Evaluation of the underestimation of citrinin content in Hongqu using hydrolysis treatments and UPLC-FLD

Yaqi Hou,Jiao Liu,Yanchun Shao,Xitian Peng,Dawen Zhang,Lifang Hu,Fusheng Chen,Youxiang Zhou

Modified mycotoxins can cause an underestimation of exposure risk due to their untraceable characteristics. Hongqu , a traditional Chinese fermented product, is widely used as food colorants , fermenters and functional foods for about 2000 years. Though there were several methods to reduce citrinin (CIT) content in Hongqu , the risk assessment and prevention of modified CIT remain inadequate, causing potential risk to human health. The current study firstly identified the occurrence of modified CIT in Hongqu via acid, alkaline and enzyme hydrolysis combined with chromatographic and mass spectrometric approaches. Then 8 Hongqu samples and 5 Monascus products were treated using the optimized acid hydrolysis method and analyzed by ultra-high-performance liquid chromatography with fluorescence detector. The results showed that CIT content in the CIT positive Hongqu increased by 35.28%–458.13% after acid hydrolysis, demonstrating there could be a underestimation risk in the CIT positive Hongqu samples, and the modification of CIT might not be chemical modified, but matrix associated.

Quantification of sialic acids in red meat by UPLC-FLD using indoxylsialosides as internal standards

Yao Hong L.,Conway Louis P.,Wang Mao M.,Huang Kun,Liu Li,Voglmeir Josef

Herein we describe a UPLC-FLD-based method for the quantification of the sialic acid content of red meat, using a synthetic neuraminic acid derivative as an internal standard. X-Gal-α-2,6- N -propionylneuraminic acid was synthesized via a chemoenzymatic pathway and its hydrolytic stability was characterized. Known quantities of this compound were incubated with samples of red meat under sialic acid-releasing conditions. The released sialic acids were derivatized, analyzed by UPLC-FLD, and the Neu5Ac/Neu5Gc content of the meat sample was determined by comparison with the internal standard. A number of red meats were analyzed by this method with the following results (Neu5Ac μg/g tissue, Neu5Gc μg/g tissue ± s.d.): pork (68 ± 3, 15.2 ± 0.7), beef (69 ± 8, 36 ± 5), lamb (46 ± 2, 33 ± 1), rabbit (59 ± 2, 0.4 ± 0.4), and hare (50 ± 4, 1 ± 1). We envisage that this methodology will find application in investigating the health effects of dietary Neu5Gc. ᅟ

Electromembrane extraction coupled with UPLC-FLD for the determination of ochratoxin A in food crops

Ming Lv,Haidong Gao,Shucai Gao,Yeru Liu,Jingjing Yao,Hongwei Liu,Fayun Wang,Mantang Chen,Lianjun Song,Tianlin Wang,Libin Wan

Ochratoxin A (OTA) contamination in food crops has sparked global concern for potential health risk to humans and animals. A simple, accurate and sensitive method was developed for the determination of OTA using electromembrane extraction (EME) coupled with ultra-high performance liquid chromatography-fluorescence detector (UPLC-FLD). By optimizing the important factors, EME showed good performance for the extraction of OTA in spiked corn flour sample. Using the developed EME-UPLC-FLD method, good linearity for OTA was achieved in the range of 1–100 ng mL −1 , and the limit of detection (LOD) was 0.02306 μg kg −1 , which was well below the established maximum residue limit. The proposed method was successfully applied for the determination of trace OTA in multiple matrices (corn flour extract, corn flour slurry and soybean milk) with satisfactory recoveries ranging from 80.01 % to 116.7 % at four spiked levels, which has great potential to analyze trace chemical contaminant in food crops.

Automatic Pretreatment of Dispersive Liquid Liquid Microextraction Based on Immunomagnetic Beads Coupled with UPLC-FLD for the Determination of Zearalenone in Corn Oils

Baoxia Ni,Jin Ye,Zhihong Xuan,Li Li,Xiangrui Wen,Zongwang Li,Hongmei Liu,Songxue Wang

Sample pretreatment is a vital step in the detection of mycotoxins, and traditional pretreatment methods are time-consuming, labor-intensive and generate much organic waste liquid. In this work, an automatic, high-throughput and environmentally friendly pretreatment method is proposed. Immunomagnetic beads technology and dispersive liquid–liquid microextraction technology are combined, and the zearalenone in corn oils is directly purified and concentrated under the solubilization effects of surfactant. The proposed pretreatment method allows for the batch pretreatment of samples without pre-extraction using organic reagents, and almost no organic waste liquid is produced. Coupled with UPLC-FLD, an effective and accurate quantitative detection method for zearalenone is established. The recovery of spiked zearalenone in corn oils at different concentrations ranges from 85.7 to 89.0%, and the relative standard deviation is below 2.9%. The proposed pretreatment method overcomes the shortcomings of traditional pretreatment methods and has broad application prospects. Keywords:immunomagnetic beads;dispersive liquid liquid microextraction;pretreatment;zearalenone;corn oils;UPLC-FLDKey Contribution:An automatic and eco-friendly pretreatment method of zearalenone (ZEN) in corn oils was developed based on immunomagnetic beads with dispersive liquid–liquid microextraction technology. The proposed pretreatment method, coupled with UPLC-FLD, met the accuracy and sensitivity detection requirements of ZEN in corn oils.
