In order to effectively avoid the side effects induced by multiple components and tedious synthesis process, a simple therapy system based on one material to simultaneously realize both photothermal therapy (PTT) and photodynamic therapy (PDT) under single laser irradiation will promote the overall phototherapeutic efficiency and make the PTT/PDT system easier to operate. Here, by using transferrin (Tf) as protein template, ultrasmall [email protected] nanodots ( [email protected] NDs) were successfully synthesized through a facile one-pot protein-based biomineralization method. The obtained [email protected] NDs exhibited not only excellent photothermal conversion ability (34.4%) but also high photoactivated formation of reactive oxygen species (ROS) upon 980 nm near-infrared (NIR) irradiation. By loading the drug doxorubicin (DOX) to [email protected] NDs, a synergistic therapy system with multiple therapeutic effects combined PTT, PDT, chemotherapy (CT) and tumor targeting properties could be perfectly implemented together by [email protected] NDs without any complicated post-modification process. Results from the in vitro cell experiments confirmed that these [email protected] NDs could produce excellent effect on cancer cells with 88.5% cell inhibition rate. In comparison with the complicated systems based on “multiple-components-in-one” strategy, this therapy system based on one single material but possess multifunctional purpose is easy to operate and more suitable for clinical applications.

Patulin is a toxic secondary metabolite produced by several moulds, which contaminates fruits and their products posing serious threats to human health. Though several microorganisms and enzymes have been reported to effectively degrade patulin, separation of them from fruit juice challenges the commercial applications. Here, a Pseudomonas aeruginosa strain TF-06 was isolated, its patulin degradation mechanism and optimum conditions for enzyme immobilization were investigated. The results indicated that TF-06 could degrade patulin into non-cytotoxic E/Z-ascladiol mainly by the activity of intracellular enzymes. For easy separation of enzymes, calcium alginate was selected for immobilization of intracellular enzymes from TF-06. The immobilized enzyme beads were effective in detoxification of patulin in apple juice. The mitigation rate was reached 95%, while there was no negative effect on juice quality. The study provides a promising way to resolve the issue of enzyme separation during mycotoxin biological detoxification in fruit juice.

Background and Objective To address the high mortality, slow onset, and suboptimal efficacy of current treatments for comorbid anxiety and depression (CAD), this study developed and systematically evaluated a novel combination formulation. Methods The anxiolytic tandospirone (TDS) was combined with fluoxetine (FLU), escitalopram, or milnacipran for cell-based screening. The optimal combination was selected for pharmacokinetic studies in rats, followed by a 49-day pharmacodynamic evaluation and mechanistic investigation in a CAD mouse model. Results Two optimal TDS-FLU combinations, TF-1 (TDS:FLU = 1.2:1,w/w) and TF-2 (TDS:FLU = 2.4:1,w/w), were obtained, both exhibiting good neuroprotective effects. Pharmacokinetic results showed that TF-1 and TF-2 significantly increased drug exposure and prolonged half-life compared with monotherapies of TDS and FLU. Pharmacodynamically, TF-1 and TF-2 exhibited superior anxiolytic-antidepressant efficacy compared with monotherapies after 28-day and 49-day interventions. At day 28, both formulations significantly increased open-field central-zone activity and sucrose preference, and reduced immobility time (all P < 0.05). Their therapeutic benefits were further augmented at day 49, with sustained and improved behavioral improvements upon long-term treatment. Mechanistic studies showed that both combinations elevated plasma and hippocampal 5-HT and BDNF levels, upregulated hippocampal 5-HT1AR and TrKB expression, and enhanced neuronal plasticity. Conclusion TF-1 and TF-2 were superior to single agents in pharmacokinetics and long-term CAD efficacy, acting via 5-HT modulation and BDNF-TrKB activation, supporting their development as CAD combination therapies.

