Optimized degradation of bisphenol A by immobilized laccase from Trametes versicolor using Box-Behnken design (BBD) and artificial neural network (ANN)

Abdul Latif,Ahsan Maqbool,Runzhan Zhou,Muhammad Arsalan,Kai Sun,Youbin Si

This study represents one of the first attempts towards bisphenol A (BPA) degradation by Trametes versicolor laccase immobilized on Ba-alginate beads. The effect of input variables including temperature (20–40 °C), BPA concentration (2–10 mg/L) and time (10–90 min) were studied using Box-Behnken design (BBD) and artificial neural network (ANN). The maximum BPA degradation of 84.34% was obtained when the temperature was 40 °C, BPA concentration 2 mg/L and time 50 min, which was accurately predicted by BBD (83.48%) and ANN (84.33%), proving the accuracy of prediction for both models. The values of R 2 and MSE for BBD were found to be 0.98, 9.88, while for ANN were 0.97, 38.25, respectively. Based on higher R 2 value and lower MSE, BBD was slightly better than ANN. The immobilized laccase showed higher storage stability than free laccase by retaining 68.64% and 44.62% of their activity at the same experimental conditions. Furthermore, BPA transformation was confirmed by Fourier-transform infrared spectroscopy (FTIR) analysis. GC-MS had detected the oxidative degradation products from BPA. Results showed that Ba-alginate immobilized laccase could be a promising biocatalyst in treating organic pollutants .

Characterization of a Trametes versicolor aflatoxin B1-degrading enzyme (TV-AFB1D) and its application in the AFB1 degradation of contaminated rice in situ

Yang Peizhou,Xiao Wei,Lu Shuhua,Jiang Shuying,Jiang Suwei,Chen Jianchao,Wu Wenjing,Zheng Zhi,Jiang Shaotong

Aflatoxin B1 (AFB1) contaminates rice during harvest or storage and causes a considerable risk to human and animal health. In this study, Trametes versicolor AFB1–degrading enzyme (TV–AFB1D) gene recombinantly expressed in engineered E. coli BL21 (DE3) and Saccharomyces cerevisiae. The TV–AFB1D enzymatic characteristics and AFB1 degradation efficiency in contaminated rice were investigated. Results showed that the size of recombinant TV-AFB1D expressing in E. coli BL21 (DE3) and S. cerevisiae was appropriately 77 KDa. The kinetic equation of TV-AFB1D was y = 0.01671x + 1.80756 (R2 = 0.994, Km = 9.24 mM, and Vmax = 553.23 mM/min). The Kcat and Kcat/Km values of TV-AFB1D were 0.07392 (s−1) and 8 M−1 s−1, respectively. The AFB1 concentration of contaminated rice decreased from 100 μg/ml to 32.6 μg/ml after treatment at 32°C for 5 h under the catabolism of TV-AFB1D. S. cerevisiae engineered strains carrying aldehyde oxidase 1 (AOX1) and Cauliflower mosaic virus 35 S (CaMV 35 S) promoters caused the residual AFB1 contents, respectively, decreased to 3.4 and 2.9 μg/g from the initial AFB1 content of 7.4 μg/g after 24 h of fermentation using AFB1-contaminated rice as substrate. The AFB1 degradation rates of S. cerevisiae engineered strains carrying AOX1 and CaMV promoters were 54 and 61%, respectively. Engineered S. cerevisiae strains integrated with TV-AFB1D expression cassettes were developed to simultaneously degrade AFB1 and produce ethanol using AFB1-contaminated rice as substrate. Thus, TV-AFB1D has significant application potential in the AFB1 decomposition from contaminated agricultural products.

