Wound healing remains a significant global health challenge, affecting millions annually and imposing substantial economic burdens. Most commercially available biomaterials for wound management primarily address external symptoms, including hemostasis, exudation, scarring, and infection. Advanced biomaterials derived from endogenous molecules aim to better replicate the native wound microenvironment, promoting enhanced repair. Since wounds frequently occur on exposed skin, which is vulnerable to UVA radiation and requires protective yet invisible materials, traditional wound care products often lack these essential features. Inspired by natural UV protection mechanisms, a novel bioscaffold is developed using recombinant human collagen XVII (rhCOL17) crosslinked with porphyra-334 (P334) to improve wound healing under UVA exposure. The resulting rhCOL17-P334 conjugate integrates extracellular matrix (ECM)-like cues with UVA-shielding properties provided by P334. This conjugate is used to construct a transparent, injectable hydrogel combining gelatin methacryloyl (GelMA) and rhCOL17-P334 (GCP). GCP significantly inhibits UVA-induced fibroblast senescence and improves wound healing outcomes by targeting integrin α6β4 through rhCOL17. Its transparency facilitates convenient wound monitoring while also addressing the aesthetic requirement for invisibility. By combining UVA shielding with wound repair capabilities, GCP presents a promising platform for advanced wound management.

Excessive ultraviolet (UV) radiation can cause human skin erythema, tanning, inflammation and other adverse reactions. Human skin can spontaneously synthesize endogenous melanin under UV radiation to defend the damage of UV, but the skin problems such as light damage and light aging cannot be avoided due to its low UV shielding efficiency. Therefore, we design a natural and biocompatible PGDA (PDA-L doped sunscreen hydrogel) sunscreen hydrogel by crosslinking melanin-like L-lysine doped polydopamine (PDA-L) and amino acid biopolymer poly-γ-glutamic acid, which displays good biosafety, efficient UVA shielding performance and excellent anti-inflammatory performance in the application of the skin protection of sun and repair after sunburn. The introduction of PDA-L to the hydrogel can improve the absorption intensity in the UV region and antioxidative performance of the hydrogel. Meanwhile, the usage of biocompatible PGDA sunscreen hydrogel can avoid the skin oxidative damage and penetrative toxicity of the conventional sunscreen.

Living organisms tend to evolve various naturally photoprotective mechanisms to avoid photodamage. Among them, polydopamine (PDA) is an effective sunscreen, a mimic of melanin, which is the main functional component of the photoprotective system of human skin. However, the concerns of its dark color, skin penetration and photoprotective efficiency remain yet to be solved. Herein, we have constructed melanin-inspired nanocomposite hydrogels (CS-PDA h -GP-HA) for photoprotection, in which PDA was prepared as hollow nanoparticles (PDA h NPs) and entrapped in a physically cross-linked hydrogel (CS-GP-HA) formed by chitosan (CS) and hyaluronic acid (HA) using β -glycerophosphate ( β -GP) as a modulator. The CS-PDA h -GP-HA hydrogels exhibit a shear-thinning flow behavior with an elastic modulus of 300 Pa with the gel-sol transition temperature maintained at about 37 °C simply by adjusting the β -GP content in the hydrogels. The CS-PDA h -GP-HA hydrogels also possess excellent resistance toward skin penetration. The photoprotective performances of CS-PDA h -GP-HA hydrogels were evaluated by the determination of sun protection factor (SPF) and in vitro UVA protection efficacy (UVAPE) along with UV–Vis spectroscopy. Compared with the TiO 2 nanoparticles in CS-GP-HA hydrogel, the CS-PDA h -GP-HA hydrogels show stronger shielding ability in both UVA and UVB regions. When protected by the CS-PDA h -GP-HA hydrogels, the cell viability of NIH-3T3 fibroblasts increases to 96% while it was only 14% in the case of non-protecting group. These results suggest that the CS-PDA h -GP-HA hydrogels could efficiently shield the UV irradiation and protect the skin from photodamage. This work introduces PDA-based nanocomposite hydrogels with safe, biocompatible and photoprotective properties, and provides a melanin-mimicking photoprotection system for the application in sunscreens.