Iron (Fe) is a crucial metal and plays a vital role in a variety of cellular processes. Studies in mammals have indicated that transferrin (TF) and transferrin receptor (TfR), jointly iron ion transport, while also contributing to antibacterial immunity. However, systematic reports on the functions and regulatory mechanisms of these components in teleost fish are lacking. In this study, TF protein was purified and identified from the serum of Nile tilapia ( Oreochromis niloticus ). A range of experimental techniques was employed to demonstrate that OnTF can inhibit the proliferation of pathogenic bacteria both in vivo and in vitro . Meanwhile, OnTF interacts with the OnTfR1 receptor on the cell surface, facilitating phagocytosis and bacterial clearance, thereby providing immune protection to the organism. Notably, OnTfR1 directly influences the secretion of OnHepc through the BMP-SMAD signaling pathway. The increased mortality of tilapia caused by downregulated OnTfR1 expression during pathogenic bacterial infection can be effectively alleviated by supplementing (r)Hepc protein. Moreover, OnTfR1 also played an indispensable role in maintaining the stability of the fish intestinal microecology and resisting pathogen infections. Overall, this study represents the first systematic investigation into the functional roles of molecules involving in iron homeostasis regulation in teleost fish. It elucidates that the OnTF-TfR1 axis, as a core pathway for iron ion uptake and metabolic regulation, plays a significant role in resisting pathogen infections and maintaining the stability of the intestinal microbiota. These findings provide new insights into understanding the functional association between iron metabolism and innate immune defense during adaptive evolution.

Safe and effective oral delivery of peptide is a challenge. Here, we used exenatide and zinc ions (Zn2+) to form a complex to explore a meaningful oral-targeted drug-delivery system. Polyethylene glycol-poly(lactic acid-co-glycolic acid) (PEG-PLGA) was used to prepare nanoparticles (NPs) to escape the degradation caused by gastrointestinal enzymes. Transferrin (Tf) was used as a targeting group. PEG-PLGA-NPs and Tf-modified exenatide-Zn2+-loaded NPs (Tf-PEG-PLGA-NPs) were uniformly sized spheres according to transmission electron microscopy. The results of pharmacodynamic and pharmacokinetic investigations in vivo were consistent with in vitro studies using Caco-2 cells. Tf enhanced NPs transport in cell-uptake and transmembrane-transport experiments. Our results showed that the relative bioavailability of Tf-exenatide-Zn2+-NPs was higher than that of exenatide-Zn2+-NPs. The relative bioavailability of Tf-exenatide-Zn2+-NPs versus subcutaneous injection of exenatide was 6.45%. This was a preliminary exploration of the oral administration of exenatide, that data from which can be used for future investigations.

This study was to explore the therapeutic effect and mechanism of puerarin (PUE) combined with PEGylated nanoparticles on a rat cerebral infarction cell model. In this context, PEG-PLGA/PUE nanoparticles were prepared by the thin-film hydration method, and the toxicity of PEG-PLGA/PUE nanoparticles to brain capillary endothelial cell (BCEC) was detected by MTT. The BCEC/TF cell model was obtained by induction of BCEC cells with TNF-α. The BCEC/TF cell model was identified by immunofluorescence; the protein expression was detected by western blotting; the expression level of miR-424 in cells was measured by RT-qPCR; the targeting relationship between miR-424 and PDCD4 was confirmed by dual-luciferase reporter assay. We found that PEG-PLGA/PUE nanoparticles prepared by the thin-film hydration method had uniform particle size, regular shape, and good stability and were not toxic to cells. The vWF was widely expressed in the cytoplasm in BCECs. The BCEC/TF cell model was obtained after TNF-α treatment, and tissue factor (TF) was widely expressed on the cell membrane of BCEC/TF cells. Furthermore, it was observed that the PEG-PLGA/PUE nanoparticles showed better therapeutic effect on the BCEC/TF cell model than PUE. PEG-PLGA/PUE nanoparticles and PUE inhibited the expression of PDCD4 protein by increasing the expression of miR-424 in BCEC/TF cells. In summary, the therapeutic effect of PEG-PLGA/PUE nanoparticles on the in vitro cell model of cerebral infarction is better than that of PUE. Moreover, PEG-PLGA/PUE inhibits the expression of PDCD4 protein by lowering the expression level of miR-424 in cells, thereby reducing the hazard of cerebral infarction.

