Acute and chronic combined effect of polystyrene microplastics and dibutyl phthalate on the marine copepod Tigriopus japonicus

Zhaochuan Li,Hao Zhou,Yang Liu,Jingjing Zhan,Wentao Li,Kaiming Yang,Xianliang Yi

Dibutyl phthalate (DBP) is a commonly used additive in plastic products, so it may potentially coexist with microplastics (MPs) in marine environment. The ingestion of MPs might affect the accumulation of DBP in marine organisms. In this study, the marine copepod Tigriopus japonicus was applied to study the combined effect of DBP and polystyrene microplastics (mPS) on the copepod through both acute mortality tests and chronic reproduction tests. The LC 50 of DBP was 1.23 mg L −1 (95% CI: 1.11–1.35 mg L −1 ), while exposure to mPS didn’t have significant lethal effect on the copepods. Adsorption to MPs led to decreased bioavailability of DBP, resulting in decreased toxicity of DBP. In contrast to the results of acute toxicity tests , DBP didn’t affect the reproduction of the copepods at lower exposure concentrations, while mPS reduced the number of nauplii and extended the time to hatch. Similar as acute toxicity tests, antagonistic interaction was observed for mPS and DBP in chronic reproduction tests, which might be attributed to promoted aggregation of mPS at presence of DBP. Overall, antagonistic toxicity effect between the two pollutants was observed for both acute and chronic tests, but the mechanisms of the interaction between DBP and mPS were different. Results of the present study highlighted the importance of long-term exposure when evaluating the toxic effect of MPs and their combined effect with other chemicals.

Effect of nano zinc oxide on the acute and reproductive toxicity of cadmium and lead to the marine copepod Tigriopus japonicus

Xianliang Yi,Tongtong Chi,Bei Liu,Chenxi Liu,Guanqun Feng,Xinyu Dai,Keke Zhang,Hao Zhou

Given the absorbability of nZnO and its inevitable coexistence with trace metals such as Cd and Pb in coastal environment, nZnO can adsorb these pollutants thereby affecting their distribution in different media of marine ecosystem. The marine copepod Tigriopus japonicus was applied in the present study to investigate the combined effect of nZnO and Cd or Pb on mortality and reproduction in marine organisms. For acute exposure, presence of 1.0 mg/L nZnO increased the toxicity of both Cd and Pb, as their LC 50 decreased from 5.9 and 75.4 mg/L to 3.95 and 48.0 mg/L, respectively. For 21 d chronic exposure, the reproduction of the copepod was influenced by Cd and Pb at environmental relevant concentrations, and the interaction between nZnO and Cd or Pb appeared to be antagonistic. The waterborne Cd and Pb concentration was affected by nZnO for neither acute nor chronic exposure, indicating no adsorption of these two metals to nZnO at relative low concentration. The overall findings of this study suggested the binary exposure to nZnO/Cd or nZnO/Pb might have potential different toxic mechanisms between acute and chronic exposure.

Combined toxicity of pristine or artificially aged tire wear particles and bisphenols to Tigriopus japonicus

Jinbo Song,Qian Meng,Hongyu Song,Xiaoming Ni,Hao Zhou,Yang Liu,Jingjing Zhan,Xianliang Yi

Tire wear particles (TWPs) are considered an important component of microplastic pollution in the marine environment and occur together with a variety of aquatic pollutants, including frequently detected bisphenols. The adverse effects of TWPs or bisphenols on aquatic organisms have been widely reported. However, the combined toxicity of TWPs and bisphenols is still unknown. In this study, the combined toxicity of both pristine (p-) and aged TWPs (a-TWPs) and four bisphenols ((bisphenol A (BPA), bisphenol F (BPF), bisphenol S (BPS), and bisphenol AF (BPAF)) to Tigriopus japonicus was evaluated. TWPs increased the toxicity of BPA and BPF but decreased the toxicity of BPAF. For BPS, there was synergistic toxic effect in the presence of p -TWPs, but slightly antagonistic effect was observed in the presence of a-TWPs. This adsorption of BPAF by TWPs resulted in a reduction of its toxicity to the copepod . A-TWPs could release more Zn than p -TWPs, and the released Zn contributed to the synergistic effect of TWPs and BPA or BPF. The aggregation formed by TWPs in certain sizes (e.g., 90–110 μm) could cause intestinal damage and lipid peroxidation in T. japonicus . The synergistic effect of p -TWPs and BPS might be due to the aggregation size of the binary mixture. The results of the current study will be important to understand the combined toxic effect of TWPs and bisphenols and the potential toxic mechanisms of the binary mixture.

Acute toxicity of tire wear particles, leachates and toxicity identification evaluation of leachates to the marine copepod, Tigriopus japonicus

Kaiming Yang,Siyuan Jing,Yang Liu,Hao Zhou,Yan Liu,Ming Yan,Xianliang Yi,Renyan Liu

