Conventional metabolomics and lipidomics typically require separate sample preparations, which can introduce systematic bias and reduce throughput, thereby hindering comprehensive elucidation of diabetic metabolic networks. In this study, we exploited the pH-responsive behavior of ZrO₂–SiO₂ composite microspheres to establish the first single-sample, dual-channel solid-phase extraction (SPE) pretreatment method: under acidic conditions, Zr⁴⁺–PO₄³⁻ coordination selectively adsorbs phospholipids, whereas under basic conditions, NH₄⁺ competes to gently elute them. The flow-through and elution fractions are used for metabolomic and lipidomic analyses, respectively. We integrated the entire workflow into an automated pretreatment platform and coupled it with high-resolution LC–IMS–QTOF detection to enable high-throughput processing. Method validation demonstrated high phospholipid recovery and excellent preservation of polar metabolites (representative results: LPC 18:0 elution recovery 100.1 % ± 3.0 %; DPPC elution recovery 94.0 % ± 2.0 %; 25-OH-VD₃ flow-through retention 99.0 % ± 2.0 %). In serum from diabetic patients and healthy controls, 390 differential metabolites were identified in the metabolome, significantly enriched in energy metabolism pathways such as galactose metabolism; lipidomics revealed disturbances in glycerophospholipid metabolism, indicating membrane signaling remodeling. Importantly, the dual-channel approach captured for the first time the coordinated disruption of the galactose–ether-lipid metabolic axis—a sequential reduction in UDP-galactose levels leading to diminished plasmalogen biosynthesis—providing direct molecular evidence for the pathogenic sequence in which disrupted glycolytic energy metabolism precipitates membrane structural dysfunction. This method not only markedly improves the consistency and throughput of omics data but also lays a technical foundation for advancing precision diagnostics and treatment of diabetes from “blood glucose management” to “metabolic network regulation.”

Anthocyanins are a group of natural water-soluble pigments in plants that contribute to the pink-purple color of a range of tissues. Because anthocyanins have various biological activities in human health, there is great research interest in the development of anthocyanin-rich foods and beverages, including purple shoot tea. Anthocyanidin 3- O -galactosides have been identified as one of the main anthocyanin components in purple shoot tea, but the enzyme responsible for their biosynthesis remains unclear. UDP-galactose anthocyanidin 3- O -galactosyltransferase (UA3GalT) is presumed to catalyze the galactosylation of anthocyanidin. Therefore, we assayed the UA3GalT activity in five tea samples with varying degrees of purple color and found that its activity was strongly positively correlated (r = 0.929, p < 0.05) with anthocyanin content. Phylogenetic analysis and sequence alignment suggested that CsUGT78A15 encoded a UA3GalT enzyme. Enzymatic assays indicated that r Cs UGT78A15 could catalyze the synthesis of cyanidin 3- O -galactoside and delphinidin 3- O -galactoside using UDP-galactose as a sugar donor, and it showed higher catalytic efficiency towards delphinidin than cyanidin. These results indicate that Cs UGT78A15 acts as a UA3GalT in vitro . Subcellular localization showed that Cs UGT78A15 was located in the endoplasmic reticulum (ER) and nucleus, consistent with the location of anthocyanin synthesis. Transient overexpression of CsUGT78A15 in the fruit of mature ‘Granny Smith’ apples showed that the upregulation of CsUGT78A15 promoted cyanidin 3- O -galactoside accumulation in apple skins. These results suggested that Cs UGT78A15 could catalyze galactosylation of anthocyanidins in planta . Our findings provide insight into the biosynthesis of anthocyanins in tea plants .

Flavonol glycosides are health-promoting phytochemicals important for human nutrition and plant defense against environmental stresses. Glycosylation modification greatly enriches the diversity of flavonols. Morella rubra, a member of the Myricaceae, contains high amounts of myricetin 3-O-rhamnoside (M3Rha), quercetin 3-O-rhamnoside (Q3Rha), and quercetin 3-O-galactoside (Q3Gal). In the present study, MrUGT78R1 and MrUGT78R2 were identified as two functional UDP-rhamnosyltransferases, while MrUGT78W1 was identified as a UDP-galactosyltransferase. Site-directed mutagenesis identified Pro143 and Asn386 as important residues for rhamnosyl transfer activity of MrUGT78R1, while the two corresponding positions in MrUGT78W1 (i.e. Ser147 and Asn370) also play important roles in galactosyl transfer activity. Transient expression data for these three MrUGTs in Nicotiana benthamiana tested the function of MrUGT78R1 and MrUGT78R2 as rhamnosyltransferases and MrUGT78W1 as a galactosyltransferase in glycosylation of flavonols. This work enriches knowledge of the diversity of UDP-rhamnosyltransferase in planta and identifies two amino acid positions important for both rhamnosyltransferase and galactosyltransferase.
