Impact of ionic strength on physicochemical stability of o/w emulsions stabilized by Ulva fasciata polysaccharide

Ping Shao,Huiling Ma,Jieyu Zhu,Qiang Qiu

The aim of this research was to investigate the effect of ionic strength (Na + 0.1–0.5 mol/L, Ca 2+ 0.01–0.05 mol/L) on the physicochemical stability of o/w emulsions stabilized by Ulva fasciata polysaccharide . Droplet diameter, Polydispersity Index (PDI), zeta-potential value, microscopic morphology and rheological properties were achieved to investigate the possible stability mechanisms. The results showed that different metal ions had significant effects on the stability of o/w emulsions. When compared the effect of Ca 2+ with that of Na + , it was found that Na + concentration did not have a significant effect on emulsions stability. The results indicated that zeta-potential decreased sharply with increasing calcium ionic strength (CaCl 2 :0.01 mol/L - 0.05 mol/L) and the minimum zeta-potential value was −27.5 mV, while zeta-potential changed slightly when NaCl (0.1 mol/L - 0.5 mol/L) were added. As for z-average size and PDI, they changed significantly with higher valence metallic ions. With increasing NaCl and CaCl 2 concentration, the retention rate of R-(+)-Limonene was reduced and emulsions exhibited non-Newtonian behavior. Another interesting result was that the addition of Ca 2+ at low concentration (0.01 mol/L - 0.02 mol/L) managed to improve the stability of emulsions .

The role of hydroponic solution of Ulva fasciata in modulating the antibiotic resistance of Vibrio alginolyticus

Feng Lijuan,Yu Shuping,Jin Weimei,Xiao Changyan,Qiao Yan,Yang Guangfeng

AimsThis study aimed to ascertain the active substances in the hydroponic solution of Ulva (U.) fasciata (HSUF) and their effects on antibiotics resistance of pathogenic bacteria Vibrio alginolyticus (Va.) at environmental levels. Methods and ResultsAnalytical results of HSUF by solid-phase extraction and gas chromatography-mass spectrometry showed that HSUF contained rich active substances and dominated by 2,4,6-tribromophenol, acetal, mono (2-ethylhexyl) phthalate and 2- (4-hydroxybenzene) ethanol, etc. Thereinto, 2,4,6-tribromophenol had the lowest minimal inhibitory concentrations (MICs) of 64–128 μg mL−1 for Va. strains. The rich antibacterial substances of HSUF inhibited 47%-63% of isolated Va. strains. After 20 generations of subculture for Va. strains in three dilutes (1/2 (HT), 1/20 (MT) and 1/50 (LT)) of HSUF (20 g L−1), the resistance to streptomycin of Va. S1 and Va. M3 changed from intermediate resistance ability to susceptible level. The large promotion of reactive oxygen species was observed in different HSUF levels, but the biofilm formation of Va. S1 did not changed significantly. Transcriptome sequencing of Va. S1 demonstrated that antibiotic resistance gene lpxA was down-regulated at different HSUF levels.ConclusionsLive U. fasciata excreted a variety of active secondary metabolites (SMs) in HSUF, which exhibited strong inhibitory effects on three isolated Va. strains. HSUF changed the antibiotic resistance of Va. Strain, especially at a higher HSUF concentrations.

Physical stability of R-(+)-Limonene emulsions stabilized by Ulva fasciata algae polysaccharide

Ping Shao,Huiling Ma,Qiang Qiu,Weiping Jing

The physical stability of R-(+)-Limonene emulsions stabilized by Ulva fasciata polysaccharide (UFP) was investigated in this study. Emulsion physical stability was evaluated under different polysaccharide concentrations (1%–5%, wt/wt) and pH values (3.0–11.0). The stability of R-(+)-Limonene emulsions was demonstrated by droplet size distribution, rheological properties, zeta potential and visual phase separation. R-(+)-Limonene emulsions displayed monomodal droplet size distributions, high absolute values of zeta potential and good storage stability when 3% (wt/wt) UFP was used. The rheological properties and stability of R-(+)-Limonene emulsions appeared to be dependent on polysaccharide concentration. The emulsion stability was impacted by pH. Higher zeta potential (−52.6   mV) and smaller mean droplet diameter (2.45   μm) were achieved in neutral liquid environment (pH 7.0). Extreme acidity caused the flocculation of emulsions, which was manifested as phase separation, while emulsions were quite stable in an alkaline environment. Through comparing the stabilities of emulsions stabilized by different emulsifiers (i.e. UFP, GA and Gelatin), the result suggested that UFP was the best emulsifying agent among them.

