Isolation, identification, and pathogenic effects of Trichoderma spp. from Auricularia auricula

Dang Hui,Kong Qianqian,Winchester William,Wan Xin,Lei Yu,Zhang Haisheng,Zhao Yu,Liu Xinyu,Xu Ben Bin,Zhang Baoshan,Wang Zhe

Auricularia auricula , one of the most important edible mushrooms, is affected heavily by Trichoderma . We collected the diseased samples from the main A. auricula cultivation regions to characterize the pathogen and study the effect of Trichoderma spp. on A. auricula species. We identified one Trichoderma species, T. pleuroticola , based on the internal transcribed spacer and morphology characteristics, and two types of A. auricula strains, Heiwei 15 (HW 15) and Hei 29 (H 29), were tested in this work. The growth rate of T. pleuroticola was 3.26–3.52 times higher than that of A. auricula and advantageously competed for living space and nutrients. In confrontation culture, T. pleuroticola completely inhibited the mycelium growth of A. auricula and grew on it, resulting in a diverse impact on HW 15 and H 29. In addition, T. pleuroticola can produce metabolites with antibacterial activity. The inhibition rate of volatile metabolites to H-29 and HW 15 was 13.46% and 10.44%, and the inhibition rate of nonvolatile metabolites to H-29 and HW 15 was 36.04% and 31.49%, respectively. Further analysis showed that these antifungal activities inhibiting Auricularia auricula growth were mainly attributed to the organic compounds from T. pleuroticola , nonanal, tyrosine, beta-sitosterol, and wortmannin. In short, T. pleuroticola was a highly pathogenic fungi in the production of A. auricula . Graphical Abstract T. pleuroticola can affect the normal growth of A. auricula hypha, and its metabolites also have inhibitory effects on A. auricula hypha.

Regulation of cellulase production via calcium signaling in Trichoderma reesei under PEG8000 stress

Liu Shuai,Quan Lin,Yang Mei,Wang Dan,Wang Yong-Zhong

In this study, the effect of polyethylene glycol 8000 (PEG8000) stress on cellulase biosynthesis in Trichoderma reesei CICC2626 via calcium signaling was investigated, and a plausible mechanism by which intracellular Ca 2+ regulates the transcription of cellulase genes was proposed. The results indicated that the total cellulase (filter paper-hydrolyzing activity [FPase]), endoglucanase (carboxymethyl cellulase activity [CMCase]), and β-glucosidase activities of the strain were 1.3-, 1.2-, and 1.3-fold higher than those of the control (no PEG8000 addition) at a final concentration of 1.5% (w/v) PEG8000. Moreover, the transcriptional levels of cellulase genes, protein concentrations, and biomass increased. With the synergistic use of commercial cellulase and T. reesei CICC2626 cellulase to hydrolyze alkali-pretreated rice straw, the released reducing sugar concentration reached 372.7 mg/g, and the cellulose content (22.7%, 0.32 g) was significantly lower than the initial content (62.5%, 1.88 g). Transcriptome data showed that 12 lignocellulose degradation–related genes were significantly upregulated in the presence of 1.5% PEG8000. Furthermore, the addition of Ca 2+ inhibitors and deletion of crz1 (calcineurin-responsive zinc finger 1-encoding gene, which is related to the calcium signaling pathway) demonstrated that calcium signaling plays a dominant role in PEG8000-induced cellulase genes overexpression. These results revealed a link between PEG8000 induction and calcium signaling transduction in T. reesei CICC2626. Moreover, this study also provides a novel inducer for enhanced cellulase production. Key points • Cellulase biosynthesis in Trichoderma reesei could be enhanced by PEG8000 • PEG8000 could induce a cytosolic Ca 2+ burst in Trichoderma reesei • The activated calcium signaling was involved in cellulase biosynthesis Graphical abstract

Treatment of amoxicillin-containing wastewater by Trichoderma strains selected from activated sludge

Yixiang Cai,Hong Yu,Liheng Ren,Yingjuan Ou,Shilin Jiang,Youzheng Chai,Anwei Chen,Binghua Yan,Jiachao Zhang,Zhiyong Yan

