Five new sulphur-containing amides from Glycosmis lucida with antifeedant activity against Tribolium castaneum

Kai Yang,Shan-Shan Guo,Zhu-Feng Geng,Chun-Xue You,Wen-Juan Zhang,Yin-Ping Li,Cheng-Fang Wang,Shu-Shan Du,Zhi-Wei Deng

Five new sulphur-containing amides ( 2 , 5 – 8 ) and three known analogue compounds ( 1 , 3 – 4 ) were isolated from the leaves of Glycosmis lucida which were collected in Yunnan province, China. Their structures were elucidated with NMR spectroscopy and mass spectrometry. The compounds were identified as N -( p -hydroxyphenethyl)-3-(methylsulfonyl)-propenamide ( 1 ), N -[2-(4- hydroxyphenyl)-ethyl]-3-methanesulfonyl- N -methyl-propionamide ( 2 ), methylgerambullin ( 3 ), methylisogerambullone ( 4 ), methylgerambullindiol ( 5 ), N -{2-[4-(6,7-hydroxy-3,7-dimethyl-oct- 2-enyloxy)-phenyl]-ethyl}-3-methanesulfonyl- N -methyl-acrylamide ( 6 ), N -{2-[4-(6,7-dihydroxy- 3,7-dimethyl-oct-2-enyloxy)-phenyl]-ethyl}-3-methanesulfonyl- N -methyl-propionamide ( 7 ), ( E )- N -{2-[4-(6-hydroxy-7-dimethyl-oct-2-enyloxy)-phenyl]-ethyl}-3-methanesulfinyl- N -methyl- acrylamide ( 8 ). Feeding deterrent activities of these surphur-containing derivatives against Tribolium castaneum were assayed for the first time. Compound 2 , 3 , 6 possessed significant feeding deterrent activities against T. castaneum adults (EC 50   =   311.6, 244.9 and 108.3   ppm, respectively). These results demonstrated that some sulphur-containing amides are promising candidates as insect antifeedant agents.

Glutathione S-transferase TcGSTu1 contributes to defense against eucalyptol in Tribolium castaneum

Gao Shanshan,Cao Yizhuo,Miao Wenbo,Li Dongyu,Zhou Can,Zhang Kunpeng

Eucalyptol is one of the major insecticidal active ingredients in a variety of plant essential oils, and has good killing and avoidance effects on Tribolium castaneum . The presence of detoxifying enzymes glutathione S-transferase (GST) in T. castaneum makes it resistant to a variety of insecticides. However, whether GST is involved in regulating the sensitivity of eucalyptol by T. castaneum is not well understood. In our previous study, a glutathione S-transferase, TcGSTu1 , was significantly up-regulated in RNA sequencing data when T. castaneum was exposed to eucalyptol. Therefore, in this study, the role of TcGSTu1 in the regulating the sensitivity of T. castaneum to eucalyptol was studied. The enzyme activities of GST and the transcription levels of TcGSTu1 were significantly increased following stimulation with eucalyptol. When using RNA interference technology knockdown TcGSTu1 heightens the sensitivity of T. castaneum to eucalyptol, demonstrating a link between TcGSTu1 and eucalyptol detoxification metabolism. Furthermore, TcGSTu1 is expressed in all developmental stages of T. castaneum , with higher expression levels observed particularly in the late egg stage. There was significant expression of TcGSTu1 in various tissues of different organisms, including larval head, fat body, and adult head. This observation indicated a possible connection between high TcGSTu1 expression and eucalyptol detoxification. The present findings suggest that TcGSTu1 may be involved in regulating the sensitivity and response of T. castaneum to treatment with eucalyptol, providing new research insight into pest control.

Toxicity and Synergistic Effect of Elsholtzia ciliata Essential Oil and Its Main Components against the Adult and Larval Stages of Tribolium castaneum