Conditions Optimizing and Application of Laccase-mediator System (LMS) for the Laccase-catalyzed Pesticide Degradation

Jin Xiaoting,Yu Xiangyang,Zhu Guangyan,Zheng Zuntao,Feng Fayun,Zhang Zhiyong

A high capacity of laccase from Trametes versicolor capable of degrading pesticides has been revealed. The conditions for degrading of five selected pesticides including chlorpyrifos, chlorothalonil, pyrimethanil, atrazine and isoproturon with the purified laccases from Trametes versicolor were optimized. The results showed that the optimum conditions for the highest activity were pH at 5.0 and temperature at 25 °C. The best mediators were violuric acid for pyrimethanil and isoproturon, vanillin for chlorpyrifos, and acetosyringone and HBT for chlorothalonil and atrazine, respectively. The laccase was found to be stable at a pH range from 5.0 to 7.0 and temperature from 25 to 30 °C. It was observed that each pesticide required a different laccase mediator concentration typically between 4.0–6.0 mmol/L. In the experiment, the degradation rates of pyrimethanil and isoproturon were significantly faster than those of chlorpyrifos, chlorothalonil and atrazine. For example, it was observed that pyrimethanil and isoproturon degraded up to nearly 100% after 24 hours while the other three pesticides just reached up 90% of degradation after 8 days of incubation.

Degradation of pharmaceuticals and personal care products (PPCPs) in municipal wastewater by white rot fungi: Neutral environment exploitation and degradation mechanisms

Yizhou Tu,Liqin Han,Xiaolu Ding,Xingqi Zhu,Guizhou Xu,Xianchuan Xie,Qing Zhou,Chendong Shuang,Aimin Li

Refractory pharmaceuticals and personal care products (PPCPs) represented by diclofenac, ibuprofen, sulfamethoxazole, and carbamazepine have become risk compounds in municipal wastewater, and they possessed generalized antimicrobial properties along with numerous electron-withdrawing groups, difficult to biodegrade through bacterial-based systems. In contrast, fungi could thrive even in environments contaminated by antibiotics, among which the white rot fungus Trametes versicolor ( T. versicolor ) possessed enzymatic degradation systems represented by laccase, capable of degrading complex chemical bonds. However, almost all white rot fungi reactors were conducted at pH 4.5, which limited their application. In this study, the neutral degradation environment was developed for T.   versicolor , and the pH-dominated laccase degradation mechanism was clarified. It was found that T. versicolor could possess both laccase activity and production at pH 6.5, with better application potential compared to acidic conditions. Accordingly, fluidized bed T.   versicolor reactors were developed, and their application potential under neutral pH was validated for the first time in municipal wastewater. Compared with reactors without any adjustment, the four refractory PPCPs removal by T.   versicolor reactors was increased from 26% to 72%, and T.   versicolor abundance was increased from almost zero to 27% by adding carbon and nitrogen sources and adjusting pH to 6.5. Metabolic pathway analysis revealed the cytochrome P450 and amide hydrolase functional genes of T. versicolor , which were critical for PPCPs degradation. Nine transformation products of PPCPs were identified, further validating the multiple capabilities of T.   versicolor for both hydroxylation and complex chemical structures (carbon–nitrogen bonds, epoxides, carbon–sulfur bonds) degradation. Furthermore, these findings also provided new ideas for the degradation of more complex aminobenzene or chlorinated aniline wastewater.

Cu2+-assisted laccase from Trametes versicolor enhanced self-polyreaction of triclosan

Kai Sun,Shunyao Li,Jialin Yu,Rui Gong,Youbin Si,Xiaohong Liu,Gang Chu

Laccase-mediated humification processes (L-MHPs) can be used to polymerize and transform phenolic pollutants in water. However, the mechanism on Cu 2+ impacts the self-polymerization of multi-purpose antimicrobial agent triclosan during L-MHPs is less understood. Here, the influence of divalent metal ions (DMIs) on Trametes versicolor laccase activity was investigated. Particularly, the performance of Cu 2+ -assisted laccase in polymerizing and transforming triclosan was systematically characterized. Compared with DMI-free, the activity of laccase was obviously accelerated with Cu 2+ present due to copper is a vital component of laccase catalytic center. It was found that Cu 2+ -assisted laccase was effective in transforming triclosan, and the enzymatic reaction kinetic constants increased from 0.28 to 0.73 h −1 as the Cu 2+ concentration increased (0–3.0 mM). Identification of intermediate products revealed that laccase oxidation predominantly generated triclosan dimers, trimers , and tetramers. The presence of Cu 2+ reinforced self-polymerization of triclosan via forming more triclosan oligomers relative to the Cu 2+ -free, which likely attributed to the enhancement of laccase activity and stability with Cu 2+ present in L-MHPs. A possible transformation mechanism was proposed as follows: Laccase initially catalyzed the oxidation of triclosan to generate phenoxy radical intermediates, which self-coupled to each other subsequently by radical-mediated C C and C O C covalent binding, forming oligomers and polymers. The growth inhibitory assays of freshwater microalgae ( Chlamydomonas reinhardtii and Scenedesmus obliquus ) demonstrated that the self-polymerized triclosan by L-MHPs had lower toxicity than the parent compound. These findings implied that Cu 2+ -assisted laccase was an effective strategy for rapidly self-polyreaction and detoxication of triclosan from Cu 2+ -triclosan combined polluted wastewater.