To overcome the inherent drawbacks of poly(propylene carbonate) (PPC), such as poor thermal stability, low mechanical strength, and high surface energy, this study introduced, for the first time, 1,1,1-trifluoro-2,3-epoxypropane (TF-PO) as a third monomer into the metal-free TEB/PPNCl catalytic system for the terpolymerization with carbon dioxide (CO2) and propylene oxide (PO), successfully synthesizing a series of fluorinated PPC (PPCF). The optimal polymerization conditions (60°C, 2.0 MPa, 12 h, n(PO):n(TF-PO) = 100:4) were determined through systematic optimization. Comprehensive structural characterization (FT-IR, NMR, XPS) confirmed the successful incorporation of TF-PO into the polymer backbone. Property evaluation revealed that the PPCF materials exhibited substantial improvements in thermal stability, mechanical strength, hydrophobicity, and dielectric properties compared to unmodified PPC. The optimal sample, PPCF4, achieved a5%weight-loss temperature (Td,−5%) of242°C, a glass transition temperature (Tg) of42°C, a tensile strength of 21.5 MPa, and a Young modulus of 296 MPa. With a5%TF-PO feed ratio, the material’s water contact angle increased to 102°, and its dielectric constant reached 6.01 at104Hz. Furthermore, density functional theory (DFT) calculations elucidated the Lewis acidity of the TEB catalyst and the reactive sites of the monomers, leading to a proposed mechanism for the ternary alternating copolymerization. This work provides an effective synthetic strategy and theoretical foundation for preparing high-performance and functionalized PPC materials through molecular structure design.

Molecular weight serves as a critical parameter governing the adhesive properties of polymer adhesives. Nevertheless, its influence on the reversible adhesion of azopolymers exhibiting photoinduced reversible solid-to-liquid transitions remains poorly understood. Here, we systematically elucidate how molecular weight modulates the reversible adhesion behavior of azopolymers. Three azopolymers (P1, P2, and P3) were synthesized by ring-opening metathesis polymerization (ROMP), with molecular weights strategically designed below, near, and above the entanglement molecular weight (Me), respectively. UV irradiation induces trans-to-cis isomerization of azobenzene moieties, while visible light triggers cis-to-trans isomerization. The azopolymers exhibit photoswitchable glass transition temperatures (Tgs) and viscous flow temperatures (Tfs). Crucially, molecular weight dictates the mechanism of photocontrolled reversible adhesion. For P1 and P2, the trans azopolymers exhibit stronger adhesion than the cis form does. However, this trend is reversed for P3, where the cis P3 surpass trans P3 in terms of adhesive performance. These findings establish molecular weight engineering as a critical strategy for developing high-performance stimuli-responsive reversible adhesives.

In this study, air nanobubbles (NBs) generated with a new device were stabilised by a complex of arabic gum (AG) and stevioside (STE). Some basic metrics of NBs produced by biosurfactants at different concentrations were characterized to determine the stability of NBs. The mixture solution rich in NBs was assisted by ultrasonic disruption (UD) to extract total phenols (TP) and total flavonoids (TF) from 7 citrus pericarp. We evaluated four key factors affecting the TF extraction of ‘ Citrus reticulata 'Chachiensis '’ (‘Chazhigan’) pericarp by one-way test. The optimal extraction conditions were further predicted and validated by Box-Behnken response surface experiments. Under the conditions of 135 w capacity, 4.1 C mixture solution concentration, 30.5°C water bath temperature, and 31min ultrasonic time, the TF content of ‘Chazhigan’ pericarp (20.82±0.85 mg/g DW) was significantly better than that extracted by 80% ethanol (12.22±0.90 mg/g DW). Finally, fourier transform infrared (FT-IR) spectroscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were utilised to provide a preliminary exploration of the potential mechanisms involved in the extraction process. The flavonoid extracts from the pericarp of ‘Chazhigan’ were qualitatively analysed. And the differences in the chemical antioxidant activity of their extracts were investigated before and after the application of traditional alcoholic extraction and novel green extraction. In conclusion, this study established a novel approach that is eco-friendly, which could effectively extract flavonoids from citrus peels.