Influences of Ulva fasciata polysaccharide on the rheology and stabilization of cinnamaldehyde emulsions

Ping Shao,Jiamei Shao,Yike Jiang,Peilong Sun

Emulsifying properties of water soluble polysaccharides from Ulva fasciata (UFP) were evaluated in cinnamaldehyde/water emulsions in terms of droplet size distribution, rheological properties, visual phase separation, and zeta-potential. The cinnamaldehyde/water (10%, wt/wt) emulsions were formulated and stabilized by different concentrations of UFP (0.1–4%, wt/wt). The obtained emulsions showed monomodal droplet size distributions with average droplet size (D[3,2]) below 1.0   μm, when 3% (wt/wt) UFP was added as the emulsifying agent under a homogenization pressure of 75   MPa. The rheological properties and zeta-potential of the emulsions appeared to be dependent on the UFP concentration. Furthermore, the UFP exhibited better emulsifying and stabilizing properties in the investigated system when compared to other commercial polysaccharides of gum Arabic and gum Ghatti. The results also suggested that the emulsifying and stabilizing mechanism of the UFP may not only be ascribed to its surface-active protein moiety, but also to the hydrophobicity of the polysaccharide itself. These findings provided a theoretical basis for potential utilization of UFP as a novel hydrocolloid emulsifying agent.

Physical and chemical stabilities of β-carotene emulsions stabilized by Ulva fasciata polysaccharide

Ping Shao,Yueqiong Zhu,Weiping Jin

The objectives of this study were to investigate the effect of Ulva fasciata polysaccharide (UFP) on the physical and chemical characteristics of β-carotene oil-in-water (O/W) emulsions. The β-carotene (O/W) emulsions were prepared by combining 95 wt% UFP (1–5%, wt/wt) solutions with 5 wt% soybean oil containing β-carotene (0.05 wt% in the final emulsion). The stability of β-carotene emulsions was evaluated by droplet size and distribution, viscosity, zeta-potential, visual phase separation, light microscopy imaging and β-carotene degradation of the emulsions during storage. In addition, the influences of ionic strength and pH on physical stabilities of emulsions were characterized. Emulsions stabilized with high concentration (>2% wt/wt) of UFP had an average droplet size (D [3, 2]) less than 1.0 μm, and the average droplet size significantly decreased until the polysaccharide concentration reached 3%. Light microscopy images and emulsion zeta-potentials demonstrated 3% (wt/wt) UFP to be the optimum emulsifying agent concentration. UFP revealed better emulsifying properties and improved physical stabilities of β-carotene emulsions when compared to other commercial polysaccharides, including gum Arabic and beet pectin.

Antibacterial activities of R-(+)-Limonene emulsion stabilized by Ulva fasciata polysaccharide for fruit preservation

Ping Shao,Haoya Zhang,Ben Niu,Ligang Jiang

It was previously found that R-(+)-Limonene emulsion could be stabilized by Ulva fasciata polysaccharide . In the present study, emulsions of R-(+)-Limonene were developed for coating of strawberries to improve the shelf life and microbiological safety. Edible coatings with at least 0.15% w/w of R-(+)-Limonene improved the microbial stability of the strawberries, resulted effective in the decontamination of external pathogens such as Escherichia coli , Bacillus subtilis and Staphylococcus aureus . Changes in weight loss, total soluble solid (TSS), pH, total phenolic, ascorbic acid , textural properties and surface color of strawberries during storage were evaluated. R-(+)-Limonene emulsions were effective for decreasing water loss of all the fruit samples tested. The decrease in TSS values was higher in the control strawberries. Initial decreases followed by increases of pH during storage of strawberries. The amount of total phenols detected in control fruit sharply increased during the first 6 days then remained steady, whereas the total soluble phenols content of treated strawberries gradually increased during storage. Strawberries coated with emulsion had higher L* and a* values as compared to control fruits. The R-(+)-Limonene coatings have demonstrated the potential to inhibit foodborne pathogen contamination of strawberries, and prolong their shelf life.