This study screened a Trichoderma strain ( Trichoderma pubescens DAOM 166162) from activated sludge to solve the limitation of traditional biological processes in the treatment of amoxicillin (AMO) containing wastewater. The mechanism of the removal of AMO wastewater by T. pubescens DAOM 166162 (TPC) was studied. AMO resulted in a higher protein percentage in the extracellular polymeric substances (EPS) secreted by TPC, which facilitated the removal of AMO from the wastewater. Fourier transform infrared spectroscopy and excitation-emission matrix were used to characterize EPS produced by metabolizing different carbon sources. It was found that the hydroxyl group was the primary functional group in EPS. The life activity of TPC was the cause of the pH rise. The main pathway of degradation of AMO by TPC was the hydroxyl group uncoupling the lactam ring and the hydrolysis of AMO in an alkaline environment . The removal efficiency of AMO in wastewater by TPC was >98 % (24 h), of which the biodegradation efficiency was 70.01 ± 1.48 %, and the biosorption efficiency was 28.44 ± 2.97 %. In general, TPC is an effective strain for treating wastewater containing AMO. This research provides a new idea for AMO wastewater treatment .

Metformin regulates cellulase production in Trichoderma reesei via calcium signaling and mitochondrial function

Wang Jiajia,Chen Yumeng,Cong Jiawei,Wang Wei

Background Trichoderma reesei is renowned for its cellulase-producing ability and is used for biofuel production from lignocellulose. In plants and fungi, cellulase production is induced by cellulose and suppressed by glucose; however, whether metformin can enhance cellulase production and mitochondrial function in T. reesei remains unclear. Metformin reduces blood glucose levels by inhibiting hepatic gluconeogenesis; therefore, it is worth investigating whether metformin transmission modulates cellulase biosynthesis in T. reesei . Results Metformin increased cellulase production and the transcription of cellulase-related genes. It also enhanced the concentrations of Ca 2+ in the cytosol and mitochondria and regulated the transcription levels of cellulase-related genes by modulating calcium homeostasis in T. reesei QM6a. In addition, metformin was identified as an antioxidant that can enhance cellulase activity by reducing reactive oxygen species (ROS). Our results demonstrated that metformin influences the state of mitochondria by enhancing mitochondrial activity and membrane potential to promote cellulase production. Conclusion Collectively, these results indicate that metformin enhances cellulase production, scavenges ROS, and protects mitochondrial activity in T. reesei . Graphical Abstract

N,N-dimethylformamide induces cellulase production in the filamentous fungus Trichoderma reesei

Chen Yumeng,Wu Chuan,Shen Yaling,Ma Yushu,Wei Dongzhi,Wang Wei

Background The filamentous fungus Trichoderma reesei produces cellulase enzymes that are widely studied for lignocellulose bioconversion to biofuel. N,N-dimethylformamide (DMF) is a versatile organic solvent used in large quantities in industries. Results In this study, we serendipitously found that biologically relevant concentrations of extracellular DMF-induced cellulase production in the T. reesei hyper-cellulolytic mutant Rut-C30 and wild-type strain QM6a. Next, by transcriptome analysis, we determined that plc-e encoding phospholipase C was activated by DMF and revealed that cytosolic Ca2+ plays a vital role in the response of T. reesei to DMF. Using EGTA (a putative extracellular Ca2+ chelator) and LaCl3 (a plasma membrane Ca2+ channel blocker), we demonstrated that DMF induced a cytosolic Ca2+ burst via extracellular Ca2+ and Ca2+ channels in T. reesei, and that the cytosolic Ca2+ burst induced by DMF-mediated overexpression of cellulase through calcium signaling. Deletion of crz1 confirmed that calcium signaling plays a dominant role in DMF-induced cellulase production. Additionally, 0.5–2% DMF increases the permeability of T. reesei mycelia for cellulase release. Simultaneous supplementation with 1% DMF and 10 mM Mn2+ to T. reesei Rut-C30 increased cellulase activity approximately fourfold compared to that without treatment and was also more than that observed in response to either treatment alone. Conclusion sOur results reveal that DMF-induced cellulase production via calcium signaling and permeabilization. Our results also provide insight into the role of calcium signaling in enzyme production for enhanced cellulase production and the development of novel inducers of cellulase.

