In this paper, emulsions stabilized byTremella fuciformispolysaccharides (TFP) were prepared and the physiochemical properties were assessed. Results showed that the TFP emulsions illustrated the highest emulsifying activity (EAI) and emulsifying stability (ESI) when the concentration of TFP and oil were 0.8% and 10% (wt%). The higher pH value was in favor of the emulsifying properties, while the addition of NaCl impaired the stability, and the greater the concentration, the lower the EAI and ESI. Besides, the emulsifying and rheological properties and stability analysis were evaluated in comparison with gum arabic, pectin, and carboxymethyl cellulose emulsions. It was discovered that TFP illustrated better storage and freeze-thaw stability, which was proved by the result of zeta-potential and particle size. The rheological measurement revealed that all the emulsions behaved as pseudoplastic fluids, while TFP displayed a higher viscosity. Meanwhile, TFP emulsions tended to form a more stable network structure according to the analysis of the parameters obtained from the Herschel–Bulkley model. FTIR spectra suggested that the O-H bond could be destructed without the formation of new covalent bonds during the emulsion preparation. Therefore, this study would be of great importance for the research of emulsions stabilized by TFP as a natural food emulsifier. Keywords:Tremella fuciformispolysaccharides;emulsifying properties;rheological properties;stability

Polysaccharides from Tremella spp. are known to have various functions and applications. To fully understand the polysaccharide compositions of fruiting body of T. fuciformis , cold water extract (CWE), hot water extract (HWE), dilute alkaline extract (ALE), and dilute acid extract (ACE) were sequentially prepared and characterized. Their physicochemical properties, chemical composition, micromorphology, and functional properties were determined. Firstly, the four extracts showed significant difference in chemical compositions. Secondly, in the structural aspect, liquid chromatographic elution curves of the extracts had only one main peak except that CWE had two peaks. Fucose , galactose (or glucose), xylose , and mannose were detected in CWE and HWE. Differently, ALE had fucose, glucose, and xylose, and ACE had only galactose and glucose, which agreed with the information obtained from Fourier transform infrared spectra. Thirdly, in the physicochemical and nutritional aspects, the thermal stability of the four extracts decreased in the order of: ACE>HWE≈ALE>CWE. HWE and ALE had similar dynamic viscoelasticity , exhibiting viscous characteristics at low frequencies and elastic characteristics at higher vibration frequencies. The four extracts were digestive-resistant after in vitro mimic experiments. This study could provide some theoretical data support for the scientific research and its application in food , medicine, and cosmetics of tremella polysaccharides.

Cuproptosis affects osteosarcoma locally, and the exploitation of cuproptosis-related biomaterials for osteosarcoma treatment is still in its infancy. We designed and synthesized a novel injectable gel of Cu ion–coordinated Tremella fuciformis polysaccharide (TFP–Cu) for antiosteosarcoma therapy. This material has antitumor effects, the ability to stimulate immunity and promote bone formation, and a controlled Cu 2+ release profile in smart response to tumor microenvironment stimulation. TFP–Cu can selectively inhibit the proliferation of K7M2 tumor cells by arresting the cell cycle and promoting cell apoptosis and cuproptosis. TFP–Cu also promoted the M1 polarization of RAW264.7 cells and regulated the immune microenvironment. These effects increased osteogenic gene and protein expression in MC3T3-E1 cells. TFP–Cu could significantly limit tumor growth in tumor-bearing mice by inducing tumor cell apoptosis and improving the activation of anti-CD8 T cell-mediated immune responses. Therefore, TFP–Cu could be a potential candidate for treating osteosarcoma and bioactive drug carrier for further cancer-related applications.

Tremella fuciformis polysaccharides (TFPS) belong to natural bioactive macromolecule with both edible and medicinal value, possessing much bioactivities such as anti-tumor, antioxidant, antidiabetics, and immunomodulatory. Currently, the production of TFPS through submerged fermentation (TFPS-1) is gradually replacing the polysaccharides extracted from the fruiting body due to improved fermentation efficiency and reduced separation costs. However, it is still unclear about the effect of TFPS-1 on gastrointestinal digestion and gut microbial fermentation, which is directly related to the function of its biological activity. This study aimed to illustrate the effect of TFPS-1 on digestive process through in vitro digestion and fecal fermentation. TFPS-1 was indigestible during the simulated gastrointestinal tract. But TFPS-1 can be digested by intestinal flora leading to alterations in the total polysaccharides content, molecular weight, and apparent viscosity. Moreover, TFPS-1 regulated the composition of gut microbial, lowering the proportion of Firmicutes to Bacteroidetes and enhancing the abundances of Parabacteroides and Bacteroides. The change of intestinal flora produced more short chain fatty acids and lowered the pH. The KEGG metabolic pathway analysis indicated that TFPS-1 enriched lipid metabolism, glycan biosynthesis and metabolism, and biosynthesis of other secondary metabolites. This study demonstrated that T. fuciformis fermented polysaccharide can be a potential prebiotic to optimize intestinal flora homeostasis and maintain host intestinal health.