Jun-Yu Liang,Jie Xu,Ying-Ying Yang,Ya-Zhou Shao,Feng Zhou,Jun-Long Wang

Investigations have indicated that storage pests pose a great threat to global food security by damaging food crops and other food products derived from plants. Essential oils are proven to have significant effects on a large number of stored grain insects. This study evaluated the contact toxicity and fumigant activity of the essential oil extract from the aerial parts ofElsholtzia ciliataand its two major biochemical components against adults and larvae of the food storage pest beetleTribolium castaneum. Gas chromatography–mass spectrometry analysis revealed 16 different components derived from the essential oil ofE. ciliata, which included carvone (31.63%), limonene (22.05%), andα-caryophyllene (15.47%). Contact toxicity assay showed that the essential oil extract exhibited a microgram-level of killing activity againstT. castaneumadults (lethal dose 50 (LD50) = 7.79 μg/adult) and larvae (LD50= 24.87 μg/larva). Fumigant toxicity assay showed LD50of 11.61 mg/L air for adults and 8.73 mg/L air for larvae. Carvone and limonene also exhibited various levels of bioactivity. A binary mixture (2:6) of carvone and limonene displayed obvious contact toxicity againstT. castaneumadults (LD50= 10.84 μg/adult) and larvae (LD50= 30.62 μg/larva). Furthermore, carvone and limonene exhibited synergistic fumigant activity againstT. castaneumlarvae at a 1:7 ratio. Altogether, our results suggest thatE. ciliataessential oil and its two monomers have a potential application value to eliminateT. castaneum. Keywords:Elsholtzia ciliata;Tribolium castaneum;essential oil;carvone;limonene;insecticidal activity;synergistic effect

Response of xenobiotic biodegradation and metabolic genes in Tribolium castaneum following eugenol exposure

Zhang Yonglei,Gao Shanshan,Zhang Ping,Sun Haidi,Lu Ruixue,Yu Runnan,Li Yanxiao,Zhang Kunpeng,Li Bin

Eugenol, a plant-derived component possessing small side effects, has an insecticidal activity to Tribolium castaneum ; however, the underlying molecular mechanisms of eugenol acting on T. castaneum are currently unclear. Here, a nerve conduction carboxylesterase and a detoxifying glutathione S-transferase were significantly inhibited after eugenol exposure, resulting in the paralysis or death of beetles. Then, RNA-sequencing of eugenol-exposed and control samples identified 362 differentially expressed genes (DEGs), containing 206 up-regulated and 156 down-regulated genes. RNA-seq data were validated further by qRT-PCR. GO analysis revealed that DEGs were associated with 1308 GO terms of which the most enriched GO terms were catalytic activity, and integral component of membrane; KEGG pathway analysis showed that these DEGs were distributed in 151 different pathways, of which some pathways associated with metabolism of xenobiotics or drug were significantly enriched, which indicated that eugenol most likely disturbed the processes of metabolism, and detoxication. Moreover, several DEGs including Hexokinase type 2 , Isocitrate dehydrogenase , and Cytochrome b-related protein , might participate in the respiratory metabolism of eugenol-exposed beetles. Some DEGs encoding CYP , UGT , GST , OBP , CSP , and ABC transporter were involved in the xenobiotic or drug metabolism pathway, which suggested that these genes of T. castaneum participated in the response to eugenol exposure. Additionally, TcOBPC11/ TcGSTs7 , detected by qRT-PCR and RNA-interference against these genes, significantly increased the mortality of eugenol-treated T. castaneum , providing further evidence for the involvement of OBP/GST in eugenol metabolic detoxification in T. castaneum . These results aid eugenol insecticidal mechanisms and provide the basis of insect control.

Contact toxicity and transcriptomic analysis of terpinen‐4‐ol exposure in Tribolium castaneum

Shan-shan Gao,Yong-lei Zhang,Kun-peng Zhang,Xing-yun Wang,Qing-bo Tang,Yuan-chen Zhang