Enhanced biodegradation of benzo[a]pyrene with Trametes versicolor stimulated by citric acid

Zhang Zhou,Wang Litao,Liang Hong,Chen Guanyu,Tao Huayu,Wu Jing,Gao Dawen

Polycyclic aromatic hydrocarbons (PAHs) are a class of persistent organic pollutants with carcinogenic, mutagenic and teratogenic effects. The white-rot fungi in the fungal group have significant degradation ability for high molecular weight organic pollutants. However, exogenous fungi are easily antagonized by indigenous microorganisms. Low molecular weight organic acids, a small molecular organic matter secreted by plants, can provide carbon sources for soil microorganisms. Combining organic acids with white rot fungi may improve the nutritional environment of fungi. In this study, immobilized Trametes versicolor was used to degrade benzo[a]pyrene in soil, and its effect on removing benzo[a]pyrene in soil mediated by different low molecular weight organic acids was investigated. The results showed that when the degradation was 35 days, the removal effect of the experimental group with citric acid was the best, reaching 43.7%. The degradation effect of Trametes versicolor on benzo[a]pyrene was further investigated in the liquid medium when citric acid was added, and the effects of citric acid on the biomass, extracellular protein concentration and laccase activity of Trametes versicolor were investigated by controlling different concentrations of citric acid. In general, citric acid can act as a carbon source for Trametes versicolor and promote its extracellular protein secretion and laccase activity, thereby accelerating the mineralization of benzo[a]pyrene by Trametes versicolor . Therefore, citric acid can be used as a biostimulant in the remediation of PAHs contaminated soil with Trametes versicolor .

Catalyzed degradation of polycyclic aromatic hydrocarbons by recoverable magnetic chitosan immobilized laccase from Trametes versicolor

Jibao Deng,Hefei Wang,Haisheng Zhan,Chenxi Wu,Yi Huang,Bing Yang,Ahmed Mosa,Wanting Ling

The capability of laccase to oxidate a broad range of polyphenols and aromatic substrates in vitro offers a new technological option for the remediation of polycyclic aromatic hydrocarbon (PAH) pollution with high cytotoxicity. However, laccase application in the remediation of PAH-contaminated sites mainly suffers from a low oxidation rate and high cost because of the difficulty in its recovery. In this study, laccases were immobilized on magnetic Fe 3 O 4 particles coated with chitosan (Fe 3 O 4 @SiO 2 -chitosan) to improve the operational stability and reusability in the treatment of PAH pollution. The enzyme fixation capacity reached 158 mg g −1 , and 79.1% of free laccase activities were reserved under the optimum immobilized condition of 4% glutaraldehyde, 1.0 mg mL −1 laccase, 2 h covalent bonding time, and 6 h fixation time. The degradation efficiencies of anthracene (ANT) and benzo[a]pyrene (B(a)P) by Fe 3 O 4 @SiO 2 -chitosan immobilized laccase in 48 h were 81.9% and 69.2%, respectively. Furthermore, it is very easy to magnetically recover the immobilized laccase from reaction systems and reuse it in a new batch. The relative activities of immobilized laccase were over 50% for the degradation of ANT and B(a)P in three catalytic runs, reaching the goal of substantially reducing cost in practice. According to the results from quantum calculations and mass spectrum analyses, the degradation products of ANT and B(a)P by laccase were anthraquinone and B(a)P-dione, respectively. The findings from this study provide valuable insight in promoting the application of immobilized laccase technology in the remediation of PAH contamination.

