Hyperuricemia (HUA) has emerged as a global metabolic disorder that poses significant risks to human health. To investigate the effects and mechanisms of Weizmannia coagulans BC99 in alleviating hyperuricemia, we established a hyperuricemia mouse model. The results demonstrate that BC99 significantly downregulates the expression of uric acid (UA) reabsorption proteins in the kidneys and intestines, while upregulating the expression of UA excretion proteins. This modulation leads to a reduction in UA synthesis, mediated through the Nrf2/NLRP3 pathway. Additionally, BC99 intervention restored gut microbiota intestinal dysbiosis in HUA mice, increased the beneficial short-chain fatty acid (SCFA)-producing bacterial genera, and corrected disturbances in amino acid, purine, and pyrimidine metabolism. Collectively, our findings suggest that BC99 exhibits strong anti-hyperuricemic effects and holds promise as a dietary supplement for lowering uric acid levels.

The purpose of this study was to prepare a pH-responsive microgel for co-delivering Weizmannia coagulans 99 (BC99) and procyanidins (PCs) to enhance the survival rate and tolerance of probiotics in complex micro-environment. The effects of different concentrations of PCs on the properties of microgels were optimized, and found that the spherical microgels had higher encapsulation efficiency (90.27 ± 2.51%) and smaller size when the concentration of PCs was 20 μg/mL. The interaction among PCs, pectin and protein could effectively improve the survival rate of BC99 under different pH, bile salt, digestive enzyme and temperature conditions, maintain their stability in acidic gastric fluid, and realize the release of probiotics in neutral intestinal fluid. Moreover, the microgel was able to protect BC99 against H 2 O 2 and antibiotics. This work provides a pH-responsive co-loaded microgels for BC99 and PCs, and has the potential in the loading and delivery of other probiotics and polyphenols.

The increased emission and accumulation of microplastics pose a severe threat to humans and the environment. As effective biological agents for alleviating the effects of microplastics, the mechanism of action of probiotics remains unclear. In this study, based on the successful establishment of a reproductive virulence model ofCaenorhabditis elegans(C. elegans), we explored the effect and mechanism ofWeizmannia coagulansCGMCC 9951 (W. coagulansCGMCC 9951) on the reproductive toxicity ofC. elegans. Our results showed that the gonad area and the number of offspring increased but the number of germ cells undergoing apoptosis decreased by 14% and 24% inC. elegans, after CGMCC 9951 treatments. Antioxidant test results showed that CGMCC 9951 increased the activity of Superoxide Dismutase (SOD), Catalase (CAT), and the content of Glutathione (GSH) inC. elegans. In addition, it was found by qPCR and mutagenesis experiments verified that CGMCC 9951 alleviated reproductive toxicity through the DNA checkpoint signaling pathway. Our findings suggested that CGMCC 9951 could alleviate the reproductive toxicity of polystyrene microplastics inC. elegansby enhancing antioxidant capacity and inhibiting DNA damage checkpoint signaling pathway. The above results suggest that probiotics can be used as a potential approach to alleviate the reproductive toxicity induced by polystyrene microplastics in humans.

Helicobacter pylori is a highly prevalent pathogen in human gastric mucosa epithelial cells with strong colonization ability. Weizmannia coagulans is a kind of active microorganism that is beneficial to the improvement of host gut microbiota balance and can prevent and treat intestinal diseases. We investigated the beneficial effects of W. coagulans BC99 in H. pylori infected mice and measured inflammation response, oxidative stress, and gut microbiota. Results showed that BC99 could alleviate the gastric inflammation, inhibit the increasing of inflammation parameters endotoxin, interleukin-10, transforming growth factor-β, and interferon-γ and oxidative stress myeloperoxidase and malondialdehyde, promote the levels of superoxide dismutase and catalase. Furthermore, 16S rRNA gene sequencing analysis revealed that BC99 reversed the change of gut microbiota by reducing the abundance of Olsenella , Candidatus _ Saccharimonas , Monoglobus , and increasing the abundance of Tyzzerella . Meanwhile, BC99 caused elevated levels of Ligilactobacillus and Lactobacillus . In view of the beneficial effect of BC99 on the content of short-chain fatty acid, valeric acid with sodium valerate interfered with H. pylori infection in mice found that valeric acid had a good restorative effect of H. pylori infection relating inflammation and oxidative stress responses. These results suggest that W. coagulans BC99 can be used as a potential probiotic to prevent and treat H. pylori infection by regulating the inflammation, oxidative stress, and gut microbiota.