The production of androst-4-ene-3,17-dione (AD) by the steroidal microbial cell factory requires transcription factors (TFs) to participate in metabolic regulation. However, microbial cell factory lacks effective TFs that can respond to AD in its metabolic pathway. Additionally, finding and obtaining natural TFs that specifically respond to AD is a complex and onerous task. In this study, we devised an artificial TF that responds to AD, termed AdT, based on structure-guided molecular dynamics (MD) simulation. According to MD analysis of the conformational changes of AdT after binding to AD, an LBD in which the N - and C -termini exhibited convergence tendencies was used as a microswitch to guide the assembly of a DNA-binding domain lexA, a linker (GGGGS) 2 , and a transcription activation domain B42 into an artificial TF. As a proof of design, a AD biosensor was designed and constructed in yeast on the basis of the ligand-binding domain (LBD) of hormone receptor. In addition, the transcription factor activity of AdT was increased by 1.44-fold for its variant F320Y. Overall, we created non-natural TF elements for AD microbial cell factory, and expected that the design TF strategy will be applied to running in parallel to the signaling machinery of the host cell.

Toddalia asiatica (L.) Lam. (TA) is a traditional folk medicine of ethnic minorities in the southwest of China. It is widely used in the treatment of dispersing blood stasis and activating blood. However, the effective substance and pharmacological mechanism have not been fully elucidated. The zebrafish larvae were treated with Phenylhydrazine (PHZ) to establish a thrombus model, and the staining intensity of zebrafish red blood cells was analyzed. The antithrombotic activity of TA was verified for the first time, and it was found that the inhibition rate of TA on thrombosis was up to 60.85 %. The chemical ingredients of TA were collected by combining UPLC-HRMS analysis and the literature research. Network pharmacology revealed that six key targets were obtained, which including TNF, AKT1, EGFR, PTGS2, PPARG, and IFNG. It showed that the PI3K-Akt pathway was a core signaling pathway. Coagulation factor III(TF), playing an important role in the process of hemostasis and thrombosis, which ranks high in the PPI network. Moreover, the results of molecular docking showed that the active components had a strong binding force with TF, which indicated that TF might be the key target of TA in treating thrombosis. In vitro experiments showed that TA could inhibit TNF-α-induced high expression of TF in EA.hy926 cells. In addition, TA could inhibit TNF-α-activated expression of Akt, IκBα and P65 protein phosphorylation in PI3K-Akt pathway. The results showed that TA had antithrombotic activity and exerted an antithrombotic effect by inhibiting the expression of TF through the PI3K-Akt-NF-κB signaling pathway.

Rapid release and clearance of antitumor drugs in vivo are the main factors used to evade the effectiveness of chemotherapeutics. Targeted delivery and controlled release of drugs are the most pressing dilemmas in cancer therapy. Herein we report the design and fabrication of multifunctional mesoporous silica nanoparticles coated with poly(N-isopropylacrylamide)-co-acrylic acid and calcium phosphate (MSCNs) with pH-triggered chemotherapeutic release and dual-targeting functions. By decorating the nanoparticle surface with a transferrin (Tf)/RGD ligand, these nanoparticles are capable of not only recognizing the intrinsic pH difference between tumor and normal tissues, but also targeting the lesion location. It was shown that Tf/RGD-MSCNs delivered the anti-tumor drug doxorubicin more efficiently into lysosomes and the resulting DOX-loaded nanoparticles (DOX-Tf/RGD-MSCNs) showed a stronger inhibitory effect towards tumor cell growth than free DOX and DOX delivered by unmodified MSNs. Moreover, the nanoparticles are more biocompatible than uncoated mesoporous silica nanoparticles. All these results indicate that Tf/RGD-MSCNs have great potential as a novel drug carrier in therapeutic applications against cancers.
