AimsThis study aimed to ascertain the active substances in the hydroponic solution of Ulva (U.) fasciata (HSUF) and their effects on antibiotics resistance of pathogenic bacteria Vibrio alginolyticus (Va.) at environmental levels. Methods and ResultsAnalytical results of HSUF by solid-phase extraction and gas chromatography-mass spectrometry showed that HSUF contained rich active substances and dominated by 2,4,6-tribromophenol, acetal, mono (2-ethylhexyl) phthalate and 2- (4-hydroxybenzene) ethanol, etc. Thereinto, 2,4,6-tribromophenol had the lowest minimal inhibitory concentrations (MICs) of 64–128 μg mL−1 for Va. strains. The rich antibacterial substances of HSUF inhibited 47%-63% of isolated Va. strains. After 20 generations of subculture for Va. strains in three dilutes (1/2 (HT), 1/20 (MT) and 1/50 (LT)) of HSUF (20 g L−1), the resistance to streptomycin of Va. S1 and Va. M3 changed from intermediate resistance ability to susceptible level. The large promotion of reactive oxygen species was observed in different HSUF levels, but the biofilm formation of Va. S1 did not changed significantly. Transcriptome sequencing of Va. S1 demonstrated that antibiotic resistance gene lpxA was down-regulated at different HSUF levels.ConclusionsLive U. fasciata excreted a variety of active secondary metabolites (SMs) in HSUF, which exhibited strong inhibitory effects on three isolated Va. strains. HSUF changed the antibiotic resistance of Va. Strain, especially at a higher HSUF concentrations.

Protein succinylation modification is a common post-translational modification (PTM) that plays an important role in bacterial metabolic regulation. In this study, quantitative analysis was conducted on the succinylated proteome of wild-type and florfenicol-resistant Vibrio alginolyticus to investigate the mechanism of succinylation regulating antibiotic resistance. Bioinformatic analysis showed that the differentially succinylated proteins were mainly enriched in energy metabolism, and it was found that the succinylation level of phosphoenolpyruvate carboxyl kinase (PEPCK) was highly expressed in the florfenicol-resistant strain. Site-directed mutagenesis was used to mutate the lysine (K) at the succinylation site of PEPCK to glutamic acid (E) and arginine (R), respectively, to investigate the function of lysine succinylation of PEPCK in the florfenicol resistance of V. alginolyticus. The detection of site-directed mutagenesis strain viability under florfenicol revealed that the survival rate of the E mutant was significantly higher than that of the R mutant and wild type, indicating that succinylation modification of PEPCK protein may affect the resistance of V. alginolyticus to florfenicol. This study indicates the important role of PEPCK during V. alginolyticus antibiotic-resistance evolution and provides a theoretical basis for the prevention and control of vibriosis and the development of new antibiotics.

Lignin, as the abundant carbon polymer, is essential for carbon cycle and biorefinery. Microorganisms interact to form communities for lignin biodegradation, yet it is a challenge to understand such complex interactions. Here, we develop a coastal lignin-degrading bacterial consortium (LD), through “top-down” enrichment. Sequencing and physiological analyses reveal that LD is dominated by the lignin degrader Pluralibacter gergoviae (>98%), with additional rare non-degraders. Interestingly, LD, cultured in lignin-MB medium, significantly enhances cell growth and lignin degradation as compared to P. gergoviae alone, implying a role of additional outliers. Using genome-scale metabolic models, metabolic profiling and culture experiments, modeling of inter-species interactions between P. gergoviae , Vibrio alginolyticus , Aeromonas hydrophila and Shewanella putrefaciens , unravels cross-feeding of amino acids, organic acids and alcohols between the degrader and non-degraders. Furthermore, the sub-population ratio is essential to enforce the synergy. Our study highlights the unrecognized role of outliers in lignin degradation.

Vibrio alginolyticus is a ubiquitous marine opportunistic pathogen that can infect various hosts in marine environment. In the present study, V. alginolyticus strain Wz11 was isolated from diseased cuttlefish , Sepia pharaonis , with 20% of promoted death and high survival capacity in skin mucus and tissue liquid. Its growth, siderophore production, and expressions of haemolysin and swarming related genes were characterized under iron limited conditions. The minimal inhibitory concentration (MIC) of 2,2′-dipyridyl (DP) to V. alginolyticus strain Wz11 was 640 μM. While growth of V. alginolyticus strain Wz11 was inhibited by DP , production of iron-seizing substances, haemolytic activity and swarming motility were increased. Moreover, expressions of haemolysin related genes tlh , tdh and vah and flagellar related genes flgH , fliC , fliD and fliS were also characterized using real-time reverse transcriptase PCR. Expression of tdh was up-regulated to 7.7-fold, while expressions of tlh and vah were down-regulated to 0.016-fold and 0.03-fold, respectively. The expression of fliC , flgH , fliD and fliS was up-regulated to 4.9-, 3.8-, 8.6- and 4.5-fold, respectively. Concluded from our results suggested that V. alginolyticus strain Wz11 was considered as a potential pathogen of S. pharaonis , and iron level played an important role in the production of iron-seizing substances, and activities of haemolysin and bacterial swarming as well as their related gene expressions.

In this study, two strains of novel sulfadiazine (SDZ)-degrading bacteria Vibrio alginolyticus SUL-3 and SUL-5 were isolated from the SDZ contaminated mariculture wastewater. The degradation rate of two strains reached 54.11% ± 0.15% and 63.57% ±0.07% of 1 mg/L SDZ within 4 d, respectively, and could also degrade other 3 sulfonamides (sulfamethoxazole, sulfamonomethoxine, and sulfamethazine). Optimal degradation conditions were identified as: neutral pH (7.0), marine salinity (35‰), and temperature of 30 °C. The addition of exogenous carbon and nitrogen sources was found to enhance bacterial degradation of SDZ at a concentration of 10 mg/L. However, this stimulatory effect was either insignificant or even inhibitory at lower SDZ concentrations. Through bond cleavage, hydroxylation and ring opening, 12 SDZ biodegrading intermediates were verified, showing less toxic than their parent. Multidrug resistance genes were identified in SUL-5 through whole-genome analysis, including sulfonamide resistance gene sul2 , which was the most prevalent sulfonamide resistance determinant in aquatic environments. Comparing differentially expressed genes (DEGs) between groups with or without SDZ, oxidoreductases, transferases, hydrolases, and lyases were proved to participate in the biodegradation of SDZ. SUL-5 adapted to and degraded SDZ by upregulating genes encoding ATP-binding cassette (ABC) transporters, degradation-related, and multidrug efflux pump enzymes. This research provides insights into the survival and biodegradation mechanisms of Vibrio sp. and evaluates its potential for environmental bioremediation.
