Response of Vallisneria natans to aluminum phytotoxicity and their synergistic effect on nitrogen, phosphorus change in sediments

Qingwei Lin,Mingjun Fan,Xue Peng,Jianmin Ma,Yi Zhang,Fei Yu,Zhenbin Wu,Biyun Liu

Increasing aluminum (Al) use and its effects on aquatic systems have been a global issue, however the Al impacts on submerged plants and their ecological functions were poorly understood. Aquatic simulation experiments were performed to study Al-toxicity on the germination and seedling morphological and physiological characteristics of Vallisneria natans , and investigate their synergistic effect on nitrogen (N), phosphorus (P) change and microbial community in sediment. The seeds germination characteristics, growth and physiological parameters of seedlings, including root activity, were significantly affected by alum treatments and the inhibition levels increased with Al 3+ concentration. The Al accumulation in roots and leaves were significantly different. Al 3+ concentration above 0.3 mg/L showed toxic to V. natans . TN, TP, IP, Fe/Al-P contents in sediments varied markedly under co-existence of Al and V. natans . Additionally, the relative abundance of sediment microbial community related to N, P cycle was effected. Results concluded that the increasing aquatic Al-concentration inhibits growth and propagation of submerged plants and the ecological restoration effect, and exerts synergistic effect with submerged plants on N, P components in sediments. Such findings were helpful for Al ecological evaluation, and were instructive for the submerged plants restoration in shallow eutrophic lakes with Al input.

Phosphorus acquisition strategy of Vallisneria natans in sediment based on in situ imaging techniques

Yi Zhang,Cai Li,Qin Sun,Cuiling Jiang,Shiming Ding,Musong Chen,Xin Ma,Zhilin Zhong,Yan Wang,Daniel C.W. Tsang

Phosphorus (P) availability is closely related to the distributions of pH, O 2 and phosphatase activities in the rhizosphere of plants growing in soils and sediments. In this study, the P uptake processes and mechanisms of Vallisneria natans ( V. natans ) during two vegetation periods (i.e., week three and six) were revealed using three noninvasive 2D imaging techniques: planar optode (PO), diffusive gradients in thin films (DGT) and zymography. The results showed that increased phosphatase activity, O 2 concentration and root-induced acidification were observed together in the rhizosphere of root segments and tips. In week three, when V. natans was young, the flux of DGT-labile P accumulated more in the rhizosphere in comparison with the bulk sediment. This was because increased phosphatase activity (of up to 35%) and root-induced acidification (with pH decreasing by up to 0.25) enhanced P acquisition of V. natans by the third week. However, the flux of DGT-labile P turned to depletion during weeks three to six of V. natans growth, after Fe plaque formed at the matured stage. The constant hydrolysis of phosphatase and acidification could not compensate for the P demand of the roots by the sixth week. At this stage, Fe plaque become the P pool, due to P fixation with solid Fe(III) hydroxides. Subsequently, V. natans roots acquired P from Fe plaque via organic acid complexation of Fe(III).

The role of submerged macrophytes in phytoremediation of arsenic from contaminated water: A case study on Vallisneria natans (Lour.) Hara

Bin Li,Bowen Gu,Zhaoguang Yang,Ting Zhang

Arsenic contamination of water is a global concern due to its heavy threat to human health. In this study, the submerged macrophyte Vallisneria natans (Lour.) Hara was used to remove environmentally relevant concentrations of arsenic in the binary As(III)/As(V) system. The concentrations of total arsenic (tAs) and As(III) in water dropped rapidly within 3 days, while As(V) first increased slightly within 3 days and then gradually decreased. About 1.2% dimethylarsinate (DMA) was detected at the 14th day of treatment. These findings indicated that As(III) could be oxidized to As(V) and methylated to DMA in water with V. natans . In relation to V. natans , both tAs and As(V) were much higher in roots compared to leaves. Arsenate was the predominant species (≥ 95.65 ± 0.10%) in roots, and As(III) was only found at the 14th day (3.45–6.96 mg kg –1 ). In leaves, As(III) significantly increased ( P  < 0.05) as the treatment duration increased. The proportions of As(V) (27.99–40.03%) were lower than those of As(III) and arsenobetaine (AsB) was detected (0.52–1.87 mg kg –1 ) after 7 d. The results of arsenic speciation demonstrated that the transformation of arsenic species in V. natans included As(V) reduction and As(III) methylation to AsB. There were a decrease in chlorophyll content, and an increase in MDA level and antioxidant enzymes (SOD, CAT , and POD) activities. The MDA level was much higher in leaves than roots, whereas the activities of SOD, CAT, and POD were the opposite, suggesting their possible role in arsenic resistance and detoxification. Our results indicate the potential of V. natans in phytoremediation of arsenic-contaminated water.