Mn2+ modulates the expression of cellulase genes in Trichoderma reesei Rut-C30 via calcium signaling

Chen Yumeng,Shen Yaling,Wang Wei,Wei Dongzhi

Background The filamentous fungus Trichoderma reesei Rut-C30 is one of the most vital fungi for the production of cellulases, which can be used for biofuel production from lignocellulose. Nevertheless, the mechanism of transmission of external stimuli and signals in modulating cellulase production in T. reesei Rut-C30 remains unclear. Calcium is a known second messenger regulating cellulase gene expression in T. reesei. Results In this study, we found that a biologically relevant extracellular Mn2+ concentration markedly stimulates cellulase production, total protein secretion, and the intracellular Mn2+ concentration of Rut-C30, a cellulase hyper-producing strain of T. reesei. Furthermore, we identified two Mn2+ transport proteins, designated as TPHO84-1 and TPHO84-2, indicating that they are upstream in the signaling pathway that leads to cellulase upregulation. We also found that Mn2+ induced a significant increase in cytosolic Ca2+ concentration, and that this increased cytosolic Ca2+ might be a key step in the Mn2+-mediated regulation of cellulase gene transcription and production. The utilization of LaCl3 to block plasma membrane Ca2+ channels, and deletion of crz1 (calcineurin-responsive zinc finger transcription factor 1) to interrupt calcium signaling, showed that Mn2+ exerts the induction of cellulase genes via calcium channels and calcium signaling. To substantiate this, we identified a Ca2+/Mn2+ P-type ATPase, TPMR1, which could play a pivotal role in Ca2+/Mn2+ homeostasis and Mn2+ induction of cellulase genes in T. reesei Rut-C30.Conclusion sTaken together, our results revealed for the first time that Mn2+ stimulates cellulase production, and demonstrates that Mn2+ upregulates cellulase genes via calcium channels and calcium signaling. Our research also provides a direction to facilitate enhanced cellulase production by T. reesei.

Shu-Hua Zhang,Xian-Lin Yue,Xue Zhao,Jing Tang,Yu Yang,Rui Xu,Hao Ma,Shuai-Ming Zhu,Fu-Yao Luo,Qin Zhang,Guo-Gang Zhang,Chang-Wei Li

Longibrachiamide A ( 1 ), a new 20-residue peptaibol, along with three known ones ( 2 – 4 ), were isolated from the fungus Trichoderma longibrachiatum Rifai DMG-3-1-1, isolated from a mushroom Clitocybe nebularis (Batsch) P. Kumm, which was collected from coniferous forest of northeast China in our previous work. The structure of longibrachiamide A ( 1 ) was determined by its NMR and ESI-MS/MS data, the absolute configuration of 1 was further determined by Marfey's analyses. And the complete NMR data of 2 – 4 were also reported for the first time. The similar CD spectra of 1 – 4 showed that they all had mixed 3 10 -/α-helical conformations. Compounds 1 – 4 showed strong cytotoxicities against BV2, A549 and MCF-7 cells, and also showed moderate inhibitory effects against the tested Gram-positive bacteria, including MRSA T144 and VRE-10. Camptothecin-producing endophytic fungus Trichoderma atroviride LY357: isolation, identification, and fermentation conditions optimization for camptothecin production

Pu Xiang,Qu Xixing,Chen Fei,Bao Jinku,Zhang Guolin,Luo Yinggang

Camptothecin (CPT), the third largest anticancer drug, is produced mainly by Camptotheca acuminata and Nothapodytes foetida . CPT itself is the starting material for clinical CPT-type drugs, but the plant-derived CPT cannot support the heavy demand from the global market. Research efforts have been made to identify novel sources for CPT. In this study, three CPT-producing endophytic fungi, Aspergillus sp. LY341, Aspergillus sp. LY355, and Trichoderma atroviride LY357, were isolated and identified from C. acuminata . Most CPT produced by these fungi was found in the fermentation broth, and their corresponding CPT yields were 7.93, 42.92, and 197.82 μg l −1 , respectively. The CPT-producing capability of LY341 and LY355 was completely lost after repeat subculturing. A substantial decrease of CPT production was also observed in the second generation of LY357. However, a stable and sustainable production of CPT was found from the second generation through the eighth generation of LY357. The fermentation medium, time, pH, temperature, and agitation rate were optimized for CPT production. Methyl jasmonate and XAD16 were proven to be an optimum elicitor and adsorbent resin, respectively, in view of that CPT yield was increased 3.4- and 11-fold through their use. A 50- to 75-fold increase of CPT yield was obtained when the optimized fermentation conditions, elicitor, and adsorbent resin were combined and applied to the culture of the seventh and eighth generations of LY357, and the highest CPT yield was 142.15 μg l −1 . The CPT-producing T. atroviride LY357 paves a potential to uncover the mysteries of CPT biosynthesis.