The terpene, terpinen-4-ol (T4ol), exhibits contact toxicity in Tribolium castaneum . However, the molecular mechanisms underlying this toxicity have not been elucidated. This study examined changes in the expression of four classic enzymes after exposure of T. castaneum to T4ol. Acetylcholinesterase and glutathione S-transferase activities were markedly inhibited after exposure to T4ol, while that of the detoxifying enzyme cytochrome oxidase P450 increased markedly. Carboxylesterase activity did not show significant changes. Furthermore, RNA sequencing revealed 260 differentially expressed genes (DEG) between the T4ol-treated and control samples, and qRT-PCR was used to validate the RNA-Seq data. The Gene Ontology analysis classified the DEGs into 36 functional groups, including the immune system processes, response to stimulus, and developmental processes. T4ol altered the response to stimulus and the immune system process of beetles by inducing the expression of the genes Stabilin-1, Attacin 1, and Defensin 1. Furthermore, the DEGs receptor tyrosine kinase Torso-like protein ( RTKTsl ), Frizzled 4 ( Fz4 ), Protein Wnt-5b, Ecdysone-induced protein 78C ( E78 ), Zinc finger protein GLIS1 ( ZFPGLIS1 ) were classified as participating in beetle development, and Fz4 and Protein Wnt-5b also mapped to the Wnt signaling pathway. This indicated that pathways associated with development are inhibited after exposure to T4ol. T4ol also induced CYP9Z6 / GSTs7 overexpression, and RNAi targeting these genes significantly increased larvae mortality on T4ol exposure, supporting the participation of CYP9Z6 / GSTs7 in the response to T4ol in T. castaneum . The results of this study will facilitate understanding of the toxic mechanisms of T4ol and provide a basis for controlling the pests of stored products.

Odorant binding protein C12 is involved in the defense against eugenol in Tribolium castaneum

Shanshan Gao,Ruixue Lu,Yonglei Zhang,Haidi Sun,Siying Li,Kunpeng Zhang,Ruimin Li

Tribolium castaneum ( T. castaneum ) is a worldwide pest of stored grain that mainly harms flour, and not only causes serious loss of flour quality but also leads to deterioration of flour quality. Chemical detection plays a key role in insect behavior, and the role of odorant-binding proteins (OBPs) in insect chemical detection has been widely studied. However, the mechanism of OBPs in insect defense against exogenous toxic substances is still unclear. In this study, biochemical analysis showed that eugenol , the active component of A. vulgaris essential oil , significantly induced the expression of the OBP gene OBPC12 from T. castaneum ( TcOBPC12 ). The mortality of late larvae treated with eugenol was higher than that of the control group after RNA interference (RNAi) against TcOBPC12 , which indicates that the OBP gene is involved in the eugenol defense mechanism and leads to a decrease in sensitivity to eugenol. Tissue expression profiling showed that the expression of TcOBPC12 in the epidermis, hemolymph, and intestine was higher than in other larval tissues, and TcOBPC12 was expressed mainly in the epidermis, head, and fat body of adults. The developmental expression profile showed that the expression of TcOBPC12 in late eggs, early and late larval stages, and late adult stages was higher than in other developmental stages. These data suggest that TcOBPC12 may be involved in the absorption of exogenous toxic substances by the larvae from T. castaneum . Our results provide a theoretical basis for the metabolism and degradation mechanism of exogenous toxic substances and help explore more potential target genes of insect pests .

Functional differentiation of ecdysis triggering hormone receptor variants in development and reproduction of Tribolium castaneum

Huiling Zhou,Wenjing Zhang,Jia Xu,Shuyan Feng,Peng Chen,Chengjun Li,Bin Li

Understanding the functional differentiation of hormone receptor variants is crucial for unraveling the complexity of endocrine regulatory networks and developing targeted pest management strategies. By integrating genetic manipulation with phenotypic analysis, we provided multiple lines of evidence for the distinct roles of ecdysis triggering hormone (ETH) receptor splice variants in the development and reproduction of Tribolium castaneum . The study revealed that the TcETH receptor gene was alternatively spliced into 2 variants, TcETHRA and TcETHRB , both of which were highly expressed in early and late pupae, early adults, and the central nervous system. The steroid hormone 20-hydroxyecdysone enhanced the expression of TcETH and its receptors via its nuclear receptor TcEcR . Functional validation experiments demonstrated that TcETHRA was the primary functional receptor. Both TcETH and TcETHRA promoted the shedding of the old cuticle, which was essential for successful molting, pupation, and eclosion. In addition, TcETH and TcETHRA specifically regulated female reproduction by influencing the expression of juvenile hormone acid methyltransferase ( TcJHAMT3 ) and vitellogenin-related genes ( TcVg1 , TcVg2 , and TcVgR ). In contrast, knockdown of TcETHRB had no significant impact on development but significantly reduced female embryo hatchability. These findings clarify the functional divergence of receptor splice variants in insect development and reproduction, providing new insights into the plasticity of arthropod endocrine networks and potential targets for pest control strategies.