Tremella fuciformis is an edible and medicinal mushroom. Polysaccharides from T. fuciformis have received increasing attention due to their diversely pharmacological activities. In this study, the digestive behavior and fermentation characteristics of T. fuciformis polysaccharides (TFP) were studied. The results revealed that the reducing sugar content, chemical composition, molecular weight, rheological property, constituent monosaccharide, and FT-IR spectrum of TFP were not altered after the in vitro simulated digestion, indicating that it was indigestible under different simulated digestion conditions. However, the physicochemical characteristics of TFP, including reducing sugar content, molecular weight, constituent monosaccharide, and free monosaccharide released, were obviously altered after the in vitro fermentation for 48 h, indicating that it was remarkably utilized by intestinal microbiota in human feces. Notably, TFP could obviously modulate the microbial composition via promoting the relative abundances of Phascolarctobacterium , Bacteroides , and Lachnoclostridium . Moreover, TFP could also increase the production of short-chain fatty acids, including acetic, propionic, n -butyric, and n -valeric acids, after the in vitro fermentation for 48 h. These results showed that TFP was stable under the simulated digestion conditions, but could be utilized by intestinal microbiota in human feces, and might possess the potential to improve intestinal health.

Tremella fuciformis , one of the most approved mushrooms worldwide, possesses both edible and medicinal value. However, low transformation efficiency and high false positive rate, long verification period, and low expression of heterologous proteins hinder further genetic research. In this study, a polyethylene glycol-mediated genetic transformation system was established by using the homologous resistance marker carboxin in T. fuciformis . By optimizing the protoplast quality and transformation conditions, the transformation efficiency reached 8.23 CFU/μg, and the positive rate reached 64.95%. Moreover, Driselase-Heating-Freezing screening system was developed for rapid transformants verification, which took only 35–45 min and made the colony PCR 100% accurate. Considering the rationality of tRNA use frequency in heterologous genes, the coordinated relative synonymous codon usage (RSCU) strategy was adopted to recode the heterologous gene sequence, which ensured the protein expression level. This study comprehensively enhances molecular biology tools in T. fuciformis , and lays a foundation for further development.

Six representative small-molecule constituents were isolated and structurally characterized from the edible mushroomTremella fuciformis:cis-oleic acid (1), 1-oleoylglycerol (2), cerevisterol (3), a cerebroside (4),trans-homoaconitic acid (5), and uridine (6). Compound5, a tricarboxylic acid derivative, is reported for the first time as abundantly accumulated in this fungus. Aging-relevant assays showed that 1 potently inhibited α-glucosidase (IC50= 0.02 mM, surpassing acarbose) and moderately inhibited acetylcholinesterase (IC50= 0.29 mM);5competitively inhibited alkaline phosphatase (IC50= 5.47 mM) and suppressed GluN2A-containing NMDA receptor currents (IC50= 0.40 mM), the first such dual report for a fungal tricarboxylic acid; and4inhibited tyrosinase (IC50= 0.80 mM) comparably to kojic acid. HPLC confirmed meaningful recovery under mild hydro-alcoholic extraction compatible with functional food processing. These multifunctional small molecules target carbohydrate metabolism, cognition, and skin homeostasis, supportingT. fuciformisas a source of healthy-aging food ingredients.

The objective of this research was to develop and analyse a chitosan- Tremella fuciformis polysaccharide (CS-TFP) edible film using chitosan (CS) and Tremella fuciformis polysaccharide (TFP) as the primary raw materials. The findings revealed significant improvements in the thickness, solubility, opacity, mechanical properties and barrier properties (water vapour and oxygen permeability) of the chitosan film upon the addition of TFP, but water content and hydrophobicity were reduced. Moreover, TFP addition markedly enhanced the antibacterial and antioxidant activities of the chitosan films. Structural analysis of the CS films and CS-TFP edible films indicated that the formation of hydrogen bonds and high compatibility between CS and TFP. Specifically, TFP increased crystallinity, reduced roughness and improved the compactness of the edible film. This increase in compactness was directly linked to enhanced physical properties, whereas increased crystallinity contributed to better thermal stability. Furthermore, the application of CS-TFP-3 edible films in chicken preservation proved that they could significantly delay the deterioration of chicken. Findings in this study suggested that CS-TFP edible film as a bioactive packaging material showed great potential to preserve chicken meat and extend the shelf life.

