DAF-2) mutants. However, the anti-ACR toxicity effect of THLE in daf-16 (-) mutants (deficit in DAF-16 that homologous to FOXO family in human) was weakened. Our results indicated that THLE exhibited protective effects against ACR-induced toxicity both in HepG2 cells and C. elegans , while DAF-16/FOXO gene is involved in THLE’ protective effect via regulating the expression levels of downstream antioxidant genes.

Phytochemical analysis and safety evaluation of the extracts from Tetrastigma hemsleyanum Diels et Gilg leaves

Zili Guo,Lingfeng Zhou,Hongle Li,Yuhao Huang,Chao Xu,Lili Niu

As the market of Tetrastigma hemsleyanum Diels et Gilg (TH) products rapidly expands and consumer demand increases, identifying the chemical components and evaluating its safety can help ensure human health during their usage. In this study, GC-MS and LC-MS were used to investigate the phytochemical characteristics of the TH leaf extracts, and the safety of TH extracts was evaluated using zebrafish embryos and larvae, with specific endpoints including the hatching rate, mortality rate, and the overall morphology. It revealed that TH leaves were rich in vitamins, flavonoids, terpenoids, etc. A total of 31 and 42 metabolites was identified in the n-hexane extract and the 60 % methanol aqueous extract of TH leaves, respectively, using GC-MS and LC/Q-TOF-MS techniques. DL-alpha-tocopherol was discovered for the first time in TH leaves. Seven exposure levels (0.02–6.4 mg/mL) of the aqueous extracts were employed in safety evaluation using zebrafish embryos. Our results indicated that the highest concentration (6.4 mg/mL) led to the lowest hatching rate (1.3 ± 1.8 %, P < 0.001) and the highest cumulative mortality rate (22.5 ± 9.4 %, P < 0.001). Subsequently, five exposure levels (0.02–1.6 mg/mL) were employed to evaluate the safety using zebrafish larvae. The results indicated that the treatment had no significant impact on the mortality rate, overall morphology, or specific organs (heart, liver, kidneys) of zebrafish larvae, nor did it induce morphological abnormalities. However, a concentration-dependent increase in heart rate was observed. This study provides a reference for consumers to use TH leaf safely and rationally.

Tetrastigma hemsleyanum Polysaccharides as Natural Inhibitors of Platelet Thrombosis via GPIIb/IIIa and MAPK Pathways

Kewei Li,Chenjun Shen,Peihao Xu,Leilei Zhu,Mingyuan Zhou,Baodong Ye,Yingzhi Shen,Zhishan Ding

Background Thrombosis, driven by abnormal platelet activation and fibrin clot formation, is a key pathological basis of cardiovascular disorders. Polysaccharide-based agents, such as heparin, possess distinct antithrombotic activities. Tetrastigma hemsleyanum polysaccharide (THP) exhibits notable anti-inflammatory properties. However, its antithrombotic potential remains unexplored. Purpose This work intended to explore the antithrombotic and antiplatelet activities of THP5, a novel acidic glucuronic acid-containing galactomannan isolated from THP, and to elucidate its molecular targets and mechanisms of platelet interaction. Methods The antithrombotic activity of THP5 was tested in vivo using a mouse model of κ-carrageenan-induced thrombosis. In vitro platelet function assays determined its antiplatelet effects. Platelet protein targets were identified via pull-down, liquid chromatography-mass spectrometry (LC-MS/MS), and surface plasmon resonance (SPR) analyses. Comparative studies with the GPIIb/IIIa inhibitor tirofiban were performed in κ-carrageenan- and FeCl 3 -induced thrombosis models. Proteomic profiling and bleeding risk assessments were conducted to investigate underlying mechanisms and safety. Results THP5 markedly reduced thrombus formation, attenuated inflammatory responses, and alleviated vascular injury in vivo. In vitro , it protected endothelial cells from LPS-induced damage and strongly inhibited platelet activation. GPIIb/IIIa was identified as a direct binding target of THP5, and proteomic analysis revealed modulation of the MAPK signaling pathway. Compared with tirofiban, THP5 demonstrated comparable efficacy with a lower bleeding risk. Conclusion THP5 exerts potent antithrombotic effects by directly targeting GPIIb/IIIa and modulating PKCα-MAPK-dependent platelet activation, supporting its potential as a safe and effective therapeutic candidate for thrombotic disorders.

Tetrastigma hemsleyanum leaf flavones have anti-NSCLC ability by triggering apoptosis using the Akt-mTOR pathway

Yonglu Li,Yiru Zhang,Yaxuan Wang,Xin Yu,Ting Yu,Xiaodong Zheng,Qiang Chu

With high morbidity and mortality, non-small cell lung cancer (NSCLC) has become a challenge for humans, because its clinic chemotherapies still remain unsatisfactory. In this study, the leaf part of a traditional Chinese plant, Tetrastigma hemsleyanum (TL), was studied, and its main flavones compounds were extracted and purified (TLF), which were composed of 5-caffeoylquinic acid, 3-caffeoylquinic acid, 4-caffeoylquinic acid, quercetin-3-O-rutinoside and kaempferol-3-O-rutinoside. To study the anti-NSCLC ability of TLF, in vitro and in vivo studies were done. TLF inhibited the proliferation and migration of A549 cells and caused mitochondrial dysfunction. TLF significantly suppressed the tumor weight and volume, and down-regulated Ki67, proliferation cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) productions. To determine the intrinsic mechanism, proteins alternations were measured in solid tumors, and results showed that TLF blocked the Akt/mTOR pathway, and thereby induced autophagy, triggering Bcl and Caspase-dependent pathways, resulting in A549 cells’ apoptosis. This study indicated that TLF is a potential candidate to be an anti-NSCLC drug, or a supplementary resource for chemotherapy.

Multi-omics analyses revealed key factors involved in fluorescent carbon-dots-regulated secondary metabolism in Tetrastigma hemsleyanum

Peng Xin,Xie Zhuomi,Wang Xiuhua,Zhao Yuxiang,Yang Chuyun,Zhang Zhongyi,Li Mingjie,Zheng Jianping,Wang Yuhui

Background Luminescent nanomaterials (LNMs), especially newly-exploited fluorescent carbon-dots (CDs), have demonstrated promising candidates for sunlight harvesting and enhanced photosynthesis efficiency of crops. However, most of the studies focus on the design and synthesis of LNMs and primary metabolism in biomass acceleration, secondary metabolism that closely associated with the quality ingredients of plants is rarely mentioned. Results UV-absorptive and water-soluble NIR-CDs were harvested via a facile microwave-assisted carbonization method. The effect and regulatory mechanism of NIR-CDs on the secondary metabolism and bioactive ingredients accumulation in Tetrastigma hemsleyanum were explored. A total of 191 differential secondary metabolites and 6874 differentially expressed genes were identified when the NIR-CDs were adopted for enhancing growth of T. hemsleyanum . The phenolic acids were generally improved, but the flavonoids were more likely to decrease. The pivotal differentially expressed genes were involved in biosynthesis of secondary metabolites, flavonoid biosynthesis, porphyrin and chlorophyll metabolism, etc. The gene-metabolite association was constructed and 44 hub genes highly related to quality ingredients accumulation and growth were identified, among which and the top 5 genes of the PPI network might be the key regulators. Conclusion This research provided key regulatory genes and differentially accumulating quality ingredients under NIR-CDs-treatment, which could provide a theoretical basis for expanding the applications of nanomaterial in industrial crop agriculture.