Polyethyleneimine-modified mesoporous silica nanoparticles loaded with methoprene increased the insecticidal efficacy against Tribolium castaneum

Shiyuan Miao,Suisui Wang,Kaiwei Wang,Yonglin Ren,Simon J. McKirdy,Yujie Lu

Surface-modified mesoporous silica nanoparticles (MSNs) are regarded as efficient and promising nanocarriers in novel pesticide delivery. In this study, MSNs were synthesized and functionalized with low molecular weight polyethyleneimine (PEI), after which methoprene was loaded by adsorption. Detailed investigations showed that MSNs-PEI exhibited excellent encapsulation efficiency (53.33 %) and loading efficiency (21.03 %) for methoprene. The release behaviors of methoprene from Met@MSNs-PEI showed an initial rapid release stage followed by a sustained release stage. Methoprene was released faster in alkaline solutions compared with in acidic and neutral media. The kinetic study revealed that the release data fit best with the Korsmeyer-Peppas model. Additionally, Met@MSNs-PEI exhibited negligible toxicity towards S2 cells and low acute toxicity to zebrafish. Bioassay results showed that Met@MSNs-PEI exhibited significantly better insecticidal activity against Tribolium castaneum larvae than methoprene alone under the same doses of active ingredient applied, which obviously enhanced the utilization efficiency. Moreover, fluorescence microscope observations confirmed the successful internalization and distribution patterns of FITC-labeled MSNs-PEI in T. castaneum larvae, indicating that MSNs-PEI could serve as nanocarriers to deliver insecticides. RNA-seq analysis and qRT-PCR validation showed that MSNs-PEI-loaded methoprene resulted in the downregulation of genes related to detoxification, hormone synthesis, and cuticle biosynthesis, which inhibited insect growth and development. Therefore, MSNs-PEI nanoparticles provided an effective delivery system for methoprene.

Discovering potential fumigants against Tribolium castaneum from essential oils using GC-MS and chemometric approaches

Yan Yan Lin,Min Xing Zhang,Miao Sun,Lu Sheng Wan,You Zhou,Jing Sheng Chen

Tribolium castaneum is recognized as one of the most significant global pests affecting stored grains. Due to their general safety, essential oil (EO)-based biopesticides have garnered increasing interest worldwide. To provide a more rapid and objective method for the identification of individual components with potential fumigants from EOs, a GC-MS-based metabolomics strategy was applied in this study. Five EOs— Eucalyptus globulus , Dendranthema indicum , Cinnamomum cassia , Pinus massoniana , and Cinnamomum camphora —demonstrated remarkable toxicity, achieving corrected mortality rates exceeding 90% against Tribolium castaneum . A total of 41 potential fumigants were screened, with 21 of these identified potential fumigants corroborated by existing literature or preliminarily bioassay experiments. Key components included (−)-myrtenol, β-pinene, p -cymene, γ-terpinene, sabinene, β-myrcene, β-ocimene, and α-terpinene. These findings not only highlight promising EO-based fumigants against Tribolium castaneum but also provide lead compounds for future structural optimization. Additionally, this discovery strategy serves as a valuable reference for similar studies in natural products and biopesticides.

Odorant binding protein TcOBPC02 contributes to phytochemical defense in the red flour beetle, Tribolium castaneum

Shanshan Gao,Mengyi Guo,Weicheng Yu,Wenbo Miao,Haofei Ya,Dongfeng Liu,Ruimin Li,Kunpeng Zhang

The red flour beetle, Tribolium castaneum , is an agricultural and storage pest with a global distribution. Studies have shown that eucalyptol has strong contact toxicity against larvae of this beetle, whereas odorant binding proteins (OBPs) are known to contribute to larval defenses against this phytochemical toxin. However, the mechanisms underlying the protective effect of insect OBP s against eucalyptol remain unclear. Here, TcOBPC02 from T . castaneum was cloned and characterized. Gene expression profile analysis showed that TcOBPC02 is highly expressed at early larval and early pupal stages. Additionally, tissue expression profiling revealed that, in the adult, TcOBPC02 was most highly expressed in the head, followed by the epidermis, whereas in larvae, TcOBPC02 was mainly expressed in hemolymph and the epidermis. These developmental stages and tissues that exhibit high TcOBPC02 expression are closely related to the detoxification of heterologous substances. Furthermore, the mRNA level of TcOBPC02 was significantly increased after exposure to eucalyptol, whereas TcOBPC02 -targeted RNA interference increased the susceptibility of T. castaneum to eucalyptol, indicating that TcOBPC02 participates in the tolerance of this beetle to eucalyptol. Additionally, recombinant TcOBPC02 was expressed in Escherichia coli and isolated, enabling a straightforward fluorescence competition binding assay. In combination, these results have demonstrated that TcOBPC02 is required for defenses against phytochemicals in T. castaneum . This study provides a theoretical basis for understanding the mechanisms underlying the degradation of exogenous toxicants in insects and adds to the repertoire of potential target genes for pest control.