Tire wear particles (TWPs) have been characterized as microplastics in recent years, and many of these TWPs will be eventually deposited in coastal areas, leading to adverse effects to marine organisms. Results of the acute toxicity test in this study showed that the 96-h LC50 values of the particles and leachate were 771.4 mg/L (95% CI = 684.4–869.6 mg/L) and 5.34 g/L (95% CI = 4.75–6.07 g/L), respectively. The chemical constituents of TWP and the leachate are very complex, and little research has been conducted to determine which of these constituents contribute to the toxicity of TWP leachate to marine organisms. Therefore, the composition of the TWP and leachate was analyzed, and a variety of chemicals were identified, including metals (Mn, Zn, etc.) and organic compounds (cyclohexanthiol, 4-ethyl-1,2-dimethylbenzene, benzothiazole, stearic acid , N-(1,3-dimethylbutyl)-N′-phenyl-p-phenylenediamine, etc.). In addition, the marine copepod Tigriopus japonicus was applied as a model species in the toxicity identification evaluation study to characterize, identify and confirm the toxicity-causing substances in the TWP leachate. Zn was identified and confirmed as the main toxicant contributing to the toxicity. Furthermore, Zn concentrations in the leachate over time were investigated. The release of Zn from TWPs to the aquatic environment was slow, and conformed to a parabolic model with a release constant k of 2.06. The organic component, benzothiazole, exhibited an antagonistic effect with zinc in the acute toxicity of the TWP leachate.

Toxic effect of triphenyltin in the presence of nano zinc oxide to marine copepod Tigriopus japonicus

Xianliang Yi,Keke Zhang,Gaorui Han,Mingyue Yu,Tongtong Chi,Siyuan Jing,Zhaochuan Li,Jingjing Zhan,Minghuo Wu

Marine organisms are naturally exposed to different environmental pollutants including organic pollutants and nanoparticles . The interactive effects between nanoparticles and other chemicals on aquatic organisms have raised concerns regarding the potential of nanomaterials as the vector for other chemicals. In the present study, the effect of nano zinc oxide (nZnO) on the bioavailability of triphenyltin chloride (TPTCl) was studied, and their combined acute and reproductive toxicity to the marine copepod Tigriopus japonicus were evaluated. At experimental concentration ranges of nZnO in this study, the percentage of dissolution of Zn 2+ was relative stable (from 62% to 66%), and nZnO did not affect the bioavailability of TPTCl to the copepods. The acute toxicity of binary mixtures of nZnO/TPT was equivalent to that of the mixture of Zn 2+ /TPT. In agreement with the decrease in TPTCl's LC 50 values at the presence of nZnO, their interacting effect was synergistic based on response addition response surface model, and the interacting parameter was modelled to be −1.43. In addition to acute toxicity test , reproductive toxicity tests revealed that exposure to nZnO and TPTCl didn't affect the successful mating rate and the number of nauplii in the 1st brood, but they extended the time for the eggs to hatch from 2.53 days to 3.94 and 3.64 days, respectively. The exposure to nZnO/TPTCl mixture delayed the time to hatch to 5.78 days.

Effect of Multi-walled Carbon Nanotubes on the Toxicity of Triphenyltin to the Marine Copepod Tigriopus japonicus

Yi Xianliang,Yu Mingyue,Li Zhaochuan,Chi Tongtong,Jing Siyuan,Zhang Keke,Li Wentao,Wu Minghuo

Marine organisms are often exposed to a mixture of various pollutants in marine environment (i.e., nanoparticles, organic pollutants). The present study investigated the potential effects of multi-walled carbon nanotubes (MWCNTs) on the toxicity of triphenyltin chloride (TPTCl). The results revealed an antagonistic interaction between MWCNTs and TPTCl on the copepod through 96 h acute exposure, which was attributed to the adsorption of TPTCl to MWCNTs and aggregation of MWCNTs in the test solutions. Results of 21 days’ chronic exposure showed that the effect concentration of MWCNTs could be 100 times lower than that of acute exposure. The exposure to binary mixture of MWCNT (1.0 mg/L) and TPTCl (0.3 µg/L) caused a reduction by 94% for the 3rd time spawning and 83% for the total number of hatched nauplii. The ingestion and exterior attachment of MWCNTs to the copepod might be the main reasons causing the adverse effect in reproduction.

Herbicide prometryn adversely affects the development and reproduction of Tigriopus japonicus by disturbing the ecdysone signal pathway and chitin metabolic pathway

Dong Wang,Guangxin Yang,Shaoguo Ru,Zhenzhong Zhang,Yuejiao Li,Jun Wang

Prometryn, a widely used triazine herbicide in agriculture and aquaculture, has been commonly detected in marine environments, but its effects on the marine copepod are unknown. In this study, marine copepod Tigriopus japonicus was chronically exposed to environmentally relevant concentrations of prometryn to investigate its impacts and potential mechanism of action. The results showed that 0.5, 5, and 50 μg/L prometryn delayed the first spawning time and hatching time, reduced the fecundity, and inhibited the population growth rate. Moreover, exposure to 0.5, 5 and 50 μg/L prometryn decreased food ingestion, the content of C and N elements, nutrient accumulation and body size, but increased the content of 20-hydroxyecdysone (20E). Transcriptome analysis showed that 50 μg/L prometryn down-regulated 1431 genes, which were mainly enriched in lysosome pathway and chitin binding and cuticle construction process. The results of qRT-PCR showed that the expression of key genes involved in juvenile hormone synthesis and chitin metabolic pathways were also inhibited after prometryn exposure. Molecular docking revealed that prometryn could bind to ecdysone receptor (EcR) and UDP-N-acetylglucosamine pyrophosphorylase (UAP), components of the ecdysteroid nuclear receptor complex. Therefore, environmental relevant prometryn delayed the molting and development of T. japonicus by disrupting the ecdysone signal pathway and chitin metabolic pathway through binding to EcR and UAP. This study provides new insights into toxic effects and molecular mechanisms of prometryn on marine copepods.