Chemical Stability and in vitro release properties of β-carotene in emulsions stabilized by Ulva fasciata polysaccharide

Ping Shao,Qiang Qiu,Jie Xiao,Yueqiong Zhu,Peilong Sun

The aim of the present study was to examine how factors (pH, temperature, iron chelators, and free radical scavengers) influence on the chemical stabilities of β-carotene encapsulated by Ulva fasciata polysaccharide (UFP)-stabilized emulsions and the bioaccessibility under in vitro oral-gastro-intestinal digestion conditions. The concentration of β-carotene in emulsions under different environments was determined. Changes in droplet properties, particle size and microstructure, throughout the oral-gastric-intestinal model were measured and the rate and extent of lipid digestion were recorded in the simulated small intestine fluid. Our results suggested that the pH had a significant effect on the stability of β-carotene, a rapid degradation at acidic environment and a relatively higher stability at high pH (6.0 and 7.0) were observed. β-carotene in emulsions was highly sensitive to extreme temperature (80   °C, 90   °C, and 100   °C). Addition of EDTA or α-tocopherol significantly increased the stability of β-carotene. Meanwhile, digested UFP-stabilized emulsions showed higher lipid digestion rate than GA-stabilized emulsions and BP-stabilized emulsions. UFP was capable of increasing the bioaccessibility of β-carotene in emulsions compared to GA and BP. Therefore, UFP-stabilized emulsion is a promising delivery system to promote applications of β-carotene in functional food and beverage system.

The fate of antibiotic resistance genes, microbial community, and potential pathogens in the maricultural sediment by live seaweeds and oxytetracycline

Lijuan Feng,Changyan Xiao,Yuqin Luo,Yan Qiao,Dongzhi Chen

Three common seaweeds including Ulva fasciata, Codium cylindricum and Ishige okamurai were used for the remediation of maricultural wastewater and sediment in the presence/absence of trace level of oxytetracycline (OTC) in lab-scale experiments. Higher NO 3 − -N and PO 4 3- -P removal rates were achieved due to the presence of seaweeds, and trace OTC also had a positive effect on NO 3 − -N removal. A slight variation of 2.10–2.15% were observed in the total relative abundances of antibiotic resistance genes (ARGs) of different sediment samples after one-month operation. However, the variation of ARGs profiles by the co-existence of different seaweeds and OTC was in the descending order of Ishige okamurai  >  Codium cylindricum  >  Ulva fasciata , which was in accordance with the variation of microbial hosts at genus level. The abundance of dominant tetracycline resistance genes promoted by the co-existence of different seaweeds and OTC in compared with the presence of single seaweed or OTC via metagenomic sequencing and qPCR analysis, and the co-existence of Ishige okamurai and OTC exhibited the largest impact. The potential pathogens were more sensitive to the co-existence of seaweed and OTC than single seaweeds. Meanwhile, a variety of ARGs were enriched in the pathogens, and the dominant pathogenic bacteria of Vibrio had 133 Vibrio species with 28 subtypes of ARGs. The variation of ARGs profiles in the sediment were strongly related with the dominant phyla Proteobacteria, Actinobacteria, Firmicutes, Planctomycetes and Cyanobacteria . Besides, Nitrate level exhibited more significant effect on ∑ARGs, ARGs resistant to vancomycin and streptogramin _a, while phosphate level exhibited more positively significant effect on ARGs resistant to fosmidomycin, ATFBT and cephalosporin.