Sesquiterpenes from two Compositae plants as promising inhibitors to nuclear hormone receptor 3 of Tribolium castaneum

Wenjuan Zhang,Zimeng Zhang,Cun Li,Xiaoxue Yu,Qing He,Chunxue You,Daowen Li,Qianying Liu,Jiaqi Zhang

Essential oils (EOs) and their volatile secondary metabolites have been proved to be effective on storage pests control, while restricted on the application due to unclear mechanism. Molecular dynamics (MD) simulations and binding free energies analysis provided an effective approach to reveal mechanism on conformational calculation. In this work, the insecticidal and repellent capacities of Praxelis clematidea and Ageratum houstonianum oils and their main components identified by gas chromatography–mass spectrometry (GC–MS) were scientifically measured. Interestingly, P. clematidea oil exhibited strong fumigant toxicity against Tribolium castaneum (LC 50  = 7.07 mg/L air). Moreover, two EOs exhibited over 80% repellent rate against T. castaneum at the highest concentration of 78.63 nL/cm 2 . Furthermore, hundreds of enzymes related to the regulation of biological processes of T. castaneum were screened to explore the underlying molecular mechanism and develop promising insecticides. Besides, top hits were subjected to MD simulations and binding free energies analysis to elucidate complex inter-molecular stability and affinity over simulated time. The results demonstrated that isolongifolene, δ -cadinene, β -caryophyllene and caryophyllene oxide were prioritized as they were establishing conserved and stable interactions with residues of nuclear hormone receptor 3 ( TcHR3 ) of T. castaneum , which suggested that the four sesquiterpenes have potential to be promising insecticides on storage pests control.

AhR/ARNT-mediated regulation of detoxification metabolism and enzyme genes in the red flour beetle, Tribolium castaneum

Anxiang Zhuang,Xu Zhao,Xiaoxue Lu,Shengyi Wang,Xi Zeng,Xiaoyu Li,Siquan Yu,Meiqi Xie,Yang Mei,Haibin Yuan,Xiangkun Meng

BACKGROUND The aryl hydrocarbon receptor (AhR) and its dimerization partner ARNT are well-established regulators of xenobiotic metabolism in vertebrates, but their functional roles in insect detoxification remain poorly characterized. This study investigates the involvement of TcAhR/TcARNT in the detoxification of insecticides and plant allelochemicals in the red flour beetle, Tribolium castaneum . RESULTS We found that the expressions of TcAhR and TcARNT were significantly induced upon exposure to imidacloprid, deltamethrin, or (−)-4-terpineol. RNA interference (RNAi)-mediated silencing of TcAhR and TcARNT dramatically increased larval susceptibility to these chemicals, and concurrently suppressed the activities of key detoxification enzymes, including P450s, CarEs, GSTs and UGTs. Conversely, treatment with the AhR agonist β-naphthoflavone upregulated TcAhR / TcARNT expression, enhanced detoxification enzyme activities, and decreased chemical susceptibility. Transcriptome analysis following TcAhR / TcARNT knockdown identified 34 co-regulated detoxification enzyme genes, the vast majority of which were downregulated. Among these, TcFE4 and TcUGT2B7 were functionally validated to be critical for detoxification, as their individual silencing significantly heightened larval chemical susceptibility. Furthermore, bioinformatic analysis revealed that 32 co-regulated genes contained potential AhR/ARNT binding motifs in their promoters, and several were physically clustered and consistently downregulated on chromosomes. CONCLUSION These results demonstrate that the TcAhR/TcARNT pathway plays a central role in regulating a battery of detoxification genes in T. castaneum , thereby modulating its tolerance to synthetic insecticides and plant allelochemicals. This study provides fundamental insights into the molecular mechanisms of insect detoxification and resistance. © 2025 Society of Chemical Industry.