Growth and physiological responses in a submerged clonal aquatic plant and multiple-endpoint assessment under prolonged exposure to ciprofloxacin

Pei Fan,Chunhua Liu,Zhen Ke,Wei Zhou,Zhonghua Wu

Ciprofloxacin is ubiquitous and poses a potential threat to aquatic ecosystems. However, the comprehensive effect of prolonged ciprofloxacin exposure on the submerged clonal plant Vallisneria natans (Lour.) Hara remains unknown. Growth and physiological responses in V. natans exposed to ciprofloxacin at concentrations of 0, 0.05, 0.25, 1.25, 2.5, 5 and 10 mg/L were repeatedly evaluated on Days 7, 14, 28, 42 and 56. V. natans maintained good growth properties under 0.05–0.25 mg/L ciprofloxacin treatments, while the inhibition effect on plant growth induced by higher-concentration treatments increased over time. The IC50 values of ciprofloxacin for growth endpoints ranged from 1.6 mg/L to 5.3 mg/L and displayed time-dependent decreases. Pigment contents were significantly stimulated by ciprofloxacin on Day 7 but decreased to varying degrees as the exposure time was extended. Soluble protein and hydrogen peroxide content rose significantly over the first 14 days of treatment with 0.25–10 mg/L ciprofloxacin but decreased under 1.25–10 mg/L ciprofloxacin treatments since Day 28. Antioxidants including superoxide dismutase, catalase, guaiacol peroxidase, ascorbate peroxidase and proline functioned well in mitigating oxidative stress under different ciprofloxacin concentrations, lowering the comprehensive toxic effects of ciprofloxacin on V. natans during the period from Day 14 to Day 42, as evidenced by decreased IBR (integrated biomarker response) values. However, the toxic pressure of ciprofloxacin on V. natans peaked on Day 56. These findings suggest that exposure time can influence the responses of V. natans exposed to ciprofloxacin and that IBR can be employed to evaluate the integrated impacts of prolonged ciprofloxacin contamination in aquatic settings.

Potential mechanism of humic acid attenuating toxicity of Pb2+ and Cd2+ in Vallisneria natans

Suzhen Huang,Zhikai Wang,Qixuan Song,Jun Hong,Tianyu Jin,Haiqing Huang,Zheng Zheng

Humic substances are widely present in aquatic environments. Due to the high affinity of humic substances for metals, the interactions have been particularly studied. To assess the effect of humic acid (HA) on submerged macrophytes and biofilms exposed to heavy metal stress, Vallisneria natans was exposed to solutions containing different concentrations of HA (0.5–2.0 mg·L −1 ), Pb 2+ (1 mg·L −1 ) and Cd 2+ (1 mg·L −1 ). Results suggested that HA positively affected the plant growth and alleviated toxicity by complexing with metals. HA increased the accumulation of metals in plant tissues and effectively induced antioxidant responses and protein synthesis. It was also noted that the exposure of HA and metals promoted the abundance and altered the structure of microbial communities in biofilms. Moreover, the positive effects of HA were considered to be related to the expression of related genes resulting from altered DNA methylation levels, which were mainly reflected in the altered type of demethylation. These results demonstrate that HA has a protective effect against heavy metal stress in Vallisneria natans by inducing effective defense mechanisms, altering biofilms and DNA methylation patterns in aquatic ecosystems.