Trichoderma-secreted anthranilic acid promotes lateral root development via auxin signaling and RBOHF-induced endodermal cell wall remodeling

Yu Chen,Yansong Fu,Yanwei Xia,Youzhi Miao,Jiahui Shao,Wei Xuan,Yunpeng Liu,Weibing Xun,Qiuyan Yan,Qirong Shen,Ruifu Zhang

Trichoderma spp. have evolved the capacity to communicate with plants by producing various secondary metabolites (SMs). Nonhormonal SMs play important roles in plant root development, while specific SMs from rhizosphere microbes and their underlying mechanisms to control plant root branching are still largely unknown. In this study, a compound, anthranilic acid (2-AA), is identified from T. guizhouense NJAU4742 to promote lateral root development. Further studies demonstrate that 2-AA positively regulates auxin signaling and transport in the canonical auxin pathway. 2-AA also partly rescues the lateral root numbers of CASP1 pro :shy2-2 , which regulates endodermal cell wall remodeling via an RBOHF -induced reactive oxygen species burst. In addition, our work reports another role for microbial 2-AA in the regulation of lateral root development, which is different from its better-known role in plant indole-3-acetic acid biosynthesis. In summary, this study identifies 2-AA from T. guizhouense NJAU4742, which plays versatile roles in regulating plant root development.

Confrontation assays and mycotoxin treatment reveal antagonistic activities of Trichoderma and the fate of Fusarium mycotoxins in microbial interaction

Ye Tian,Dianzhen Yu,Na Liu,Yan Tang,Zheng Yan,Aibo Wu

Mycotoxins are toxic fungal metabolites, contaminating cereal grains in field or during processing and storage periods. These environmental contaminants pose great threats to humans and animals’ health due to their toxic effects. Type A trichothecenes, fumonisins and fusaric acid (FA) are commonly detected mycotoxins produced by various Fusarium species. Trichoderma spp. are promising antagonists in agriculture for their activities against plant pathogens , and also regarded as potential candidates for bioremediation of environmental contaminants. Managing toxigenic fungi by antagonistic Trichoderma is regarded as a sustainable and eco-friendly strategy for mycotoxin control. However, the metabolic activities of Trichoderma on natural occurring mycotoxins were less investigated. Our current work comprehensively explored the activities of Trichoderma against type A trichothecenes, fumonisins and FA producing Fusarium species via co-culture competition and indirect volatile assays. Furthermore, we investigated metabolism of type A trichothecenes and FA in Trichoderma isolates. Results indicated that Trichoderma were capable of bio-transforming T-2 toxin, HT-2 toxin, diacetoxyscirpenol and neosolaniol into their glycosylated forms and one Trichoderma strain could bio transform FA into low toxic fusarinol. These findings proved that Trichoderma isolates could manage toxigenic Fusarium via direct competition and volatile-mediated indirect inhibition. In addition, these antagonists possess defensive systems against mycotoxins for self-protection, which enriches our understanding on the interaction mechanism of Trichoderma spp. on toxigenic fungus.

Antagonistic and Detoxification Potentials of Trichoderma Isolates for Control of Zearalenone (ZEN) Producing Fusarium graminearum

Tian Ye,Tan Yanglan,Yan Zheng,Liao Yucai,Chen Jie,De Boevre Marthe,De Saeger Sarah,Wu Aibo

Fungi belonging to Fusarium genus can infect crops in the field and cause subsequent mycotoxin contamination, which leads to yield and quality losses of agricultural commodities. The mycotoxin zearalenone (ZEN) produced by several Fusarium species (such as F. graminearum and F. culmorum) is a commonly-detected contaminant in foodstuffs, posing a tremendous risk to food safety. Thus, different strategies have been studied to manage toxigenic pathogens and mycotoxin contamination. In recent years, biological control of toxigenic fungi is emerging as an environment-friendly strategy, while Trichoderma is a fungal genus with great antagonistic potentials for controlling mycotoxin producing pathogens. The primary objective of this study was to explore the potentials of selected Trichoderma isolates on ZEN-producing F. graminearum, and the second aim was to investigate the metabolic activity of different Trichoderma isolates on ZEN. Three tested Trichoderma isolates were proved to be potential candidates for control of ZEN producers. In addition, we reported the capacity of Trichoderma to convert ZEN into its reduced and sulfated forms for the first time, and provided evidences that the tested Trichoderma could not detoxify ZEN via glycosylation. This provides more insight in the interaction between ZEN-producing fungi and Trichoderma isolates.
