Hyperuricemia, a metabolic disease characterized by elevated uric acid (UA) levels due to abnormal purine metabolism, may benefit from a multi-target therapeutic approach using lactic acid bacteria (LAB). In this study, 49 strains of LAB were screened from plant and animal samples, with the top 10 strains selected via principal component analysis (PCA) based on their lipid-lowering, glucose-lowering, and obesity-inhibiting abilities to improve energy metabolism. After evaluating the hemolytic, DNase-, gelatin-, and amine-producing abilities of these strains, one strain X1 was elected for its high UA lowering ability based on the in vitro xanthine oxidase (XO) inhibition and in vivo (zebrafish model) XO reduction assay. X1 was then identified, its probiotic characteristics were investigated, and its potential anti-hyperuricemia mechanism was explored. The results showed X1 was Weissella cibaria with probiotic potential, and exhibited an in vitro XO inhibitory capacity of 57.77 ± 10.34% and reduced UA levels and XO activities by 47.30 ± 1.33% and 41.29 ± 5.26%, respectively, in a hyperuricemia zebrafish model. The anti-hyperuricemia mechanism of X1 may involve the degradation of inosine (45.26%) and guanosine (99.23%), along with the regulation of Organic anion transporter 1 (OAT1), UOX (urate oxidase), Hepatocyte nuclear factor 4 alpha (HNF4A), and Hypoxanthine-guanine phosphoribosyltransferase (HPRT) expression levels. These findings indicated that X1 has potential as a probiotic for treating hyperuricemia.

An exopolysaccharide (EPS)-producing bacterium, identified as Weissella cibaria based on 16S rDNA sequencing, was isolated from Chinese traditional rice wine koji. Whole-genome sequencing (WGS) revealed 2261 core genes and 3 plasmids, with 95 carbohydrate-active enzymes identified in the genome. The EPS produced by W. cibaria ZJU.1 was primarily composed of glucose, with a molecular weight of 144.5 kDa, as determined by high-performance anion-exchange chromatography (HPAEC) and high-performance gel chromatography (HPGPC). Structural analysis indicated that the EPS predominantly consists of α-(1→6) glycosidic bonds (88.32%) and α-(1→3) branches (11.68%). Microscopic examination revealed an interleaved fibrous structure at low concentrations and a lamellar structure at higher concentrations. Thermal analysis demonstrated good stability, with the carbon ring structure intact up to 264.12°C. Furthermore, the EPS exhibited excellent dispersion stability and rheological properties at higher concentrations. Activity prediction using a zebrafish model suggested potential therapeutic benefits, including alleviation of diabetes, anti-tumor, and anti-epileptic effects. This study highlights the promising food and pharmaceutical applications of W. cibaria ZJU.1 EPS.

The uric acid (UA) degrading Weissella cibaria X1 was employed for the fermentation of safflower ( Carthamus tinctorius L.). Compared with naturally fermented safflower, X1 fermented safflower significantly enhancing the antioxidant capacity and xanthine oxidase (XO) inhibition rate, decreased UA levels by 81.30%, XO activity by 99.02%, and creatinine levels by 27.08% based on a hyperuricemia (HUA) zebrafish model. Additionally, it upregulated the genes inhibiting UA synthesis—hypoxanthine-guanine phosphoribosyltransferase 1 (HPRT1 gene) 1.91 fold, and those promoting UA excretion—urate oxidase (UOX) gene 5.40 fold, organic anion transporter 1 (OAT1) gene 4.80 fold, hepatocyte nuclear factor 4 alpha (HNF4A gene) 3.29 fold, and polyvalent PDZ domain 1 (PDZK1) gene 57.46 fold. Subsequently, non-targeted metabolomics analysis showed that X1 fermentation downregulated the content of L-glutamine and modulated purine metabolism than the safflower fermented naturally, thus decreasing the levels of purines and increasing those of scolymoside. Our research provided a potential HUA therapy via both pharmaceutical and nutraceutical strategies.

Introduction The intensive farming of crucian carp ( Carassius auratus ) often compromises fish health and increases disease susceptibility, driving interest in probiotic interventions as sustainable alternatives to antibiotics. However, the in vivo efficacy and mechanistic basis of Weissella cibaria strains in freshwater fish remain poorly understood. Methods A bacteriocin-producing strain, Weissella cibaria XN-2a, was isolated from fish viscera and supplemented in the diet of crucian carp at an initial viable dose of 2.74 × 10 10 CFU/g feed (freshly prepared and replaced every 3 days) for 30 days, followed by a challenge with Aeromonas hydrophila . Growth performance, immune parameters, intestinal histology, and gut metagenomic profiles were compared between the probiotic-fed group (XS) and the control group (XD). Results Probiotic supplementation significantly improved weight gain (28.17% vs. 13.98%), feed conversion ratio (1.88 ± 0.32 vs. 2.58 ± 0.27, p < 0.05), and post-challenge survival (50% vs. 0%) ( p < 0.001). The XS group exhibited enhanced intestinal morphology (higher mucosal fold height and FH/IFD ratio, lower inter-fold distance), elevated IgM and SOD activities, and reduced post-challenge IL-6 and MDA levels. Metagenomic analysis revealed that the probiotic reshaped the gut microbiota from a pathogen-associated virulence profile (LPS, flagella, enterobactin) toward a commensal profile enriched in T6SS/T2SS and alternative siderophores, without increasing the overall antibiotic resistance burden. The butanoate metabolism pathway showed differences consistent with increased butyrate synthesis potential, with enrichment of butyrate-synthetic enzymes (e.g., enoyl-CoA hydratase, acetolactate decarboxylase) in XS and butyrate-diverting enzymes in XD. CAZy profiling further showed enrichment of antibacterial (GH25/GH73) and antioxidant (AA4) gene families in XS, with Weissella as a major contributor. Discussion These findings suggest that dietary Weissella cibaria XN-2a enhances growth performance, intestinal health, and disease resistance in crucian carp, potentially through a dual mechanism involving bacteriocin-mediated microbiota modulation and enhanced butyrate synthesis potential. Further investigations, including direct metabolite quantification, are warranted to validate these mechanistic inferences.