Vallisneria natans drives pesticide removal from agricultural waters: The role of bioaccumulation and epiphytic bacteria

Hongyu Guo,Songhe Zhang,Jiajia Zhang,Jianwei Cui,Yuyu Li,ShuYou Zhang,Hongwei Yu

Pesticide mixtures are prevalent in aquatic systems within agricultural regions, but their transport and removal mechanisms in the presence of submerged macrophytes remain unclear. This study investigated changes in water quality, plant physiology, epiphytic microbial communities, and pesticide distribution in wetland substrates during 56 days of repeated exposure to a mixture of 12 pesticides at environmental (0.5 and 1 μg/L) and high (5 and 10 μg/L) concentrations. The average removal rates of total pesticides ranged from 58.61% to 75.52% and decreased with increasing pesticide addition and exposure time. Bioconcentration factors in plants were significantly higher than sediment-water partition coefficients for all pesticides. Notably, triazole pesticides (paclobutrazol, tebuconazole and hexaconazole) tended to accumulate in the system compared to neonicotinoid insecticides (dinotefuran, imidacloprid, and thiamethoxam). Pesticide addition reduced nutrient removal efficiency and induced oxidative damage in V. natans leaves. A total of 3, 14, and 17 biomarkers were identified in Con, PG1, and PG10, respectively, suggesting that PG1 and PG10 disturbed the epiphytic bacterial community. Co-occurrence network analysis revealed potential associations between pesticide residues and epiphytic bacterial taxa in PG10. PICRUSt2-based functional prediction indicated that compared to control, the predicted potential for xenobiotics biodegradation and metabolism was higher in PG1, whereas predicted genes related to starch and hemicellulose decomposition increased and denitrification-related predicted genes decreased in PG10. These data highlight that V. natans can accumulate pesticides and survive repeated pesticide exposure during the 56-day experiment, although high concentration pesticides had adverse effects on V. natans -epiphytic bacterial community.

Synergism or Antagonism in Toxicity Induced by Co-Exposure to Polyamide Microplastics and Cadmium Is Dose-Dependent in the Submerged Macrophyte Vallisneria natans

Yuqi Feng,Xuerong Wang,Ruiming Han,Pengcheng Zhou,Jiakang Mu,Qinghui Jiang,Shaoting Chen,Jiasheng Ma,Lilin Zheng,Wei Wei,Mingxi Zhou

The contamination of microplastics (MPs) and heavy metals (HMs) in water has caused widespread concern, while their effects on submerged macrophytes have rarely been reported. Experiments were carried out to investigate the toxic effects of polyamide microplastics (PAMPs; 0.1%, 0.3%, and 1.0%w/w) and cadmium (Cd; 0.3 and 1.0 mg/L), alone or in combination, on the submerged macrophyteVallisneria natans(V. natans). The results showed that PAMPs significantly reduced Cd accumulation in leaves (decrease of 2.38%~26.12%) but elevated Cd accumulation in roots. Both Cd exposure and high PAMP exposure alone inhibited plant growth. The combined stress showed concentration-dependent effects: the low Cd concentration (0.3 mg/L) and PAMPs synergistically exacerbated toxicity (synergism, MDR > 1.3), as PAMPs disrupted the sediment structure and enhanced the bioavailability of Cd, whereas whenV. natanswas co-exposed to the high Cd concentration (1.0 mg/L) and PAMPs, the PAMPs blunted the toxicity of Cd by efficiently adsorbing it (antagonism, MDR < 0.7). Both individual and combined exposures decreased chlorophyllaand chlorophyllbsynthesis and increased superoxide dismutase (SOD) and peroxidase (POD) activities as well as malondialdehyde (MDA) content in plant tissues. However, exposure to low and medium concentrations of MPs (0.1% and 0.3%w/w) alone had positive effects on plant growth and photosynthesis systems, while combined exposures exacerbated the damaging effects of PAMPs on the antioxidant defense system inV. natans. These results allow for a better understanding of the synergistic effect of co-contamination of microplastics and heavy metals in freshwater ecosystems, and highlight the necessity of ecological risk assessment during phytoremediation using submerged macrophytes.