Myco–macromolecular symbiosis: Chitosan–folate ZnO nanoplatforms from Trametes versicolor for dual-functional oncological and antibacterial therapy

Mojtaba Ranjbar,Masumeh Ahmadpour,Mahmoud Kiani,Mostafa Govahi

The increasing prevalence of antibiotic-resistant bacteria and cancer necessitates the development of novel, targeted therapeutic strategies. This study aimed to develop a multifunctional nanoplatform combining antibacterial and anticancer properties through green synthesis and strategic surface functionalization. Zinc oxide nanoparticles (ZnO NPs) were biosynthesized using Trametes versicolor extract as a reducing and capping agent, then surface-functionalized with chitosan (Cs) for enhanced biocompatibility and conjugated with folic acid (FA) for targeted delivery. The NPs were characterized using multiple analytical techniques and evaluated for antibacterial activity against Gram-positive ( Bacillus subtilis , Staphylococcus aureus ) and Gram-negative ( Escherichia coli ) bacteria. Cytotoxicity was assessed in cancer cells (MDA-MB-231) and normal cells (MCF-10 A), followed by gene expression analysis of key oncogenic and apoptotic markers. Characterization confirmed the formation of crystalline, core-shell structures with successful ligand attachment. Cs-ZnO NPs demonstrated significantly enhanced antibacterial activity against all tested bacterial strains compared to bare ZnO NPs. FA-Cs-ZnO NPs exhibited selective cytotoxicity toward cancer cells while maintaining biocompatibility with normal cells. Gene expression analysis revealed down-regulation of cancer stemness genes ( CD44 , SOX2 ) and proliferation markers ( mTOR , β-catenin ), coupled with up-regulation of the apoptotic marker Caspase 3 . This green-synthesized, dual-functional nanoplatform demonstrates promising potential for combined antibacterial and targeted anticancer therapy, warranting further in vivo evaluation for clinical translation.

Adsorption of Trametes versicolor laccase to soil iron and aluminum minerals: Enzyme activity, kinetics and stability studies

Yue Wu,Ying Jiang,Jiaguo Jiao,Manqiang Liu,Feng Hu,Bryan S. Griffiths,Huixin Li

Laccases play an important role in the degradation of soil phenol or phenol-like substance and can be potentially used in soil remediation through immobilization. Iron and aluminum minerals can adsorb extracellular enzymes in soil environment. In the present study, we investigated the adsorptive interaction of laccase, from the white-rot fungus Trametes versicolor , with soil iron and aluminum minerals and characterized the properties of the enzyme after adsorption to minerals. Results showed that both soil iron and aluminum minerals adsorbed great amount of laccase, independent of the mineral specific surface areas. Adsorbed laccases retained 26–64% of the activity of the free enzyme. Compared to the free laccase, all adsorbed laccases showed higher K m values and lower V max values, indicating a reduced enzyme-substrate affinity and a lower rate of substrate conversion in reactions catalyzed by the adsorbed laccase. Adsorbed laccases exhibited increased catalytic activities compared to the free laccase at low pH, implying the suitable application of iron and aluminum mineral-adsorbed T. versicolor laccase in soil bioremediation, especially in acid soils. In terms of the thermal profiles, adsorbed laccases showed decreased thermal stability and higher temperature sensitivity relative to the free laccase. Moreover, adsorption improved the resistance of laccase to proteolysis and extended the lifespan of laccase. Our results implied that adsorbed T. versicolor laccase on soil iron and aluminum minerals had promising potential in soil remediation.

Trametes versicolor laccase-assisted oxidative coupling of estrogens: Conversion kinetics, linking mechanisms, and practical applications in water purification