Weizmannia coagulans with high temperature tolerance and stability was selected for the production of exopolysaccharides (EPS) at the fermentation temperature of 52 °C. The weak basic anion exchange resin was employed in the fermentation for the construction of buffer system to neutralize some lactic acid and form intermolecular hydrogen bonds with hydroxyl groups on capsular polysaccharides of bacteria via tertiary amine groups, resulting in the increased growth space for capsular polysaccharides. In addition, the fermentation at relatively high temperature significantly decreased the adsorption of Ca 2+ derived from CaO used for pH adjustment to the resin and sustained the ion exchange capability of the resin available for the formation of intermolecular hydrogen bonds with capsular polysaccharides. The concentration of EPS obtained achieved up to 19.8 g/L, which was increased by approximately 20-fold as compared to the concentration of EPS obtained by conventional fermentation. The conversion rate of carbohydrates to EPS reached up to 35.1%. The EPS produced under the optimized fermentation condition included neutral and acidic heteropolysaccharides with a molecular mass of 3.4 × 10 4 –1.5 × 10 6 Da. The concentration and molecular mass distribution of EPS exhibited good reproducibility. This study provides a novel method for the industrial production of EPS.

Introduction: Hyperuricemia (HUA) refers to the presence of excess uric acid (UA) in the blood, which increases the risk of chronic kidney disease and gout. Probiotics have the potential to alleviate HUA.Methods: This study established a hyperuricemia model using Caenorhabditis elegans (C. elegans), and studied the anti-hyperuricemia activity and potential mechanisms of Weizmannella coagulans BC99 (W. coagulans) at different concentrations (107 CFU/mL BC99, 108 CFU/mL BC99). Subsequently, we utilized UPLC-Q-TOF/MS to investigate the impact of BC99 on endogenous metabolites in C. elegans and identified pathways and biomarkers through differential metabolomics analysis. Results: The results of this study showed that BC99 treatment significantly reduced the expression of P151.2 and T22F3.3 (p < 0.05), reduced the levels of UA and xanthine oxidase (XOD) in nematodes (p < 0.05), while extending their lifespan and movement ability (p < 0.05). Mechanistically, BC99 activates the transcription factors DAF-16 and SKN-1, thereby inducing the expression of stress response genes, enhancing the activity of antioxidant enzymes and tolerance to heat stress in the body, and reducing the production of ROS (p < 0.001). This effect was most significant in the H-BC99 group. Furthermore, non-targeted metabolomics indicated that BC99 predominantly regulated pathways associated with amino acid metabolism (Carnosine), glycerophospholipid metabolism, and purine metabolism.Discussion: These results underscore BC99 as an effective and economical adjunct therapeutic agent for hyperuricemia, providing a scientific basis for further development and application.

A colon-targeted and adhesion-colonized BC99 and PC co-delivery system was prepared. • Microgels with high embedding rate could effectively maintain the survival rate of BC99. • Microgels improved the mechanical, immune and microbial barriers in UC mice. • Microgels improved the levels of brain neurotransmitters and metabolites. • Gut-microbiota-brain axis is an important pathway to alleviate UC and its comorbidities. Two-way communication between gut and brain can cause people with ulcerative colitis (UC) to exhibit notable mental disorders, such as anxiety and depression. In this study, Weizmannia coagulans BC99 and procyanidins co-delivery systems with adhesive colonization and colon targeting functions was prepared, and the mechanism for alleviating colitis and its accompanying anxiety-depression symptoms was revealed by regulating gut-microbiota-brain axis. The prepared microgels were spherical with 95.18% encapsulation efficiency, and could effectively maintain the survival rate of probiotics under simulated digestion conditions. The microgel also had mucosal adhesion, colon targeting and intestinal colonization capabilities. Subsequently, a UC mouse model was constructed, and the colon length of mice increased by 41.42% after SH-BP@PWC microgel intervention. SH-BP@PWC effectively inhibited the degree of oxidative stress, reduced the release of pro-inflammatory cytokines, maintained the mechanical barrier integrity of gut, regulated the imbalance of gut microbiota and arginine biosynthesis pathway. Moreover, the microgel could effectively relieve anxiety and depression-like behaviors in UC mice, improve the level of brain neurotransmitters, and relieve depressive symptoms by regulating metabolites, such as arginine. Correlation analysis further proved the interactions among gut microbiota, oxidative stress, inflammatory cytokines, brain neurotransmitters, and metabolites, highlighting the central role of gut-microbiota-brain axis, especial arginine metabolic pathway. This study designs a probiotic multifunctional co-delivery system that can provide new insights into alleviating colitis and its accompanying anxiety and depression from gut-microbiota-brain axis. A colon-targeted and adhesion-colonized Weizmannia coagulans -procyanidins delivery system was successfully designed and prepared, which could effectively alleviate colitis and its accompanying anxiety and depression via gut-microbiota-brain axis.