Degradation of pyrene by Xanthobacteraceae bacterium strain S3 isolated from the rhizosphere sediment of Vallisneria natans: active conditions, metabolite identification, and proposed pathways

Ge Huanying,Liu Xinghao,Lu Denglong,Yang Zhaoguang,Li Haipu

Polycyclic aromatic hydrocarbons (PAHs) were typical environmental contaminants that accumulated continuously in sediment. Microbial degradation is the main way of PAH degradation in the natural environment. Therefore, expanding the available pool of microbial resources and investigating the molecular degrading mechanisms of PAHs are critical to the efficient control of PAH-polluted sites. Here, a strain (identified as Xanthobacteraceae bacterium ) with the ability to degrade pyrene was screened from the rhizosphere sediment of Vallisneria natans . Response surface analysis showed that the strain could degrade pyrene at pH 5–7, NaCl addition 0–1.5%, and temperature 25–40 °C, and the maximum pyrene degradation (~ 95.4%) was obtained under the optimum conditions (pH 7.0, temperature 28.5 °C, and NaCl-free addition) after 72 h. Also, it was observed that the effect of temperature on the degradation ratio was the most significant. Furthermore, eighteen metabolites were identified by mass spectrometry, among which (2Z)-2-hydroxy-3-(4-oxo-4H-phenalen-3-yl) prop-2-enoic acid, 7-(carboxymethyl)-8-formyl-1-naphthyl acetic acid, phthalic acid, naphthalene-1,2-diol, and phenol were the main metabolites. And the degradation pathway of pyrene was proposed, suggesting that pyrene undergoes initial ortho-cleavage under the catalysis of metapyrocatechase to form (2Z)-2-hydroxy-3-(4-oxo-4H-phenalen-3-yl) prop-2-enoic acid. Subsequently, this intermediate was progressively oxidized and degraded to phthalic acid or phenol, which could enter the tricarboxylic acid cycle. Furthermore, the pyrene biodegradation by the strain followed the first-order kinetic model and the degradation rate changed from fast to slow, with the rate remaining mostly slow in the later stages. The slow biodegradation rate was probably caused by a significant amount of phenol accumulation in the initial stage of degradation, which resulted in a decrease in bacterial activity or death. Effects of Aluminum Salts on Bacterial Community in Epiphytic Biofilms on Vallisneria Natans Leaves: Characteristics of Nitrogen and Phosphorus Fractions and Bacterial Community Composition

Peng Xue,Zhang Xinyi,Zhang Shuxian,Lin Qingwei,Zhang Haokun,Zhang Xiaowen,Zhang Yi,Xu Steven Jing-Liang,Lee Fred Wang-Fat,Tam Nora Fung-yee,Wu Zhenbin,Liu Biyun

The widespread use of aluminum-based coagulants in ecological restoration projects has raised increasing concerns regarding the potential ecological impacts of residual aluminum (Al) in aquatic ecosystems. Epiphytic biofilms on submerged plants play a vital role in regulating nutrient cycling and energy flow in aquatic environments. However, the effects of Al on the biological and abiotic components of epiphytic biofilms on submerged macrophyte leaves remain poorly understood. In this study, we investigated the Al content in epiphytic biofilms on Vallisneria natans and its relationships with nitrogen, phosphorus and bacterial composition in Hangzhou West Lake, China. This location was selected due to the potential for residual Al into the lake from the daily pretreatment of the Qiantang River water diversion project—a water quality improvement project. Our results showed that Al contents in epiphytic biofilms decreased with increasing distance from the inlet and were positively correlated with total phosphorus, Chlorophyll-a and ash-free dry mass ( P  < 0.05), but showed no significant correlation with total nitrogen. Al accumulation enhanced Chl-a levels in biofilms by increasing the inorganic phosphorus content. Furthermore, Al content was positively correlated with the abundance of Cyanobacteria and negatively correlated with the abundance of Proteobacteria. Al also showed a significant negative correlation with key denitrifying bacteria, such as Vogesella and Phreatobact er, suggesting that elevated Al levels might affect the nitrogen cycling function in epiphytic biofilms. These findings provide important reference data for for ecological risk assessment and inform management strategies for aquatic systems undergoing restoration or exposed to aluminum-based treatments.