Kai Sun,Dan Hong,Jie Liu,Abdul Latif,Shunyao Li,Gang Chu,Wenxiu Qin,Youbin Si

The potential application of fungal laccase-assisted bioremediation for estrogen elimination has caused tremendous attention. Herein, naturally-occurring Trametes versicolor laccase ( Tv lac) was capable of significantly removing 17 β -estradiol (E2) and 17 α -ethynylestradiol (EE2) at pH 5. A lower (or higher) pH caused decline in the velocity constants ( k prcs ) by obstructing the single-electron oxidation of estrogen at Tv lac T1-Cu site (or intercepting the intramolecular-electron transfer between Tv lac T1-Cu and T2/T3-Cu sites). Humic acid (HA) and natural phenolic compounds (PCs) are omnipresent in water and can influence the conversion kinetics of estrogen in Tv lac-triggered reactions. Compared with HA-free, the k prcs values of E2 and EE2 in presence of HA respectively reduced 58.61%–83.72% and 69.72%–95.62% at different pH levels. Additionally, the k prcs values for estrogen were also hampered obviously by O -dihydroxyphenol structure of model PCs, owning to the created monomeric/polymeric O -quinones inverted estrogen phenoxy radicals. The generated carbon‑carbon/oxygen self-linking aggregates during Tv lac-mediated estrogen oxidative coupling were characterized, and the polymerization mechanisms were speculated. A fluid-bed reactor with cross-linked-entrapped Tv lac in Ca-alginate beads was established to treat E2 and EE2, respectively. Compared with free Tv lac, immobilized Tv lac prominently exhibited a higher pH and temperature stability. Especially, the fluid-bed reactor with immobilized Tv lac could be reused, keeping its high conversion efficiencies of E2 and EE2 in natural water, only changing from 99.2% to 73.6% and from 98.5% to 70.9% after 1 to 5 cycles, respectively. These findings present an alternative immobilized fungal laccase-based clean biotechnology for continuously handling estrogen-contaminated waterbodies at a large-scale.

Activity enhancement of Trametes versicolor aflatoxin B1-degrading enzyme (TV-AFB1D) by molecular docking and site-directed mutagenesis techniques

Peizhou Yang,Shuhua Lu,Wei Xiao,Zhi Zheng,Suwei Jiang,Shaotong Jiang,Shuying Jiang,Jieshun Cheng,Danfeng Zhang

Aflatoxin B 1 (AFB 1 ) is highly carcinogenic and teratogenic as a secondary metabolite of Aspergillus flavus and Aspergillus parasiticus . Trametes versicolor aflatoxin B 1 -degrading enzyme (TV-AFB 1 D) has capability of AFB 1 degradation. In this study, in order to improve the catalytic performance of TV-AFB 1 D, the molecular docking and site-directed mutagenesis techniques were applied to modify the key amino acid sites. Three mutation sites of Y305A, E436A, and H554A were selected for the directed mutation of TV-AFB 1 D. Four TV-AFB 1 D combinations of mutant H554A, Y305A/H554A, E436A/H554A, Y305A/E436A/H554A had potential to modify the catalytic performance according to the value of de-folding free energy. The TV-AFB 1 D mutations were expressed in engineering E. coli BL21 (DE3) with the size of approximately 77 kDa. The optimal temperature and pH value of TV-AFB 1 D were 32 °C and pH 7, respectively. E436A/H554A mutant represented the highest enzyme activities among these four mutants. The specific activity of E436A/H554A mutant (36 U/mg) was 1.84-fold in comparison with wild-type TV-AFB 1 D (19.6 U/mg) under the conditions of pH 7 and 32 °C. This study would contribute to the activity increase of TV-AFB 1 D by the performance improvement of AFB 1 degradation by site-directed mutagenesis approach.

Immobilization of Trametes Versicolor laccase on Cu-alginate beads for biocatalytic degradation of bisphenol A in water: Optimized immobilization, degradation and toxicity assessment

Abdul Latif,Ahsan Maqbool,Kai Sun,Youbin Si

The present work investigated the immobilization of laccase ( Trametes versicolor ) on Cu-alginate beads by entrapment method and its utilization to degrade bisphenol A (BPA) in aqueous medium. The immobilized laccase demonstrated higher storage stability as compared to free enzyme. Box-Behnken experimental design was used to optimize the immobilization conditions (sodium alginate concentration, CuSO 4 molarity, and hardening time). The optimal conditions were found to be 3% sodium alginate concentration (W/V), 0.141 mM CuSO 4 and 90 min hardening time. Experimental data exhibited that immobilized laccase (0.5 g) could degrade 96.12% of BPA (10 mg/L) at pH 5.0, 30°C, 150 rpm and 1 h of reaction. The degradation products of BPA were identified by LC-MS analysis; hence the degradation pathways were suggested. The toxicity of BPA products was judged by phytotoxicity test. Immobilized laccase could also degrade BPA in real water, hence could be considered as an efficient candidate for removing BPA in water.