BACKGROUND Because of their diverse biological activities, polysaccharides derived from Tremella fuciformis have received growing attention. This study aimed to investigate the structural characterization of a purified polysaccharide (designated as PTP-3a) derived from T. fuciformis and explore its interaction with gut microbiota in vitro . RESULTS The findings revealed that PTP-3a had a molecular weight of 1.22 × 10 3 kDa and consisted of fucose, glucose, xylose, mannose and glucuronic acid in a molar ratio of 0.271:0.016:0.275:0.400:0.038. The primary linkage types identified in PTP-3a were 1,3-linked-manp, 1,4-linked-xylp and 1,2,3-linked-fucp, with corresponding ratios of 0.215:0.161:0.15. In addition, PTP-3a demonstrated notable thermal stability and exhibited a triple-helical structure. Moreover, following in vitro fermentation for 48 h, PTP-3a was efficiently utilized, resulting in a reduction in carbohydrate levels, the production of short-chain fatty acids (SCFAs) and pH adjustment. Furthermore, during in vitro fecal microbial fermentation, PTP-3a decreased the relative abundance of Firmicutes while increasing the proportions of Bacteroidetes and Proteobacteria, resulting in a significantly reduced Firmicutes/Bacteroidete s ratio. Additionally, PTP-3a stimulated the growth of beneficial bacteria such as Parabacteroides merdae , Gordonibacter pamelaeae , Bifidobacterium pseudolongum and Parabacteroides distasonis . Importantly, a strong correlation was observed between the production of SCFAs and specific microorganisms. CONCLUSION These findings suggested that PTP-3a has potential as a prebiotic for modulating the gut microbiota. © 2024 Society of Chemical Industry.

Carbon allocation trade-off enables SERS metabolic phenotyping of fungi. • Polysaccharides/carbohydrates ratio serves as metabolic SERS signatures. • A superior accuracy of 99.2% and an AUC value of 1.0 are achieved. • Portable device enables 15-min on-site fungal phenotyping traceability assay. • It upgrades SERS from chemical fingerprinting to bio-metabolic phenotyping. Background Accurate metabolic phenotyping and geographical traceability of gelatinous fungi are critical for uncovering fungal physiological adaptation mechanisms and advancing edible fungus quality supervision. However, high phenotypic similarity and subtle metabolic differences in homologous fungal samples pose formidable challenges to conventional physicochemical assays. Especially, relying on chemical fingerprinting without biological phenotypic implications, conventional surface-enhanced Raman scattering (SERS) analysis is unable to discern delicate metabolic discrepancies in fungal samples. Results Herein, we propose a metabolic characteristic peak ratio-based SERS strategy using Tremella fuciformis ( T. fuciformis ) as the model. By extracting intrinsic metabolic signatures represented by the carbon allocation trade-off between stress-resistant functional polysaccharides and storage-type carbohydrates, it delivers metabolic specificity and detection stability/reproducibility that are not possible with traditional fingerprint-based SERS fungal analysis. Integrated with machine learning, this method accurately differentiates six T. fuciformis samples, including specimens with distinct metabolic phenotypes as well as geographically differentiated samples with highly similar phenotypic features, achieving a superior accuracy of 99.2% and an AUC of 1.0. Combined with a custom-built portable Raman device we constructed, the entire detection procedure can be completed within 15 min, enabling efficient on-site analysis. Significance and novelty This study establishes a bio-signature-oriented SERS paradigm that upgrades chemical fingerprint identification to fungal metabolic phenotyping, providing a versatile technical platform for exploring fungal metabolic variation and phenotypic differentiation. The effect of deproteinized Tremella olysaccharide (DTP) on the in vitro digestion of potato starch (PS) was investigated. DTP reduced the hydrolysis rate of PS in a concentration-dependent manner, with digestion rates of 76.99 %, 72.03 %, and 29.73 % after 180 min for 0.3, 0.6, and 1.2 mg/mL DTP, respectively. DTP delayed digestion by shifting the kinetics from single-phase to biphasic behavior. It significantly inhibited pancreatic α-amylase (IC 50 = 5.76 mg/mL) by modifying the enzyme's secondary structure and reducing substrate binding. Additionally, DTP enhanced digesta viscosity, which may have contributed to the reduction in postprandial glucose release, thereby supporting its role as a potential hypoglycemic agent. However, DTP exhibited minimal glucose adsorption, binding only 1.2 % of 100 mM glucose at 1.2 mg/mL. These findings suggest that DTP reduces starch digestion primarily by inhibiting α-amylase activity and increasing digestive viscosity, providing insights into its hypoglycemic potential.

To investigate the composition, structure, and physical and chemical properties of Tremella fuciformis polysaccharide (TFPS), four new polysaccharide fractions (TFPS1, TFPS2, TFPS3, and TFPS4) were successfully isolated from hot water-extracted crude polysaccharides using a DEAE-52 cellulose column. Their structures were analyzed using FT-IR, SEM, HPGPC, monosaccharide composition, Congo red and I 2 -KI experiments. TFPS1 was found to be a heteropolysaccharide with a molecular weight of 742 Da for the major polysaccharide fragment. TFPS2, TFPS3, and TFPS4 were identified as single-component homogeneous polysaccharides with molecular weights of 350774 Da, 622675 Da, and 624724 Da, respectively. Glucose was the dominant monosaccharide of TFPS1 and TFPS2, while mannose was the major monosaccharide of TFPS3 and TFPS4. The results showed that TFPS1 and TFPS2 displayed a stable triple helix structure in aqueous solution, while TFPS3 and TFPS4 had longer side chains and more branched chains. Additionally, all four TFPSs showed in vitro antioxidant activity, as well as significant cholesterol binding and sodium taurocholate binding abilities.
